Poly(Dimethylaminoethyl Methacrylate)-Based pH-Responsive Hydrogels Regulate Doxorubicin Release at Acidic Condition

  • Stimuli-responsive biomaterials that alter their function through sensing local molecular cues may enable technological advances in the fields of drug delivery, gene delivery, actuators, biosensors, and tissue engineering. In this research, pH-responsive hydrogel which is comprised of dimethylaminoethyl methacylate (DMAEMA) and 2-hydroxyethyl methacrylate (HEMA) was synthesized for the effective delivery of doxorubicin (Dox) to breast cancer cells. Cancer and tumor tissues show a lower extracellular pH than normal tissues. DMAEMA/HEMA hydrogels showed significant sensitivity by small pH changes and each formulation of hydrogels was examined by scanning electron microscopy, mechanical test, equilibrium mass swelling, controlled Dox release, and cytotoxicity. High swelling ratios and Dox release were obtained at low pH buffer condition, low cross-linker concentration, and high content of DMAEMA. Dox release was accelerated to 67.3% at pH 5.5 for 6-h incubation at $37^{\circ}C$, while it was limited to 13.8% at pH7.4 at the same time and temperature. Cell toxicity results to breast cancer cells indicate that pH-responsive DMAEMA/HEMA hydrogels may be used as an efficient matrix for anti-cancer drug delivery with various transporting manners. Also, pH-responsive DMAEMA/HEMA hydrogels may be useful in therapeutic treatment which is required a triggered release at low pH range such as gene delivery, ischemia, and diabetic ketoacidosis.

A Review of Nanostructured Ca-aluminate Based Biomaterials within Odontology and Orthopedics

  • This presentation will give an overview of Ca-aluminate based biomaterials and their proposed use within the field of nanostructured biomaterials. The paper describes typical features of Ca-aluminate materials with regard to technology, chemistry, biocompatibility including hemocompatibility and bioactivity, and developed microstructure. Special focus will be on the developed microstructure, which is in the nanosize range. Application possibilities within odontology, orthopedics, and drug delivery are presented. The nanostructure including pore size below 5 nm in these structures opens up this material for some use in specific dental-related applications in which antibacterial and bacteriostatic aspects are of importance, and as thin coating on implants within dental and orthopaedic applications. Nanosize porosity is essential in drug delivery systems for controlled release of medicaments. The priority field for Ca-aluminate biomaterials is implant materials, which use minimally-invasive techniques to offer in vivo, on-site developed biomaterials.

Chitosan-Coated Fe3O4 Magnetic Nanoparticles as Carrier of Cisplatin for Drug Delivery

  • A synthesis method for a chitosan-coated magnetic drug-delivery system of cisplatin is proposed. Here, cisplatin was conjugated to the surface of Magnetite ($Fe_3O_4$) nanoparticles via a (3-Aminopropyl)-trimethoxysilane (APTS) coupling agent. To reduce the cytotoxic effect of cisplatin, the magnetic drug was then encapsulated in chitosan (CS-cisplatin-$Fe_3O_4$) through the water/oil (W/O) emulsion method. The CS-cisplatin-$Fe_3O_4$ nanoparticles were synthesized in a spherical shape with a diameter of 190 nm. The cytotoxicity assay was performed using HeLa cells. The cisplatin uptake of the cells was determined using High Performance Liquid Chromatography (HPLC) to calculate the drug content. The controlled release of cisplatin was demonstrated by regulating the dissolution and diffusion of the drug through the chitosan matrix.

  • We investigated the diffusive transport of bupivacaine HCl through the microchannels of microfluidic drug delivery devices. In the biodegradable microfluidic drug delivery devices developed in this research, the drug release rate can be controlled by simply modulating the geometrical parameters of the microchannels, such as the length, number, and cross-sectional area of the microchannels, when the microchannels are used as paths for drug release. However, the hydrophobic nature of a biodegradable polymer, 85/15 poly(lactic-co-glycolic acid), hinders the infiltration of a release medium (phosphate-buffered saline) through the microchannels into the reservoir of a device that contains bupivacaine HCl, at the early stage of drug release. This can have an adverse effect on the early stage release of local analgesic compounds from the device. In this study, microfluidic channels were surface-treated with surfactants such as PEG600 and Tween80, and the effects of the surfactants on the release performance are presented and analyzed.

Effect of pH on Swelling Property of Hyaluronic Acid Hydrogels for Smart Drug Delivery Systems

  • Hyaluronic acid(HA) hydrogels were synthesized by immersing HA microbeads in phosphate buffered saline solutions having different pH levels to assess the effect of pH on the swelling ratio of HA hydrogels for smart drug delivery systems. No beads were formed when the HA solution(below pH 9) was crosslinked with divinyl sulfone(DVS) because DVS is a basic solution. The variation regarding the size of the microbead was not significant, suggesting that the bead size is not a function of pH(10 ~ 14). However, the pore size of the microbeads decreased with increasing pH from 10 to 14, leading to the surface smoothness and dense network as a result of higher crosslinking. The swelling ratio of hydrogels increased when the pH rose from 2(acidic) to 6(neutral). Afterwards, it decreased with further increasing pH(basic). The lower swelling ratio may be due to the lack of ionization of the carboxyl groups. On the other hand, a higher swelling ratio is likely due to the increased electrostatic repulsions between negatively charged carboxyl groups on different chains. Experimental results suggested that pH-responsive HA hydrogels can be applicable to the controlled drug delivery systems.

Iontophoretic Delivery of Vitamine C 2-Phosphate (이온토포레시스를 이용한 2-인산 비타민 C의 피부투과)

  • In order to develop an optimum formulation for iontophoretic flux of vitamine C 2-phosphate (VCP), we have prepared three different hydrogels containing VCP, using carbopol, HPMC and poloxamer, and iontophoretic flux through hairless mouse skin from these hydrogels was carried out. Drug stability in phosphate buffer (PBS) solution (pH 7.4) with and without current application was studied. The effect of various factors, such as drug concentration, current density, and current profile on skin flux was also investigated. Stability study indicated that VCP in PBS (pH 7.4) solution was stable under the experimental condition, irrespective of the presence of current. Cathodal delivery increased the flux markedly, whereas the anodal and passive flux was negligible. Thus, cathodal delivery was used in all experiments. Flux increased as the drug concentration (2.5, 5.0, 7.5%) and current density $(0.2,\;0.4,\;0.6\;mA/cm^2)$ increased. Pulsed application of the current showed lower flux than constant current application. The results obtained suggest that VCP can be delivered into the skin and the amount delivered can be controlled by varying hydrogel, current density, drug concentration and current application profile.

  • The characteristics of the controlled drug release were studied for biodegradable transdermal drug delivery system. A biodegradable polymeric matrix was prepared from chitosan, silver sulfadiazine, and glycerine. The release behavior of silver sulfadiazine from chitosan matrix was consistent with the Higuchi's diffusion controlled model. The release time was delayed by increasing the content of silver sulfadiazine and thickness of the matrix, whereas decreased as glycerine concentration increased. The apparent constant (K) of release rate was proportional to the content of drug or glycerine and the thickness of chitosan matrix. These results indicated that chitosan matrix shows some potential as a drug delivery system for transdermal therapeutic application.

Multi-Layered Matrix Tablets with Various Tablet Designs and Release Profiles

  • Tablet dosage forms have been preferred over other formulations for the oral drug administration due to their low manufacturing costs and ease of administrations, especially controlled-release applications. Controlled-release tablets are oral dosage forms from which the active pharmaceutical ingredient (API) is released over an intended or extended period of time upon ingestion. This may allow a decrease in the dosing frequency and a reduction in peak plasma concentrations and hence improves patient compliance while reducing the risk of undesirable side effects. Conventional singlelayered matrix tablets have been extensively utilized to deliver APIs into the body. However, these conventional single-layered matrix tablets present suboptimal delivery properties, such as non-linear drug delivery profiles which may cause higher side effects. Recently, a multi-layered technology has been developed to overcome or eliminate the limitations of the singlelayered tablet with more flexibility. This technology can give a good opportunity in formulating new products and help pharmaceutical companies enhancing their life cycle management. In this review, a brief overview on the multi-layered tablets is given focusing on the various tablet designs, manufacturing issues and drug release profiles.

A Study on Drug trading countermeasures via internet and sns (인터넷과 sns를 이용한 마약거래 대응방안에 관한 연구)

  • The drug trade among the general public via the Internet and sns have been increasing, which is becoming a social problem. The general public believe that even if they do the drug trade via the Internet and sns the probability of detection is low. so they will conduct drug trade via the Internet and sns. Therefore, if the general public recognize that there is a high likelihood of disclosure, drug trade via the Internet and sns are likely to decline. If the possibility of punishment increases through specification of controlled delivery techniques and Introduction of entrapment investigator, it seems that the general public can not easily deal with drug trade via the Internet and sns. Also by further subdividing the penalties for drug offenses, for simple drug buyers through cure-oriented treatment rather than punishment drug demand be suppressed and penalties for drug suppliers should be strengthened.
