암컷 마우스 생식기관 내 수은 화합물의 위치와 시간에 따른 축적된 수은 농도 변화 (Localization and Accumulated Concentration Changes of Mercury Compound in Reproductive Organs of Female Mice with Time)

  • 본 연구에서는 암컷 생식기관 내 축적된 수은 화합물의 위치와 시간에 따른 수은의 농도 변화를 확인하기 위하여 수행하였다. Methylmercuric chloride를 일주일에 한 번씩 총 3주간 사춘기 암컷 마우스에 피하 주사하였다. 시간에 따른 수은의 농도 변화를 확인하기 위해 투여 종료 후 10일, 150일, 300일째에 희생하였다. 투여 종료 후 10일에 희생한 경우 체중에 유의한 차이가 있었지만, 그 외 경우에는 체중과 난소의 무게에 있어서 대조군과 수은 투여군 사이에 유의한 차이가 없었다. 축적된 수은의 위치를 오토메탈로그라피 방법으로 확인하였고, 자궁, 난소, 난자에 축적된 수은 화합물의 위치를 광학현미경으로 분석하였다. 자궁의 경우, 투여 종료 후 10일째에 수은이 지질 세포와 자궁바깥막의 중피에 위치하였다. 150일째에는 수은 농도가 감소하였으며 300일째에는 나타나지 않았다. 투여 종료 후 10일째 난소의 경우, 수은이 피질 부위의 지질 세포와, 난포를 둘러싸는 난포막 세포, 황체에 분포하였다. 150일째에는 수은이 난소의 수질 부위에 축적되었으며, 300일째에는 분포하지 않았다. 투여 종료 후 10과 150일째 난자의 경우, 수은이 난자의 주변부에 주로 분포하였으며, 300일째에는 수은 농도가 감소되고 난자 전체에 고르게 분포하였다. 이런 결과는 암컷 마우스에서 수은에 의해 호르몬 생성, 착상, 그리고 발생 중인 배아가 영향을 받을 수 있다는 사실을 제시해 준다.

젖소에서 $PGF_2{\alpha}$ 투여에 의한 발정 유도 후 수태율과 다른 인자와의 관계 (Relationship between the Conception Rate after Estrus Induction using $PGF_2{\alpha}$ and Other Parameters in Holstein Dairy Cows)

  • The purpose of this study was to determine the relationship between conception rate and other parameters (body condition score; BCS, progesterone concentrations and follicle size) before estrus induction with $PGF_2{\alpha}$. The conception rate in cows with (2.75, 2.75 to 3.25 and 3.25), BCS regardless of AI (artificial insemination) time was 47.5, 67.5% and 48.5% at $PGF_2{\alpha}$ injection, respectively. The conception rate regardless of BCS was 59.0% in cows inseminated based on detected estrus, and 46.2% in cows inseminated at 72 to 80 hours (timed artificial insemination, TAI) after $PGF_2{\alpha}$ injection. The conception rate regardless of AI time was 43.0% in cows with low progesterone concentrations (less than 1.0 ng/ml), and 67.5% in cows with high progesterone concentrations (more than 1.0 ng/ml) at $PGF_2{\alpha}$ injection. The conception rate regardless of progesterone concentrations was 59.9% in cows inseminated based on detected estrus, and 48.1% in cows of TAI after $PGF_2{\alpha}$ injection. The conception rate regardless of AI time was 36.0% in cows with small dominant follicles (less than 5 mm), 56.0% in cows between 5 mm to 10 mm of follicle size, and 65.5% in cows with large dominant follicles (more than 10 mm) at $PGF_2{\alpha}$ injection, respectively. The conception rate regardless of follicle size was 57.3% in cows inseminated based on detected estrus, and 47.6% in cows of TAI after $PGF_2{\alpha}$ injection. These results indicated that if the cows with BCS 2.75 to 3.25, active corpus luteum, and/or large dominant follicle (more than 10 mm) are used for estrus induction, the conception rate will be greater.

보중익기탕과 귀비탕 투여가 임신랫드의 모체 및 태자에 미치는 연구 (Toxicological Effect of Bojungiggitang and Gwibitang (Herbal prescription) in the Pregnant Rat and Fetuses -Focusing on Reproductive and developmental Toxicity-)

  • The experiments were undertaken to evaluate the effects of herbal medicine, Bojungiggitang and Gwibitang in pregnant rats and their fetuses. Female Sprague-Dawley rats were orally administered with the Bojungiggitang and Gwibitang at dose of 5ml/kg/day for 20 days. Pregnant rats were sacrificed at 20th day of gestation, and the internal and reproductive organs. Approximately live fetuses in the 20th day of gestation were randomly selected and fixed in 95% ethanol. To observe skeletal malformations, fetuses were stained with alcian blue and alizarin red S. Maternal body weights of Bojungiggitang and Gwibitang treated group has a tendency to increase compared to that of control group. There were no significant differences in internal and reproductive organs. There were no significant changes between two groups in blood chemistry and hematological values. There were no significant changes in number of corpus luteum, implantation and live fetuses. But Bojungiggitang and Gwibitang administered group showed higher implantation rate than the control group. Also, Bojungiggitang and Gwibitang administered groups showed lower early resorption rate than the control group. And Gwibitang had the higher value in all the other groups in all items. From the sex ratio, the number of females were larger than the number of males in the control group, and more males than females in Gwibitang administered group. Neonatal body weight and the number of fetus of Bojungiggitang and Gwibitang group were higher than that of control group. The fetuses of dams treated with Bojungiggitang and Gwibitang did not show external malformation. Vertebral and sternal variations were observed in Bojungiggitang and Gwibitang administered group compared to the control group. Those variations were insignificant. There were no significant changes in number of ribs, cervical, thoracic, lumbar, sacral and caudal vertebras. From these results, it can be concluded that Bojungiggitang and Gwibitang showed no toxic effects on maternal body weight and the number of live fetuses. There were no significant changes in organ weight, hematological data, and reproductive organs. Although skeletal variations were shown in vertebra and sternum, Bojungiggitang, Gwibitang were shown insignificant changes in bone malformation.

흰쥐 생식소에서 GnRH-like mRNA의 발현과 세포내 분포 (Expression and Cellular Localization of Gonadotropin-Releasing Hormone (GnRH)-like Messenger Ribonucleic Acid in the Rat Gonad)

  • 시상하부에서 합성, 분비되는 gonadotropin releasing horrnone (GnRH)의 면역반응성이 생식소를 비롯한 여러 부위에서도 검출됨이 알려졌으나, 이 펩타이트가 과연 생식소에서 국부적으로 합성되는 지에 관해서는 아직 밝혀지지 않았다. 본 연구에서는 흰쥐 생식소에서 GnRH유전자발현을 연구하기 위하여 GnRH-like mRNA와 GnRH펩타이트의 발현과 세포내 분포 양상을 조사하였다. GnRH 방사면역측정법과 GnRH를 크로마토그라피 방법으로 분리한 결과,시상하부에서 합성되는 GnRH와 유사한 GnRH 면역반은이 흰쥐 생식소 추출물에서 상당량 검출되었다. GnRH-면역반응이 흰쥐 난소의 다양한 세포군에서 나타냄에 반하여, GnRH-like mRNA는 granulsa,theca 그리고 luteal 세포에서만 주로 발현되었다. 또한 흰쥐 정소에서 GnRH면역반응성은 원시정세포, Sertoli,Leydig 세포에서만 검출된 반면에, GnRH-like mRNA는 정세관내의 Seertoli세포에서만 발현되었다. 따라서 이 연구는 생식소에 존재하는 GnRH는 생식소 내에서 국부적으로 합성, 발현되는 결과라고 사료되며, 생식소 내에서 생성된 GnRH는 생식소내 세포군간의 정보교환의 매개자로서 역활을 수행하고 있다고 추정된다.

안태음이 임신랫드와 태자에 미치는 영향 (The Effects of the Administration on Oriental Medicine, Antaeeum, in the Pregnant Rat and Their Fetuses)

  • This study have a object to found out the effects of oriental herb medicine, Antaeeum, to dams of rats and their offsprings. The Antaeeum was savaged to female Sprague-Dawley rats at a dose of 5 mg/kg/day for 3 weeks during gestation periods. Dams of rat were sacrificed at 20th day of gestation, and were observed major internal and reproductive organs. Approximately live fetuses in the 20th days of gestation were selected randomly and examined with stereo microscopes. Others offsprings were fixed with 95% ethanol for skeletal examinations. The fixed fetuses were stained with alcian blue and alizarin red S to observe skeletal variations or malformations. Maternal body weight of Antaeeum treated dams have a tendency of increasing compared with control dams. There were no significant difference in internal and reproductive organs of weight or findings. The spleenic organ relative weight of treated dams were decreased compared with the control significaltly (p<0.05). There were no significant changes between two groups in blood chemistry and hematological values. There were no significant changes in number of corpus luteum, implantation, live fetuses and implantation rate, delivery rate, late resorption rate and sex ratio. But in the Antaeeum treated group showed lower early resorption rate than that of the control dams. Fetal body weight and number of fetus a dam at Antaeeum treated group were higher than that of control group. The fetuses of dams treated with Antaeeum didn't induced external malformations. Vertebral and sternal variations were observed in Antaeeum group, but compared with the control, those variations were not significant. The ossification numbers of rib, cervical, thoracic, and lumber were normal. Fetuses treated with Antaeeum to the dams showed no significant difference in the number of caudal vertebra (P>0.01). From these results, it can be concluded that Antaeeum showed no toxicity effects on maternal side especially on body weight, early resorption rate, and number of live fetuses. Also there were no significant changes on maternal organ weights except spleen, hematological data, reproductive organs. Although skeletal variations were examined at vertebra and sternum, this Antaeeum could not induced significant choses in bone malformation.

면역형광측정법에 의한 우수정란의 성 판별 (Immunofluorescent Detection of H-Y Antigen on Preimplantation Bovine Embryos)

  • In order to determine the sex of preimplantation embryos prior to transfer in cattle, a series of experiments were carried out using 45 Holstein donor cows to examine the ovarian response on the gonadotropin and PGF2${\alpha}$, and the morphology of fresh embryos or frozen/thawed embryos after deep freezing at -196$^{\circ}C$. The sexing of embryos treated with the medium containing H-Y antiserum(10%, v/v) and FITC anti-mouse IgG(10%, v/v) were analysed by chromosomal analysis, and the sex of the embryos which survived were ascertain after delivering the pups. The results obtained were summarized as follows ; 1. The average number of developed follicle and corpus luteum per cow were 13.5 and 8.1, and the ovalation rate was 60.1%. 2. Of 220-ova recovered, 75(34.1%) were morula and 91(41.4%) were blastocyst, and the morphological normal and abnormal rate of ova recovered were 75.5% and 24.5%, respectively. 3. Of 39 frozen/thawed embryos, the scores of normal morula and blastocyst, after thawing were 79.2%(19/24) and 73.3%(11/15). The average rate of frozen/thawed embryos which appeared morphologically normal post thawing was 76.9%(30/39). 4. The sex ratio was measured using the embryos treated with immunofluorescence assay to examine the relationship between embryo developmental stage, sex ratio of morula stage embryo was 42.2%(19/45) fluorescing and 57.8%(26/45) non-fluorescing, on the other hand, the ratio switched to 46.8%(29/62) fluorescing and 53.2%(33/62) non-fluorescing embryo in blastocyst stage. The sex ratio was also measured between fresh and frozen/thawed embryos, fresh and frozen/thawed treated embryos were indicated 45.8%(38/83) fluorescing, 54.2%(45/83) non-fluorescing and 41.7%(10/24) fluorescing, 58.3%(14/24) non-fluorescing. This trend indicated the approximal sex ratio was 1 : 1. 5. The result of karyotype test showed the successful rate of sexing embryo is fluorescing and non-fluorescing was 21.2%(7/33) and 29.6%(8/27). The female to male ratio within 33 fluorescing was 28.6 : 71.4, and the ratio of 27 non-fluorescing embryos was 87.7 : 12.5. 6. Of the embryo transferred after assignment of H-Y phenotype, five of the fluorescing embryos survived to term, all was males. Whereas six non-fluorescing embryos also survived to term and the sexes of the calves were 1 male 5 female.

전자현미경에 의한 착상 전후 돼지수정란의 형태학적 변화에 관한 연구 (Microscopic Study of the Pig Peri-implantation Embryos)

  • 전자현미경에 의해 자궁부착 전후의 돼지 수정란의 형태형성 및 분화에 따른 배발생 과정을 검토하였다. 돼지 초기배는 자궁이주후 균일하게 자궁에 배분되기전 약 2~3일간은 자궁각의 proximal portion에 존재하며, 임신 4일째에 할구와 할구의 경계를 상실하는 tight한 gap junction을 가진 상실배로 발달한다. 배반포를 형성하는 시기에 estradiol 17$\beta$는 compact한 상실배를 cavitated blastocyst로 발달을 촉진시키면서, steroid hormone이 이후의 배발생을 지배한다. Hatching의 시기는 교배후 6~7일경 zona pellucida을 둘러사고 있는 glycoprotein의 thinning과 lysis에 의해 이루워지는데, hatching 과정은 embryo의 세포수와 무관하였으며, 이때의 embryo의 직경은 0.5~1.0mm인 것을 본 실험에서 확인하였다. 12일경부터는 embryo는 prostaglandins, IGF-binding protein, retinol binding protein, plasminogen activator등의 단백질이 풍부해 이들 인자가 elongation 개시 후보로 고려될 수 있었다. 또한 이 시기의 embryo는 embryonic disc로 발달시 progesterone과 estrogen을 estradiol 17$\beta$로 전활할 수 있으며, 이러한 변화와 함께 spherical stage로부터 tubular 혹은 filamentous form으로 변형되었다. Estrogen이 임신을 통해 prostagladins의 분비를 uterine lumen에 지시하는지는 알 수 없으나 13일 경을 전후해 conceptus estrogen이 uterine arterial blood flow, uterine vasular permeability을 증가시키는 것으로 나타났으며, 자궁에서 protein과 calcium, PGF2$\alpha$, plasminogen inhibitor를 증가시키는 것으로 나타났다. 이 시기의 자궁 변화와 함께 embryo의 attachment는 trophoblast와 uterine membrane사이의 느슨한 결합에 의해 개시되었으며, 18일경 uterine과 trophoblastic microvili의 interdigitation에 의해 완성된다. 이 시기에 conceptus attachment를 위해 필요한 uterine microvili에서의 glycocalyx의 형성과 endometrial epithelium의 erosion을 야기하기 위해 plasminogen activator을 분비하였으며, 반면 자궁에서 plasminogen 역할을 하는 것은 estrogen이며, blastocyst cell 표면의 lectin binding이 attachment에 중요한 역할을 한다. 이상과 같은 일련의 과정을 거친 초기배는 성공적인 임신으로 유도된다고 본다. 따라서, 본 연구는 이상과 같이 착상을 전후한 시기의 배를 전자현미경에 의해 형태형성의 변화를 특히 착상을 전후해 배 취사율이 높은 시기를 대상으로 분석하였다. 이 분석 시기중 성공적인 착상성공율은 56%(71/126)였다.

다배란처리와 발정동기화가 난소반응, 수태율 및 산자수에 미치는 영향 (Effect of Superovulation and Synchronization on Ovarian Response, Pregnancy Rate and Number of Newborn in Rabbit)

  • This study was carried out to investigate effects of superovulation and time of embryo recovery on ovarian response, recovery rate and developmental stage of embryo in donor and effects of methods of synchronization, number of corpus luteum (CL), stage of embryo and time of embryo transfer on ovarian response, conception rate and number of newborn in recipients which were transferred on 2.5, 3.5 and 4.5 days after synchronization. The results obtained are as follows; 1. The ovulation point of superovulated donor on 2.5, 3.5 and 4.5 days after copulation was 23.3, 35.3 and 23.3, respectively. The number of embryos recovered from the donors on 2.5, 3.5 and 4.5 days after copulation was 23.3, 25.8 and 19.8, respectively. The ovulation point and number of embryos recovered on 3.5 days were greater than those of 2.5 and 4.5 days. Among 232 embryos recovered on 3.5 days after copulation, 84 were blastocyst and 62 were hatching blastocyst. 2. The number of CL in recipients on 2.5, 3.5 and 4.5 days after synchronization was 3.2, 2.9 and 3.8 and showed no difference among the days. 3. When the number of CL was 0, 2-3, 4-6 and more than 7 the pregnancy raet of recipients was 0, 37.5, 66.7 and 75%, respectively. The pregnancy rate of recipients increased as the number of CL increased. 4. The pregnancy rate of transferred morula, blastocyst and hatching blastocyst was 32.0, 37.2 and 24.7%, respectively. The blastocyst nhowed highest pregnancy rate. 5. When the recipients were synchronized by HCG, the number of CL, unruptured follicle, hemorrhage, pregnancy rate and number of young were 5.5, 6.4, 3.3, 72.7% and 3.3, whereas that of GnRH were 2.3, 4.4, 2.8, 25.0% and 1.2, respectively. Recipients synchronized by HCG showed better results than GnRH. 6. When the embryos were collected on 2.5 days after copulation and transferred to the synchromized recipients, the pregnancy rate and the number of young born was 62.58% and 3.1, respectively. Those of 3.5 and 4.5 days after copulation was 57.1% and 1.3, and 37.5% and 1.6. The 2.5 days showed higher pregnancy rate and number of young born than 3.5 and 4.5 days.

Relationship between Peripheral Plasma Inhibin and Progesterone Concentrations in Sahiwal Cattle (Bos Indicus) and Murrah Buffaloes (Bubalus bubalis)

  • The present study investigated peripheral plasma immunoreactive inhibin (ir-inhibin) concentrations in relation to the stage of oestrous cycle and progesterone concentrations in cycling Sahiwal cattle (Bos indicus) and Murrah buffaloes (Bubalus bubalis). Blood samples were collected once daily for thirty-two consecutive days from cattle and buffaloes (5 each) during winter months of January and February. Mean (${\pm}$S.E.M.) plasma ir-inhibin concentrations ranged from $0.40{\pm}0.01$ to $0.59{\pm}0.03ng/ml$ in cattle and from $0.29{\pm}0.03$ to $0.52{\pm}0.05ng/ml$ in buffaloes. In cattle, ir-inhibin concentrations increased from $0.47{\pm}0.07ng/ml$ on day -4 (day 0=day of oestrus) to reach a maximum value of $0.59{\pm}0.03ng/ml$ on day -2. Thereafter, ir-inhibin concentrations showed a decline to reach a low of $0.40{\pm}0.01ng/ml$ on day 11 of the oestrous cycle. In buffaloes, ir-inhibin concentrations increased from $0.38{\pm}0.04 ng/ml$ on day -4 to reach a maximum concentration of $0.52{\pm}0.05ng/ml$ on day -2. Ir-inhibin concentrations then declined to reach a low of $0.29{\pm}0.03ng/ml on day 9 of the cycle. In both cattle and buffaloes, ir-inhibin concentrations which were lowest ($0.43{\pm}0.02$ and $0.34{\pm}0.02ng/ml$, respectively) during the mid-luteal phase of the oestrous cycle increased (p<0.05) to $0.52{\pm}0.03$ and $0.44{\pm}0.04ng/ml$, respectively, during the late luteal phase, and then further to the highest value of $0.53{\pm}0.02$ and $0.49{\pm}0.04ng/ml$, respectively, during the perioestrus phase, following which these declined to $0.50{\pm}0.02$ and $0.39{\pm}0.03ng/ml$, respectively, during the early luteal phase. The variations in peripheral plasma ir-inhibin profile in both the species appear to be related to the changes in characteristics of follicular populations during the oestrous cycle. Peripheral plasma ir-inhibin concentrations were negatively correlated with progesterone concentrations in cattle (r=-0.51, p<0.01) and buffaloes (r=-0.30, p<0.01) indicating that the corpus luteum is not a source of peripheral ir-inhibin in these species.

융모성성선자극호르몬 투여가 임신랫드의 황체및 태아에 미치는 영향 (Effects of Exogenous Human Chorionic Gonadotrophin(HCG) on the Corpus Luteum and Fetus in Pregnant Rats)

  • 임신랫드에 융모성성선자극호르몬을 투여한 후 모체의 황체와 태아에 미치는 영향을 조사하기 위하여 임신랫드에 융모성성선자극호르몬제제인 Profasai 1000(Serona, Switzland) 50 또는 100IU를 임신 4, 8, 12 또는 16일에 각각 투여한 후 임신20일에 부검하여 태아와 모체에 대한 조사를 한 결과를 요약하면 다음과 같다. 1. 황체는 모든 대조군과 HCG투여군에서 그 수가 거의 비슷한 수준이었고 임신8일의 경우에 그 수가 비교적 감소되었다. 2. 태아의 흡수는 임신 4, 8및 12일의 HCG투여군과 임신 12일의 대조군에서 나타났지만 임신4일의 경우가 발생빈도가 현저하게 높았고 임신 12일의 경우는 매우 낮았다. 3. 태아의 체중은 임신4일에 50IU의 HCG투여군에서만 감소되었다. 4. 태아의 골격검사에서 기형발생과 골화의 이상은 인정되지 않았지만 변이는 인정되었다. 변이는 14번늑골만 인정되었으며, 임신 4, 8 그리고 12일의 HCG투여군과 임신8일과 16일의 대조군에서 다양하게(5.26~8.33%) 나타났다. 5. 임신랫드에 HCG를 투여하면 태아의 흡수가 나타날 수 있지만 골격형성 과정에서 반드시 변이가 발생된다고 생각하지 않는다.
