Objectives: This study addresses the effect of using nanoparticles (np) on the antimicrobial properties of bioactive glass (BAG) when used in intracanal medicaments against Enterococcus faecalis (E. faecalis) biofilms. Materials and Methods: E. faecalis biofilms, grown inside 90 root canals for 21 days, were randomly divided into 4 groups according to the antimicrobial regimen followed (n = 20; BAG-np, BAG, calcium hydroxide [CaOH], and saline). After 1 week, residual live bacteria were quantified in terms of colony-forming units (CFU), while dead bacteria were assessed with a confocal laser scanning microscope. Results: Although there was a statistically significant decrease in the mean CFU value among all groups, the nano-group performed the best. The highest percentage of dead bacteria was detected in the BAG-np group, with a significant difference from the BAG group. Conclusions: The reduction of particle size and use of a nano-form of BAG improved the antimicrobial properties of the intracanal treatment of E. faecalis biofilms
본 연구의 목적은 돼지난자의 체외성숙, 수정 및 단위발생시 일어나는 표층과립의 분포를 살펴보고, 그것들의 역할을 규명해 보고자 실시하였다. 난자의 표층과립은 형광염색을 실시한 후 laser scanning confocal microscope를 이용하여 관찰하거나 transmission electron microscope를 사용하여 관찰하였다. Germinal vesicle 단계의 돼지난자에서는 표층과립은 난자피질에 비교적 두꺼운 형태로 발견되었는데, germinal vesicle breakdown이 일어난 직후 피질 부근으로 표층과립의 움직임이 관찰되었다. Microfilaments의 중합화를 방해하는 cytochalasin B를 처리하였을 때 표층과립의 움직임은 관찰되지 않았다. 무 처리군의 수정 및 단위발생을 유도한 난자에서는 표층과립 내용물들이 위란강내에 균질하게 관찰되었으나, cytochalasin B를 처리한 난자에서는 비정상적인 cortical granule의 움직임을 관장하고 이러한 움직임이 수정시 다정자 침입을 막고 표층과립 반응에 영향을 미치는 것으로 사료된다.
Using a line scan camera and a Galvano mirror, we constructed a high-speed line-scanning microscope that can generate 2D images ($8000{\times}8000pixels$) without any moving parts. The line scanner consists of a Galvano mirror and a cylindrical lens, which creates a line focus that sweeps over the sample. The measured resolutions in the x (perpendicular to line focus) and y (parallel to line focus) directions are both $2{\mu}m$, with a 2X scan lens and a 3X relay lens. This optical system is useful for measuring defects, such as spalling, chipping, delamination, etc., on the surface of carbon fiber reinforced plastic (CFRP) holes after machining in conjunction with adjustments in the angle of LED lighting. Defects on the inner wall of holes are measured by line confocal laser scanning. This confocal method will be useful for analyzing defects after CFRP machining and for fast 3D image reconstruction.
Collagen can be used in building artificial skin replacements for treatment of burns and towards the reconstruction of bone as well as researching cell behavior and cellular interaction. The strength of collagen in connective tissue rests on the characteristics of collagen fibers. 3D confocal imaging of collagen fibers enables the characterization of their spatial distribution as related to their function. However, the image stacks acquired with confocal laser-scanning microscope does not clearly show the collagen architecture in 3D. Therefore, we developed a new method to reconstruct, visualize and characterize collagen fibers from fluorescence confocal images. First, we exploit the tensor voting framework to extract sparse reliable information about collagen structure in a 3D image and therefore denoise and filter the acquired image stack. We then propose to segment the collagen fibers by defining an energy term based on the Hessian matrix. This energy term is minimized by a min cut-max flow algorithm that allows adaptive regularization. We demonstrate the efficacy of our methods by visualizing reconstructed collagen from specific 3D image stack.
The localization of paclitaxel was investigated in suspension culture cells of Taxus chinensis. Over 93% of the cell-associated paclitaxel were detected throughout the entire culture period. Intracellular localization of paclitaxel over the culture time was analyzed further by cell fractionation for days 21 and 42. Paclitaxel contents in intracellular organelles were decreased at day 42, while the content in the cell wall fraction was increased at day 42 compared to the value for day 21. The localization of paclitaxel in the cell wall was confirmed by using the immunocytochemical method with the aid of a confocal laser scanning microscope.

A image of $140\times120{\mu}m^2$ is obtained within 1 second by SCM because scan speed of a x-axis and y-axis are 1kHz and 1Hz, respectively. AFM is able to measure correctly hight and width of ITO and scribing depth and width of a wafer with a resolution less than 300 . However, the scan speed is slow and it is difficult to distinguish a surface composed of different nm kinds of materials. Results show that SCM is preferable to obtain a image of a sample composed of different kinds of material than AFM because the intensity of a reflected light from surface is different from each material.

압축력의 변화에 따른 균열 간극변화 양상을 측정하고 간극변화와 수리전도도와의 관계를 파악하기 위해 이 연구에서는 다섯 단계의 수직 일축압축력을 균열면에 연속적으로 가하면서 고해상도의 공초점 레이저 스캔 현미경 (confocal laser scanning microscope; CLSM)을 이용하여 간극의 크기를 측정하고 디지털 이미지를 획득하였다. 기존의 연구들과는 달리 이 연구의 측정방법은 압력변화에 대한 동일시료 간극의 반응을 연속적으로 파악할 수 있는 점이 특징이다. 측정결과는 간극크기가 일정하지 않은 불평탄한 균열형태를 매우 잘 나타내었다. 균열 조도(roughness)로 인해 압력에 따른 간극 변화량은 일정하지 않고 부분마다 다름을 보였다. 각 압력단계에서 간극변화에 따른 투수성 변화양상을 파악하고자 실내투수시험을 병행하여 실시한 결과, 각 압력단계에서의 투수성 변화도 일정한 감소율을 나타내지 않고 간극 변화율이 크더라도 투수율은 오히려 작은 경우도 관찰되었다. 현미경을 통해 측정한 물리적 간극과 실제 유체유동 경로가 되는 수리간극의 크기 차이를 파악하기 위해 계산을 한 결과, 미미한 갈이지만 물리적 간극보다 작은 크기의 수리간극이 구해졌다. 또한, 실내투수시험 결과를 이용하여 구한 투수계수는 삼승법칙을 따르지 않는 것으로 드러났으며, 이러한 사실들은 균열 양쪽이 서로 평행하지 않고 불평탄한 양상을 가짐을 의미하는 것으로서, 현미경을 통해 직접 관찰한 간극양상과 잘 일치하는 것이다.
In-depth knowledge of fabric microstructure is essential for understanding clothing comfort since it plays a significant role in heat and mass transfer between the human body and clothing. In this study, a novel method was employed for investigating 3D surfaces and solid construction characteristics of specific fabrics by using a reverse engineering technique. The surface construction data were obtained by a confocal laser scanning microscope and then manipulated by a 3D analysis program. Triangle mesh was used for connecting each 3D point, with clouds and fabric surface characteristics created by rendering techniques. For generating a 3D solid model, determinants of radius of curvature was used. According to the proposed method, actual surface expression of the real fabric was achieved successfully. The results from this methodology can be applied to the detailed analysis of clothing comfort that is highly influenced by the microstructure of the fabric.

Physical and chemical weathering degrades rock, affecting its structural properties and thus the stability of stone buildings or other structures. Confocal laser scan microscopy (CLSM) is used here to observe temporal changes in the surface roughness of rock samples under simulated accelerated weathering. Samples were pressurized to 50, 55, or 70 MPa using a pressure frame, and subjected to freeze/thaw cycling controlled by a thermostat. The temperature was cycled from -20℃ to 40℃ and back. After each 20 cycles, CLSM was used to assess the change in surface roughness, and roughness factors were calculated to quantify the progression of the surface condition over time. Variations in cross-section line-roughness parameters and surface-roughness parameters were analyzed for specific parts of the sample surfaces at 5× and 50× magnification. The result reveals that the highest and lowest values of the roughness factors are changed according to elapsed time. Freezing/thawing at high pressure caused larger changes in the roughness factor than at low pressure.
