Thin-film optical waveguide $K^{+}$-ion sensor using the evanescent field absorption (소산장 흡수를 이용한 박막 광도파로형 칼륨이온센서)

  • A thin film optical waveguide sensor has been developed to measure and analyze quantitatively some inherent optical properties of biochemical substances. In this paper, two different kinds of thickness of thin film waveguide were prepared by RF sputtering of Corning-7059 glass(n = 1.588 at ${\lambda}=\;514nm$, Ar laser) on Pyrex glass substrates. We made a sensing membrane coated on the thin film waveguide with the poly(vinyl chloride-co-vinyl acetate-co-vinyl alcohol) (91 : 3 : 6) copolymer membrane based on $H^{+}$-selective chromoionophore and $K^{+}$-selective neutral ionophore and then proposed the thin film opptical waveguide ion sensor which can select a potassium ion. This sensor based ell the absorbance change by utilizing chromoionophore and neutral ionophore, which changes their absorption spectrum in the UV-vis region upon complexation of the corresponding ionic species, have been reported. The sensitivity dependence of the proposed sensor on interaction length, waveguide thickness, and content of a chromoionophore was investigated. This sensor has the measurement range of $10^{-6}M{\sim}1M$ for $K^{+}$ concentration and 90% response time of duration within 1 min. Also, our thin film optical waveguide sensor using the evanescent field was investigated as compared with conventional transmission sensor or optode sensor by the optical fiber. The sensitivity of thin-film waveguide $K^{+}$ sensor is higher than that of the conventional transmission sensor. The proposed sensor is expected to be useful to biochemical, medical, environmental inspection and so on.

Flavonoid Component Changes and Antioxidant Activities of Fermented Citrus grandis Osbeck Peel (당유자 과피 발효물의 플라보노이드 성분 변화 및 항산화 활성)

  • In this study, we investigated the change of antioxidant activity and flavonoid contents by fermentation of Citrus grandis Osbeck peel (CGP) using the Saccharomyces cerevisiae (KCCM35053), comparing to unfermented CGP. Total flavonoid content in the fermented Citrus grandis Osbeck peel (FCGP) was 3,768 g/100 g sample and higher than that of CGP. The antioxidant activities of FCGP was determined by DPPH, hydroxyl, alkyl radicals, and hydrogen peroxide scavenging assays. FCGP showed higher activities than CGP in all scavenging assays. The $IC_{50}$ values of FCGP were 261.3 ${\mu}g$/mL for DPPH; 1,474 ${\mu}g$/mL for hydroxyl; 90.9 ${\mu}g$/mL for alkyl and 1,195 ${\mu}g$/mL for $H_2O_2$ in respective scavenging assays. Flavonoid compositions of both samples were determined by liquid chromatography/mass spectrometry (LC/MS). In the spectrum FCGP was similar to CGP in the contents of neohesperidin, naringin and an unknown No. 7 compound, but some unknown compounds (No. 1, 2, 4, 5, 6) were higher than CGP in each flavonoid contents. Therefore, the fermentation of CGP could increase the contents of unknown compound and improved antioxidant activities.

In vitro studies of anti-inflammatory and anticancer activities of organic solvent extracts from cultured marine microalgae

  • Marine microalgae are a promising source of organisms that can be cultured and targeted to isolate the broad spectrum of functional metabolites. In this study, two species of cyanobacteria, Chlorella ovalis Butcher and Nannchloropsis oculata Droop, one species of bacillariophyta, Phaeoductylum tricornutum Bohlin, and one species of Dinophyceae, Amphidinium carterae (Hulburt) were cultured and biomasses used to evaluate the proximate comical compositions. Among the determined proximate chemical compositions of the cultured marine microalgae, the highest content of crude proteins and lipids were exhibited in P. tricornutum and A. carterae, respectively. Solvent-solvent partition chromatography was subjected to fractionate each of the cultured species and separated n-hexane, chloroform, ethyl acetate, and aqueous fractions. Nitric oxide production inhibitory level (%) and cytotoxicity effect on lipo-polysaccharide-induced RAW 264.7 macrophages were performed to determine the anti-inflammatory activity. N. oculata hexane and chloroform fractions showed significantly the strongest anti-inflammatory activity at $6.25{\mu}g\;mL^{-1}$ concentration. The cancer cell growth inhibition (%) was determined on three different cell lines including HL-60 (a human promyelocytic leukemia cell line), A549 (a human lung carcinoma cell line), and B16F10 (a mouse melanoma cell line), respectively. Among the extracts, C. ovalis ethyl acetate and A. carterae chloroform fractions suppressed the growth of HL-60 cells significantly at 25 and $50{\mu}g\;mL^{-1}$ concentrations. Thus, the cultured marine microalgae solvent extracts may have potentiality to isolate pharmacologically active metabolites further using advance chromatographic steps. Hence, the cultured marine microalgae can be described as a good candidate for the future therapeutic uses.

Effects of Compatibilizer and Graphene Oxide on the Impact Strength of PC/ABS Blend (PC/ABS의 충격강도에 미치는 상용화제와 그래핀 옥사이드의 영향)

  • In this study, effects of both the grafted ABS-g-MAH and the added graphene oxide (GO) on the impact strength of polycarbonate (PC)/poly(acrylonitrile-butadiene-styrene) (ABS) blends were discussed. The PC/ABS blends and PC/ABS/GO composites were fabricated by using twin screw extruder with ABS-g-MAH as a compatibilizer. The ABS-g-MAH was prepared by melting extrusion of ABS and maleic anhydride (MAH) with DCP (dicumyl peroxide) as an initiator using twin screw extruder and the synthesis of ABS-g-MAH was confirmed by the presence of carbonyl group (C=O) peak at $1780cm^{-1}$ of FT-IR spectrum. According to the thermal, rheological, and impact properties of PC/ABS blends, 5 phr (parts per hundred resin) of compatibilizer was chosen as an optimum content for the PC/ABS/GO composites. It was observed that the thermal decomposition of ABS/PC/GO composites increased with GO contents, but there was no significant changes or a decrease in the impact strength. Also the composite fabricated by ABS/GO showed small increase in the impact strength. From the result of the dynamic rheometer to observe the processing properties, the complex viscosities of PC/ABS blend including the compatibilizer increased, but the complex viscosities of composites added GO were not changed.

Characteristics of Polysaccharide Isolated from the Fruit Body and Cultured Mycelia of Phellinus linteus IY001 (Phellinus linteus IY001의 자실체와 균사체 배양물로부터 분리한 다당류의 물리화학적 특성 비교)

  • This study was conducted to investigate the characteristics of polysaccharides isolated from the fruit body and cultured mycelia of Phellinus linteus IY001. All fractions were extracted by hot water, followed by ethanol precipitation (F-THE and M-HE) or ultrafiltration (M-HU) (F-TH, F-THE; fruit body, M-HE, M-HU; cultured mycelia). Among these fractions, F-TH fraction was obtained at the highest yields of 6.83% and yield of F-THE was at the level 2.79%. The carbohydrates of these fractions was found to be a heteroglucan composed of glucose, galactose, mannose, fructose, ribose and xylose by analysis of gas chromatography. The total carbohydrate contents of M-HE and M-HU fractions were 99.2%, and 86.0% respectively. The glucose content of M-HE, M-HU and F-THE ranged from 54 to 84.8% of the total monosaccharide. Amino acid pattern showed that all fractions contained a large amount of aspartic acid, glycine, glutamic acid, alanine. Serine and threonine were found to be involved in the linkage, O-linked type. These fractions, except F. TH, contained polysaccharides with the molecular weights of 12 kD and showed the characteristics of IR absorption for ${\beta}-glucosides$ at $890\;cm^{-1}$.

  • In this thesis, both soap and enzymatic degumming method were adopted and the optimum degumming conditions were obtained. Difference between the two degumming methods in silk fiber state was investigated and analyzed on the basis of the results of physical testings, polarizing microscopy, scanning electron microscopy, viscosity measurement, (${\alpha}$$\varepsilon$) amino group contents measurement, birefringence measurement, amino acid analysis, thermal analysis, infrared spectroscopy and x-ray diffraction analysis. The results obtained were summarized as follows; Physical test results of the degummed silk fiber showed that the tenacity and the elongation of enzymatic degummed silk fiber were lower than those of soap degummed fiber. But SEM observation and amino acid analysis showed almost the same tendency in the two degumming methods. The viscosity of enzymatic degummed silk fiber was lower than that of soap degummed fiber, but (${\alpha}$$\varepsilon$) amino group contents was higher in the enzymatic degummed fiber. It can be suggested that the enzymatic degummed silk fibroin was more degraded than the soap degummed fibroin. The birefringence, endothermic temperature of DSC spectrum, IR crystallinity and X-ray lateral order factor of enzymatic degummed silk fiber were higher than those of soap degummed fiber. It seems that the enzymatic degummed silk fiber has the higher crystallinity than that of soap degummed one according to the above results. However, it can be inferred that these differences between soap and enzymatic degummed fiber would be lessened if pretreatment and aftertreatment were included in the enzymatic degumming process.

  • Biochemical and histological effects of ozone gas (0.3 ppm) on rice plant were discussed. After ozone expoure, damage symptom, percentages of destroyed leaves, activities of peroxidase and polyphenoloxidase, and the contents of flavonoid, protein and sugar were examined on two rice varieties (Seokwangbyeo, Jinjubyeo), on tillering stage, and at different exposure time (0, 1, 3, 5 hr). The result were as followed. 1. The ozone-injured cell adjoining stomata become pigmented red-brown. 2. The percentage of injured leaves in Jinjubyeo was higher than that in Seokwangbyeo. 3. The activities of peroxidase and polyphenoloxidase increased on ozone-injured leaves. 4. The peroxidase activity increased with time in Jinjubyeo compared to Seokwangbyeo. 5. Peroxidase isozyme spectrum was altered after ozone exposure. 6. The content of flavonoid and reducing sugar in the rice leaves was increased, but the contents of protein was reduced.

  • Human neutrophil elastases (HNElastase, EC 3.4.21.37), a causative factor of inflammatory diseases, are regulated by plasma proteinase inhibitors, alpha-proteinase inhibitor and ${\alpha}_2-macroglobulin$. Under certain pathological conditions, however, released enzymes or abnormal function of inhibitors may cause various inflammatory disease. NSAIDs have been clinically applied for treatment of inflammatory diseases. Inhibition of cyclooxygenase is a known mechanism of action of NSAIDs in the treatment of inflammatory disease. In in vitro experiments, HNElastase was inhibited by naproxen, phenylbutazone, and oxyphenbutazone, but ibuprofen, ketoprofen, aspirin, salicylic acid, and tolmetin did not inhibit elastase. HNElastase was also inhibited by chelating agents, EDTA & EGTA, and tetracyclines. Removal of divalent metal ions by EDTA caused inhibition of elastase, and reconstitution of the metal ions recovered the enzyme activity to a certain level. Frequencies and contours in the Raman spectra of various conditions of human neutrophil elastase undergo drastic changes upon partial removal and/or reconstitution of calcium and zinc ions. The metal ion content dependent activities and change of the contour of the Raman spectrogram suggest us that the mechanism of action of a chelator or chelator-like agents on neutrophil elastase may be related to the conformational change at/or near the active site, especially -C=O radical or -COOH radical.

A Study on Physical Dispersion and Chemical Modification of Graphene (Graphene의 물리적 분산과 화학적 표면 개질 연구)

  • Graphene has a wide spectrum on its application field due to various and excellent physical properties. However, it is very difficult to apply that graphene exists as lump or fold condition in general organic solvents. Besides, graphene was difficult to maintain as uniform condition due to chemical inert and distributions with various size and shapes. Therefore, this study was focused to study dispersion and modifying methods of aggregated graphene. The dispersion methods contain as follow: i) physical milling using glass bead, ii) co-treatment of glass bead and ultrasonic waves, iii) dispersion in organic solvents, iv) modifying with dry-ice. Milling using glass bead with size 2.5 mm was effective to be size decrease of 36.4% in comparison with control group. Mixed treatment of glass bead (size 2.5 mm) and ultrasonic waves (225W, 10 min) showed relative size decrease of 76%, suggesting that the size decrease depends on the size of glass bead, intensity of ultrasonic waves and treatment time. Solvents of Ethyl acetate (EA) and Isoprophyl alcohol (IPA) were used in order to improve dispersion by modifying surface of graphene. IPA of them showed a favorable dispersion with more -CO functional groups in the FT-IR analysis. On the other hand, the oxygen content of graphene surface modified by dry-ice was highly increased from 0.8 to 4.9%. From the results, it was decided that the favorable dispersion state for a long time was obtained under the condition of -CO functional group increase in IPA solvent.

Chemical Properties and Spectroscopic Characteristics of Humic Fractions Isolated from Commercial Organic Fertilizers (국산(國産) 유기질비료(有機質肥料)의 부식조성(腐植組成) 및 분광학적(分光學的) 특성(特性))

  • Humic substances of 17 organic fertilizers available on the market were the objects of study. The list of ingredients for formulation of them comprised fish meal. bone meal, oil-cakes, brewer's grains, peat, sawdust, wood bark, zeolite, soil conditioner, live-stock droppings, amino acid fermentation byproduct, chaff, limestone and others. Humic and fulvic acids were isolated from those substances and given chemical and spectroscopic analyses. Nutritional values of the organic fertilizers showed big diversity. Humification of organic matter was incomplete for some of the fertilizers as indicated by a high C/N ratio. Extractable humic acid percentage was higher, in general, than that of fulvic acid. Also the relative content of humin increased with advanced humification. Total acidity was closely related to phenolic hydroxyl groups. Relationships between carboxyl and hydroxyl groups. and carboxyl and alcoholic hydroxyl groups were very significant. Ultraviolet and visible light absorption spectra of humic and fulvic acids were substantially similar. The types of humic acids were B. P, and Rp. Two humic acids of the 17 samples belonged to B type. 3 to P type and all the rest to Rp type.
