Immunomodulatory Effects of Aureobasidium pullulans SM-2001 Exopolymers on Cyclophosphamide-Treated Mice

  • The immunomodulatory effects of Aureohasidium pullulans SM-2001 exopolymers containing $\beta$-1,3/1,6-glucan were evaluated in cyclophosphamide (CPA)-treated mice. To induce immunosuppression, 150 and 110 mg/kg of CPA were intraperitoneally injected 3 days and 1 day, respectively, before beginning administration of the test material. Exopolymers were delivered subcutaneously or orally, four times, in a volume of 10 ml/kg at 12-h intervals beginning 24 h after the second CPA treatment. Changes in thymus and spleen weights, splenic amounts of tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-$1{\beta}$, and IL-10, and numbers of CD3+, CD4+, CD8+, and TNF-$\alpha+$ thymus and spleen cells were monitored in CPA-treated mice. As a result of CPA treatment, dramatic decreases in the number of CD3+, CD4+, CD8+, and TNF-$\alpha+$ cells were detected in the thymus and spleen, along with decreases in thymus and spleen weights. In addition, splenic TNF-$\alpha$, IL-$1{\beta}$, and IL-10 contents were also decreased on observation with flow cytometry. However, oral and subcutaneous treatments with exopolymers effectively reduced the immunosuppressive changes induced by CPA. Therefore, it is concluded that exopolymers of A. pullulans SM-2001 can effectively prevent immunosuppression through, at least partially, the recruitment of T cells and TNF-$\alpha+$ cells or enhancement of their activity, and can provide an effective component of prevention or treatment regimens for immunosuppression related to cancer, sepsis, and high-dose chemotherapy or radiotherapy.

폐경 후 여성의 골밀도에 따른 영양소 섭취상태와 혈청 구리, 아연, 망간 함량에 관한 연구 (A Study on Nutrient Intakes and Serum Levels of Copper, Zinc and Manganese in Korean Postmenopausal Women with Different Bone Mineral Density)

  • The purpose of this study was to investigate the relationships among nutrient intakes, BMD, serum copper, zinc and manganese contents in Korean postmenopausal women residing in Seoul and Kyunggi-do. Nutrient intakes analyses and anthropometric measurements were performed for 64 postmenopausal women. Also, fasting blood samples were collected. The BMDs of the lumbar spine (L2-L4) and femoral neck were measured by dual energy X-ray absorptiometry (DEXA). Subjects were classified into three groups: osteoporosis (T-score ${\leq}$ -2.5, n = 20), osteopenia (-2.5 < T-score ${\leq}$ -1, n = 24), and normal (T-score) -1, n = 20). The results are summarized as follows; the mean age, height and weight were 62.1 yrs, 153.8 em, and 56.1 kg, respectively, with no significant difference among three groups. On the other hand, the mean BMI $(23.7\;g/m^2)$ of normal group was significantly higher than those of other two groups. The mean energy intake was 1,395.3 kcal (77.5% of Dietary referance intakes for koreans (KDRls)). The average daily intakes of vitamin $B_2$, calcium and zinc did not reach KDRIs. The mean intakes of iron, copper, zinc and manganese were 10.9 mg, 1.6 mg, 7.8 mg and 5.1 mg respectively, with no significant difference among three groups, The mean serum levels of copper, zinc and manganese were 96.4 mg/dl, 91.5 mg/dl and 39.5 ng/dl, respectively with no significant differences among three groups. The BMD of lumbar spine showed a significantly negative correlation intakes of animal protein, fat and iron intakes after adjusting for age, Bill. However, the BMD of lumbar spine was not significantly correlated with animal iron intakes after adjusting for animal protein, and fat intakes, indicating animal food intake is a possible confounding factor. Proper intakes of vitamin $B_2$, vitamin C are required to protect osteoporosis in postmenopausal women. These results suggest that excessive animal food intakes may have adverse effect on bone mineral density. Further studies arc necessary to elucidate the role of serum minerals in BMD.

Effect of the Inhibition of PLA2 on Oxidative Lung Injury Induced by $Interleukin-1{\alpha}$

  • In order to understand the pathogenetic mechanism of adult respiratory distress syndrome (ARDS), the role of phospholipase A2 (PLA2) in association with oxidative stress was investigated in rats. $Interleukin-1{\alpha}\;(IL-1,\;50\;{\mu}g/rat)$ was used to induce acute lung injury by neutrophilic respiratory burst. Five hours after IL-1 insufflation into trachea, microvascular integrity was disrupted, and protein leakage into the alveolar lumen was followed. An infiltration of neutrophils was clearly observed after IL-1 treatment. It was the origin of the generation of oxygen radicals causing oxidative stress in the lung. IL-1 increased tumor necrosis factor (TNF) and cytokine-induced neutrophil chemoattractant (CINC) in the bronchoalveolar lavage fluid, but mepacrine, a PLA2 inhibitor, did not change the levels of these cytokines. Although IL-1 increased PLA2 activity time-dependently, mepacrine inhibited the activity almost completely. Activation of PLA2 elevated leukotriene C4 and B4 (LTC4 and LTB4), and 6-keto-prostaglandin $F2{\alpha}\;(6-keto-PGF2{\alpha})$ was consumed completely by respiratory burst induced by IL-1. Mepacrine did not alter these changes in the contents of lipid mediators. To estimate the functional changes of alveolar barrier during the oxidative stress, quantitative changes of pulmonary surfactant, activity of gamma glutamyltransferase (GGT), and ultrastructural changes were examined. IL-1 increased the level of phospholipid in the bronchoalveolar lavage (BAL) fluid, which seemed to be caused by abnormal, pathological release of lamellar bodies into the alveolar lumen. Mepacrine recovered the amount of surfactant up to control level. IL-1 decreased GGT activity, while mepacrine restored it. In ultrastructural study, when treated with IL-1, marked necroses of endothelial cells and type II pneumocytes were observed, while mepacrine inhibited these pathological changes. In histochemical electron microscopy, increased generation of oxidants was identified around neutrophils and in the cytoplasm of type II pneumocytes. Mepacrine reduced the generation of oxidants in the tissue produced by neutrophilic respiratory burst. In immunoelectron microscopic study, PLA2 was identified in the cytoplasm of the type II pneumocytes after IL-1 treatment, but mepacrine diminished PLA2 particles in the cytoplasm of the type II pneumocyte. Based on these experimental results, it is suggested that PLA2 plays a pivotal role in inducing acute lung injury mediated by IL-1 through the oxidative stress by neutrophils. By causing endothelial damage, functional changes of pulmonary surfactant and alveolar type I pneumocyte, oxidative stress disrupts microvascular integrity and alveolar barrier.

아파트 공사에서의 건설기술자 역량 분석 연구 - 공무업무를 중심으로 - (Analyzing the Competence of Construction Engineers in Apartment Construction)

  • 국내 건설 산업에서 국내 상위 10위권의 건설 회사들이 대규모 주택 건설 프로젝트를 진행하고 있으며, 건설기술자의 역량이 프로젝트의 성공을 위한 요소라고 말하고 있다. 우리나라도 개인의 능력을 중요시함에 따라 산업현장에서 직무를 수행하기 위해 요구되는 지식 기술 태도 등의 내용을 국가 차원에서 체계화 한 국가직무능력표준(NCS, National Competency Standards)이 있지만, 건설현장구성원 각자의 구체적인 역량 수준에 대한 분석이 이루어지지 않은 상태이다. 본 연구에서는 건설현장의 대표 업무 중 하나인 공무업무를 중심으로 하여 세부역량을 정의하고, AHP 분석방법과 분산분석(ANOVA)을 사용하여 건설기술자들이 현재 보유하고 있는 역량 수준과 이들이 생각하는 세부역량의 중요 정도를 알아보고자 했다. 그 결과, 아파트 공사현장 기술자들의 현재 보유 역량이 그들이 생각하는 역량의 중요 정도보다 미흡한 것으로 나타났으며 근무경력에 따라, 직급에 따라 보유하고 있는 역량이 다른 것을 알 수 있었다. 이는 경력이 쌓이면 자연스럽게 보유하고 있는 역량이 달라진다는 뜻으로 근무경력이 짧고, 직급이 낮은 건설기술자들의 역량을 향상시킬 방안이 필요할 것으로 판단된다.

페노피브레이트 유연물질 표준품 대체시험법 개발 (Development of an Alternative Analytical Method without Related Substance Reference Standards for Fenofibrate in Korean Pharmacopoeia)

  • Analytical method for related substances can be categorized into two methods depending on the necessity of reference standard (RS). The analytical method of related substances with RS is fast and accurate, but it's very expensive and technically difficult to synthesize RS due to their complicated structure. Another method is using relative retention time (RRT) and relative response factor (RRF) which are already validated with RS. Validation of this method is not easy and time consuming, but once it has been developed, it can save cost and time. In this study, we developed the analytical method for related substances of fenofibrate using RRT and RRF. We validated the method by evaluating specificity, linearity, accuracy and precision according to the "Manual for Guideline Application for Validation of Analytical Procedures" of MFDS. Also, we calculated RRT and RRF between fenofibrate and fenofibrate related substances. The results of this study showed high specificity for fenofibrate and fenofibrate related substances. Correlation coefficient(r) of all substances were more than 0.99, and the recovery of fenofibrate, fenofibrate related substance I, II and III were 99.44%, 100.84%, 99.14% and 101.58%, respectively. Precision of fenofibrate and its related substances were ranged between RSD 0.29% and 0.93%. Quantification limits of fenofibrate, fenofibrate related substance I, II and III were determined to be $0.03{\mu}g/ml$, $0.05{\mu}g/ml$, $0.04{\mu}g/ml$ and $0.02{\mu}g/ml$, respectively by confirming signal to noise ratio of each chromatogram. The RRT for fenofibrate related substance I, II and III were determined to be 0.35, 0.41 and 1.34, respectively. Also, the RRF for fenofibrate related substance I, II and III were determined to be 1.28, 0.98 and 0.79, respectively. The developed method was applied to determine contents for fenofibrate related substances in commercial fenofibrate (active pharmaceutical ingredient). As a result, developed analytical methods of related substances will be used for revising the monograph of fenofibrate in Korean Pharmacopoeia revision and contribute quality control of drugs by improving cost and time consuming problem of RS.

${\beta}-amylase$와 transglucosidase의 처리가 홍삼 extract의 품질에 미치는 영향 (Effects of ${\beta}-amylase$ and Transglucosidase on the Qualities of Red Ginseng Extract)

  • 홍삼 exoact를 첨가한 액상 제품에서의 침전 생성을 감소시키기 위하여 홍삼 exoact에 ${\beta}-amylase$와 transglucosidase를 처리한 후 품질 특성의 변화를 조사하였다. 이소말토올리고당인 isomatose 가 $5.25\%$ 생성되었고 glucose의 함량이 증가하였으며, ginsenoside는 $Rb_1$$Rb_2$가 감소하였고 Rd의 함량이 증가하였다. 효소처리 한 홍삼 exDact는 무처리구에 비하여 Bifdus속균과, Lactobacizlus속균과 같은 장내유용세균에 대한 증식효과가 있었으며 Bifdus속균은 Lactobacizlus속균보다 대체로 흥삼 extract 첨가에 의한 생육도가 낮았다. 효소처리한 홍삼 extract는 단맛과 신맛이 증가하였으나 홍삼 extrac의 좋지 않은 맛으로 평가되는 쓴맛과 떫은맛이 감소하여 향미가 개선되었다. 효소처리 한 홍삼 extract를 첨가하여 $pH\;3.0\~4.5$의 음료를 제조한 다음 $40^{\circ}C$에서 1개월간 저장하였을 때 무처리구에 비하여 침전량이 현저히 감소하였으나 pH 3.0에서는 효소처리 한 홍삼 extract첨가구에서 $17.7\%$, 무처리구에서 $30.3\%$의 침전이 형성되었다. 따라서 인삼 음료의 침전을 감소시키기 위하여는 홍삼 exoact에 P-amylase와 transglucosidase를 처리하고 동시에 음료의 pH를 3.5 이상으로 유지하는 것이 요구되었다.

Anticancer and Immunopotentiating Activities of Crude Polysaccharides from Pleurotus nebrodensis on Mouse Sarcoma 180

  • Pleurotus nebrodensis is an edible and commercially available mushroom in Korea. This study was conducted in order to evaluate the anticancer and immunopotentiating activities of crude polysaccharides, extracted in methanol, neutral saline, and hot water (hereafter referred to as Fr. MeOH, Fr. NaCl, and Fr. HW, respectively) from the fruiting bodies of P. nebrodensis. ${\beta}$-Glucan and protein contents in Fr. MeOH, Fr. NaCl, and Fr. HW extracts of P. nebrodensis ranged from 23.79~36.63 g/100 g and 4.45~6.12 g/100 g, respectively. Crude polysaccharides were not cytotoxic against sarcoma 180, HT-29, NIH3T3, and RAW 264.7 cell lines at a range of $10{\sim}2,000{\mu}g/mL$. Intraperitoneal injection with crude polysaccharides resulted in a life prolongation effect of 11.76~27.06% in mice previously inoculated with sarcoma 180. Treatment with Fr. NaCl resulted in an increase in the numbers of spleen cells by 1.49 fold at the concentration of $50{\mu}g/mL$, compared with control. Fr. HW improved the immuno-potentiating activity of B lymphocytes through an increase in alkaline phosphatase activity by 1.65 fold, compared with control at $200{\mu}g/mL$. Maximum production of nitric oxide ($14.3{\mu}M$) was recorded in the Fr. NaCl fraction at $200{\mu}g/mL$. Production of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), and interleukin-6 (IL-6) was significantly higher, compared to control, and IL-6 production was highest, in contrast to TNF-${\alpha}$, IL-$1{\beta}$, and positive control, concanavalin at the tested concentration of the various fractions. Results of the current study suggest that polysaccharides extracted from P. nebrodensis have a strong anticancer effect and may be useful as an ingredient of biopharmaceutical products for treatment of cancer.

경남지역 밭 토양 화학성분이 미생물 생태에 미치는 영향 (Impacts of Chemical Properties on Microbial Population from Upland Soils in Gyeongnam Province)

  • 경남지역 밭 토양 25개소를 대상으로 2009년에 토양 화학성분과 미생물 다양성을 검토하고 토성, 지형 및 작물별 주요 변동요인을 주성분분석으로 해석하였다. 토양 유효인산 및 치환성 칼륨과 칼슘 함량은 적정수준 보다 높았으며 대부분의 토양 화학성분이 최소값과 최대값의 차이가 크게 나타났다. 지형적으로 산록경사지는 토양 유기물 함량과 치환성 칼륨 함량이 유의적으로 높았고 (p<0.05) 토성별 유기물 함량은 양토에서 높은 반면에 염류농도와 유효인산 함량은 사양토에서 높았으며 고구마 재배지의 치환성 칼슘함량은 감자 재배지에 비해 유의적으로 높았다. 지형별 밭토양의 곰팡이 개체수는 곡간 및 선상지가 하성평탄지 보다 유의적으로 많았고 토양 미생물체량과 탈수소효소 활성은 산록경사지가 다른 지형에 비해 유의적으로 높았다 (p<005). 토성에 따른 토양의 coliform 그룹은 미사질양토가 사양토와 양토에 비해 유의적으로 높았고 미생물체량은 양토가 미사질양토 보다 유의적으로 높았으나 (p<005) 작물별 미생물상과 토양 미생물체량 및 탈수소효소 활성은 유의적인 차이가 없었다. 주성분 분석결과 제1주성분이 32.2%, 제2주성분이 24.2%로서 전체 56.4%의 자료를 설명할 수 있었으며 제1주성분은 토양의 유효인산 함량이 가장 크게 기여하였다. 토양 바실러스균과 곰팡이균은 토양의 유기물 함량과 정의 상관관계를 보였으며 토양 pH는 형광성 슈도 모나스균과 정의 상관관계를 나타냈다.

흰목이버섯 추출물의 미백 및 주름개선 효과 (Whitening and Anti-Wrinkle Effects of Tremella Fuciformis Extracts)

  • Background : The white jelly mushroom (Tremella fuciformis), one of the most popular edible fungi, has medicinal properties. However, the effects of T. fuciformis in skin whitening or anti-wrinkle efficacy has not been defined to date. The aim of the present study was to investigate the effects of T. fuciformis extracts on whitening and anti-wrinkle efficacy in skin cells. Methods and Results :We prepared T. fuciformis extracts with water. The extracts ($80^{\circ}C$) contained 12.11 mg/g polyphenol and 8.54 mg/g flavonoid concentration. T. fuciformis extracts markedly decreased melanin contents and tyrosinase activity in ${\alpha}$-MSH-stimulated melanocytes (B16F10 cells). In addition, the mRNA expression of melanin formation factors, such as microphthalmia-associated transcription factor (MITF), tyrosinase-related protein-1 (TRP-1) and tyrosinase-related protein-2 (TRP-2) were significantly down-regulated in ${\alpha}$-MSH-stimulated melanocyte. Furthermore, T. fuciformis extracts increased the synthesis of type I procollagen and reduced mRNA expression of matrix metalloproteinase 1 (MMP-1) in the human dermal fibroblast (HDFn cells). These data indicated that T. fuciformis extracts induce repression of cellular melanogenesis and protect against wrinkles caused by UVB-stimulated damage. Conclusions : Thus T. fuciformis extracts could be a cosmetic candidate for skin whitening and anti-wrinkle effects.

육아지원기관의 특별활동 이용 현황 및 지출 결정요인 분석 (An Analysis of Extra-Curricular Activities in Childcare Facilities and the Factors Affecting on Expenses of Extra-Curricular Activities)

  • 본 연구는 정부의 보육료 지원 증가에도 불구하고, 가구에서 체감하는 교육 및 보육료 부담이 감소되지 못한다는 문제점에서 비롯하여 추가비용으로 지출되는 특별활동 비용에 미치는 요인을 살펴보는 것을 목적으로 하였다. 한국아동패널 4차년도 자료를 사용하여 분석한 결과, 기관 내 특별활동 여부는 있음이 68.5%였으며, 평균적으로 2.9개의 특별활동을 이용하는 것으로 나타났다. 월평균 지출비용은 약 50천원이었다. 지역에 따라 특별활동 이용개수 및 지출비용을 살펴본 결과 지역 간의 차이는 통계적으로 유의하였다. 변수를 단계적으로 투입하여 특별활동 비용 지출에 영향을 미치는 요인을 분석한 결과, 모 학력, 모 취업, 육아지원기관 유형, 지역변인이 유의한 것으로 나타났으며, 이중 지역변인이 가장 큰 설명량을 차지하였다. 연구결과를 바탕으로 몇 가지의 정책적 시사점을 도출하였다. 첫째, 특별활동 과목 내용 및 비용에 대한 구체적인 실태파악과 관리감독이 이루어져야 한다. 둘째, 중앙정부차원에서의 필요경비에 대한 구체적인 기준제시가 필요하다. 셋째, 어린이집뿐만 아니라 유치원 등 아동이 이용하는 육아지원기관 전반에 걸쳐 특별활동 과목, 강사, 비용, 강사의 경력 등을 공개하고 주기적으로 모니터링해야 한다. 넷째, 취약계층 아동에 대한 특별활동 지원이 요구된다.
