Ecological studies have indicated that the essential fatty acids in maternal and umbilical cord blood samples are associated with gestational length and birth weight. The objectives of this study were to examine serum fatty acid concentration, particularly $\omega$3 fatty acids, in maternal and umbilical cord blood and to investigate the relationship of serum fatty acid levels in the blood of the mother and of the umbilical cord. Subjects consisted of 30 full-term and 30 pre-term mothers and neonates of both groups. Serum levels of fatty acids were measured by gas chromatography. The concentration of total saturated fatty acids in pre-term pregnant women was significantly higher than that of the full-term group (p<0.05), however, the maternal level of $\omega$3 fatty acids in the pre-term group was significantly lower than that of the full-term pregnant women (p<0.05), Moreover, the concentrations of $\alpha$-linolenic acid and eicosapentaenoic acid in full-term pregnant women were significantly higher than those of the pre-term group. In umbilical cord blood, the levels of total $\omega$3 fatty acid and arachidonic acid were significantly lower in the pre-term group than in the full-term group (p<0.05). Based on the coefficient of correlation between serum fatty acids in the mother and the umbilical cord, it turned out that in the full-term group, the newborn's umbilical cord serum fatty acids were not influenced by the levels of serum fatty acids in the mother. However, in the pre-term group, it seems to have positive correlations in terms of the levels of SFA, MUFA, PUFA and $\alpha$-linolenic acid. This study suggests that a lower status of $\omega$3 fatty acids in maternal and umbilical cord blood probably is a risk factor for pre-term birth.
Apgar score is most widely used evaluating indicator of newborn health, but it is very Subjective. Umbilical cord blood gas analysis is more objective and effective than Apgar score in evaluation of newborn status. Cord blood gas was changed slightly by processing of time after fetal birth. This study was undertaken to observe objectiveness and effectiveness of umbilical arterial blood gas analysis and effects of time interval of cord clamping to newborn health with 122 pregnant women and their babies. We observed following results : 1. There were poor correlation between Apgar score and umbilical cord arterial blood analysis in evaluating of newborn health (P>0.05). 2. There was no clinical significance of Apgar score or umbilical arterial blood gas analysis as single indicator in evaluating of newborn health (P>0.05). 3. Gas analysis and pH of umbilical arterial blood was more helpful in evaluating of newborn health than Apgasr score. 4. There were no significant effects of time interval of umbilical cord clamping to newborn health. 5. If there were no indications of early umbilical cord clamping, cord blood sampling at immediately after birth without cord clamping was more effective to evaluate newborn status.
Cord blood (CB) has been used as an important and ethical source for hematopoietic stem cell transplantation (SCT) as well as cell therapy by manufacturing mesenchymal stem cell, induced pleuripotential stem cell or just isolating mononuclear cell from CB. Recently, the application of cell-based therapy using CB has expanded its clinical utility, particularly, by using autologous CB in children with refractory diseases. For these purposes, CB has been stored worldwide since mid-1990. In this review, I would like to briefly present the historical development of clinical uses of CB in the fields of SCT and cell therapy, particularly to review the experiences in Korea. Furthermore, I would touch the recent banking status of CB.
Objective : Matrix metalloproteinases (MMPs), especially MMP-2 and MMP-9 have been known to play an important role in secondary inflammatory reaction after spinal cord injury (SCI). The aim of this study was to investigate the expression and activity of MMP-2 and MMP-9 and to determine their relationship with disruption of endothelial blood-barrier after photochemically induced SCI in rats. Methods : Female Sprague-Dawley rats, weighing between 250 and 300 g (aged 8 weeks) received focal spinal cord ischemia by photothrombosis using Rose Bengal. Expressions and activities of MMP-2 and MMP-9 were assessed by Western blot and gelatin zymography at various times from 6 h to 7 days. Endothelial blood-barrier integrity was assessed indirectly using spinal cord water content. Results : Zymography and Western blot analysis demonstrated rapid up-regulation of MMP-9 protein levels in spinal cord after ischemic onset. Expressions and activities of MMP-9 showed a significant increased at 6 h after the photothrombotic ischemic event, and reached a maximum level at 24 h after the insult. By contrast, activated MMP-2 was not detected at any time point in either the experimental or the control groups. When compared with the control group, a significant increase in spinal cord water content was detected in rats at 24 h after photothrombotic SCI. Conclusion : Early up-regulation of MMP-9 might be correlated with increased water content in the spinal cord at 24 h after SCI in rats. Results of this study suggest that MMP-9 is the key factor involved in disruption of the endothelial blood-barrier of the spinal cord and subsequent secondary damage after photothrombotic SCI in rats.

Objectives: This study aims to analyze manganese (Mn) concentrations in maternal and cord bloods at delivery and to estimate the Mn exposure risk for fetuses whose mothers were occupationally exposed to Mn. Materials and Methods: Forty-six pairs of maternal and cord blood samples were collected at delivery from mothers who were occupationally unexposed to Mn. Mn concentrations of blood were analyzed by graphite furnace atomic absorption spectrometer. Mn exposure levels for fetuses of female workers were estimated by simulating two working exposure scenarios. Results: The geometric mean concentration of Mn in maternal and cord blood were $27.0(1.34){\mu}g/L$, $46.6(1.25){\mu}g/L$, respectively. Transfer ratios of Mn from maternal to cord blood were $1.81{\pm}0.62$, which indicated that the Mn concentrations in cord blood were higher than those in maternal blood. Mn concentrations in cord blood for the worse or general scenarios were estimated to $22.3-1,881{\mu}g/L$ and $1.59-308{\mu}g/L$, respectively. The probabilities of exceeding $74{\mu}g/L$, which was adopted as a reference level reported in a previous study, were 95% and 44% for the two scenarios, respectively. Conclusions: Comparable levels of Mn exposure in maternal or cord blood to those in this study have shown various health effects in previous studies. This suggests that Mn exposure levels in mothers and fetuese in Korea need to be monitored and managed. In addition, female workers who are occupationally exposed to Mn should be protected from the exposure since their fetuses can be exposed to Mn at risky levels during their pregnancy.

Objective: Highly sensitive haptoglobin measurement should be used in neonates because the haptoglobin concentration in neonates is lower than that of adults. The aim of this study was to establish the reference values of haptoglobin levels in the cord blood of uninfected neonates. Methods: The cord blood of 29 preterm and 51 term babies was collected, and data from the mother and the newborn were recorded. The haptoglobin concentrations of 80 cord blood samples were simultaneously measured by enzyme-linked immunosorbent assay (ELISA; Assaypro, St Charles, MO, USA) and immunoturbidimetry assay (Roche Diagnostics, Basel, Switzerland). C-reactive protein (CRP) was also measured by immunoturbidimetry assay (Roche Diagnostics, Switzerland). Results: Mean values of CRP and ELISA haptoglobin were not significantly different between preterm and term babies. The 2.5 percentile and 97.5 percentile values of ELISA haptoglobin concentration were as follows: 80 neonates, 0.01 mg/dL and 0.59 mg/dL; 29 preterm babies, 0.08 mg/dL and 0.18 mg/dL; and 51 term babies, 0.07 mg/dL and 0.23 mg/dL. There were no differences in ELISA haptoglobin concentration according to maternal underlying diseases, delivery method, usage of antibiotics or steroids before delivery, gestational age, gender of baby, or twin gestation. Conclusion: A highly sensitive haptoglobin method should be used to determine the haptoglobin concentration in Korean newborns because the reference values of cord blood haptoglobin concentration in Korean newborns are less than the lower detection limit for commonly used immunoturbidimetric haptoglobin measurement methods.
To investigate the blood lead concentration, their interrelation, correlation factor and influence on pregnant women and newborn, lead concentration in the maternal blood and umbilical cord blood were determined. Samples were collected from 130 mothers who were living in the Taegu City, during March, 1989. Blood lead concentration was estimated using the Atomic Absorption Spectrophotometer(IL. 551) equipped with Flameless Furnace Atomizer (IL. 665). The mean lead concentration of maternal and cord blood were $17.47{\pm}7.92{\mu}g/d{\ell}$, $15.31{\pm}7.98{\mu}g/d{\ell}$, respectively. A significant correlation was observed between the lead concentration of maternal and cord blood, r=0.663, Y=0.667X+3.646. No significant correlation was observed between previous spontaneous abortion and obstetric complication of mother and maternal blood lead concentration. Similarly, no significant correlation was observed between the sex, gestational age of neonate and cord blood lead concentration. But the birth weight of neonate had some negative correlation with cord blood lead concentration. The blood lead concentration of mother who had engaged in manufactures were higher than others and the longer working years were, the higher blood lead concentration were. Significant correlation was observed between husband's occupational exposure to lead and maternal blood lead concentration.
This study was carried out to find out PSS(Porcine Stress Syndrome) with the PSE(Pale, Soft, Exudative) in different piglets. These experiments were accomplished with the aid of PCR-RFLP(Polymerase Chain Reaction-Restriction Fragment Length Polymorphism). The samples were collected and examined from umbilical cord blood of piglets of Yorkshire, Landrace and Crossbred. And then, the PCR products were digested by restriction enzyme, Hha I. The results obtained were as follows; The PCR products of the blood genomic DNA of ryanodine receptor gene were length of 1 .8kb in umbilical cord blood. Normal type(NN), heterozygous type(Nn) and recessively homozygous type(nn, PSS) as a result of digestion of restriction enzyme, Hha 1, were 90.0%, 10.0% and 0.0% in Yorkshire piglets, 76.2%, 19.0% and 4.8% in Landrace, 69.1%, 23.8% and 7.1% in crossbred, respectively. As already showing the above results, the blood from piglets umbilical cord can be availably used for the determination of genotypes of PSS because of easiness of blood collection without stress in live piglets.
Folate and Vitamin $B_{12}$ are essential nutrients important during pregnancy. This study was conducted to evaluate the folate and vitamin $B_{12}$ nutritional status of Korean pregnant women and to investigate the relationship between maternalumbilical cord serum folate and vitamin B12 levels and pregnancy outcomes. Dietary intakes of the pregnant women were estimated by 24 hour-recall (3 times). Serum folate and vitamin B12 levels in maternal blood and umbilical cord of 27 pregnant women at 1'st-, 2'nd-, 3'rd-trimester and delivery were measured by RIA (radioimmuno assay), respectively. Means of folate and vitamin $B_{12}$ intake were $283.53\pm58.01{\mu}g/day\;and\; 2.99\pm1.32mg/day$, respectively. Maternal mean serum folate levels of the trimester and delivery were $9.75\pm3.60ng/ml,\;10.46\pm4.63ng/ml,\;10.71\pm4.14ng/ml\;and\;15.05\pm7.04ng/ml$. Those maternal levels were significantly lower than that of umbilical cord blood $(23.99\pm9.42ng/ml)$. Serum vitamin $B_{12}$ levels of maternal trimester and delivery were $479.07\pm137.56 pg/ml,\;310.96\pm137.56pg/ml,\;308.22\pm74.65pg/ml,\;and\;295.67\pm93.36pg/ml$, which were significantly lower than those of umbilical cord blood $(500.13\pm185.60ng/ml)$. This finding indicates that the uptake of folate and vitamin $B_{12}$ in the fetus may be due to an active placental transport mechanism. Maternal serum level correlated positively with those of umbilical cord blood, showing that folate and vitamin $B_{12}$ concentration of umbilical cord blood might be affected by maternal status. There was no significant correlation between the serum folate levels in maternal-umbilical cord blood and the pregnancy outcomes. However, maternal vitamin $B_{12}$ level at l'st trimester was significant positive correlation between the gestational age except for birth weight and weight gain.
