Purpose: Although several reports have described the relationship between periodontal disease and cardiovascular disease, information about the association between periodontal disease and the progression of degenerative aortic stenosis (AS) is lacking. Therefore, we performed a retrospective, single-center, pilot study to provide insight into this potential association. Methods: Data from 45 consecutive patients (19 men; median age, 83 years) with mild or moderate degenerative aortic stenosis were analyzed for a mean observation period of 3.3±1.9 years. The total amount of Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis and titers of serum immunoglobulin G (IgG) against periodontal bacteria and high-sensitivity C-reactive protein (hs-CRP) were evaluated. Aortic valve area (AVA), maximal velocity (Vmax), mean pressure gradient (mean PG), and the Doppler velocity index (DVI) were evaluated. The change in each parameter per year ([ParameterLATEST-ParameterBASELINE]/Follow-up Years) was calculated from the retrospective follow-up echocardiographic data (baseline vs. the most recently collected data [latest]). Results: No correlation was found between the concentration of periodontopathic bacteria in the saliva and AS status/progression. The anti-P. gingivalis antibody titer in the serum showed a significant positive correlation with AVA and DVI. Additionally, there was a negative correlation between the anti-P. gingivalis IgG antibody titer and mean PG. The hs-CRP concentration showed positive correlations with Vmax and mean PG. Meanwhile, a negative correlation was observed between the anti-P. gingivalis IgG antibody titer and ΔAVA/year and Δmean PG/year. The hs-CRP concentration showed positive correlations with Vmax and mean PG, and it was significantly higher in patients with rapid aortic stenosis progression (ΔAVA/year <-0.1) than in their counterparts. Conclusions: Our results suggest that periodontopathic bacteria such as A. actinomycetemcomitans and P. gingivalis are not directly related to the status/progression of degenerative AS. However, inflammation and a lower immune response may be associated with disease progression.

Taurine, a ${\beta}-amino$ acid, plays an important role as a neuromodulator and is necessary for the normal development of the brain. Since de novo synthesis of taurine in the brain is minimal and in vivo studies suggest that taurine dose not cross the blood-brain barrier, we examined whether the choroid plexus, the blood-cerebrospinal fluid (CSF) barrier, plays a role in taurine transport in the central nervous system. The uptake of $[^3H]-taurine$ into ATP depleted choroid plexus from rabbit was substantially greater in the presence of an inwardly directed $Na^+$ gradient taurine accumulation was negligible. A transient in side-negative potential gradient enhanced the $Na^+-driven$ uptake of taurine into the tissue slices, suggesting that the transport process is electrogenic, $Na^+-driven$ taurine uptake was saturable with an estimated $V_{max}$ of $111\;{\pm}\;20.2\;nmole/g/15\;min$ and a $K_M\;of\;99.8{\pm}29.9\;{\mu}M$. The estimated coupling ratio of $Na^+$ and taurine was $1.80\;{\pm}\;0.122.$$Na^+-dependent$ taurine uptake was significantly inhibited by ${\beta}-amino$ acids, but not by ${\alpha}-amino$ acids, indicating that the transporter is selective for ${\beta}-amino$ acids. Since it is known that the physiological concentration of taurine in the CSF is lower than that in the plasma, the active transport system we characterized may face the brush border (i.e., CSF facing) side of the choroid plexus and actively transport taurine out of the CSF. Therefore, we examined in vivo elimination of taurine from the CSF in the rat to determine whether elimination kinetics of taurine from the CSF is consistent with the in vitro study. Using a stereotaxic device, cannulaes were placed into the lateral ventricle and the cisterna magna of the rat. Radio-labelled taurine and inulin (a marker of CSF flow) were injected into the lateral ventricle, and the concentrations of the labelled compounds in the CSF were monitored for upto 3 hrs in the cisterna magna. The apparent clearance of taurine from CSF was greater than the estimated CSF flow (p<0.005) indicating that there is a clearance process in addition to the CSF flow. Taurine distribution into the choroid plexus was at least 10 fold higher than that found in other brain areas (e. g., cerebellum, olfactory bulb and cortex). When unlabelled taurine was co-administered with radio-labelled taurine, the apparent clearance of taurine was reduced (p<0.0l), suggesting a saturable disposition of taurine from CSF. Distribution of taurine into the choroid plexus, cerebellum, olfactory bulb and cortex was similarly diminished, indicating that the saturable uptake of taurine into these tissues is responsible for the non-linear disposition. A pharmacokinetic model involving first order elimination and saturable distribution described these data adequately. The Michaelis-Menten rate constant estimated from in vivo elimination study is similar to that obtained in the in vitro uptake experiment. Collectively, our results demonstrate that taurine is transported in the choroid plexus via a $Na^+-dependent,saturable$ and apparently ${\beta}-amino$ acid selective mechanism. This process may be functionally relevant to taurine homeostasis in the brain.
가토 신장 피질에서 Percoll density gradient방법으로 분리한 basolateral membrane vesicle (BLMV)에서 rapid filtration technique을 이용하여 succinate의 이동 특성을 관찰하였다. $Na^+$은 succinate의 이동을 증가시켜 "overshoot"현상을 보였으며 이러한 효과는 $K^+,{\;}Li^+,{\;}Rb^+,{\;}choline$과 같은 다른 양이온들에 의해 나타나지 않았다. $Na^+$농도변화에 따른 succinate의 이동율은 sigmoid모양을 보였고, $Na^+$에 대한 Hill coefficient는 2.0이었다. soccinate의 이동은 vesicle 내부가 음전압일 때 더욱 증가되었다. BLMV에서 succinate이동은 용액내 pH변화에 따라 영향을 받았으나 brush border membrane vesicle (BBMV)에서는 영향을 받지 않았다. 동력학적 분석결과 succinate의 Km값은 $15.5{\pm}0.94{\;}{\mu}M$이었고 Vmax는 $16.22{\pm}0.25{\;}n{\;}mole/mg{\;}protein/min$이었다. succinate의 이동은 $4{\sim}5$탄소를 가진 dicarboxylate들에 의해 강력하게 억제되었으나 monocarboxylate나 다른 유기음이온들에 의해 영향을 적게 받거나 받지 않았다. succinate의 이동은 DIDS, SITS, furosemide와 같은 음이온 이동 억제제와 harmaline과 같은 $Na^+$ 이동 억제제에 의해 억제되었다. 이들 결과들은 BLMV에서 succinate는 $Na^+$에 의존하여 이동하며 다른 Krebs cycle중간 산물들과 동일한 운반기전을 이용함을 가르킨다. 또한 BLMV에서 succinate의 이동은 그 기질특이성에 있어서 다른 연구자에 의해 보고된 BBMV에서 이동특성과 유사함을 보였다.
For evaluating the quality of ginseng, simple and fast analysis methods are needed to determine the ginsenoside content of the ginseng products. The aim of this study was therefore to optimize conditions for fast analysis of the ginsenosides, the active ingredients in extracts of Korean red ginseng. When tandem HPLC mass spectrometry (HPLC-MS/MS) was used, four forms of ginsenoside, Rb1, Rb2, Rc, and Re, were readily separated in seven minutes using a gradient mobile phase (acetonitrile and water containing acetic acid). This is the shortest separation time reported among the studies of major ginsenoside analysis. When gradient HPLC with UV detection was used, the detection limit was high, but separation of these four ginsenosides required 25 minutes using acetonitrile and water containing formic acid as a mobile phase. HPLC-MS/MS was able to separate ginsenoside Rg1 easily regardless of the mobile phase condition, but the HPLC-UV could not separate Rg1 because acetonitrile concentration in the mobile phase had to be maintained below 20%. Ginsenoside peaks were clearer and had more sensitive detection limits when Korean red ginseng extract was analyzed by the HPLC-MS/MS, but the UV detection was useful for chromatographic fingerprinting of all four major ginsenosides of the extract: Rb1, Rb2, Rc, and Re. Extracts were found to contain 2.17 mg, 1.51 mg, 1.29 mg, and 0.46 mg of ginsenoside Rb1, Rb2, Rc, Re, respectively, per gram weight. The ratios of each ginsenoside in the extracts were 1.0 : 0.7 : 0.6 : 0.2, respectively. Taken together, the results indicate that HPLC-MS/MS spectrometry could be the most useful method for rapid analysis of even small amounts of major ginsenosides, while HPLC with UV detection could also be used for rapid analysis of major ginsenosides and for quality control of ginseng products.
옥수수 뿌리로부터 분리한 13,000g pellet과 13,000~80,000g pellet 내에 있는 membrane-bound ATPases의 특성을 구명하였다. 13,000g pellet과 13,000~80,000g pellet의 membrane-bound ATPases의 최적 pH는 5와 9였다. Discontinuous sucrose gradient centrifugation에 의한 13,000g pellet의 분획중 Fraction C는 pH 5에서, Fraction D, E 및 F는 pH 5에서보다 pH 9에서 더높은 활성을 나타냈다. 13,000~80,000g pellet의 분획에서 보면, Fraction A, C는 pH 9보다 pH 5에서, Fraction B, D, E 및 F는 pH 5보다 pH 9에서 더 높은 활성을 가겠다. pH 5와 pH 9에서 membrane-bound ATPases의 기질포화 농도는 3~5 mM이며 ATP에 대한 Km 값은 모두 0.25 mM이였다. Vmax 값은 8.0~55.6 $\mu$M Pi/mg membrane protein/hr의 범위에 있었다. Membrane-bound ATPase의 활성은 $K^+$ 이온에 의해 증가되었다.
본 연구는 건강한 식이에 대한 관심이 늘어나면서 그 수요량이 증가하고 있는 녹즙에 대한 미생물학적 품질평가를 목적으로 실시되었다. 또한 날것으로 섭취하는 과채류의 장출혈성 대장균의 오염으로 인한 집단식중독 사고가 많이 보고되고 있는 상황에서 E. coli O157:H7이 녹즙에 오염되었을 경우 보관온도, 보관 시간별 미생물 커뮤니티의 변화를 정량 분석법과 DGGE분석을 통해 확인하여 잠재적인 식중독 위험성을 확인해 보고자 하였다. 정량 분석 결과 초기부터 총 호기성 세균수가 식품위생법상 기준치($1.0{\times}10^5CFU/mL$)를 초과한 결과를 보였으며, E. coli O157:H7에 대한 세균수 측정결과를 보면 $5^{\circ}C$와 $10^{\circ}C$에서 균수가 감소하다가 증가하는 경향을 보였다. DGGE결과를 보면 보관시간이 길고 보관온도가 높을수록 더 많은 미생물 분포도를 보였고, 저장시간이 후반으로 갈수록 부패균인 젖산균이 다수 출현하는 것을 확인할 수 있었다. 이 외에 Rahnella aquatilis와 Citrobacter, Pseudomonas 등의 기회감염성 세균이 검출되었다. 이에, 위 녹즙시료는 잠재적인 식중독의 위험성을 보이므로 제품의 생산, 유통에 더욱 주의를 기울일 필요가 있다고 생각된다. 또한, 제품 구입 후 냉장저장을 하더라도 가능한 한 빠른 섭취가 권장되는 바이다. E. coli O157:H7에 위 제품이 오염되었을 경우 냉장과 실온조건 모두에서 세균수가 증가하였고, 특히 실온에 방치하였을 경우 그 증가폭이 크므로, 이에 특히 주의를 기울일 필요가 있다고 생각된다.
감잎으로부터 tyrosinase 억제물질의 추출을 위하여 60% ethanol을 사용하여 추출한 경우 추출물의 phenol성물질 함량이 21.91 mg/g로 가장 높았고, tyrosinase 억제효과도 37%의 억제율로 가장 높게 나타났다. 감잎 추출물로부터 tyrosinase 저해 활성 물질을 정제하기 위하여 Sephadex LH-20 column과 MCI-gel CHP-20 column을 이용하여 gradient로 용출한 결과 tyrosinase 저해 활성을 가지는 2가지 순수한 compound를 정제하였으며, 정제된 2가지 compound를 구조 동정한 결과, (+)-gallocatechin과 prodelphinidin B-3이었다. 정제물의 tyrosinase 저해활성을 측정한 결과 (+)-gallocatechin과 prodelphinidin B-3는 29.5, 40.2%의 저해활성을 나타내었으며, melanoma 세포에서의 멜라닌 생합성 저해효과를 측정한 결과 (+)-gallocatechin과 prodelphinidin B-3는 각각 32.5, 46.7%의 저해활성을 나타내어 멜라닌생성억제효과가 매우 뛰어남을 알 수 있었다. 정제된 미백물질을 이용하여 제조한 essence 제품의 안정성을 살펴본 결과, 미백 essence제품의 pH는 10,000 ppm의 농도에서 4.90~4.95였으며, 점도는 23,000~26,000 cP로 60일 동안의 저장기간 동안 큰 변화를 나타내지 않았다. 일반적 보존 시험인 온도에 따른 안정성과 광안정성(인공광, 자연광) 검사에서도 안정하였으며, 특수, 가혹 보존 시험인 온도순환에 따른 안정성 검사에서도 상의 분리와 변색, 변취 없이 모두 안정함을 확인할 수 있었다.
