Dyeing of Tencel with Coptis chinensis Franch (황련에 의한 Tencel직물의 염색)

  • The dyeing of Tencel fabric with Coptis chinensis Franch was studied. The colorant was extracted with distilled water under various concentrations of dyestuff, times, mordants. UV-VIS spectra were obtained to find the maximum wavelength and absorbance of colorant. Tencel fabric was dyed with different mordants in pre and post-mordanting methods. Maximum wavelength of spectrum was 343 nm, and K/S value of pre-mordant was higher than post-mordant. Aluminium and stannic mordants changed color from yellow to blue-greenish and red-bluish. Colorfastnesses of light, and washing were low grade, but that of perspiration was increased.

Screening of Plant Extracts with Cholinesterase Inhibition Activity (콜린 에스테라제 저해효과 보유 식물 추출물 탐색)

  • This study was conducted to select candidates from plant resources for the purpose of improving or treating Alzheimer's disease, a type of dementia. One hundred and eighty-four plant extracts at a final concentration of $100{\mu}g/ml$ were screened to determine their capacity to inhibit acetylcholinesterase (AChE) by in vitro assay. From this AChE assay, seven plant extracts - including methanol ext. and water ext. of Phellaodendron amurense Rupr. (bark), methanol ext. of Nelumbo nucifera Gaertn (stamen/ovary), methanol ext. of Persicaria tinctoria H. GROSS (flower), methanol ext. of Coptis chinensis (rhizome), ethanol ext. of Cinnamomum cassia Blume(bark) and ethanol ext. of Carthamus tinctorius L. (fruit) - showed effective inhibition activity ranging from 18.7% to 63.1%. The selected extracts were testified their inhibition activities on AChE and BuChE (butyrylcholinesterase) at concentrations of 25, 50, 100, $200{\mu}g/ml$. In the AChE assay, five extracts including methanol ext. of Nelumbo nucifera Gaertn. (stamen/ovary), methanol ext. of Persicaria tinctoria H. GROSS (flower), methanol ext. of Coptis chinensis (rhizome), methanol ext. and water ext. of Phellaodendron amurense Rupr. (bark) showed inhibition activity of 15.0%~73.5%, 19.5%~63.5%, 81.6%~58.5%, 69.9%~80.5%, and 54.8%~78.3%, respectively, at concentrations of 25, 50, 100, $200{\mu}g/ml$. In the BuChE assay, the extracts of Nelumbo nucifera Gaertn. (stamen/ovary), Persicaria tinctoria H. GROSS (flower), and Coptis chinensis (rhizome) showed inhibitory capacities of 58.9~81.6%, 45.8%~72.4%, and 33.1%~55.4% at concentrations of 25, 50, 100, $200{\mu}g/ml$, respectively. In conclusion, it is suggested that Nelumbo nucifera Gaertn. (stamen/ovary), Persicaria tinctoria H. GROSS, Coptis chinensis (rhizome) and Phellaodendron amurense Rupr. (bark) could be selected as candidate materials for improving or treating Alzheimer's disease on the basis of further study.

Changes in the Keeping Quality of Strawberry and Cucumber treated with Korean Medical Herb Extracts (대황 및 황련 추출물을 처리한 딸기 및 오이의 저장중 품질 변화)

  • Strawberry and cucumber were dipped with Korean medical herb extracts such as Rheum palmatum L. and Coptis chinensis Franch. to maintain the keeping quality during storage at 5$^{\circ}$C and 10$^{\circ}$C, respectively. Treatment with 500 ppm solutions gor strawberry retarded weight loss, microbial load increase, and decay during storage. Decay ratio was shown 68.8% for control, 45.0% for Rheum palmatum L. and 42.5% for Coptis chinensis Franch. at 10 days. Strawberry treated with Coptis chinensis Franch. showed a higher hardness and soluble solid content than others. Cucumber treated with 1,000 ppm solutions revealed a different O$_2$ and CO$_2$ concentrations after 16 days. Korean medical herb extracts showed an antimicrobial activity for the quality retention of cucumber during storage. Ascorbic acid content was generally increased till 8 days, but decreased thereafter.

Molecular Modeling and Docking Studies of 3'-Hydroxy-N-methylcoclaurine 4'-O-Methyltransferase from Coptis chinensis

  • Coptis chinensis 3'-hydroxy-N-methylcoclaurine 4'-O-methyltransferase (HOMT), an essential enzyme in the berberine biosynthetic pathway, catalyzes the methylation of 3'-hydroxy-N-methylcoclaurine (HMC) producing reticuline. A 3D model of HOMT was constructed by homology modeling and further subjected to docking with its ligands and molecular dynamics simulations. The 3D structure of HOMT revealed unique structural features which permitted the methylation of HMC. The methylation of HMC was proposed to proceed by deprotonation of the 4'-hydroxyl group via His257 and Asp258 of HOMT, followed by a nucleophilic attack on the SAM-methyl group resulting in reticuline. HOMT showed high substrate specificity for methylation of HMC. The study evidenced that Gly117, Thr312 and Asp258 in HOMT might be the key residues for orienting substrate for specific catalysis.

Fractionated Coptis chinensis Extract and Its Bioactive Component Suppress Propionibacterium acnes-Stimulated Inflammation in Human Keratinocytes

  • Coptis chinensis (CC) is widely used in Asian countries to treat inflammatory diseases. We investigated the anti-inflammatory activity of the aqueous fraction separated from CC extract and of berberine, its key bioactive component, in human keratinocytes and the possible molecular mechanisms underlying this. Treating HaCaT keratinocytic cells with heat-killed Propionibacterium acnes induced nitric oxide and proinflammatory cytokine (e.g., tumor necrosis $factor-{\alpha}$, interleukin $(IL)-1{\beta}$, and IL-8) production and their mRNA expression; these effects were suppressed by pretreatment with the aqueous fraction or berberine, which also suppressed the phosphorylation of ERK, JNK, and p38 kinases and the nuclear expression of nuclear factor $(NF)-{\kappa}B$ p65 in P. acnes-stimulated cells. Thus, the aqueous fraction and berberine effectively exerted anti-inflammatory activities by suppressing mitogen-activated protein kinase and $NF-{\kappa}B$ signaling pathways in human keratinocytes and may be used for treating P. acnes-induced inflammatory skin diseases.

Packagng of Fresh Curled Lettuce and Cucumber by Using Low Density Polyethylene Films Impregnated with Antimicrobial Agents (항균소재를 함유시킨 저밀도폴리에틸렌 필름에 의한 상추와 오이의 포장)

  • Low density polyethlene(LDPE0 films of 50${\mu}{\textrm}{m}$ thickness were fabricated with addition of antimicrobial agents of Rheum palmatum extract, Coptis chinensis extract, sorbic acid and Ag-substitude inorganic zirconium matrix in 1% concentration. The films were compared in physical properties, tested in antimicrobial activity against some selected microorganisms on the agar plate medium and then applied for packaging fresh curled lettuce and cucumber to preserve their qualities. The films with Rheum palmatum extract, Coptis chinensis extract, and Ag-substituted inorganic zirconium matrix did not show any antimicrobial activity on the disk test against Escherichia coli, Staphylococcus aureus, Leuconostoc mesenteroides, Saccharomyces cerevisiae, Aspergillus niger, Aspergillus oryzae, Penicilluium chrysogenum, while film with sorbic acid did against E. coli, S. aureus and L. mesenteroides. The added antimicrobial agents changed the color and light transmittance of the films, but did not affect their mechanical tensile strength, heat shrinkage and wettability. For the packaged curled lettuce and cucumber stored at 5$^{\circ}C$ and 1$0^{\circ}C$, all the LDPE films impregnated with antimicrobial agents showed the reduced growth of total aerobic bacteria in the vegetables compared with control film without any additive until it reached the level around 108/g. They did not give any negative effect on other quality attributes during storage.

Insecticidal Activity of Coptis chinensis Extract Against Myzus persicae (Sulzer)

  • In view of the environmental and health hazards posed by synthetic insecticides, the use of plant products as botanical insecticides has gained increasing in recent years. In this study, we reported the insecticidal activity of extracts isolated from Coptis chinensis. On crude extraction, among the various solvent types tested (water, 1% (w/v) of sodium hydroxide, 70% ethanol), the 70% ethanol extract showed the best insecticidal activity (36.5%). Three different fractions (n-hexane, chloroform and ethyl acetate) were obtained from crude extract (70% ethanol) of the chloroform fraction and found to have noteworthy insecticidal activity (62.9%) by filter paper contact bioassay. Their chemical structures were identified as 2-methoxy-4-vinylphenol and aniline by head space-GC-MS analysis. Both compounds displayed a dose-dependent insecticidal activity of Myzus persicae (Sulzer). Insecticidal activity at the lowest concentration tested (500 ppm) approached 85.4% in the aniline compared with 79.9% in the 2-methoxy-4-vinylphenol. The insecticidal activity was greater for the aniline than 2-methoxy-4-vinylphenol. It is believed that the insecticidal activity is due mainly to the presence of aniline.

A Study for the isolation of the Berberine-type Alkaloid from Coptidis Rhizoma and for their Antitumor Activities

  • The purpose of this study is the separation of biologically active ingredients from Coptidis Rhizoma which has been widely used as one of oriental herbal medicine for body fever. In this study, berberine-type alkaloids were tested on their biological activities in the aspect of antibacterial, antitumor, anti-herpetic and anti-HIV activity. Contents of five major alkaloids for the various origin of Coptidis Rhizoma were assayed by HPLC. As the results, the content of berberine from Coptis chinensis and Coptis japonica were 6.78% and 7.09%, respectively. The contents of coptisine, jatrorrhizine, berberastine from Coptis chinensis were higher than those of Coptis japonica. The amount of palmatine from both species were almost the same. Surprisingly for antitumor experiment, all compounds have been shown remarkable activity, especially against SNU-l(human stomach cancer) cell line. Among the compounds purified through column chromatography, palmatine, coptisine, and jatrorrhizine inhibited the growth of K-562(human chronic myelogenous leukemia) cell line whereas jatrorrhizine has been shown the effective inhibition of A-549 (human lung) cell line at the same time.

Optimization of the Extraction Process and Antioxidant Capacity of Coptis chinensis Franch Extract through Cluster Analysis (클러스터 분석을 이용한 황련 추출물 항산화 활성 최적화 추출공정)

  • Coptis chinensis Franch is a valuable traditional oriental medicinal plant used for the treatment of various diseases. The major factors affecting the content of bioactive compounds and the relationship between bioactive compounds and antioxidant capacities of Coptis chinensis Franch were poorly understood. Thus, effects of the solvent, temperature, and extraction time on the extraction yields of bioactive compounds and the antioxidant activity of C. chinensis Franch extracts were investigated in this work. Our cluster analysis indicated that the hydroalcoholic solvent (50% ethanol : 50% water) at $35^{\circ}C$ for 30 min (extract time) was the best extraction condition for a factory use because the highest level of bioactive compounds and antioxidant activities was achieved. Multiple linear regression analysis revealed that total phenolic content (TPC) contributed to the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity, while both total alkaloid content (TAC) and total flavonoid content (TFC) were responsible for ferric reducing antioxidant power (FRAP) activity. These results indicated that extraction conditions controlled the yield of bioactive compounds and the antioxidant activity of C. chinensis Franch, which can provide important information for the industrial extraction.

The Effect of Three kinds of Coptis Rhizoma on the Contraction of Isolated Rat Tracheal Smooth Muscle (삼종(三種) 황연류(黃連類)의 기관지(氣管支) 평활근(平滑筋) 이완효과(弛緩效果)에 관(關)한 연구(硏究))

  • This study was carried out to investigate the tracheal muscle relaxation of water extracts of three kinds of Coptis rhizoma and also compare their effects with berberine and palmatine, which are the important alkaloid components of Coptis rhizoma, in isolated rat tracheal smooth muscle. As a result Coptis chinenis showed significant tracheal muscle relaxation on KCI-induced contraction and more markedly on carbachol-induced contraction in dose-dependent manner. Coptis japoinin exhibited also tracheal muscle relaxation on both contraction with slightly weaker activities than those of Coptis chinensts. The activities of the root hair of Coptis chinensis were the most weakest in three rhizomas. Berberine and palmatine had strong tracheal muscle relaxation, especially, on carbachol-induced contraction, however, their activities on KCI-induced contraction were weaker than that of Coptis chinenis. The inhibitory effects of three kinds of Coptis rhizoma on the contraction by KCI and carbachol were investigated to explain the process of their tracheal muscle relaxation. All rhizomas inhibited the contraction by both agents, among them, Coptis japonica showed the most significant effect, so indicating that the tracheal muscle relaxation of Coptis rhizoma may be concerned with the blocking of muscarine receptor and $Ca^{2+}$channel.
