청과물 저장에 관한 연구(제3보) -환경압력변화가 Tomato 과실의 저장에 미치는 영향에 대하여- (A Study on the Storage of Fresh Fruits and Vegetables(III) -Effects of the Storage of Tomato fruits by controlled atmospheric pressure-)

  • 1. 품종은 노지에서 재배한 복수2호를 사용하였으며 1971년과 1972년 2회에 걸쳐 Tomato 과실의 성숙시기별 호흡량변황 및 저장중 성분변화에 대하여 조사하였다. 2. 환경압력조절에 의한 Tomato 과실의 호흡억제효과는 SAP 구중 660 Torr. 구가 가장 효과적이었다. 3. 성장중 성숙시기별로 채취한 Tomato의 호흡량 변화는 각채취시기별로 차이가 있었으며 Tomato의 일대에 있어서 시기별 숙기에 이른 과실을 경과일수에 따라 호흡량을 조사하면 전체적으로 호흡량의 증감이 sigmoid 형으로 나타났으며 하나의 climacteric pattern을 나타내 주고 있다. 4. 성장중 성숙시기별로 채취한 Tomato 과실의 환경압력변화에 의한 영향은 어느시기에서나 NAP구보다 SAP구의 호흡량이 적었으며 전 기간중 동일한 경향으로 호흡억제 효과가 있었다. 5. 저장중 Tomato 과실의 중량변화는 NAP 구가 SAP 구보다 현저하며 산, 당 및 Vitamine-C의 변화에 있어서는 SAP구가 NAP구 보다 감소율이 적었다.

비닐하우스 토마토의 온실유형에 따른 장기평균비용을 고려한 적정 생산규모 분석 (An Analysis of Optimal Production Scales by Greenhouse Types using Long-run Average Cost in Controlled Tomato)

  • The optimal greenhouse scales for controlled tomato should be studied because the increase of oil price and labor costs following the shortage of workforce makes greenhouse cultivation hard to gain profits. The purpose of this study is to estimate optimal production scales by greenhouse types for controlled tomato. The translog cost function is estimated based on the production cost survey data. The results can be summarized as follows: First, the average production cost of controlled tomato per kg decreases as the production scale increases. Second, according to the tomatoes farm of standard farming income data of RDA, the minimum production scale is 23 ton. Third, the estimated output of single-span greenhouse considering production scale with minimum average cost is 345 ton and production cost per kg is 1,476 won. The corresponding figures of multi-span greenhouse are 415 ton and 936 won, respectively. The study results can be used as basic materials for efficient decision making of tomato farmhouses and novice farmers. Also, the study shows that multi-span greenhouse should be encouraged to be built, since it requires lower marginal cost than single greenhouse. The results of this paper will help increase the income of farmhouses and cut expenses for the coming years.

Screening assay for tomato plants resistant to Fusarium oxysporum f. sp. lycopersici race 2 using the expression of the avr2 gene as a selection marker

  • Fusarium wilt disease of tomato plants caused by Fusarium oxysporum f.sp. lycopersici (FOL race2) is one of the most important diseases of tomatoes worldwide. In the competition between tomato and FOL, the FOL can win by overcoming the immune system of tomato plants. Resistant interaction between the FOL race2 and tomato plants is controlled by avirulence genes (AVR2) in FOL and the corresponding resistance genes (I2) in tomato plants. In this study, 7 FOL isolates (KACC) were used to test their pathogenicity, and FOL race2 was selected because it is a broad problem in Korea. The Fol40044 isolates showed the most severe pathogenicity, and the avr2 gene was also isolated and identified. Moreover, to select resistance, 20 tomato varieties were inoculated with the Fol40044, and the degree of pathogenicity was evaluated by analyzing the expression of the avr2 gene. As a result, three resistant tomato varieties (PCNUF73, PCNUF101, PCNUF113) were selected, and the expression of the avr2 gene was much lower than that of the control Heinz cultivar. This result shows that the screening assay is very efficient when the avr2 gene is used as a marker to evaluate the expression level when selecting varieties resistant to tomato wilt disease. Based on these results, it is possible to isolate the I2 gene, which exhibits resistance and molecular biological interactions with the AVR2 gene from the three tomato-resistant varieties. The I2 gene provides breeders more opportunities for Fusarium disease resistance and may contribute to our understanding of their interactions with the FOL and host plant.

Probiotication of Tomato Juice by Lactic Acid Bacteria

  • This study was undertaken to determine the suitability of tomato juice as a raw material for production of probiotic juice by four lactic acid bacteria (Latobacillus acidophilus LA39, Lactobacillus plantarum C3, Lactobacillus casei A4, and Lactobacillus delbrueckii D7). Tomato juice was inoculated with a 24­h-old culture and incubated at $30^{\circ}C$. Changes in pH, acidity, sugar content, and viable cell counts dur­ing fermentation under controlled conditions were measured. The lactic acid cultures reduced the pH to 4.1 or below and increased the acidity to $0.65\%$ or higher, and the viable cell counts (CFU) reached nearly 1.0 to $9.0\times10^9/ml$ after 72 h fermentation. The viable cell counts of the four lactic acid bacteria in the fermented tomato juice ranged from $10^6\;to\;10^8\;CFU/ml$ after 4 weeks of cold storage at $4^{\circ}C$. Pro­biotic tomato juice could serve as a health beverage for vegetarians or consumers who are allergic to dairy products.

토마토 분말 첨가 시폰 케이크의 제조조건 최적화 및 품질 특성 (Processing Optimization and Antioxidant Activity of Chiffon Cake Prepared with Tomato Powder)

  • The purpose of this study was to determine the optimal mixing condition of tomato powder and sugar for producing chiffon cake. The experiment was designed according to the central composite design of response surface, which yielded ten experimental points, including two replicates. Physiochemical and sensory properties were measured, and theses values applied to mechanical models. A canonical form and perturbation plot showed the influence of each ingredient on the final product mixture. The results of the physiochemical analysis of each sample showed significant differences in sweetness (P<0.01), color L (P<0.001), color a (P<0.001), color b (P<0.05), hardness (P<0.05), and cohesiveness (P<0.01). The sensory measurements were significantly different in color (P<0.05), appearance (P<0.05), flavor (P<0.05), sweetness (P<0.01), moistness (P<0.05), and overall acceptability (P<0.05). The optimal formulation, calculated using the numerical and graphical method, was determined to be 59.27 g tomato powder and 285.66 g sugar. The sensory evaluation showed significantly higher preferences in the color, flavor, appearance, texture, sweetness, tenderness, moistness and overall quality of the optimized chiffon cake compared to the controlled chiffon cake. The optimized chiffon cake also showed a high antioxidative activity compared to the controlled chiffon cake. Our results show that chiffon cake prepared with tomato powder enhances sensory characteristics and antioxidative activity.

Biocontrol Activity of Acremonium strictum BCP Against Botrytis Diseases

  • Biological control activity of Acremonium strictum BCP, a mycoparasite on Botrytis cinerea, was examined against six plant diseases such as rice blast, rice sheath blight, cucumber gray mold, tomato late blight, wheat leaf rust, and barley powdery mildew in growth chambers. The spore suspension of strain BCP showed strong control activities against five plant diseases except against wheat leaf rust. On the other hand, the culture filtrate of A. strictum BCP was effective in controlling only cucumber gray mold and barley powdery mildew. Further in vivo biocontrol activities of A. strictum BCP against tomato gray mold were investigated under greenhouse conditions. Control efficacy of the fungus on tomato gray mold increased in a concentration-dependent manner. Treatment of more than $1{\times}10^6$ spores/ml significantly controlled the disease both in tomato seedlings and in adult plants. The high disease control activity was obtained from protective application of the strain BCP, whereas the curative application did not control the disease. Foliar infections of B. cinerea were controlled with $1{\times}10^8$ spores/ml of A. strictum BCP applied up to 7 days before inoculation. In a commercial greenhouse, application of A. strictum BCP exhibited the similar control efficacy with fungicide procymidone (recommended rate, $500{\mu}g/ml$) against strawberry gray mold. These results indicate that A. strictum BCP could be developed as a biofungicide for Botrytis diseases under greenhouse conditions.

인공광원이 방울토마토(Lycopersicon esculentum var. cv. 'CF Jelly') 생육에 미치는 영향 (Artificial Light Sources Influence Cherry Tomato (Lycopersicon esculentum var. cv. 'CF Jelly') Growth and Development)

  • BACKGROUND: Selection an suitable light source is essential in the year-round production of horticultural crops in closed production systems such as plant factory with controlled environments. This study was investigated to confirm the effects of artificial light sources on growth of cherry tomato'CF Jelly'(Lycopersicon esculentum var.) under high-pressure sodium lamps (HPS), metal-halide lamps (MH), and LEDs. METHODS AND RESULTS: Light intensity of the light sources was controlled at 220±30 µmol/m2/s with 12 hrs of photoperiod for a day. Flower development was significantly faster in HPS and MH treatments compared to the LEDs. There was no significant difference between the leaf number and leaf shape under the HPS and MH treatments. Reproductive growth of cherry tomato was significantly promoted by the LEDs treatment of blue plus red lights. Fruit yield per plant also increased under the LEDs compared to the others. CONCLUSION(S): Growth, flowering, and fruit setting of the cherry tomato were accomplished by the artificial lights under plant factory conditions. The HPS treatment showed negative effect on fruit quality in terms of blossom-end rot incidence compared to the LEDs or MH treatment. Effect of the LEDs on promotion of fruit weight and yield was also proved. Additional research should be carried out for improving sugar metabolism or decreasing disease in the fruits under plant factory system using only artificial lights.

Variation of Microbial Communities with Crop Species in Controlled Horticultural Soils of Gyeongnam Province

  • In this study, we examined the chemical properties and microbial community characteristics in 25 controlled horticultural soils (CHS) sampled from Gyeongnam Province by fatty acid methyl ester (FAME) method. The electrical conductivity of watermelon CHS was significantly (p < 0.05) higher than those of red pepper CHS, pumpkin CHS, and strawberry CHS. The amounts of total FAMEs, total bacteria, gram-negative bacteria, gram-positive bacteria, and fungi were significantly (p < 0.05) higher in red pepper CHS than those in strawberry CHS and pumpkin CHS. In addition, higher (p < 0.05) ratios of cy19:0 to $18:1{\omega}7c$ were detected in tomato CHS than those in watermelon CHS, pumpkin CHS, and red pepper CHS. This implied that microbial communities of tomato CHS were stressed more than other species of cultivation soils. Actinomycetes community in red pepper CHS was significantly (p < 0.05) higher than those in tomato CHS, strawberry CHS, and watermelon CHS. Differences in soil microbial community composition were highly associated with cultivated crop species which might result from the management inputs such as fertilizer, herbicide, and irrigation.

잎마름역병 저항성 육종을 위한 토마토 유전자원의 저항성 평가 (Evaluation of Tomato Genetic Resources for the Development of Resistance Breeding Lines against Late Blight)

  • Phytophthora infestans에 의해 발생하는 토마토 잎마름 역병은 전 세계적으로 발생하여 토마토 생산에 막대한 피해를 준다. 이 병의 방제를 위하여 저항성 품종의 이용은 매우 중요하다. 저항성 품종육종의 초기 및 본 육종에서 토마토 유전자원의 저항성 평가는 매우 중요하다. 본 연구는 2009년에 유묘 및 성체식물을 대상으로 78종의 토마토 시판 품종과 계통에 대한 잎마름역병 저항성을 검정하였다. Legend를 포함한 모든 시판 품종은 감수성이지만, California 대학에서 분양 받은 KNU-2, KNU-6-1, KNU-11, KNU-13, KNU-14-1 계통과 저항성 유전자 Ph-3를 가지고 있는 L3708, $AV107-4{\times}L3708$, $07-15{\times}L3708$, $BS67{\times}L3708$$06-9-62A{\times}06-9-62A$ 계통은 고도의 저항성인 것으로 나타났다. 본 연구에서 선발된 토마토 잎마름역병에 고도의 저항성을 가지는 10계통은 저항성 품종육종을 위한 유용한 자원으로 활용될 수 있다.

CAPS Marker Linked to Tomato Hypocotyl Pigmentation

  • Tomato hypocotyl can generally be one of two colors, purple or green. Genetically, this trait is controlled by a single dominant gene. Hypocotyl tissue specific color expression is one of many visible genetic marker sources used to select tomato progeny. However, the visible marker does not show a clear distinction between homozygous genotype and heterozygous genotype from the breeding lines. Therefore, to identify a hypocotyl pigmentation related marker, we screened DNA polymorphisms in thirteen tomato lines showing purple or green hypocotyls. The markers used for screening consisted of primer set information obtained from anthocyanin related genes, conserved ortholog set II (COS II) marker sets localized near anthocyanin related genes, and restriction fragment length polymorphism (RFLP) markers localized near COS II markers, which produce polymorphisms between purple and green tomatoes. One primer from a RFLP fragment resulted in a polymorphism on agarose gel electrophoresis. From the RFLP fragment, a cleaved amplified polymorphic sequence (CAPS) marker was developed to distinguish between purple and green hypocotyls. The genotypes of 135 $F_2$ individuals were analyzed using the CAPS marker, and among them, 132 individuals corresponded to the phenotypes of hypocotyl pigmentation.
