Somatic embryos on growth regulator-free medium can be produced directly from cotyledon explants of Panax ginseng C. A. Meyer. When the embryo developmental stage was torpedo and cotyledon, the germination rate of embryos was quite high on MS medium supplemented with gibberellic acid ($GA_3$). However, the percentage of plantlet formation at the cotyledon stage was higher than that at the torpedo stage. This result demonstrates that the embryo at the cotyledon stage was the most appropriate for increasing germination by $GA_3$. Embryos cultured on medium including four levels of $GA_3$ concentrations (3, 5, 10, or 20 mg/$\ell$) showed all quite high germination rates (87-91%). When the well-developed embryos were continuously cultured on media including high concentrations of $GA_3$ from 10 to 20 mg/$\ell$, the percentage of formation of normal plantlets was lower than that seen under low concentrations from 3 to 5 mg/$\ell$. This treatment of high concentrations resulted in shoots with abnormal shape. The optimal $GA_3$ treatment provides a basis for the efficient method obtaining healthy plantlets derived from ginseng somatic embryos.

When dehulled apple seeds (Malus pilmila var. domestica) were induced to germinate, the rate of germination, which was increased by piercing both the radicle and cotyledon, was related to the site of injury. Also, the germination rates of an embryo with ¾ cotyledon (¾ CE), embryo with ½ cotyledon (½ CE), embryo with ¼ cotyledon (¼ CE), and embryo without cotyledon (NCE) gradually decreased compared to an embryo with a normal cotyledon, but it significantly recovered on the agar media containing MS solution and/or 80 mg/L gibberellin (GA3) as nutrition. The results show that the germination of the embryo was inhibited by the partial removal of cotyledon and also demonstrated that the rate of germination could be significantly increased by adding GA3 (80 mg/L) and/or MS medium.
Hypocotyls explants of melon seedling were cultured on Murashige and Skoog's (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D) and 0.5 mg/L benzyl aminopurine (BA) for 6 weeks to produce somatic embryos. In somatic embryos produced through intervening bright yellow friable (BYF) from the explants, somatic embryos with two-cotyledon (26%) and horn-type cotyledon (74%) were observed. The procambial strand of cotyledons was originated from circular procambial tissues of lower hypocotyls. The circular procambial independently divided into two procambial strand at the edge of cotyledonary-node, and then connected to each cotyledon to form somatic embryos with two-cotyledon. When cotyledon was horn-type, the circular procambial strand in lower hypocotyls would continuously remain connected to the cotyledon. However, somatic embryos with two or horn type cotyledon formed an abnormal shoot apex without the tunica-corpus structure or dome shape in the inter-cotyledonary area. These results demonstrated that the variation of cotyledon in somatic embryos was closely related to procambial tissue differentiation and shoot apical formation.
To study for germination rate of apple artificial seeds encapsulated with 2% alginic acids, the rates of germination for artificial seeds of various embryos with partial elimination of cotyledon measured by adding 80 mg/L gibberellic acid or/and nutritional media. The germination rate of artificial seeds produced embryo with 3/4 cotyledon (3/4 CE), embryo with 1/2 cotyledon (1/2 CE), embryo with 1/4 cotyledon (1/4 CE), and embryo without cotyledon (NCE) were gradually decreased with compare to normal embryo, but it's considerably recovered on the alginic acid containing MS solution or/and 80 mg/L GA3. In special, the low frequency (55.81%) of germination for artificial seeds of 1/2 cotyledon embryo with alginic acid was increased by treatment with combination of alginic acid, 3% sucrose, and 80 mg/L gibberellic acid (98.74%). In case of embryo without cotyledon, the low frequency of embryo without cotyledon (24.44%) in alginic acid almost recovered to 82.45% in alginic acid with MS solution and 80 mg/L gibberellic acid. The results shown that the germination of embryo was decreased by the removal of cotyledon, and demonstrated that the low rate of germination could be considerably increased by adding GA3 (80 mg/L) or/and nutrition.
The present study investigated the effects of triacontanol (TRIA) on plant growth and peroxisomal enzyme activities in radish seedlings. The optimum concentration of TRIA with respect to radish seedling bioassay was decided to 1.0mg $1^{-1}$. In comparison to untreated controls (including Tween 20 treatment), 1.0mg $1^{-1}$ TRIA treatment caused an increase in seed germination rate and root growth, but no stimulation in hypocotyl growth. Chlorophyll accumulation in cotyledon during carly development stage increased rapidly, and degradation of chlorophyll in later stage due to the cotyledon senesence was noticeably retarded. Increase of soluble protein content in cotyledon at early period of development was observed. Isocitrate lyase and catalase activity was not significantly different in both the treated and the untreated plants. But, glycolate oxidase activity was inhibited by TRIA down to 20% against controls. Also, the increase of the activity of peroxidase, a leaf-senescence marker enzyme, was continuously retarded over control for 8 days of development. Based on above results, TRIA was found to be active in both the growth and the peroxisomal enzyme activities of radish seedlings.
Immature embryos of Glycine max L. was cultured on Murashige and Skoog's (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D). After 6 to 8 weeks of culture, immature embryos produced somatic embryos. Of somatic embryos, two cotyledonary embryo (14%), one cotyledonary embryo (37%), fused cotyledonary embryo (43%), and stunted globular embryos (6%) were observed. The procambial strand of cotyledons originated from circular procambial tissues of lower hypocotyl. The circular procambial tissues were independently divided into one or two procambial strand at the edge of cotyledonary-node, and then connected to each cotyledon to form somatic embryos with one or two cotyledons. When cotyledon was a fused type, the circular procambial strand in lower hypocotyl was continuously connected to the cotyledon. Also, somatic embryos with two cotyledons developed a functional shoot apex with the tunica-corpus structure. In contrast, somatic embryos with one or fused cotyledon formed an abnormal shoot apex without the tunica-corpus structure or with non-dome shape in the inter-cotyledonary area. These results indicated that the variation of cotyledon in somatic embryos is closely related to procambial differentiation and shoot apical meristem development.

A successful, efficient system for multiple shoot induction and plant regeneration of soybean (Glycine max) was established. Four soybean genotypes were compared for organogenic responses on various media cultured under light conditions. The adventitious shoots (98%, 2.6 shoots/cotyledon) directly from one-day-old cotyledon after germination induced by the hormone treatment and its efficiency was higher than any other conditions. The optimal medium for the induction of multiple shoots from cotyledon in Pungsannamulkong(shoot formation rate, 98%), Lx 16 (83%) and IIpumgeomjeongkong(63%) was MS medium supplemented with 2 mg/L BAP, but for Alchankong(75%), MS medium supplemented with 1mg/L zeatin and 1mg/L IAA, 3% sucrose, 4% Phytagel. Higher root induction (88%) was observed from the shoots placed on rooting medium (hormone-free MS basal). Plantlets were transferred onto the same medium supplemented with 1% activated charcoal for further development. With this treatment, regenerated plantlets were obtained within 7-8 weeks (shoot induction for 4 weeks, rooting and shoot elongation for 3-4 weeks).

BACKGROUND: Before generating transgenic plant using the CRISPR-Cas9 system, the efficiency test of sgRNAs is recommended to reduce the time and effort for plant transformation and regeneration process. The efficiency of the sgRNA can be measured through the transient expression of sgRNA and Cas9 gene in tomato cotyledon; however, we found that the calculated efficiency showed a large variation. It is necessary to increase the precision of the experiment to obtain reliable sgRNA efficiency data from transient expression. METHODS AND RESULTS: The cotyledon of 11th, 15th, 19th, and 23rd-day-old tomato (Solanum lycopersicum cv. Micro-Tom) were used for expressing CRISPR-Cas9 transiently. The agrobacterium harboring sgRNA for targeting ALS2 gene of tomato was injected through the stomata of leaf adaxial side and the genomic DNA was extracted in 5 days after injection. The target gene edition was identified by amplifying DNA fragment of target region and analyzing with Illumina sequencing method. The target gene editing efficiency was calculated by counting base deletion and insertion events from total target sequence read. CONCLUSION: The CRISPR-Cas9 editing efficiency varied with tomato cotyledon age. The highest efficiency was observed at the 19-day-old cotyledons. Both the median and mean were the highest at this stage and the sample variability was also minimized. We found that the transgene of CRISPR-Cas9 system was strongly correlated with plant leaf development and suggested the optimum cotyledon leaf age for Agrobacterium-mediated transfection in tomato.

Seedling development among reciprocal FI hybrids and their parents, Lotus comiculatus and L renuis, were studies using a simple method of measuring cotyledon area. Cotyledon area attached to the plant could be measured with simple rapid method. Cotyledons expanded very rapidly during the first week, then slightly expanded until maximum size was reached around 3 weeks after emergence. Cotyledon area upon emergence was correlated with seed size (P=0.05), but after emergence there was a divergence in cotyledon expansion rate. Rapidity of photosynthetic area development was determinant factor for seedling growth of trefoil species. 2 $\times$ m2 and 1 $\times$ m2 among the hybrids promised the possibility increasing seedling vigor of birdsfoot trefoil.
We investigated the expression of cucumber genes involved in lipid mobilization and metabolism during cotyledon development to compare gene activity and to study the direction of carbon (acetyl unit) transport between glyoxysomes and mitochondria. The core metabolic pathway involving 10 genes was examined in four intracellular compartments: glyoxysomes (peroxisomes), mitochondria, chloroplasts, and cytosol. Additionally, we tested the early germination response of dark-grown seedlings and the immediate light response for a further 3 days. According to the reverse transcription polymerase chain reaction (RT-PCR), 3-L-ketoacyl-CoA thiolase 2 (Thio2), isocitrate lyase (ICL), and malate synthase (MS), the genes involved in storage lipid mobilization showed a similar and consistent pattern of gene expression in seedling development. Furthermore, coordinate expression of the A BOUT DE SOUFFLE (BOU) gene with ICL and MS during seedling emergence pointed to a possible secondary route of acetyl unit (acetyl-CoA) transport between peroxisomes and mitochondria in cucumber. The expression of the BOU gene was light dependent, as shown by BOU activity in Arabidopsis, suggesting that the dark condition also results in weak membrane biogenesis. In addition, several genes were active throughout the development of the green cotyledon, even during senescence. In conclusion, this study summarizes oil-seed germination and gene expression during cucumber cotyledon development and proposes an additional route for acetyl unit transport.
