Production of Xylooligosaccharides by Yeast Cell Surface-Displayed Endoxylanase (효모 세포 표면 발현된 Endoxylanase를 이용한 Xylooligosaccharides의 생산)

  • The yeast surface expression system, pCTXYN (6.8 kb), of Bacillus endoxylanase gene (xynB, 642 bp) was constructed and introduced into Saccharomyces cerevisiae EBY100 cell. The transformed yeast cell showing the highest endoxylanase activity was selected through the active staining of colonies grown on YPDG medium containing xylan. With the yeast transformant, EBY100/pCTXYN, grown on galactose containing medium, it was found that the endoxylanase was successfully displayed on the yeast cell surface and the xylooligosaccharides were efficiently produced from xylan. The most of endoxylanase activity was detected in the cell fraction and reached about 1.9 unit/mL after 48 h cultivation. The optimized conditions for xylooligosaccharides production from xylan were determined as follows: substrate and its concentration, oat spelt xylan 6%; concentration of yeast whole-cell, 5 unit/mL; temperature, $50^{\circ}C$, and reaction time $2{\sim}4\;h$. When the oat spelts xylan and corncob xylan were hydrolyzed by treatment with cell surface-displayed endoxylanase, xylotriose was formed as a main product.

Two-Step Process Using Immobilized Saccharomyces cerevisiae and Pichia stipitis for Ethanol Production from Ulva pertusa Kjellman Hydrolysate

  • We established a two-step production process using immobilized S. cerevisiae and P. stipitis yeast to produce ethanol from seaweed (U. pertusa Kjellman) hydrolysate. The process was designed to completely consume both glucose and xylose. In particular, the yeasts were immobilized using DEAE-corncob and DEAE-cotton, respectively. The first step of the process included a continuous column reactor using immobilized S. cerevisiae, and the second step included a repeated-batch reactor using immobilized P. stipitis. It was verified that the glucose and xylose in 20 L of medium containing the U. pertusa Kjellman hydrolysate was converted completely to about 5.0 g/l ethanol through the two-step process, in which the overall ethanol yield from total reducing sugar was 0.37 and the volumetric ethanol productivity was 0.126 g/l/h. The volumetric ethanol productivity of the two-step process was about 2.7 times greater than that when P. stipitis was used alone for ethanol production from U. pertusa Kjellman hydrolysate. In addition, the overall ethanol yield from glucose and xylose was superior to that when P. stipitis was used alone for ethanol production. This two-step process will not only contribute to the development of an integrated process for ethanol production from glucose-and xylose-containing biomass hydrolysates, but could also be used as an alternative method for ethanol production.

Optimal Condition for Simultaneous Saccharification and Fermentation Using Pretreated Corncob by Oxalic Acid (옥살산 전처리 옥수숫대를 이용한 동시당화발효 최적 조건 탐색)

  • In this study, we determined optimal conditions for simultaneous saccharification and fermentation (SSF) using corncob biomass pretreated with oxalic acid. The effect of SSF temperature ($25.8{\sim}34.2^{\circ}C$) and agitation speed (80~220 rpm) were significant at a 99% confidence level in its effect on ethanol production. The highest ethanol production was expected when SSF was performed at $30^{\circ}C$, 170 rpm (22.5 g/L). The ethanol production was improved by mixture of yeast extract (1.25 g/L) and urea (1.25 g/L) as nitrogen source. However, addition of trace metal components and vitamin for SSF was not affected in the ethanol production. Optimal concentration of $KH_2PO_4$, $MgSO_4{\cdot}7H_2O$ for SSF was 1 g/L, 0.25 g/L respectively.

Influence of Mucor mucedo immobilized to corncob in remediation of pyrene contaminated agricultural soil

  • In recent years, immobilization agents were introduced into organic contaminated soil remediation and more and more materials were screened and used as the immobilizing carrier. However, effect of the decomposition of the immobilizing carrier on the bioremediation was rarely concerned. Therefore, the decomposition experiment of immobilizing carrier -corncob was carried out in the lab with the efficient degradation fungi - Mucor mucedo (MU) existing, and polycyclic aromatic hydrocarbons (PAHs) residues E4/E6 of the dissolved organic matter and microbial diversity during the decomposition process were studied. The results showed that: a) during the decomposition, the degradation of pyrene (Pyr) was mainly in the first 28 d in which the content of extractable Pyr decreased rapidly and the highest decrease was in the treatment with only MU added. b) Anslysis of E4/E6 changes showed that rich microorganisms could promote aromatization and condensation of humus. c) From the diversity index analysis it can also be seen that there is no significant difference in effects of PAHs on the uniformity of microorganisms. These results will not only be useful to have a better understanding of the bioavailability of contaminants adsorbed to biodegradable carriers in PAHs contaminated soil remediation, but also be helpful to perfect the principle of immobilized microbial technique.

Production of Xylose from Xylan by Endoxylanase and ${\beta}-Xylosidase$ Expressed in Yeast

  • The endoxylanase (642 bp; 213 amino acids) and ${\beta}-xylosidase$ (1,602 bp; 533 amino acids) genes from Bacillus sp. were amplified by PCR and separately inserted downstream of the yeast ADH1 promoters, resulting in the pAEDX-1 and pAEX plasmid. When the yeast transformants, S. cerevisiae SEY2102 harboring pAEDX-1 or pAEX, were grown on YPD medium, the total activities of the enzymes reached about 9.8 unit/mL for endoxylanase and 2.9 unit/mL for ${\beta}-xylosidase$. When the three kinds of xylan from oat spelts, birch wood, and corncob were hydrolyzed by treatment of recombinant endoxylanase and ${\beta}-xylosidase$, it was found that xylose, xylobiose and xylotriose were produced and xylose was the major product after 12 h reaction. In addition, with the higher amount of enzymes, the more amount of xylose was produced.

Selection of an Substitute Sawdust Material in Pleurotus ostreatus by Bottle Cultivation (느타리버섯 병재배의 톱밥 대체 배지 재료 선발)

  • In this study, we attempted to find substitute materials, swelling rice hull, cocopeat, corncob and coconut sawdust, for sawdust in bottle cultivation of Pleurotus ostreatus. Chemical characters of mixture substrates with four substitute materials for pine sawdust were not different significantly. By comparison in mycelial growth and yield of fruitbody, mixture substrate of cocopeat were showed the same level in P. ostreatus. Therefore, it is suggested that cocopeat was substituted for sawdust for cultivation of P. ostreatus.

Studies on Artificial Cultivation of Pleurotus eryngii (De Canolle ex Fries) Quel. (큰느타리(Pleurotus eryngii)의 인공재배에 관한 연구)

  • This study was carried out to investigate the artificial cultivation of Pleurotus eryngii on the optimal medium vessel, periods of cultivation and the optimal method of pinheading for both yield and quality of fruiting body were also performed. The optimal composition of sawdust medium in polypropylene(PP) bottle was combination of sawdust(70%) and corncob(30%) but increased amount of corncob delayed the period of mycelial growth. The mycelial growth and the yield of fruiting body in the medium with beat pulp were worse than that without beat pulp. The optimal composition of nutrients for both yield and quality of fruit body tuned out to be a combination of rice bran(12%), wheat bran(12%) and cottonseed cake(6%). Additions of zeolite, shell lime and bean curd dregs were not effective in mycelial growth and yield of fruit body. When testing size of PP bottle for cultivation, the larger of bottle mouth is, the more pinheading number found, but the number of available fruit body is not significantly different. The culture in $1100\;ml-{\phi}75\;mm$ bottle is the best in the yield and quality of fruit body than those in $555\;ml-{\phi}50\;mm,\;850\;ml-{\phi}58\;mm,\;850\;ml-{\phi}65\;mm\;and\;1100\;ml-{\phi}65\;mm$ bottle. Using the PP bag for cultivation, a square shaped bag was better than a round shaped and black square shaped in mycelial growth and yield of fruit body. The most suitable period of incubation was 35 days after inoculation at $22{\pm}2^{\circ}C$. When the incubation periods was decreased less than 35 days, the pileus formation and yields were very bad but a pinheading condition looked similar, For an optimal pinheading, turning upside down was better than standing and covering.

  • 432 samples of raw goat milk were analyzed for sodium, calcium, potassium, chloride, and ashes content. The samples came from 16 pure breed goats, 2 to 4 years of age kept in semi-feedlots. They were basically fed soy bran, cotton bran, triturated Guandu hay and disintegrated corn with its straw and corncob (according to requirements for due preservation and production). The mean values found were: sodium, $45{\pm}10.2mg%$ (24.5-65.6 mg%); potassium, $206{\pm}44.7mg%$ (116.6-295 mg%); calcium, $111{\pm}22mg%$ (66.5-155.4 mg %); ashes, $0.82{\pm}0.06mg%$ (0.68-0.95%); chloride, $235{\pm}39mg%$ (157-313 mg%). Results were related to: a) breed, the Brown and the Saanen revealed superior values for calcium and ashes, b) climate, which in the rain season led to higher contents of sodium, potassium, and calcium when compared to the local dry season, c) time of lactation which increasingly affected the contents of calcium, sodium and ashes, and d) the time between milking, when higher concentration of chloride and ashes were mostly concentrated in the evening and calcium in the morning. These results have important implications in the handling and management of the goat herd.

Optimum mixing rate of used media for saving the production cost of Flammulina velutipes (팽이버섯(Flammulina velutipes) 생산비 절감을 위한 폐배지 적정 혼합비율)

  • These experiments were conducted to find the optimum mixing rate of used media for saving the production cost of Flammulina velutipes. The materials for media formation was used needle-leaf tree sawdust, media that finished 1th cultivation of F. velutipes, corncob meal, and rice bran, wheat bran as nutrition source. We inoculated the F. velutipes in 14 kinds of different media types and checked the spawn growth speed, fruit body quality and quantity. Two nutrition agents, which is rice bran and wheat bran, did not affected the incubation period, but the effective stem number, quality and quantity of fruit body was better at rice bran than wheat bran. The quality of fruit body produced at mixed 20% of used media (needle-leaf tree sawdust 60% + used media 20% + rice bran 20%) was similar to control plot (needle-leaf tree sawdust 80% + rice bran 20%), but the yield was improved 10% than control plot 130g. According as the used-media mixing amount increases, quality and quantity of fruit body became low remarkably. Therefore, the optimum mixing amount of used-media was 20% and it increased 10% of fruitbody yield.
