Microbial Strains and Bioactive Exopolysaccharide Producers from Thai Water Kefir

  • The aims of this novel work were to determine the microbial strains and exopolysaccharide (EPS) producers in water kefir from Nakhon Ratchasima Province, Thailand. Thirty-three microbial strains were identified using 16S rRNA gene analysis consisting of 18 bacterial strains, as 9 strains of acetic acid bacteria (AAB), 9 strains of lactic acid bacteria (LAB), and 15 yeast strains. All bacteria were able to produce EPS with a diverse appearance on agar media containing different sugars at a concentration of 8%. Culture supernatants from AAB and LAB showed 31-64% 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity with the highest antioxidant activity of 64% from Acetobacter pasteurianus WS3 and WS6. Crude EPS from A. pasteurianus WS3 displayed the highest ferric reducing antioxidant power at 280 mM $FeSO_4/g$ EPS, greatest anti-tyrosinase activity at 20.35%, and highest EPS production of 1,505 mg EPS/L from 8% sucrose. These microbes offer beneficial health implications and their EPSs can be used as food additives and cosmetic ingredients.

  • Many techniques have been developed for reconstruction of the hand; however, less attention has been paid to foot reconstruction techniques. In particular, reconstruction of the forefoot and big toe has been considered a minor procedure despite the importance of these body parts for standing and walking. Most of the weight load on the foot is concentrated on the forefoot and big toe, whereas the other toes have a minor role in weight bearing. Moreover, the forefoot and big toe are important for maintaining balance and supporting the body when changing directions. Recently, attention has been focused on the aesthetic appearance and functional aspects of the body, which are important considerations in the field of reconstructive surgery. In patients for whom flap reconstruction in the forefoot and big toe is planned, clinicians should pay close attention to flap survival as well as functional and cosmetic outcomes of surgery. In particular, it is important to assess the ability of the flap to withstand functional weight bearing and maintain sufficient durability under shearing force. Recovery of protective sensation in the forefoot area can reduce the risk of flap loss and promote rapid rehabilitation and functional recovery. Here, we report our experience with two cases of successful reconstruction of the forefoot and big toe with a sensate anterolateral thigh flap, with a review of the relevant literature.

  • Background: Mandibular deficiency leading to facial asymmetry causes cosmetic deformity as well as psychological stigma for the patient. Correction of these mandibular asymmetries is a major challenge. The study investigates the efficacy of bidirectional mandible distraction for the treatment of mandibular deficiency. Methods: This prospective study included six individuals aged between 17 and 24.4 years. Five patients had hemifacial microsomia and one had unilateral temporomandibular joint ankyloses. All patients underwent mandibular distraction osteogenesis. Postoperative skeletal changes in affected mandible, and changes in occlusal plane and oral commissure cant were evaluated using three-dimensional reconstruction. Patient satisfaction and understanding of the procedure were assessed through three questionnaires administered during pre-distraction, distraction and post-distraction phases. Results: In pre-distraction phase, aesthetic appearance seemed to be the primary indication for surgery. In distraction phase, pain while chewing was the primary handicap. In post-distraction phase all patients were satisfied with the aesthetic outcome. The facial deformity was improved through mandibular distraction osteogenesis. On the affected side in all the patients, height and length of the mandible increased. Canting of the occlusal plane and oral commissure was corrected. Conclusion: Bidirectional mandible distraction is an effective treatment for correction of mandible deformities in adult patients.

  • Chronic progressive external ophthalmoplegia (CPEO) is a complex slowly progressive mitochondrial disorder characterized by extraocular muscle weakness with or without multisystem involvement. The mainstay of therapy in a patient with CPEO is supportive. However, in moderate cases, surgery might be indicated including surgeries for ptosis and strabismus. In this article, we report a Saudi patient with CPEO due to compound heterozygous variants in the DNA polymerase gamma (POLG) gene c.2246T>C p.(Phe749Ser) and c.1735C>T p.(Arg579Trp), which are classified as pathogenic. Proper diagnosis with genetic testing confirmation is important to guide the management and counsel the patient about the prognosis and the management options. The patient was successfully managed with bilateral frontalis sling and illustrates the importance of surgical intervention to improve vision and cosmetic appearance in patients with CPEO. We emphasize the importance of multidisciplinary care in the management of cases of mitochondriopathy, especially CPEO.

A Study on the Mechanical Change of Emulsion-Treated Hair by Color

  • With the increasing interest in the expression of individuality and appearance of modern people, it is time to conduct research and development of novel hair coloring from various angles. Therefore, taking into account the order of discoloration of hair pigments, we selected a creative and novel emulsion as a novel material for hair coloring, rather than a cosmetic material such as hot water extract using natural products dealt with in previous studies, commercially available hair manicure, and oxidation hair dye for hair. Thus, the change in tensile strength and elongation of hair samples by color was studied. As a result of the study, hair with green emulsion paint had a significantly higher maximum load, maximum stress, maximum elongation and breaking load, breaking stress, breaking elongation values are shown. Maximum in terms of modulus, green emulsion applied hair and the control group were higher in the 0-15s strain and 15-145s sections, respectively, and the tangential modulus value was much higher in the control group than the experimental group hairs in all the 0-145s sections. This study, which analyzes the dynamic changes of hair samples that extend the daily color gamut, will greatly contribute to the development of innovative hair coloring materials in the research and production of hair beauty works, and it is judged that it will also contribute to the development of the beauty industry.

  • Various flaps are used to reconstruct skin and soft tissue defects of the vulva following resection of malignancies. Whenever possible, reconstruction using local flaps is the standard treatment. Here, we describe vulvar defect reconstruction using keystone flaps. Standard keystone flaps are based on randomly located vascular perforators. However, we designed a keystone flap that includes perforators of three named arteries (the anterior labial artery of the external pudendal artery, cutaneous branches of the obturator artery, and posterior labial artery of the internal pudendal artery) and the pudendal nerve, which accompanies the internal pudendal artery. Four patients with squamous cell carcinoma and extramammary Paget's disease of the vulva underwent radical vulvectomy and keystone flaps including perforators of three arteries. Depending on the morphology of the defects, keystone flaps were used with different designs. For elliptical and unilateral vulvar defects, a standard keystone flap was designed, and for defects on both sides of the vulva, a double opposing keystone flap was used. For oval defects, the omega variant keystone flap was designed, and when the morphology of the defect needed rotation of the flap, a rotational keystone flap was designed. All the patients showed good function and sensation, with an acceptable cosmetic appearance.

  • Today, men's appearance is an important part of the global fashion industry, including Korea, with such fashion concepts as the Metrosexual and Ubersexual. Domestic and overseas beauty and cosmetics related organizations are using this phenomenon for marketing and researching to improve the function of cosmetics. The purpose of this study was to investigate the effect of Prunus persica L. Batsch leaf extract on the skin condition after clinical application to male subjects with oily skin to determine the possibility of functional cosmetics utilizing this extract. The cleansing bars were prepared by the MP method and the cleansing packs were prepared using natural ingredients. After 8 weeks of application, moisture, oil, erythema, melanin, pH, elasticity and tactile sensation were measured using MPA5. The results showed that there was a statistically significant difference in T-zone moisture and T-zone oil after using the natural cleansing bar, but there was no significant difference between the two groups for other items. However, the statistically significant changes were found in the individual skin condition change items of the subjects in each group, and it was judged that the two kinds of formulations could be used at the same time to have a higher skin improvement effect. Thus, Prunus persica L. Batsch leaf extract demonstrates potential to be used as a functional cosmetics item.

Functional Cosmetic Characteristics of the Oxalidaceae Extracts (괭이밥과 추출물의 기능성화장품 소재로서의 특성)

  • In this study, methanol extracts of the Oxalidaceae were tested with a potential functional cosmetic agent. As cosmetic agent tests, cell toxicity, polyphenol content, antioxidation, anti-wrinkle, and whitening effects were measured. Cell toxicity of the extracts was weak up to $1,000{\mu}g/mL$. Polyphenol contents of Oxalis corniculata L., Oxalis obtriangulata Maximowicz and Oxalis articulata Savigny were $116.036{\pm}0.37mg/g$, $54.72{\pm}0.52mg/g$ and $88.18{\pm}1.15mg/g$, respectively. Oxalis corniculata L., Oxalis obtriangulata Maximowicz and Oxalis articulata Savigny extracts showed 89%, 80% and 88% of antioxidation effects at $1,000{\mu}g/mL$ concentration using DPPH free radical scavenging assay. Oxalis corniculata L., Oxalis obtriangulata Maximowicz and Oxalis articulata Savigny extracts indicated 81%, 51% and 57% of antiwrinkle effects at $1,000{\mu}g/mL$ concentration using elastase inhibition assay. Oxalis corniculata L. extract was particularly excellent in elastase inhibition effect. Whitening effect using tyrosinase inhibition assay was relatively weak. Lotion formulation including 1% Oxalis obtriangulata Maximowicz extract was stable based on the temperature stability test for 28 days in terms of pH, viscosity and appearance. However, Lotion formulation including 1% Oxalis corniculata L. extract and Oxalis articulata Savigny extract need formulation improvement. From the research, methanol extract of Oxalis corniculata L. seems to be good candidate for antiwrinkle functional cosmetic agent.

Evaluation of the Potential of Cellobiose as a Material for Whitening Cosmetics based on Autophagy and Melanin Production Efficacy in Melanocytes (셀로비오스의 미백화장품 소재 가능성 평가를 위한 멜라닌 세포에서 자가포식 및 멜라닌 생성 효능 연구)

  • Cellobiose is a dissacharide constituted by two glucose units joined by a β-('1,4') glycosidic bond that is produced by the decomposition of cellulose. This product exists naturally in plants and has been utilized in different industries as a food sweetener, and as a cosmetic and pharmaceutical material. In this study, the potential of cellobiose as a whitening cosmetic product was evaluated by analyzing autophagy induction and the inhibition of melanin production. A cytotoxicity test conducted in the human melanin-producing cell line MNT-1 with increasing concentrations of cellobiose revealed that this compound did not cause cytotoxicity at 20 mg/mL or less. Based on this, autophagy was firstly evaluated by immunostaining with the autophagy marker microtubule-associated protein 1 light chain 3 (LC3) after treatment with 20 mg/mL of cellobiose. The subsequent confocal microscopy analysis revealed an increase in LC3 puncta, indicating induction of autophagy. In addition, autophagy was further confirmed by western blot analysis, which demonstrated that cellobiose converted LC3-I to LC3- ∏ in a concentration- and time-dependent manners. An analysis of melanin contents after cellobiose treatment at a concentration of 20 mg/mL during 7 days revealed that melanin production was reduced by more than 50%. Additionally, the expression levels of melanogenesis-related proteins TYR and TYRP1 were markedly decreased after cellobiose treatment. Based on these studies, a cosmetic cream formulation containing cellobiose was prepared and the change in formulation was tested for 4 weeks, and it was confirmed that the appearance changed to liquid form at high temperature, but the pH did not change. In conclusion, the present research demonstrated that cellobiose activates autophagy and inhibits melanin production, and showed the potential of this product as a material for whitening cosmetics.

Anti-aging Effects of the Extracts from Leaf. Stem, Fruit and Seed of Yew (Taxus cuspidata Sieb) by Solvent Extraction Method (용매추출법에 의한 주목의 잎, 줄기, 과실 추출물의 항 노화 효과)

  • Yew (Taxus cuspidata Sieb.) chose that grow as medicine, food, decorative plant in Korea's Kyong-Gi province surroundings. Extracts of yew extracted leaf of 250g and stems of 300g with 1,3-butylene glycol (l,3-BG), propylene glycol (PG) and water. As results, external appearance of leaf extract of yew was slightly brown clear extract. The pH was 5.3${\pm}$0.5, and specific gravity was 1.012${\pm}$0.05, and refractive index was l.375${\pm}$0.05. Also, appearance of stem's extract was slightly brown clear extract, and the pH was 5.4${\pm}$0.5, and specific gravity was 1.016${\pm}$0.05, and refractive index was 1.358${\pm}$0.05. Oil of yew separated from seeds, and extracted polysaccharide high purity from fruits. As a result, specific gravity of oil was 0.987, and obtained 40.0% of yield. Total polyphenols amount of yew extract is detected 0.563% in leaves, 0.325% in stems, whereas total tannins amount contained 0.054% and 0.037% each in leaves and stems. As effect in cosmetics, the anti-oxidative effect by DPPH method is 75.0% in leaves, and stems was 64.0%. Collagen synthesis rate was shown high activity by 54.16% in stem's extract, 33.18% in leaves' extract. Also, PPE-inhibitory activities were 13.7% and 23.5% each in leaves and stems. Anti-inflammatory effect of yew seed oil displayed superior effect of 41% than control. Polysaccharide's molecular weight that is gotten from fruits was 5${\times}$10$^4$-3${\times}$10$\^$5/ dalton, and got 20.0${\pm}$5% of yield.
