Effects of Trifoliate Orange (Poncirus trifoliata) Extract on Inflammatory Responses in LPS-induced Shock Rats and RAW 264.7 Cells

  • This study examined the effects of trifoliate orange extract (TOE) on inflammatory reactions at the time of an LPS shock by performing experiments on rats injected with trifoliate orange extract and in Raw 264.7 cell cultures, with the aim of developing a new anti-inflammatory medicine. The IL-1β, IL-6, and TNF-α concentrations were lower in all of the groups treated with TOE than in the control group after 5 h of LPS treatment. The IL-10 concentration was higher in the 300-㎎/㎏ TOE group than in the control group after 2 h and 5 h of LPS treatment. The liver concentrations of cytokines IL-1β and IL-6 decreased more in the groups treated with TOE than in the control group and the IL-6 concentration did not differ significantly between the 100-㎎/㎏ TOE group than in the control group. The TNF-α and IL-10 concentrations did not differ significantly between the TOE groups and the control group. In the experiments involving Raw 264.7 macrophage cultures subjected to LPS shock, the productions of IL-1β, IL-6, and TNF-α decreased in all of the groups treated with TOE compared to the control group. The IL-10 concentration did not differ significantly between the groups treated with TOE and the control group. Together the findings of this study suggest that TOE contains functional substances that can influence inflammatory reactions.

The Effect of Foot Massage on Post operative Pain in Patients Following Abdominal Surgery (복부수술 환자의 수술후 통증에 미치는 발마사지의 효과)

  • More and more non-injured operations are being implemented these days, thanks to the development of medical technology. Still, however, most operations leave direct scars on patient' bodies, as well as accompanying pain. The massage as an independent nursing intervention can stimulate the circulation of the blood of tissue and muscle and increase the relationship between a patient and a nurse. The purpose of this study is to investigate the effect of foot massage on pain in post abdominal operative patients. The nonequivalent control group, pre-post test design is used for this study. From July 7, 2000 to February 20, 2001, the 40 patients who were operated under general anesthesia in a university hospital in Seoul were studied. They were divided into two groups ; 20 patients were part of the experimental group, and the others, in the control group. In order to evaluate the effect of foot massage, severity of pain was checked with the VAS (Visual Analog Scale) and also each patients' vital signs were measured with pulse rate, systolic blood pressure and diastolic blood pressure. The collected datas were processed by SAS version 6.12 program and analyzed by the Chi-square, Fisher's exact test, t-test and repeated measures ANOVA. The results of this study were as follows. 1. The severity of pain decreased significantly in the experimental group as compared to the control group following foot massage (t=-3.317, p= .002). 2. Measured vital signs in the experimental group had more reduction of that than in the control group following foot massage. - The pulse rate in the experimental group was lower than that in the control group following foot massage (F=7.73, p=.008). - The systolic blood pressure in the experimental group was lower than that in the control group following foot massage (F=25.75, p=.000). - The diastolic blood pressure in the experimental group was lower than that in the control group following foot massage (F=15.27, p=.000). In conclusion, foot massage is an effective dependent nursing intervention for pain control of post abdominal operative patients.

Effects of $\beta$-Carotene Supplementation on Lipid Peroxide Levels and Antioxidative Enzyme Activities in Diabetic Rats ($\beta$-Carotene 첨가식이가 당뇨쥐의 지질과산화물 수준과 항산화효소 활성에 미치는 영향)

  • This study investigated the effect of dietary $\beta$-carotene supplementation on lipid peroxidation and anti oxidative enzyme activity as indices of oxidative stress in diabetic rats. Fifty Sprague-Dawley male rats aging 7 weeks were used as experimental animals, which were divided into the non-diabetic control group and the diabetic group. The diabetic group received an intraperitoneal injection with streptozotocin to induce diabetes. Then the diabetic rats were divided into four dietary groups which contained different amounts of $\beta$-carotene; 0%, 0.002%, 0.02%, or 0.2% of the diet. The diabetic rats were fed the experimental diets and the non-diabetic rats were fed the basal diet without $\beta$-carotene supplementation for 2 weeks and then sacrificed. The diabetic group had a significantly higher blood glucose level than the non-diabetic group. However, blood glucose level were not significantly changed by the level of dietary $\beta$-carotene supplementation. Compared to the non-diabetic control group, the diabetic control group indicated a significant increase of plasma thiobarbituric acid reactive substance (TBARS). Liver TBARS level also tended to be higher in diabetic control group, although it was not significant. The $\beta$-carotene supplementation did not reduce plasma TBARS level. However, Liver TBARS level was significantly decreased when 0.02% or more $\beta$-carotene was supplemented in the diet. The liver lipofuscin level in the diabetic control group was higher than in the non-diabetic control group, but the effect of $\beta$-carotene supplementation did not show any differences. Superoxide dismutase activity was significantly lower in the diabetic group, but it was increased in groups receiving 0.02% or more $\beta$-carotene. Compared to the non-diabetic control group, lower activities of catalase and glutathione peroxidase were observed in the diabetic control group, although it was not significant. Catalase and glutathione peroxidase activities tended to increase as the levels of $\beta$-carotene supplementation increased, although it was not statistically significant. Therefore, it seems that dietary $\beta$-carotene supplementation might reduce diabetic complications by partly decreasing the lipid peroxidation and increasing the activity of antioxidative enzyme in diabetes.

  • To clarify the anesthetic effect of acupoint injection(aquapupuncture) using general anasthetics in dogs, 18 mongrel dogs were divided into control and two experimental groups(Tian-ping+Bai-hui : Tian-ping group and San-yang-luo+Gong-sun group : San -yang- lux group). Control group was intramuscularly injected with ketamine hydrochloride, 22 mg/ kg of body weight into the thigh and experimental groups were injected into each acupoint with half volume of dosage, respectively. Clinical findings(recumbency time, induction time of anesthesia, time of head lift and standing time) and changes of vital sign(temperature, heart rate and respiration rate) were investigates at pre-anesthesia, during anesthesia and poststanding, respectively. Preventive Effects on Porcine Epidemic Diarrhea(PED) Using by PEDV Antiserum II. Clinical Sign, Histopathological Lesion and Immunohistochemical Finding (항혈청 투여에 따른 돼지 유행성 설사병 예방효과 II. 임상증상, 병리조직학적 검사, 면역조직학적 검사)

  • The purpose of this study was to investigate to protective effects against porcine epidemic diarrhea virus (PEDV) infection in piglets by administration of the PEDV antiserum orally at 2 hrs, 24hrs and 36hrs after birth. six piglets administered the antiserum were experimentally infected with PEDV at five-day-old. Control group were four piglets infected with PEDV only. Clinical signs and gross, histopathological lesion and immunohistochemical findings were examined. The results obtained were as follows; 1. In clinical signs, piglets of the control group appeared the typical signs of severe watery diarrhea, depression and anorexia but piglets of the PEDV antiserum treated group recovered progressively. In mortality, control group showed 75%, but PEDV antiserum treated group showed 16.7%, respectively. 2. In gross findings, piglets of the control group appeared the typical findings of congestion, distension of lumen, containing curdes of undigested milk in stomach. But piglets of the PEDV antiserum treated group appeared milder than those of control group. 3. In histopathological findings, piglets of the control group appeared the typical findings of villous atrophy and fusion, congesion, exfoliation, vacuolation, squamation, loss of cilia and proliferation of crypt. But piglets of the PEDV antiserum treated group appeared milder than those of control group. 4. In immunohistochemical findings, piglets of the PEDV antiserum treated group showed more intensive in reaction for IgG and IgG than those of control group. The recation for IgA was stronger than that of IgG. It was concluded that oral administration of PEDV antiserum to piglets was effective in preventing PEDV infection and reduced their mortality.

Effects of Curculiginis Rhizoma on anti-thrombotic activity (선모(仙茅) 열수(熱水) 추출물(抽出物)의 항혈전(抗血栓) 효능 연구)

  • Objectives : An aim of study is to investigate effects of curculiginis rhizoma in vitro (factor Xa (FXa) inhibitor assay, prothrombinase assay, prothrombin time (PT) assay, activated partial thromboplastin time (aPTT) assay) and in vivo experiment (blood clotting time, thromboxane B2 content assay in serum and weight of thrombus by AV-shunt rat model). Methods : We gained a human serum and used serum in vitro study such as factor X activity (FXa) inhibition, prothrombinase inhibition, prothrombin time (PT) and activated partial thromboplastin time. Fifteen SD rats were divided into three groups (intact control group and two experimental group treated with extract of Curculiginis Rhizoma(ECR)). Rats were orally administrated DW (intact control group), 600 mg/kg concertration of ECR and 200 mg/kg concertration of ECR. After one hour, we anesthetized rats and made arteriovenous (AV) shunt rat models to study weights of thrombus, took a hole blood to study content of thromboxane B2 and blood clotting time. Results : In vitro, ECR increased a inhibitory activity of FXa, prothrombinase and aPTT compared than intact control group. Especially ECR made significant increase of FXa and prothrombinase inhibitory activity (p<0.05, p<0.01). And PT were increased in ECR control group compared with intact control group. In vivo, a blood clotting time of experiment group treated with ECR 600 mg/kg were significantly increased compared with that of intact control group (p<0.05) and content of thromboxane B2 was significantly decreased in group treated with ECR 600 mg/kg in seum. The weight of thrombus were significantly reduced in group treated with ECR 600 mg/kg compared with intact control group (p<0.05). But in vivo experiment study, those of group treated with ECR 200 mg/kg were reduced compared with those of intact control group without statistical significance. Conclusions : ECR has a antithromboic activity in internal course with inhibitory activity of FXa and prothrombinase in vitro, it required to research more study for effective compounds.

Effect of Medium and Long Chain Triglyceride Diet on the Serum Lipids of Rats Fed with Cholesterol (중쇄(中鎖)와 장쇄지방(長鎖脂肪) 식이(食餌)가 Cholesterol급여(給與) 흰쥐의 혈청지질(血淸脂質)에 미치는 영향(影響))

  • Because of its more complete hydrolysis and rapid absorption, MCT is expected its usefulness in the diets of patients with malabsorption syndrom. Also, several authors reported that serum cholesterol level was lower after MCT feedings. In this study, rate of each experimental group were fed for 4weeks with 20% MCT, 20% corn oil, mixed diet of 10% MCT and 10% corn oil, mixed diet of 17% corn oil and 3% shortening. After experimental diet, it was measured to growth rate and serum cholesterol, triglyceride and phospholipid level. The results were as follows; 1. The body weight gain was the lowest in MCT group and others were lower than control group. 2. Serum total cholesterol level was the lowest in corn oil group and others were significantly lower than control group. Free cholesterol level was lower in all experimental groups than control group too. 3. Serum HDL-cholesterol level was significantly higher in all experimental groups than control group. 4. The ratio of VLDL, LDL-Cholesterol to HDL-Cholesterol was significantly lower in all experimental groups than control group. 5. Serum triglyceride level was higher in all experimental groups than control group. Serum phospholipid level was significantly in only mixed diet group of corn oil and shortening than control group. 6. The ratio of total cholesterol to phospholipid was significantly lower in corn oil diet group than control group.

A Study on the Anti-oxidative Effect of Electroacupuncture at Yangreungcheon(GB34) in rats (양릉천 전침의 항산화 효과에 대한 실험적 연구)

  • Objectives : The purpose of this study is to observe the anti-oxidative effects of electroacupuncture at GB34(Yangreungcheon) in rats. Methods : The authors performed several experimental items including measurements of body weight, relative liver weight, levels of albumin, total bilirubin, LDL-cholesterol, LDH, GOT and GPT in blood serum, levels of SOD, glutathione, catalase, NO and MDA in liver, and histological analysis of liver. The conclusions are as follows. Results : 1. In the GB34-EA group, liver index was lower than those of the control group and the holder group. 2. In the GB34-EA group, the levels of albumin and total bilirubin in serum were increased significantly compared to those of the control and the holder group. LDL-cholesterol level was decreased significantly compared to those of the control group and the holder group. 3. In the GB34-EA group, liver SOD, glutathione, catalase activity were increased, and NO concentration in liver was decreased significantly compared to those of control and the holder group. 4. In the GB34-EA group, the density of liver tissue was more similarly maintained to that of the normal group compared to those of the control group, holder group and sham-EA group. Conclusions : These results suggest that electroacupuncture at GB34 may have an antioxidant effect in rat.

The Effect of Hyangbujapalmultang on The Serum Catecholamine Contents and Body Weight of Rats in Immobilization Stress (향부자팔물탕(香附子八物湯)이 구속(拘束) Stress 흰쥐의 체중(體重) 및 혈장(血漿) Catecholamines 함량(含量)에 미치는 영향(影響))

  • In order to study the anti-stress effect of hyangbujapalmultang, several measures of stress, including body weight changes, organ weight changes and catecholamine changes. The following result have been obtained: 1. The weight loss of contol group was $28.5{\pm}1.8g$, that of sample group was $20.3{\pm}1.6g$. This differance was statistcally significant. 2.The organ weight(Liver,Spleen,Kidney,Adrenal gland) was the only significant change in the spleen, in sample group comparing to control group. 3. The norepinephrine contents of control group was $695.5{\pm}22.7pg/ml$, that of sample group was $607.4{\pm}21.7pg/ml$. This shows significant difference in sample group comparing to contol group. 4. The epinephrine contents of control group was $212.8{\pm}9.8pg/ml$, that of sample group was $182.6{\pm}8.4pg/ml$. This shows significant differance in sample group comparing to control group. 5. The dopamine contents of control group was $504.5{\pm}31.3$, that of sample group was $463.4{\pm}27.8pg/ml$. This shows the value decreased, but none of it is significant. Based on avove results, it may be conculed that hyangbujapalmultang has anti-stress effects.

The Effects of Ozone Gas on Wound Healing in Skin (오존 가스가 피부창상의 치유에 미치는 영향)

  • This study was performed to determine the optimal dose of ozone for wound healing. Twenty Korean black goats were allocated to 4 groups. The skin wound with $1{\times}1cm$ was induced over the hoof. For 5 minutes per day, 0.1 ppm (Group I), 1 ppm (Group II) and 6 ppm (Group III) ozone gas was applied. In control group, 5 ml of normal saline was applied daily on the wound. Wound square was determined using color image analyzer at day 0, 4 and 14. Soft tissue depth of wound was determined using radiograph. Before the application of ozone gas, $1{\times}1cm$ of skin tissue was resected and observed for histopathologic findings. To determine the healing effect, $2{\times}1cm$ of wound tissue was resected and observed for histopathologic findings at day 14. In mean squares of wound, all experimental groups revealed more reduced mean squares than control group at day 4, and especially Group II (p<0.05) and Group III (p<0.05) revealed significant reduction. At day 14, Group I and Group II (p<0.05) revealed more reduced mean squares than control group, but Group III revealed less reduced mean squares than control group. In soft tissue depth of wound, all experimental groups and control group revealed swelling at day 4, compared with day 0. At day 14, all experimental groups and control group revealed reduction, compared with day 4, and especially Group II (p<0.05) revealed significant reduction. In histopathological findings, inflammation findings in epidermis and dermis, hemorrhagic finding in epidermis were revealed in control group. In Group I, inflammation finding was reduced, compared with control group. Formation of scab and many reepithelialization of epidermis were revealed. In Group II, inflammation findings in epidermis and dermis which were observed in control group were not found, normal skin tissue was revealed. In Group III, inflammation findings in epidermis and dermis were revealed, reepithelialization of epidermis was not founded. Slow healing process was observed, compared with Group I and II. It was concluded that Group II has excellent promotion effect of wound healing.
