The Effect of Children's Beverages on Degradation of Dental Resin-Based Pit and Fissure Sealant (어린이 음료가 레진계 치면열구전색제의 화학적 분해에 미치는 영향)

  • The consumption of beverages among children is rising. The purpose of this study was to examine the effect of kid's drink on dental resin-based pit and fissure sealant. Pororo, I-kicker, Sunkist kids were included in the experimental groups, and Samdasu was included in the control group. A conventional dental sealant material ($Clinpro^{TM}Sealant^{(R)}$) was selected for this study. Resin specimens (8 mm in diameter and 1 mm in thickness) were prepared according to manufacturers' instructions and the initial roughness (Ra) was then measured. The pH of all the four groups was measured using a pH meter. The specimens were individually immersed in 5 ml of the experimental solutions and stored at $37^{\circ}C$ for 72 hours. Following this, the surface roughness of the resin specimens was measured by Surftest. The concentration of residual monomer released was determined by high performance liquid chromatography (HPLC). The surface morphology of the resin specimen was evaluated before and after storage by scanning electron microscopy (SEM). Data were statistically analyzed using Kruskal-Wallis and Duncan's test. The results showed that all the children's beverages examined in this study contained citric acid. The pH of I-kicker was the lowest ($3.03{\pm}0.01$), followed by that of Sunkist kids ($3.26{\pm}0.02$) and Pororo ($3.47{\pm}0.02$). We observed an increase in the surface roughness of resin specimens after 72 h of immersion in all the beverages tested (p<0.05). There was matrix degradation after immersion, visualized on SEM image, in all the beverage groups. Bisphenol-A-glycidyl methacrylate was not detected after 72 hours, but triethylene glycol dimethacrylate levels were increased in all the beverages tested during the 72 hours by HPLC. These results suggest that intake of beverages containing acid can cause degradation of the resin-based pit and fissure sealants in children.

Effects on Ginseng Growth and Ginsenoside Content in ICT-based Process Cultivation and Conventional Cultivation (ICT 기반의 공정재배와 관행재배에 있어서 인삼 생장 및 진세 노사이드 함량에 미치는 영향)

  • This study conducted an experiment with EC 1.0ms/cm ratio and excellent soil conditions for germination in ICT-based ginseng process cultivation. The first growth survey was conducted before transplantation of ginseng 1-year roots grown by seeding ginseng in the process cultivation, conventional cultivation and a second growth comparison survey was conducted after 3 months of growth. In the results, it was confirmed that ginseng grown in the process cultivation grew more than in the field. As a result of comparing the contents of 11 ginsenosides of 1-year and 2-year-old ginsenosides in the process cultivation and conventional cultivation ginseng, it was confirmed that the content of the process cultivation ginseng was higher than that of practice cultivation ginseng. In conclusion, conventional cultivation ginseng grows due to various factors under the natural cultivation environment, but process cultivation can secure the growth stability of ginseng by allowing stable soil and environmental control, so continuous research is needed in the future.

Enzymatic Synthesis of Low-trans Fats Containing Conjugated Linoleic Acids and Their Physicochemical Characteristics (Conjugated Linoleic Acid(CLA)를 함유한 기능성 저트랜스 유지의 효소적 합성 및 이화학적 특성 연구)

  • Scale-up production of low-trans fat containing conjugated linoleic acid (CLA-TFO) was performed through lipase-catalyzed synthesis. Blend of fully hydrogenated soybean oil, olive oil containing conjugated linoleic acid and palm oil with 1:2:7 ratio was interesterified through Lipozyme RM IM in the 1 L-batch type reactor at $65^{\circ}C$ for 12 hrs, and the physicochemical and melting properties of CLA-TFO were compared with conventional (high trans fat) or commercial low-trans fat shortening. The trans fatty acids content in the conventional shortening (48.8 area%) was much higher than that of low-trans shortening (0.4 area%) and CLA-TFO (0.3 area%+CLA; 7.6 area%). Acid, saponification and iodine values of CLA-TFO were 0.4, 173.9 and 59.0, respectively. Their ${\alpha}$-, ${\gamma}$-tocopherol contents showed 4.7, 1.0 mg/100 g. Differences were observed in the solid fat contents (SFC), melting point of the conventional or low-trans fat and CLA-TFO. Each SFC of conventional, low-trans fat and CLA-TFO was 32.0, 29.3 and 30.4% with melting point of 38.5, 43.0 and $39.5^{\circ}C$ at $35^{\circ}C$, respectively. In texture profile analysis, hardness of conventional, low-trans fat and CLA-TFO was 111.7, 75.2 and 63.8 g.

Development of Near-Critical Water Reaction System for Utilization of Lignin as Chemical Resources

  • Plant biomass has been proposed to be an alternative source for petroleum-based chemical compounds. Especially, phenolic chemical compounds can be obtained from lignin by chemical depolymerization processes because lignin consists of complex aromatic polymer such as trans-p-coumaryl, coniferyl and sinapyl alcohols, etc. Phenolic chemical compounds from lignin were usually produced in super critical water. However, we applied Near-critical water (NCW) system because NCW is known as a good solvent for lignin depolymerization. Organic matter like lignin can be solved in NCW system and the system has a unique acid-base property without conventional non-eco-friendly chemicals such as sulfuric acid and sodium hydroxide. In this work, we tried to optimize the NCW depolymerization system by adjusting the processing variables such as reaction time, temperature and pressure. Moreover, the amount of additional phenol was optimized by changing the molar ratio between water and phenol. Phenol was used as capping agent to prevent re-polymerization of active fragment such as formaldehyde. Alkali-lignin was used as a starting material and characterized by a Solid State 13C-NMR, FT-IR and EA (Elemental Analysis). GC-MS analysis confirmed that o-cresol, p-cresol, anisole and 4-hydroxyphathalic acid were the main product and they were quantitatively analyzed by HPLC.

Effects of High-Hydrostatic Pressure on Ginsenoside Concentrations in Korean Red Ginseng

  • The effects of high-hydrostatic pressure (HHP) on the ginsenoside concentration in Korean red ginseng were investigated. HHP-pretreated Korean red ginseng samples were compared to samples produced by a conventional method. Six-year-old Korean fresh ginseng (Panax ginseng C.A. Meyer) samples were vacuum-packaged in polyethylene film and treated at room temperature for 1 min with HHP (200-600 MPa) and steamed at $98^{\circ}C$ for 3 hr. Major ginsenosides of red ginseng were analyzed by HPLC. HHP-pretreated red ginseng showed a 45% higher level of total major ginsenosides than conventionally prepared red ginseng. The levels of 4 protopanaxadiol-type ginsenosides increased 34-43% and the levels of 5 protopanaxatriol-type ginsenosides increased 45-66%. Scanning electron microscopy and electrical conductivity spectrum analysis showed that HHP pretreatment damaged ginseng plant cells and increased extraction efficiencies of ginsenosides from red ginseng products.

Quantitative Source Estimation of PM-10 in Seoul Area (서울시 PM-10 오염원의 정량적 기여도 추정)

  • Recently in Korea, due to the significant drop of lead and bromine levels as a marker of autoemission source in the urban areas, the conventional application of receptor methods has many difficulties to properly apportion mass contribution of some sources. It is then needed to urgently develop alternative source profiles and identify new emission markers. Thus, the study has extensively examined the results obtained from using PAHs and elemental data for receptor modeling and has provided an opportunity to identify alternative source compositions and to determine a proper number of the ambient emission sources in Seoul area. The purpose of the study is to identify the sources of PM-10 and to estimate their mass contributions in Seoul area. Thus, a receptor model, target transformation factor analysis(TTFA) has been massively applied. The TTFA offers the possibility of determining the number of sources and their mass contributions. The input data used in this study are composed of two separate sets: fine (d$_{p}$ < 2.5.mu.m) and coarse (2.5.mu.m < d$_{p}$ < 10.mu.m) mode aerosol samples. Each sample was simultaneously collected by a PM-10 dichotomous sampler during the daytime(8 AM to 8 PM) and the nighttime(8 PM to 8 AM) from February to October 1993 on the Sungdong-Gu, Seoul. All the samples were analyzed to determine the levels of 10 inorganic elements by an XRF system as well as 14 PAHs by a HPLC. However, only 8 inorganic elements and 7 PAHs were used for the various statistical analysis.sis.

A Rapid Method of Ginsenoside Analysis in HPLC by Pretreatment through the reverse-phase minicolumn (역상소형컬럼 전처리를 이용한 Ginsenoside의 신속정량법)

  • The solvent separation step in the conventional method for quantitative analysis of ginsenosides was substituted by purification through a small reverse-phase $C_18$-column resulting in the decrease of analysis time by one fourth. New method showed high recovery of total ginsenosides but low recovery in protopanaxatriol-ginsenosides. Sugars did not affect the recovery by the amount in usual root sample. Coefficient of variation in recovery of ginsenosides was lower in the rapid minicolumn method. Optimum load of ginsenosides to minicolumn was 10 to 15 mg. The rapid minicolumn method showed highly significant correlation with the solvent separation method for dried root and red ginseng. For the rapid minicolumn method a small acryl device was used for the simultaneous extraction of 8 samples. This method appeared to be beneficial in cost and for the health of analyst.

Advanced Lipid Extraction Method for the Determination of the Phospholipase D Activity

  • Phospholipase D is a ubiquitous enzyme that plays an important role in various lipid mediated cellular signaling pathways and produces rare phospholipids, phosphatidylethanol or phosphatidylbutanol, instead of phosphatidic acid with unique catalytic activity transphosphatidylation in the presence of primary alcohols. The reaction products, phosphatidylethanol or phosphatidylbutanol are used as markers of in vitro phospholipase D activity in many studies. For the sensitive detection of the phospholipase D products, we developed an advanced lipid extraction method that facilitates recovery of the compounds. With the new method, the activity change of phosaholipase D by agonists could be detected more easily and the recovery rate was also increased. The increase of detected enzyme activity change was about double fold compared to the conventional lipid extraction method. This method provides selective force for the phospholipase D products in the extraction procedure.

On-Line Measurement System for the Determination of Chemical Oxygen Demand (화학적 산소 요구량 측정을 위한 On-Line 측정 시스템에 관한 연구)

  • A simple on-line measurement system consisting of a conventional peristaltic pump, a HPLC-type heater, and a flow-through spectrophotometer is introduced for the determination of chemical oxygen demand(CODI. The system was configured such that the reaction mixture in the highly concentrated surffuric acrid medium flowing through the PTFE reaction tubing was heated at 150℃ and the absorbance of dichromate was continuously moutored at 445 m. The same addation principle as in the standard procedure was employed akcept the use of CoSO4 as a new effective catalyst. To test the system, potassium hydrogen phthalate was selected as a COD standard material. With suitably optimized reaction condition, the applicable concentration range depends on the concentration of potassium dichromate in the oxidizing reagent. With 2.0×10-3 M and 5.0×10-4M dichromate, the linear dynamic range was observed up to 400 ppm and 100 ppm, respectively. The standards in the Unear ranges were shown to be completely oxidized, which was confirmed with sodium oxalate or Mohr's salt. In all cases, the typical reproduclbility for betweenruns was 2% or less. The proposed measurement system provides the valuable in- formation for the further development of automated analysis system based on the present standard procedure.

Comparative Effects of Microwave-Assisted Process Under Atmospheric Pressure Condition and Conventional Process on Extraction Efficiencies Ginseng Components (상압조건의 마이크로파 공정과 현행 방법에 의한 인삼근 유용성분의 추출효율 비교 연구)

  • Microwave assisted extraction(MAE) is known as a more environmental friendly process with economic advantages in terms of less time, less solvent, less energy and less wastes than the current time consuming reflux method. It was applied to develop a rapid extraction method for soluble ginseng components that are major materials used for the processing of ginseng products. In a comparative study between pre established MAE(ethanol 60%, power 80 W, process time 4 min$\times$5) and current extraction method(ethanol 80%, temp. 85oC, time 8 hr$\times$5), MAE was more efficient than the current method to obtain an extract yield(soluble solid), but it was insufficient to extract individual ginsenosides, total phenols, reducing components and acidic polysaccharides. MAE with 80% ethanol by 5 times showed, however, that its extraction efficiency on soluble solid, crude saponin, major ginsenosides, and the other components was equal or superior to that of the current method, indicating that ethanol concentration is one of the critical parameters influencing the MAE process. The quality of ginseng extracts from MAE was assured by evaluating the corresponding standards and by comparing TLC and HPLC patterns with the control.
