Modulation of Cyclic AMP-PKA System on Inhibitory and Excitatory Herbs-induced Ion Currents (Cyclic AMP-PKA System이 억제성 및 흥분성 한약재에 의해 유발된 이온전류에 미치는 영향)

  • Modulatory effect of cAMP-PKA system on inhibitory- and excitatory herbs-induced ion currents were investigated by nystatin-perforated patch clamp method under voltage-clamp condition. Ion currents induced by Bupleuri radix and Coptidis rhizoma were not affected by cAMP-PKA system. Ion current induced by Ecliptae herba was partially inhibited by cAMP-PKA system. Ion currents induced by Aconiti tuber and Boshniakiae herba were inhibitory modulated and ion current induced by Zingiberis rhizoma was excitatory modulated by cAMP-PKA system. Modulation of cAMP-PKA system on ion currents induced by Ginseng radix was fluctuated. In this results, it can be seen generally that excitatory herbs-induced ion currents were modulated by cAMP-PKA system while cAMP-PKA system did not affect inhibitory herbs-induced ion currents.

  • Cancer is one of the most important cause of death. So recently, investigation of cancer progress prosperously all over the world. Cancer in the present medicine correspond to You-Am, Sin-Am, Young-Soon, Sel-Gyun, Sil-Young, Young-Lyoo, Seg-Je, Seg-Young, Seg-Ha, Jerk-Chui(積聚), Jing-Ha, Oel-Gyek, Ban-Oui, Bi-Gi, Bok-Lyang, Jang-Dan, Hyen-Bek in the oriental medicine. Among these, generally Jerk-Chui(積聚) is expressed to cancer. So to develop of new drugs of cancer in the present medicine, bibliographic investigation of mass-prescriptions was studied in the oriental medicine-books. According to the bibliographic study of Jerk-Chui-prescriptions, the results run as follows. 1. According to the analyses of three hundred sixty eight Jerk-Chui-prescriptions in the twenty-seven kinds of literature, the frequency number of the used drugs were Pericarpium Citri Nobilis Viride 140 times, Pericarpium Citri Reticulatae 135 times, Rhizoma Scirpi 124 times, Radix Aucklandie 115 times, Rhizoma Zedoariae 114 times, Cortex Magnoliae Officinalis 111 times, Radix Glycyrrhizae 106 times, Rhizoma Zingiberis 100 times, Rhizoma Coptidis 94 times, Radix Ginseng 93 times, Poria 86 times, Rhizoma Pinelliae 85 times, Semen Arecae 83 times, Rhizoma Cyperi 82 times, Radix Angelicae Sinensis 80 times, Rhizoma Atractylodis 74 times, Massa Fermentata Medisinalis 67 times, Radix Et Rhizoma Rhei 66 times, Fructus Aurantii 62 times, Fructus Hordei Genninatus 55 times, Conex Cinnamomi 54 times, Fructus Evodiae 51 times, Fructus Aurantii Immaturus 49 times, Fructus Crataegi 49 times, Rhizoma Cnidii 46 times, Radix Platycodi 44 times, Semen Tiglii 44 times, Radix Aconiti 43 times, Fructus Amoni 38 times, Semen Raphani 37 times, Radix Aconiti Praeparata 36 times, Radix Scutellariae 35 times, Pericarpium Zanthoxyli 35 times, Rhizoma Corydalis 33 times, Rhizoma Acori Graminei 31 times, Carapax Amydae 31 times, Fructus Foeniculi 31 times, Semen Persicae 30 times, Radix Bupleuri 30 times. 2. The clinical-botanic classifications of the used drugs in the Jerk-Chui-prescriptions were regulating the flow of Qi drugs, warm-heating drugs, promoting blood circulation drugs, killing mass drugs, resolving drugs, purgative drugs, Qi and blood tonics drugs, heat clearing drugs, removing dampness by promoting diures is drugs, phlegm eliminating drugs, allaying pain drugs. 4. According to the nature and taste in the drugs, warm and heating recipes were used most, heatclearing recipes were used a few times assistantly. 5. The Jerk-Chui-prescription used frequently was Bun-Don-Tang, which was used 13 times ; Bok-Oyang-Hoan 12 times, Bi-Gi-Hoan(肥氣丸) 12 times, Sik-Boon-Hoan 12 times, A-Uie-Hoan 12 times, Bi-Gi-Hoan 12 times, Dai-Cil-Gi-Tang 8 times, San-Cuie-Tang 8 times, Guye-Gyen-Tang 6 times, On-Baig-Won 5 times, So-Jek-Jeng-Ouen-San 5 times, Jin-In-Hoa-Cel-Tang 5 times, Byel-Gab-San 5 times, Sng-Hong-Hoan 5 times, Ji-Sil-San 4 times, So-A-Oie-Hoan 4 times, Hyang-Rng-Hoan 4 times.

A Study on a paradigm of Radix Aconiti(附子) in the treatment of heart-systemic disease(心系疾患) due to heat syndrome(熱證) (심계영역질환(心系領域疾患) 화열증(火熱證)에 사용된 부자(附子)의 활용(活用)에 대한 문헌적(文獻的) 연구(硏究))

  • We inquired into the prescription including Radix Aconiti(附子) on the records of the past. According to the role of Radix Aconiti(附子), we can divide into four group. (1) In the case of excess in the upper and deficiency in the lower(上盛下虛), heat in the upper and cold in the lower(上熱下寒), Radix Aconiti(附子) supplies the fire from the gate of life(命門火) and the fire of deficiency type(虛火) is recurred into the origine(根源) by it. (2) In the case that Yang(陽) is kept externally by Yin(陰)-excess in the interior(陰盛格陽), and there is heat syndrome in the exterior with cold syndrome in the interior(裏寒外熱), Radix Aconiti(附子) can remove cold(祛寒) and revitalizes(回陽). (3) In the case of the heat of excess type syndrome(實熱證), Radix Aconiti(附子) treats heat-syndrome(熱證) with heat-clearing(淸熱) drug. (4) In the case of wind-syndrome(風證) and phlegm-syndrome(痰證), Radix Aconiti(附子) supplies the kidney-Yang(元陽) and promotes movement of the other drugs. According to frequency in use of drugs, we can divide into four group. (1) In e case of the fire of deficiency type(虛火), Radix Aconiti(附子) is used with drugs of invigorating-Yin(補陰), invigorating-Yang(補陽) and diuresis(利水), such as Rhizoma rehmanniae(熟地黃), Cortex cinnamomi(肉桂), Fructus comi(山茱萸), Poria, etc. (2) In the case that Yang is kept externally by Yin(陰)-excess in the interior(陰盛格陽), Radix Aconiti(附子) is used with drugs of dispelling cold(祛寒) and revitalization(回陽), such as Rhizoma zingiberis siccatum(乾薑), Radix glycyrrhizae(甘草), Radix ginseng(人蔘), etc. (3) In the case of the heat of excess type syndrome(實熱證), Radix Aconiti(附子) is used with drugs of heat-clearing(淸熱), such as Rhizoma coptidis(黃蓮). (4) In the case of wind-syndrome(風證) and phlegm-syndrome(痰證), Radix Aconiti(附子) is used with drugs of dispersing(發散), invigorating(補氣), enriching the blood(補血), removing the phlegm(去痰), heat-clearing(淸熱). such as Radix oxterici koreani(羌活), Radix ledebouriellae(防風), Radix ginseng(人蔘), Radix angelicae gigantis(當歸), etc. Especially in many of cases, Radix Aconiti(附子) is used with drugs of nourishing-Yin(滋陰), but it is hardly used with drugs of treating middle-jiao energy(中氣).

Inhibitory Effects of Oriental Medicinal Herbs on Enzymatic Activity of Sialyltransferases (ST3Gal I and ST6Gal I) Associated with Cancer

  • In many cases of human cancer, the appearance of hypersialylated glycan structures is related to a precise stage of the disease ; this may depend on the elebated sialyltransferase activity during carcinogenesis. The aim of this study was to investigate the inhibitory effects of Oriental medicinal herbs on enzymatic activities of two kinds ofsialyltransferase, Gal $\beta$ 1,3GalNAc$\alpha$2,3-sialyltransferase(ST3Gal I) and Gal $\beta$ 1,4GlcNAc $\alpha$2,6-sialyltransterases(ST6Gal I), which are well known as glycosyltransterases associated with cancer. The aqueous extracts of Scutellaria Baicalensis Georgi, Coptidis Rhizoma, Glycyrrhiza urlensis Fisch, Bupleuri Radix and Platycodi Radix were prepared and tested, respectively. At concentration of 100$\mu$g, Glycyrrhiza uralensis Fisch showed the highest inhibitory effects(about 42% and 57%, respectively) on ST3Gal Iand ST6Gal Iactivities. ST3GAl I was inhibited about 23% by Scutellaria baicalensi G댁햐, but not by the other samples, whereas ST6Gal I was inhibited about 20% and 40%,respectively, by Scutellaria baicalensis Georgi and Bupleuri Radix. All inhibitory effects were obtained in a concentration-dependent manner.

Specification and Testing Method of WHW ex. for Chronic Renal Failure (만성신부전 치료제 개발을 위한 WHW 엑스의 기준 및 시험방법 설정)

  • This study is to specify the criteria and testing methods of WHW extract which has a potency as a therapeutic agent for chronic renal failure. The determination of specifications of WHW extract is mostly important because of the quality assurance. Three batches of WHW extract were obtained by the extraction at $98^{\circ}$C for 3 hr using water from mixture of 15 herbal medicines including Codonopsis Pilosulae Radix, Salviae Radix, Pinelliae Rhizoma, Coptidis Rhizoma, Evodiae Fructus, Epimedii Herba, Rhei Rhizoma, Perillae Herba, Glycyrrhizae Radix, Artemixiae capillaris Herba, Alimatis Rhizoma, Hoelen, Atractylodies Rhizoma alba, Polyporus and Cinnamomi Ramulus, subsequently, vaccum-dried for 15 hr. The yield of WHW extracts was 24.53% on the average. The identification of each herbal medicine of WHW extract was performed by modification of Korean Pharmacopeia IX (KP IX). The assay of WHW extract was performed using standard such as berberine, icariin, glycyrrhizin, and cinnamic acid of indicative herbal medicines by modification of KP IX, too. As well as, paticle size classification test was carried out to indicate the boundary of particle size of WHW extract and the particle size of WHW extract more than 50% showed the 140 ${\mu}$m. Taken together, WHW extract could be prepared reproducibly and assurable if follows the presented extraction and drying steps and its specifications were satisfied with the indicated criteria.

  • Atopic dermatitis is frequently encountered in the Dermatology clinic and the treatments are very various but the effects are not good enough. Especially in case of infants, taking herb med or acupuncture treatments are difficult so we would like to treat the atopic dermatitis using external therapy. We chose the oriental medicine textbooks and the oriental medicine journals that external therapy was used and divided into Yugobub(油膏法), Buchubbub(敷貼法), Sechukbub(洗滌法). The results were as follows: 1. In the frequency of medicine was Coptidis Rhizoma(黃連), Phellodendri Cortex(黃栢), Sophorae Radix(苦蔘), Cnidii Fructus(蛇床子), Allumen(枯礬) etc. 2. The effects of medicine were Chungyul(淸熱), Haedok(解毒), Zosub(燥濕), salchung(殺蟲), Gupung(祛風), Jiyang(止痒) etc.

  • This study was progressed for oriental medical care of neonates by fetal toxicosis removal therapy, and the results as follows: 1. The purpose of fetal toxicosis removal therapy : It removes dirty blood in neonatal mouth and prevents diseases. 2. Kinds of fetal toxicosis removal therapy : Radix Glycyrrhizae(甘草), Rhizoma Coptidis(黃連), Cinnabaris-Mel(朱蜜), Calculus Bovis(牛黃), Sojae Semen Praepartum, Juice of Semen Allii Tuberosi(非汁) and mercury(汞粉). 3.   • A Literatual Study on the external treatment of eye diseases, the result were as follows. 1. The external treatment of eye diseases used eyedrops(點,滴), steam bath(熏), washing(洗), plaster(敷). 2. Eyedrops used filterated decoction. 3. Eye diseases used Bomeolum(永片), Coptidis Rhiwma(黃連), Angelicae gigantis Radix(當歸) with clearing away heat and toxic materials, clearing away swelling, pain, ithing and convulsion, clearing away nebula 4. The Growth Inhibiting Effect of Vibrio parahaemolyticus KCTC 2471 by the Natural Products Bearing Antioxidative Capacity (Vibrio Parahaemolyticus KCTC 2471에 대한 항산화 천연산물의 성장 억제 효과)

  • The growth of V. parahaemolyticus was inhibited by the combination of natural products having antioxidative capacity. The growth of V. parahaemolyticus was controlled effectively by each or the combination of Paeonia japonica, Acanthodanacis cortex, Phellodendri cortex, Mori cortex radicis, Theae folium, Galla rhois, Coptidis rhizomn and Astragali radix. The growth inhibition ratio of the combination of Astragali radix, Phellodendri cortex, Coptidis rhizotna and Paeonia japonica was 95%, and its antioxidative capacity was also 5.4 times higher, and the elimination ratio of hydroxyl radical was more than 80% compared with control. As natural products might activate some parts of human body, they could be utilized as pharmaceuitcal applications, functional foods, antiaging tea, and multifunctional fermentative beverages.

Exploring the Effective Herbal Prescription for Cognitive Disorder Treatment among Licensed Herbal Medicines in Korea - A Preliminary Study for Clinical Trial of Cognitive Disorders (기허가 한약제제를 대상으로 한 인지장애 치료 유효 약물 탐색 - 인지장애 임상연구를 위한 예비연구)

  • It is necessary to investigate the efficacy of commercial Korean herbal medicine for cognitive disorder. The purpose of this study is to select candidates among licensed herbal medicines that are expected to be effective in the treatment of cognitive disorders and can be used in future clinical trial. From laboratory experiments, we first found individual single herbal drugs that could be effective for cognitive disorder, and then through experts recommendation, selected five priorities of single herbal drug and obtained the information of the best herb as a combination of each herbal drug. To derive the final herbal prescriptions, we searched the KFDA drug information system for licensed herbal medicines containing each drug and its combination. As a result of laboratory experiments and experts recommendation, we found that the five effective single herbal drugs for cognitive disorders. They are Ginseng Radix, Acori Graminei Rhizoma, Cyperi Rhizoma, Coptidis Rhizoma, Pinelliae Rhizoma, Hoelen cum Pini Radix, and Rehmanniae Radix Preparata(Pinelliae Rhizoma, Hoelen cum Pini Radix, and Rehmanniae Radix Preparata were tied for $5^{th}$). And licensed herbal prescriptions derived from the planned search are Palbohoichoon-tang, Taehwa-hwan, Bosim-hwan, and so on. Among these, in consideration of the feasibility of research and possibility of success in development, Yukgunja-tang, Samhwangjichul-hwan can be selected as future study subjects. Through experimental studies and expert recommendations, we have derived herbal prescriptions that can be effective in treating cognitive disorders from licensed herbal medicines.
