Effects of Culture Condition on Solubilization of Coal by Microorganisms (배양 조건의 변화가 미생물에 의한 석탄의 액화에 미치는 영향)

  • Biosolubilization of an Australian lignite was investigated by using Streptomyces viridosporus and Poria cocos. In order to solubilize coals effectively they were pretreated by nitric acid both in surface and liquid cultures. The optimum growth pH was 7.5 for S. viridosporus and 4.5 for P. cocos. The effects of various carbon, nitrogen and metal sources on overall solubilization were also studied. Solubility increased with the addition of urea for S. viridosporus, and peptone and tryptone for P. cocos. However carbon and metal sources had little or negative effects on solubilization. Maximum amount of coal solubilized was 85%(w/w) in a batch fermentation culture. Extracellular materials produced by micro-organism were found to be responsible for the coal solubilization. Approximately 70 to 80% of coal solubilization was determined to be the result of non-enzymatic reactions, and the rest to be the result of enzymatic reactions. Characteristics of the solubilized coal were compared with those of original coal and pretreated coal by the approximate and ultimate composition analysis, and IR-spectrum analysis. The spectroscopic results showed that the mechanism of coal solubilization was caused by continuous oxidation.

Production of 10-deacetylbaccatin III in Taxus cuspidata Suspension Cell Culture (주목 현탁세포배양을 이용한 10-deacetylbaccatin III 생산)

  • In this study, enhanced production of 10-deacetylbaccatin III(10-DAB), a precursor of taxol in semisynthesis, was investigated in Taxus cuspidata cell suspension cultures. The effects of initial inoculum size and sugar concentration were examined to prove the relationship between the production of 10-DAB and cell growth. The cell growth was found to be stimulated in Schenk and Hildebrandt(SH) medium. The lower the inoculum size as well as initial sugar concentration, the faster the cell growth rate. When the initial sugar concentration was dept low, the production of 10-DAB into medium was increased. By using perfusion culture, continuous cell growth was possible until the end of culture and more than 34.67 g/L of cell concentration could by obtained. This is about 2.5 times higher level than that of control batch culture.

Plant Regeneration from Protoplasts of Suspension Cultured Cells in Arabidopsis thaliana (애기장대(Arabidopsis thaliana) 현탁배양세포의 원형질체로부터 식물체 재분화)

  • Protoplasts of Arabidopsis thaliana were easily isolated from the shoot-forming (SF) suspension-cultured cell clusters with 4 hours-shaking condition (40 rpm) on CPD enzyme solution containing 1% cellulase R-10, 0.25% pectolyase Y-23 and 0.5% driselase. Protoplasts were cultured on liquid KAO medium supplemented with 1 mg/L 2,4-D, 0.5 mg/L kinetin, 200 mg/L spermidine and 68 g/L glucose. Also, protoplasts were cultured on 0.2 $\mu$M membrane filter placed onto CP solid medium containing the suspension cells as feeder cells in the dark at $25^{\circ}C$ for 4 weeks. Protoplast-derived-SF calli were cultured on MS medium containing 0.05 mg/L IAA, 7 mg/L 2 ip and 30 g/L sucrose under the continuous illumination for four weeks. The frequency of shoot formation was about 60%. The regenerants were transferred into potting soil to grow mature plants. The regenerants formed the silques with seeds after 8 weeks of cultures.

Genetic Analysis of the Ability of Callus Formation and Plant Regeneration in Seed Culture of Rice (벼의 종자배양에서 캘러스 형성과 식물체 재분화 능력의 유전)

  • This study was conducted to determine the inheritance of the ability of callus formation and plant regeneration in seed cultures of rice. The culturabilities of three Japonica rices,'Chucheongbyeo', 'Nagdongbyeo', and 'Daeribbyeo 1', were higher than those of Tongil type cultivars, 'Milyang 23' and 'Samgangbyeo 'The frequency for callus growth in F₂ populations of the three crosses, 'Milyang 23/Chucheongbyeo', 'Milyang 23/Daeribbyeo 1', and' Samgangbyeo/Nagdongbyeo', revealed a nearly normal distribution. The broad-sense heritability estimated from the ability of callus formation in the crosses were ranged from 83.8% to 90.1%. The frequency distribution of plant regenerability in F₂ population of 'Milyang 23/Daeribbyeo 1' showed a continuous variation. But the segregation mode of plant regenerability from seed-derived callus in the F₂ progenies of 'Milyang 23/Chucheongbyeo' and 'Samgangbyeo/Nagdongbyeo' appeared to fit the expected 3 : 1 ratio for the high and low regenerability. These results suggest that the high plant regenerability of 'Chucheongbyeo' and 'Nagdongbyeo' was regulated by a single dominant gene.

  • Performance art is an integrated genre of different art areas, and make-up is a visual art area to give lively effects to the performance art, which makes actors and actresses changed into the characters of plays. It can be said to be a kind of deliverer of idea to help effectively understand the contents of the performance. When we are in the world abundant in various cultures and innumerable styles of expression of different countries, we should rediscover the value of the beauty of our traditional masks that have represented the emotion and thought of Han nation, which we can find in play culture of our ancestors that has kept our ancestors' value system. That's the way we can get continuous life force and competitive power of our traditional culture. The Korean traditional masks haven't only been for the prevention of bad luck and the prayer of fortune, but also well expressed Korean people's looks, features and personalities according to the roles. They are excellent artistic works in themselves, and many studies have been performed on the analyses and uses of the features of the masks in various fields due to their good expression of different cultural aspects, while the studies in the light of make-up are rarely performed. Therefore, this article aims to analyze the features of the Korean traditional masks and, by using this analysis, to present effective make-up designs in outdoor performances through a stage performance. In addition, this article also tries to present the orientation of make-up as an expression of artistic image, and to expand the expression area of make-up by using the Korean traditional masks in the light of make-up. For this purpose, I examined the related literature and the precedent study materials to find the origin and kinds of the Korean traditional masks, and analyzed the features of the masks. Furthermore, I established the make-up design plan using the material features of the Korean traditional masks by analyzing the characters in actual performance works, and presented the photos of works actually performed.

Evaluation of Negative Results of BacT/Alert 3D Automated Blood Culture System

  • Although automated continuous-monitoring blood culture systems are both rapid and sensitive, false-positive and false-negative results still occur. The objective of this study, then, was to evaluate negative results occurring with BacT/Alert 3D blood culture systems. A total of 1032 samples were cultured with the BacT/Alert 3D automated blood culture system, using both aerobic (BPA) and anaerobic (BPN) media, and 128 of these samples yielded positive results. A total of 904 negative blood samples were then subcultured in $5\%$ sheep blood agar, eosin methylene blue, chocolate agar, and sabouraud-dextrose agar. Organisms growing on these subcultures were subsequently identified using both Vitek32 (bioMerieux, Durham, NC) and conventional methods. Twenty four $(2.6\%)$ of the 904 subcultures grew on the subculture media. The majority $(83.3\%)$ of these were determined to be gram-positive microorganisms. Fourteen $(58.3\%)$ were coagulase-negative staphylococci, two $(8.3\%)$ were Bacillus spp., one $(4.2\%)$ was Staphylococcus aureus, and one $(4.2\%)$ was identified as Enterococcus faecium. Streptococcus pneumoniae and Neisseria spp. were isolated together in two $(8.3\%)$ vials. Gram-negative microorganisms comprised $12.5\%$ of the subcultures, of which two $(8.3\%)$ were found to be Pseudomonas aeruginosa, and one $(4.2\%)$ was Pseudomonas fluorescens. The other isolate $(4.2\%)$ was identified as Candida albicans. We conclude that the subculture of negative results is valuable in the BacT/Alert 3D system, especially in situations in which only one set of blood cultures is taken.

Optimum Salinity and Temperature Condition for Mass Culture of the Brackish Water Flea, Diaphanosoma celebensis (기수산 물벼룩, Diaphanosoma celebensis의 대량배양을 위한 최적 염분 및 수온 조건)

  • This study investigated the optimum salinity and temperature conditions for mass culture of the brackish water flea, Diaphanosoma celebensis. Community and individual cultures of flea were maintained in 1 L beakers and 3 mL vessels (of a 12-well culture plate), respectively, and fed green algae, Tetraselmis suecica. In salinity experiments ranging from 5 to 34 psu, continuous growth of flea populations was found up to 34 psu. However, the specific growth rate and life span of females showed decreasing tendencies with the increase of salinity. The highest maximum density and offspring number were 33.6 individuals (ind.)/mL and 55.3 ind. at 10 psu, respectively. In the temperature experiments ranging from 20 to $40^{\circ}C$, population growth of D. celebensis increased continuously until $35^{\circ}C$ and then decreased over $40^{\circ}C$. The specific growth rate was significantly higher at 25 and $30^{\circ}C$ than at 20 and $40^{\circ}C$. Female life span tended to decrease with temperature increase. The highest maximum density and offspring number were 52.3 ind./mL and 46.0 ind. at $30^{\circ}C$, respectively. These results suggest that the optimum salinity and temperature for mass culture of D. celebensis may be 10 psu and $30^{\circ}C$, respectively.

Negative Pressure Wound Therapy of Chronically Infected Wounds Using 1% Acetic Acid Irrigation

  • Background Negative-pressure wound therapy (NPWT) induces angiogenesis and collagen synthesis to promote tissue healing. Although acetic acid soaks normalize alkali wound conditions to raise tissue oxygen saturation and deconstruct the biofilms of chronic wounds, frequent dressing changes are required. Methods Combined use of NPWT and acetic acid irrigation was assessed in the treatment of chronic wounds, instilling acetic acid solution (1%) beneath polyurethane membranes twice daily for three weeks under continuous pressure (125 mm Hg). Clinical photographs, pH levels, cultures, and debrided fragments of wounds were obtained pre- and posttreatment. Tissue immunostaining (CD31, Ki-67, and CD45) and reverse transcription-polymerase chain reaction (vascular endothelial growth factor [VEGF], vascular endothelial growth factor receptor [VEGFR]; procollagen; hypoxia-inducible factor 1 alpha [HIF-1-alpha]; matrix metalloproteinase [MMP]-1,-3,-9; and tissue inhibitor of metalloproteinase [TIMP]) were also performed. Results Wound sizes tended to diminish with the combined therapy, accompanied by drops in wound pH (weakly acidic or neutral) and less evidence of infection. CD31 and Ki-67 immunostaining increased (P<0.05) post-treatment, as did the levels of VEGFR, procollagen, and MMP-1 (P<0.05), whereas the VEGF, HIF-1-alpha, and MMP-9/TIMP levels declined (P<0.05). Conclusions By combining acetic acid irrigation with negative-pressure dressings, both the pH and the size of chronic wounds can be reduced and infections be controlled. This approach may enhance angiogenesis and collagen synthesis in wounds, restoring the extracellular matrix.

On Ethnic Images shown in Japanese Designers' Collections - Focused on Design Comparison between Issey Miyake and Yohji Yamamoto (일본 디자이너 컬렉션에 나타난 에스닉 이미지 - 이세이 미야케와 요지 야마모토의 디자인 비교를 중심으로)

  • The globalization phenomenon of the 21st century has acted as the catalyst to accept diversity, and a new cultural code, ethnic, has emerged in the modem society by the pursuit of diversity throughout the whole society and culture. Unlike preceding studies focusing on ethnic concepts and design development, this study attempted comparative analyses on ethnic trends shown in the collections of two designers, Issey Miyake and Yohji Yamamoto, who have strong ethnic consciousness. It is considered the comparative analysis on the two designers' collections with ethic images will be a guide to indicate the fashion philosophy of the two designers in the category of Japan, and this will be useful as basic data for the establishment of the globalization identity which is needed in the future fashion industry. The study results are as follows. First, it was found that the ethnic code has been so widely accepted by the world designers in a very positive form to accept foreign cultures that the ethnic code is now showing an aspect of eclecticism. Second, designer Issey Miyake has been pursuing his own ethnic style based on his philosophy to liberate humans through continuous researches and efforts on clothes. Third, Yohji Yamamoto has been pursuing a Japanese ethnic style as a designer who has expressed the unique beauty of Japan from the characteristics of Japanese traditional clothes. Fourth, as a result of comparative analyses on collections, they both have pursued an ethnic style based on the unique national characteristics of Japan, but it was also found that their fashion philosophy has developed differently in the same category of ethnic trend.

  • The purpose of this study is to develop the elementary 'after school' smart music education program using smart devices in connection with elementary school curriculum of 2015 music and education. The research method was based on the data related to the after school education program and the music-related contents of the current elementary school curriculum. And a smart music education program was developed by applying the contents of elementary school music curriculum to practical music using smart devices. The study result suggests three programs with 16 times. Through this program, we can expect diverse, systematic and continuous music education rather than one-sided or one-off music education which are provided by the existing public education courses. Through the field experience, students will be able to directly experience arts and cultures as well as to awaken their potential artistry and creativity. In addition, it is expected that it will help to reduce the economic burden by reducing the burden of private education expenses through the after school program and to realize the qualitative growth of music education in schools.
