Detection of Dopamine and Serotonin by Competitive Enzyme-Linked Immunosorbent Assay (경쟁적 ELISA를 이용한 도파민과 세로토닌의 검출)

  • Dopamine (DA) and serotonin (5-Hydroxytryptamine, 5-HT) are neurotransmitters and hormones that exist in small amounts but have important role in the body. Serum and 24-hour urine are used as specimens, and are usually examined by HPLC-MS. In this study, we tried to detect DA and 5-HT by competitive ELISA using antigen-antibody (Ab) reaction. After immobilizing $5{\mu}g/mL$ BSA conjugate on a 96-well surface, hormone and primary Ab, which are respectively diluted to different concentrations, were treated. Then, HRP-conjugated secondary Ab and TMB were added to measure absorbance. The regression equation and $R^2$ value were calculated based on absorbance, and sensitivity of Ab to hormone as well as the correlation between hormone concentration and absorbance were determined. In DA ELISA, $R^2$, the correlation between the concentration of hormone and absorbance, was the highest by 0.91 when anti-dopamine Ab was diluted 6,000 times and 7,000 times. In 5-HT ELISA, $R^2$ was bigger than 0.90 in every concentration except 3,000 times and 6,000 times. Both DA and 5-HT were not effectively detected at low concentrations (less than $1.0{\times}10^{-7}M$); and because reference value of serum DA is lower than this, HPLC-MS was required to detect serum DA. However, competitive ELISA may be effective in detecting 24-hour urine DA, serum, and 24-hour 5-HT. Further studies are needed to detect hormones more accurately at lower concentrations.

  • In order to predict the possible fertilizer requirement from the K supplying capacity of soil, the relative K activity ratio, Kas/Kai and Gapon coefficients, KG. were determined for the soil samples before flooding and at heading stage of rice in pot experiment. These parameters assumed as the K supplying capacity of soils were discussed through correlation with other factors such as grain yields or the amounts of $K_2O$ uptake by the rice plant. The results may be summarized as follows: 1. The KGo values in soils before flooding were 7.8, 6.6, and 7.1, whereas the Kas/Kai values were 1.37, 1.26 and 2.11, respectively, in clay, loam and sandy loam soils. 2. The significant yield responses to the application of potassium fertilizer were observed whenever the KG values in soils at heading stage become larger to the original KG values, regardless of any levels of fertilizer application. 3. The linear correlations between the exchangeable cation ratios [Kex./(Ca+Mg) ex.:me/100g] in soils and the potassium activity ratios ($[K^+]/\sqrt{[Ca^{{+}{+}}+Mg^{{+}{+}}]}$: mole/l) in equilibrium solutions were observed with different linear gradients according to the soil properties. 4. The Kas/Kai in the soils, estimated prior to the experiment, showed high correlations with the grain yields or the amounts of $K_2O$ uptake in the all treatments, while the Kas/Kai and the KGo in the soils at heading stage showed high correlations with the grain yields or the amounts of $K_2O$ uptake in only N 15 Kg/10a treatments. 5. The Kas/Kai and the KGo values determined in the soil at heading stage of rice showed high negative correlation each other and they could be used as soil factors for predicting potassium fertilizer requirement.

Potassium and Clay Minerals in Upland Soils (밭 토양(土壤)의 점토(粘土) 광물(鑛物)과 가리(加里))

  • The present paper summerizes the studies on clay mineralogical characteristics of Korean soil, relationship between potassium and clay minerals, potassium release pattern of clay minerals and utilization of clay minerals for soil conditioner and fertilizers, which have been carried out in this laboratory. 1. The red yellow podzolic soil is mostly abundant in the upland of Korea and mainly consists of halloysite and weathered intermediates of mica such as illite and vermiculite. 2. With regard to soil parent material, kaolin mineral occurs abundant in soils derived from granite and granite gneiss. Mica is dominant in basaltic soil. The main clay mineral of the soil, originated from the Tertiary, is found montmorillonite and the volcanic soil of Jeju Island has plenty of allophane as its main clay mineral. 3. It is confirmed that the soil fertility depends on the composition of clay minerals. The red yellow podzolic soil, containing lot of kaolin, shows low productivity while the montmorillonite soil has higher productivity. 4. The release rate of solid phase potassium (micas and fixed potassium) follows the 1st order reaction equation in the equilibrium solution of $IN-NH_4OAc$. The potassium release constant is positively correlated with the mica content of the clay but negatively correlated with the content of $14.5{\AA}$ minerals. On the other hand, the potassium release constant has very high correlation with the ratio(Kex/Kt) of exchangeable potassium(Kex) to total potassium(Kt). 5. It is also found that Kex/Kt has rather high correlation with the content of mica and $14.5{\AA}$ minerals existed in the clay as well as the mica content of the soil.

The Prediction of Ambient Temperature and the Correlation Analysis for Carbon Dioxide, Carbon Monoxide and Relative Humidity in Gwangju (광주지역 기온변화 예측과 $CO_2$, CO, 상대습도와의 상관성분석)

  • The ambient temperature and concentration of carbon dioxide in Gwangju and the reducing method of temperature, air pollutants were investigated using the atmospheric data in Gwangju. Average ambient temperature ($T_{a-ave}$ was $13.5^{\circ}C$ during 1961 to 2008. The temperature was predicted as increasing of about $2.7^{\circ}C$ in 2108 after 100 years using the trend line of regression equation. Carbon dioxide was 370.7 and 391.4 ppm at Anmyundo, in 1999 and 2008, respectively, showing proportionally increased as ambient temperature. The temperature at Gwangju, $14.2^{\circ}C$ during 1997 to 2008, was a little higher than at neighboring counties as Naju, Damyang, Hwasoon, and Jangsung. In Gwangju, Spring will start in mid-January of 2108, Summer in mid-May, Autumn in mid-October, and Winter in last-December. The average relative humidity in the air ($RH_{a-ave}$) was gradually decreased as the temperature inversely increased. The average $CO_2$ was 457 ppm, which is 65.6 ppm higher than that in Anmyundo, korean background area of $CO_2$ in 2008. Carbon dioxide showed positive correlation, both of them, with carbon monoxide (0.87) and relative humidity (0.48).

Relationship of between blood lead level and lead related symptoms in low level lead exposure (저농도 연폭로에서 혈중 연농도와 자각증상과의 관계)

  • This study intended to obtain an useful information on the prevalence of subjective symptoms, and to clarify the interrelationships between blood lead and lead related symptoms in low level lead exposure. The 93 male workers exposed to lead and 56 male nonexposed workers were examined for their blood lead(PBB), Zinc-protoporphy(ZPP), hemoglobin(HB) and personnal history, and completed 15 questionnaires related to symptoms of lead absorption : also measured lead concentration in air (PBA) in the workplace. The results obtained were as follows ; 1. The means of blood lead (PBB), blood ZPP and hemoglobin (HB) among workers exposed to lead were $26.1{\pm}8.8{\mu}g/dl,\;28.3{\pm}26.0{\mu}g/dl$ and $16.2{\pm}1.2g/dl$ : whereas those of nonexposed workers were $18.7{\pm}5.1{\mu}g/dl,\;20.6{\pm}8.7{\mu}g/dl$ and $17.3{\pm}1.1g/dl$. The means of above three indicies between two groups showed significant difference statistically (p<0.05). 2. The means of blood lead (PBB), blood ZPP and hemoglobin of workers exposed .to different lead concentration in air were as follows : When it was below $25{\mu}g/m^3$, the indices were $24.7{\pm}79,\;26.1{\pm}26.8{\mu}g/dl\;and\;16.4{\pm}1.1g/dl$ respectively : These indices were $27.1{\pm}8.5,\;23.9{\pm}10.92{\mu}g/dl\;and\;16.2{\pm}1.3g/dl$ when the lead concentration in air was $25{\sim}50{\mu}g/m^3$ : and they were $3.4{\pm}9.3,\;42.3{\pm}31.3{\mu}g/dl\;and\;15.5{\pm}1.2g/dl$ when the concentration of lead was above $50{\mu}g/m^3$. Although there were statistical difference in blood lead and hemoglobin among three different lead concentration in air, there was no statistical difference of blood ZPP among the three groups with different exposure levels (p>0.05). 3. The most frequent by complained symptom was 'Generalized weakness and fatigue', and fewest symptom was 'Intermittent pains in abdomen' 4. Only two symptoms out of fifteen symptoms checked by themselves revealed significant difference between exposed and nonexposed groups. These were 'Intermittent pains of abdomen' and 'Joint pain or arthralgia' (p<0.05), No positive correlation was found between the levels of blood lead and symptom groups categorized as gastrointestinal, neuromuscular and constitutional symptoms, 5. Blood lead (r=0.3995) and ZPP (r=0.2837) showed statistically significant correlation with mean lead concentration in air, whereas correlations were not demonstrated between blood lead and lead related symptoms or blood ZPP and lead related symptoms. 6. Blood lead (PBB) and ZPP showed association (r=0.2466) and the equation PBB=23.75+0.0842 ZPP was derived. 7. On stepwise multiple regression, using blood lead level as a dependent variable and ZPP, hemoglobin (HB), age, work duration (WD) and symptom prevalence as a independent variables, only ZPP significantly contributed a lot to blood lead level. 8. While the ZPP measurement was found to be a good indicator in evaluating health effect of lead absorption in low level lead exposure, lead related symptoms were not sensitive enough to evaluate of lead absorption in low level exposure.

  • In the first paper of the series the five soil fertility factors were evaluated by means of principal component analysis and varimax method. They are interpreted as representing, 1) skeletal available phosporus status, 2) organnic matter status, 3) salt status 4) base status, and 5) free oxide status. In order to resynthesize such fragmented information for the overall soil fertility evaluation, the method of multiple regression analysis was adopted, using the five factor scores and yield data for Korean paddy soils as independent and dependent variables respectively. As test of linear models with different combinations of independent variables the results of t-test of regression coefficient were revealed that the organic matter status (FII) has no relevance to the yield of paddy and that the free oxides and salt supply has by it self only an insignificant contribution to the yield. The multiple correlation coefficient (R) revealed its multiple regression analysis was as low as 0.43. Introduction of quadratic terms to the linear model bettered the result. Thus multiple correlation coefficient (R) was increased as 0.59. Therefore, a coefficient of determination 0.35 was obtained by a quadratic model with interaction terms among the five fertility constituents. Generally we think that the fertility factor has more contribution to raise the rice yield in paddy and that the failure of yield prediction by fertility factor scores was caused by one of follows; 1) the roughness of the yield inspection, and 2) missextraction of fertility constituents. The second step in this study, assuming that the residuals by multiple regression analysis were due to factors other than soil fertility, we can now proceed to predicting the yield from the field characters with the classified fertility groups by means of Hayashi's theory of quantification No. 1. Such variables as fertility groups (FTYG), water availability (WATER), soil drainage (DRNG), climatic zone (CLIZ), surface soil's stickiness (STCKT), surface soil's dry consistence (DCNST), and surface soil's texture (FTEXT) are taken up as the explanatory variables. The quantification appears reasonable; the well to extremely well in soil drainage, very sticky of surface soil, inefficiency in water availability, coarse texture, and very hard to extremely hard dry consistence in soil are detrimental to the rice yield. The R was as high as 0.90 for the set of variables. But the given explanatory variables in this study were not quite effective in explaining rice yield. The method developed seems to be promising only if properly collected data are available. Conditions that should be satisfied in the yield inspection obtained from common cultivator for the purpose of deriving a prediction equation were put forward.

Comparison of TME Values of Imported Corns and ME Values Determined by In vitro Method (수입 옥수수들의 TME가와 In vitro 방법에 의해 측정한 ME가의 비교)

  • An experiment was conducted to compare the ME values of imported corns measured by In vivo(TME) and In vitro(MEn, ME and MEpc) methods and to investigate the true amino acid availability(TAAA) and the true nutrient availability(TNA) of imported corns. For TME assay, twenty four 57-weeks-old Hy-Line roosters were assigned to fasting group(4 roosters) and four corn groups(5 roosters each): USA; corn produced in the United States, ARG; corn produced in Argentina, CHN; regular corn produced in China, CHNP; premium corn produced in China. The MEn, ME and MEpc values were determined by equations based on chemical analysis. The TME value of USA(3,745kcal/kg) in as fed basis was significantly (P<0.01) higher than ARG(3,555kcal/kg) and CHNP(3,518kcal/kg) but was not significantly different from CHN(3,671kcal/kg). The TME value of USA(4,144kcal/kg) in DM basis was not significantly different from CHN(4,060kcal/kg) and CHNP(4,008kcal/kg) but was significantly(P<0.05) higher than ARG(4,001kcal/kg). There were significant differences in TAAA of phenylalanine, histidine and arginine among imported corns. Those of USA were highest but overall TAAA was not significantly different among imported corns. True availability of NFE of USA, ARG and CHN was significantly(P<0.05) higher than that of CHNP. However, true availability of crude protein, crude fat, crude fiber and crude ash were not significantly different among corns. The correlation coefficient between TME and MEn value was 0.91 which was significant at P<0.1 but correlation coefficient between TME and ME value and between TME and MEpc value was 0.90 and 0.83, respectively which was not significant at P<0.1. In conclusion, US corn was highest in TME values and Chinese premium corn was not significantly different from regular Chinese corn. The MEn value obtained by equation based on chemical analysis may be used as a tool to evaluate TME value of corn.

Estimating the Yield of Marketable Potato of Mulch Culture using Climatic Elements (시기별 기상값 활용 피복재배 감자 상서수량 예측)

  • The object of this study was to evaluate the effects of climatic elements on potato yield and create a model for estimating the potato yield. We used 35 yield data of Sumi variety produced in mulching cultivation from 17 regions over 11 years. According to the results, some climatic elements showed significant level of correlation coefficient with marketable yield of potato. Totally 22 items of climatic elements appeared to be significant. Especially precipitation for 20 days after planting (Prec_1 & 2), relative humidity during 11~20 days after planting (RH_2), precipitation for 20 days before harvest (Prec_9 & 10), sunshine hours during 50~41 days before harvest (SH_6) and 20 days before harvest (SH_9 & 10), and days of rain during 10 days before harvest (DR_10) were highly significant in quadratic regression analysis. 22 items of predicted yield ($Y_i=aX_i{^2}+bX_i+c$) were induced from the 22 items of climatic elements (step 1). The correlations between the predicted yields and marketable yield were stepwised using SPSS, statistical program, and we selected a model (step 2), in which 4 items of independent variables ($Y_i$) were used. Subsequently the $Y_i$ were replaced with the equation in step 1, $aX_i{^2}+bX_i+c$. Age Estimation by Modified Johanson Method (Johanson 방법을 응용한 연령추정)

  • Age estimation is fundamental and important in personal identification with forensic medicine and dentistry. Recently, a lot of studies using various part of the body have been done for age estimation. Age estimation with teeth is the most significant method comparing ones with other part of the body. Gustafson method and Johanson method using postmortem teeth have been authorized in accuracy and systemization and used domestically and internationally. The verification of the accuracy in above methods had been tried many times but it is still rare in Korea. Fifty-nine teeth(incisors, canine, premolars and molars) which were extracted due to periodontal diseases or orthodontic problem were collected. Present study is to 1) compare the accuracy of estimated age in applying Gustafson method and Johanson method to the teeth in Korea, 2) compare and analyze the correlation with results using Gustafson method and Johanson method by age, gender, maxilla - mandible and anterior - premolar - molar. Teeth were embedded in resin and sliced and then examined each one using Gustafson method and Johanson method. The results are as follows: 1. Actual age was a significant difference in estimated age by Johanson method. Actual age was a significant correlation in estimated age by Gustafson method and modified Johanson method. Modified Johanson method was more significant than Gustafson method. 2. In estimated age by Gustafson method, Johanson method and modified Johanson method, there was no significance with actual age by location and gender. 3. In estimated age by Gustafson method, Johanson method and modified Johanson method, there was significance with actual age by age group. Finally, Gustafson method and Johanson method can be used in Korea. To make more accurate verification, however, it needs more specimen and postmortem teeth. Johanson equation proposed by himself has to be developed by further studies.

Establishment of Economic Threshold by Evaluation of Yield Component and Yield Damages Caused by Rice Leaf Blast (Magnaporthe grisea) (벼 잎도열병 피해해석에 의한 경제적 방제수준 설정)

  • This study was conducted to decide disease incidence level of rice leaf blast required for reasonable fungicide application in paddy field. We induced the disease development by inoculating rice blast pathogens on rice seedlings (Jinmibyeo) in the greenhouse and transplanting the infected seedlings in the field two weeks after rice plants were transplanted. We scored the disease incidence, grouped and marked according to degree of percentage of diseased leaf area at maximum stage of disease development. The percent diseased leaf area (PDLA) had negative correlations with panicle number per hill, ripened grain (%), and total yield; their correlation coefficients (r), $-0.97^{**}$, $-1.00^{**}$ and $-0.96^{**}$, respectively. However, it had positive correlations with spikelets per panicle and thousand grain weight; their correlation coefficients (r), $0.98^{**}$ and $0.98^{**}$, respectively. Gain threshold (GT) calculated based on control cost and market price was estimated to be 8.35. Economic injury level (EIL) obtained based on GT and coefficient of damage of regression equation between disease incidence and the different yield components; panicle number per hill, spikelets per panical, ripened grain(%), thousand grain weight and yield were 41.8, 9.7, 19.1, 291.1 and 3.4%, respectively. Economic threshold (ET) for yield was 2.7% ($3.4%(EIL){\times}0.8$) on PDLA. These results suggest that application of fungicide is necessary when two under leaves are almost covered with lesions or contained more than twenty large lesions under leaves at maximum tillering stage.
