Atypical Actions of G Protein-Coupled Receptor Kinases

  • G protein-coupled receptor kinases (GRKs) and ${\beta}$-arrestins have been known as regulators of G protein-coupled receptors. However, it has been recently reported that GRKs and ${\beta}$-arrestins mediate receptor-mediated cellular responses in a G proteinin-dependent manner. In this scheme, GRKs work as a mediator or a scaffold protein. Among 7 members of the GRK family (GRK1-GRK7), GRK2 is the most extensively studied in vitro and in vivo. GRK2 is involved in cellular migration, insulin signaling, and cardiovascular disease. GRK6 in concert with ${\beta}$-arrestin 2 mediates chemoattractant-stimulated chemotaxis of T and B lymphocytes. GRK5 shuttles between the cytosol and nucleus, and regulates the activities of transcription factors. GRK3 and GRK4 do not seem to have striking effects on cellular responses other than receptor regulation. GRK1 and GRK7 play specific roles in regulation of rhodopsin function. In this review, these newly discovered functions of GRKs are briefly described.

  • It is a difficult and important task to measure the reverberation time of an acoustic room with a short reverberation time. This paper presents a new technique to measure the reverberation time of an acoustic room with low value of BT60. The digital signal processing technique used to do this is the wavelet filter which is very flexible to design the 1/n octave band filter and has no delay problem compared with the conventional IIR digital filter. This method is successfully applied to the measurement of the reverberation time at low frequency band of famous concert halls in Korea.

  • Recently, many membrane structure buildings are constructed with the trend of multi functional and high technological construction. The membrane structure has the characteristics such as distinguished architectural shape which can make variable space creation and can make economic use of material. Therefore, it is in the spotlight of sport complex, various concert hall, and public service buildings. However, the acoustic study of membrane structure has not been following up the increasing demand for the membrane structure. In this study, the acoustic characteristics of membrane structure will be studied and analyzed using architectural acoustic factors based on acoustic design theory And also, the differences between theoretical exhortation value and outcome of study will be studied with the basis of architectural acoustic material study.

The Kinetics of Complexation of Nickel(II) and Cobalt(II) Mandelates in Aqueous Solution

  • The rate constants for the formation and dissociation of nickel(II) and cobalt(II) complexes with mandelate have been determined by the pressure-jump relaxation study. The forward and reverse rate constants for the mandelate complex formation reactions were obtained to be $k_f=3.60{\times}10^4\;M^{-1}s^{-1}$ and $k_r=1.73{\times}10^2\;s^{-1}$ for the nickel(II), and $k_f=1.75{\times}10^5\;M^{-1}s{-1}$ and $2.33{\times}10^3\;s^{-1}$ for the cobalt(II) in aqueous solution of zero ionic strength ($(\mu{\to}0)\;at\;25^{\circ}C$. The results were interpreted by the use of the multistep complex formation mechanism. The rate constants evaluated for each individual steps in the multistep mechanism draw a conclusion that the rate of the reaction would be controlled by the chelate ring closure step in concert with the solvent exchange step in the nickel(II) complexation, while solely by the chelate ring closure step for the cobalt(II) complex.

Single-Cell Sequencing in Cancer: Recent Applications to Immunogenomics and Multi-omics Tools

  • Tumor heterogeneity, the cellular mosaic of multiple lineages arising from the process of clonal evolution, has continued to thwart multi-omics analyses using traditional bulk sequencing methods. The application of single-cell sequencing, in concert with existing genomics methods, has enabled high-resolution interrogation of the genome, transcriptome, epigenome, and proteome. Applied to cancers, these single-cell multi-omics methods bypass previous limitations on data resolution and have enabled a more nuanced understanding of the evolutionary dynamics of tumor progression, immune evasion, metastasis, and treatment resistance. This review details the growing number of novel single-cell multi-omics methods applied to tumors and further discusses recent discoveries emerging from these approaches, especially in regard to immunotherapy.

Sphingosine-1-phosphate Inhibits Human Keratinocyte Proliferation via Akt/PKB Inactivation

  • Although sphingosine-1-phosphate (S1P) is a well-known mitogen, our results show that S1P potently inhibits keratinocyte proliferation, and that this leads the inhibition of DNA synthesis. Interestingly, the prolonged activation of extracellular signal-regulated protein kinase (ERK) and the transient inactivation of Akt/protein kinase B (PKB) were also oberved in concert with the inhibition of keratinocyte proliferation by S1P. To further verify the anti-proliferative action of S1P, we examined changes in cell cycle related proteins. (omitted)

  • Lysophosphatidic acid (LPA) is a well-known mitogen in various cell types. However, we were surprised to find that LPA inhibits melanocyte proliferation. Thus, we further investigated the possible signaling pathways involved in melanocyte growth inhibition. We first examined the regulation of the three major subfamilies of mitogen-activated protein (MAP) kinases and of the Akt pathway by LPA. The activations of extracellular signal-regulated protein kinase (ERK) and c-Jun N-terminal kinase (JNK) were observed in concert with the inhibition of melanocyte proliferation by LPA, whereas p38 MAP kinase and Akt were not influenced by LPA. Endometriosis and Environmental Endocrine Disruptors

  • Endometriosis is classically defined as the growth of endometrial glands and stroma at extrauterine sites. Although it is a common gynecological problem accompanied by chronic pelvic pain, infertility, and adhesion formation, the etiology of this disease is unknown. Endometriosis pathogenesis may involve endocrine and immune dysfunction since uterine endometrial growth is regulated by sex hormones in concert with bioactive mediators produced by uterine immune and endocrine cells. Thus, exposure to environmental toxicants disrupting endocrine and immune responses potentially affect the development and progression of endometriosis.

A spectral model for human bouncing loads

  • Fourier series-based models in the time domain are frequently established to represent individual bouncing loads, which neglects the stochastic property of human bouncing activity. A power spectral density (PSD) model in the frequency domain for individual bouncing loads is developed herein. An experiment was conducted on individual bouncing loads, resulting in 957 records linked to form long samples to achieve a fine frequency resolution. The Welch method was applied to the linked samples to obtain the experimental PSD, which was normalized by the bouncing frequency and the harmonic order. The energy, energy distribution center, and energy distribution shape of the experimental PSD were investigated to establish the PSD model. The proposed model was used to analyze structural vibration responses using stochastic vibration theory, which was verified via field measurements. It is believed that this framework can evaluate the vibration capacity of structures excited by bouncing crowds, such as concert halls and grandstands.
