The Influences of Addition of Sugar with or without L. buchneri on Fermentation and Aerobic Stability of Whole Crop Maize Silage Ensiled in Air-stress Silos

  • The whole plant of crop maize was chopped and ensiled in double-layered polyethylene bags to determine the influence of residual sugar on the fermentation of lactic acid and aerobic stability by L. buchneri in whole crop maize silage made in airstress condition. There were a total of six treatments used in this experiment as follow: added 25 g de-mineralised water per kg chopped maize serving as control (con), 37.5 g glucose solution containing 12.5 g glucose ($g_1$), 75 g glucose solution containing 25 g glucose ($g_2$), 25 g, L,.buchneri suspension intended for $10^6$ cfu $g^{-1}$ (L.b.), $g_1$+L.b. and $g_2$+L.b. All silos were opened at day 91 after ensiling for measuring the pH values, microbiological enumeration, fermentative products and aerobic stability. The dry matter loss increased significantly (p<0.01) due to inclusion of sugar or L. buchneri. The lower lactic acid concentrations were observed (p<0.01) in silages inoculated with L. buchneri only or in combination with sugar addition than the correspondent uninoculated silages. Compared with control silage, ethanol production was about 3 or 6-fold higher due to addition 12.5 or 25 g glucose per kg chopped maize at ensiling. The silages added with sugar contained less acetic acid concentration (p<0.01) than control, but silages inoculated with L. buchneri showed the contrary effects (p<0.01) at different sugar levels. No butyric acid was found in uninoculated silages, silages inoculated with L. buchneri. producted more propionic acid, 1-propanol and butyric acid. Lactic acid bacteria counts increased markedly (p<0.01) due to inoculation with L. buchneri, whereas it was reduced (p<0.01) by added sugar. No significant difference was observed in count of yeast, but inoculation with L. buchneri shows a decreasing trend. Mould count in all silages was less than 2 (log cfu $g^{-1}$). The added sugar had negative effects on aerobic stability of maize silage made under air-stress conditions, whereas inoculation with L. buchneri improves (p<0.01) the aerobic stability.

Chemical and Sensory Characteristics of Boiled Soup Extracted from Crossbred Ogol Chicken as Affected by the Level of Flavourzyme (단백질 분해효소(Flavourzyme)의 첨가량에 따른 오골계 증탕액의 화학적 및 관능적 특성)

  • This study was conducted to investigate chemical and sensory characteristics of boiled soup extracted from crossbred ogol chicken as affected by the level of Flavourzyme produced from Aspergillus oryzae and composed with endo-proteinase(674U/g) and exoproteinase(8,053U/g). It was hydrolyzed by different concentrations of the protease enzyme (Flavourzyme)(0%(Control), 0.01%(T$_2$), 0.1%(T$_3$) and 0.5%(T$_4$)) at 45$^{\circ}C$ for 4hrs. Manufacture of the extract was performed by boiling treated meats with medicinal herbs(Sipchun daebo) at a higher pressure condition. Minerals, free amino acid content, sensory properties of the extract were as follows. The sodium contents were increased as the treatment levels of enzyme increased. The iron contents were lowest when the enzyme treated by 0.5% level, however there were not significantly different among the treatments. The copper and mangan contents had no significantly different among the enzyme treatment levels. Higher contents of fee amino acid were observed as the amount of the protease increased with the treatment of higher than 0.1% enzyme, no significant effect was observed. In sensory properties, the extract manufactured by addition of 0.01∼0.1% of Flavourzyme resulted in a similar or better appearance, flavor, taste and overall palatabilitycompared to control(no enzyme treatment). However, the extract manufactured with 0.5% of Flavourzyme resulted in lower scores in appearance, flavor, taste and overall palatability than the control. In addition, this product showed more off-flavor than control.

Immune Responses of Mice Intraduodenally Infected with Toxoplasma gondii KI-1 Tachyzoites

  • Toxoplasma gondii Korean isolate (KI-1) tachyzoites were inoculated intraduodenally to BALB/c mice using a silicon tube, and the course of infection and immune responses of mice were studied. Whereas control mice, that were infected intraperitoneally, died within day 7 post-infection (PI), the intraduodenally infected mice survived until day 9 PI (infection with $1{\times}10^5$ tachyzoites) or day 11 PI (with $1{\times}10^6$ tachyzoites). Based on histopathologic (Giemsa stain) and PCR (B1 gene) studies, it was suggested that tachyzoites, after entering the small intestine, invaded into endothelial cells, divided there, and propagated to other organs. PCR appeared to be more sensitive than histopathology to detect infected organs and tissues. The organisms spread over multiple organs by day 6 PI. However, proliferative responses of splenocytes and mesenteric lymph node (MLN) cells in response to con A or Toxoplasma lysate antigen decreased significantly, suggesting immunosuppression. Splenic $CD4^+$ and $CD8^+$ T-Iymphocytes showed decreases in number until day 9 PI, whereas IFN-${\gamma}$ and IL-10 decreased slightly at day 6 PI and returned to normal levels by day 9 PI. No TNF-${\alpha}$ was detected throughout the experimental period. The results showed that intraduodenal infection with KI-1 tachyzoites was successful but did not elicit significant mucosal immunity in mice and allowed dissemination of T. gondii organisms to systemic organs. The immunosuppression of mice included reduced lymphoproliferative responses to splenocytes and MLN cells to mitogen and low production of cytokines, such as IFN-${\gamma}$, TNF-${\alpha}$, and IL-10, in response to T. gondii infection.

  • The purpose of this study is to investigate the quality and sensory characteristics of ginseng Jeolim, or ginseng pickles. The moisture contents of ginseng pickles decreased significantly with increase in soy sauce and in storage period. The L-value of pickle solution decreased significantly with increase in soy sauce and in storage period while its a-value and b-value increased significantly with increase in soy sauce and in storage period. The salinity and sugar contents of pickle solution increased significantly with increase in soy sauce; however, they decreased with increase in storage period. The pH of pickle solution decreased significantly with increase in soy sauce and in storage period. As the results of acceptance test, apparence, flavor, taste, texture, and overall acceptance were higher in C3. In conclusion, ginseng Jeolim showed great preference with adding soy sauce, and the preference has increased with 20% of soy sauce added and 14 day storage period.

Effects of feeding corn naturally contaminated with aflatoxin on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of Na+, K+-ATPase in ducklings

  • Objective: A 14-d trial was conducted to determine the effects of feeding corn naturally contaminated with aflatoxin $B_1$ ($AFB_1$) on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of $Na^+$, $K^+-ATPase$ in ducklings. Methods: A total of 704 ducklings were blocked on the basis of sex and body weight (BW), and then allocated randomly to one of the following two treatments: i) CON, basal diet and ii) $AFB_1$, diets with 100% of normal corn replaced with $AFB_1$ contaminated corn. There were 22 pens per treatment and 16 birds per pen. The concentration of $AFB_1$ was 195.4 and $124.35{\mu}g/kg$ in the contaminated corn and $AFB_1$ diet, respectively. Results: The $AFB_1$ decreased average daily gain, average daily feed intake, d 7 BW, final BW in the whole trial, and feed conversion ratio (FCR) during d 8 to 14 and d 1 to 14 by 10% to 47% (p<0.05), while FCR during d 1 to 7 was increased (p<0.05). $AFB_1$ did not affect mortality to 7 d of age, and then increased to 5.8% from 8 to 14 d of age (p<0.01). Apparent ileal gross energy digestibility was reduced by $AFB_1$, whereas apparent ileal digestibility of dry matter, nitrogen, and amino acid was improved (p<0.01). Feeding $AFB_1$ diets increased serum concentration of leptin and insulin-like growth factors-1 (IGF-1) (p<0.05), but had no effect on neuropeptide Y, ghrelin, cholecystokinin-8 or insulin (p>0.05). Dietary treatments did not influence relative expression of jejunal $Na^+$, $K^+-ATPase$ gene (p>0.05). Conclusion: Taken together, feeding corn naturally contaminated with $AFB_1$ reduced growth performance, improved apparent ileal digestibility, and affected serum leptin and IGF-1 in ducklings from d 1 to 14.

Effects of Sargassumpallidum on 2,4,6-Trinitrobenzene Sulfonic Acid-Induced Colitis in Mice (해조가 2,4,6-trinitrobenzene-sulfonic acid로 유발된 염증성 장질환 동물모델에 미치는 영향)

  • Objectives : The aim of the current study was to investigate the effects of Sargassum (Sargassum pallidum (TURN.) C. AG.; SP) on the experimental colitis induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS) in mice. Methods : ICR mice were divided into 7 groups (NOR, CON, $SS50\times5$, $SP20\times3$, $SP50\times3$, $SP20\times5$, $SP50\times5$). TNBS processing was intrarectally applied to all experimental groups on the 3rd experiment day, except the normal group (NOR). For investigating the prophylactic effect, SP at doses of 20 mg/kg ($SP20\times5$) and 50 mg/kg ($SP50\times5$) were orally administered for 5 days. The SP at doses of 20 mg/kg ($SP20\times3$) and 50 mg/kg ($SP50\times3$) were orally administered for 3 days after the colitis induction in order to check the effect of treatment. As a positive control group, sulfasalazine 50 mg/kg ($SS50\times5$) was administrated. Macroscopic findings of epithelial tissue on mice were measured by colon length and macroscopic score. Histologic findings were also checked by crypt cell, epithelial cell, inflammatory cell and edema of submucosa. We measured the ability of SP to inhibit lipid peroxidation and myeloperoxidase activity. We also measured levels of the inflammatory markers, interleukin (IL)-$1\beta$ and cyclooxygenase-2 (COX-2), its transcription factor activation, phospho-NF-${\kappa}B$ (pp65), in the colon by enzyme-linked immunosorbent assay and immunoblot analysis. We measured activation of fecal bacterial enzyme, $\beta$-glucuronidase and degradation activation of fecal glycosaminoglycan (GAG), and hyaluronic acid. Results : Oral administration of SP on mice inhibited TNBS-induced colon shortening and myeloperoxidase activity in the colon of mice as well as IL-$1\beta$ and COX-2 expression. SP also inhibited TNBS-induced lipid peroxidation and pp65 activation in the colon of mice. SP inhibited $\beta$-glucuronidase activation and fecal hyaluronic acid degradation activation as well. Conclusions : SP could be a possible herbal candidate and preventive prebiotic agent for treating inflammatory bowel disease (IBD). Further experiments to differentiate effects of SP on IBD, such as other solutions and extracting times, might be promising.

Ideal Freezing Curve Can Avoid the Damage by Latent Heat of Fusion During Freezing (냉동 시 잠재용융열에 의한 피해를 최소화할 수 있는 이상냉동 곡선)

  • Background:Liquid nitrogen freezing techniques have already met with widespread success in biology and medicine as a means of long-term storage for cells and tissues. The use of cryoprotectants such as glycerol and dimethylsulphoxide to prevent ice crystal formation, with carefully controlled rates of freezing and thawing, allows both structure and viability to be retained almost indefinitely. Cryopreservation of various tissues has various con-trolled rates of freezing. Material and Method: To find the optimal freezing curve and the chamber temperature, we approached the thermodynamic calculation of tissues in two ways. One is the direct calculation method. We should know the thermophysical characteristics of all components, latent heat of fusion, area, density and volume, etc. This kind of calculation is so sophisticated and some variables may not be determined. The other is the indirect calculation method. We performed the tissue freezing with already used freezing curve and we observed the actual freezing curve of that tissue. And we modified the freezing curve with several steps of calculation, polynomial regression analysis, time constant calculation, thermal response calculation and inverse calculation of chamber temperature. Result: We applied that freezing program on mesenchymal stem cell, chondrocyte, and osteoblast. The tissue temperature decreased according to the ideal freezing curve without temperature rising. We did not find any differences in survival. The reason is postulated to be that freezing material is too small and contains cellular components. We expect the significant difference in cellular viability if the freezing curve is applied on a large scale of tissues. Conclusion: This program would be helpful in finding the chamber temperature for the ideal freezing curie easily.

Antimicrobial Activities of Corn Silk Extract of Klebsiella pneumoniae (옥수수수염 추출액의 Klebsiella pneumoniae에 대한 항균활성)

  • Klebsiella pneumoniae is found in the normal flora of the skin, mouth, respiratory tract, urinary tract, and intestines in human. However, the stain is opportunistic pathogen, which is the causative agent of community acquired pneumonia. Corn silk has been known to be effective for antimicrobial activity against pathogenic bacteria, including K. pneumoniae, Staphylococcus aureus, Bacillus subtilis, Shigella spp., Salmonella spp., Escherichia coli, Pseudomonas aeruginosa, et al. In this study we focused on the antimicrobial properties of con silk water extract of K. penumoniae. K. pneumoniae isolates K. pneumoniae ATCC 13883 and broad-spectrum β-lactamase (BSBL), exteded-spectrum β-lactamase (ESBL), carbapenemase-producers. Antimicrobial susceptibilities were determined by the disk diffusion method. Searches for bla genes were performed by PCR amplication and direct sequencing. MacConkey agar plate medium was prepared using the corn silk extracts (50% or 100%) instead of distilled water for antimicrobial activity test. The microbial growth inhibitory potential of K. pneumoniae was determined by using the MacConkey agar plate spreading method, and the plate was incubated 18 hr at 37℃. Genes encoding β-lactamases including SHV-1 (n=8), SHV-2a (n=8), SHV-5 (n=2), SHV-11 (n=2), SHV-12 (n=18), TEM-1 (n=10), CTX-M-3 (n=2), CTX-M-14 (n=2), CTX-M-15 (n=1), GES-5 (n=5), KPC-2 (n=6), KPC-3 (n=4), and NDM-1 (n=2) were detected. The corn silk extract showed significantly antimicrobial activity against K. pneumoniae ATCC 13883, but BSBLs, ESBLs, and carbapenemase producers were not. Therefore, corn silk extract is thought to be able to assist in the prevention and rapid recovery of infectious disease caused by K. pneumoniae.

Effects of Probiotic Microbes on Growth Performance, Innate Immunity, and Pathogen Sensitivity in Cultured Olive Flounder (Probiotic 기능을 가진 미생물을 함유한 양어용 생균제가 넙치의 성장, 선천성면역 및 항병능에 미치는 영향)

  • Probiotic principles can be applied in aquaculture for the purpose of growth and immunity stimulation, disease prevention and eventually better production performance. This study was to assess effects of combinations of microbes containing two Bacillus sp., plus one Lactobacillus sp. as the basal preparation (BSL-LAB), and additional Nitrosomonas sp. (nitrifying bacteria consortium, NBS) in olive flounder (Paralichthys olivaceus). The effects examined were growth parameters, hematologic parameters, innate immunity and pathogen challenge test. Fish were assigned to 4 treatments as Control (no probiotics), Group A (Bacillius and Lactobacillus to culture water), Group B (Bacillius and Lactobacillus both in water and feed), Group C (same as Group B with additional Nitrosomonas in feed). Fish were allocated to the above 4 groups, each group being composed of triplicate 30 fish, for a 7-week feeding in the laboratory. Positive effects were observed both in growth and pathogen sensitivity with all three probiotic combinations. Such effects were attributed to improved innate immune functions. This result indicates that the tested probiotic microbes are beneficial to olive flounder aquaculture.

${SO_4}^{2-}$ ion Removal from Solution by Crystal Precipitation on Surface Active Glasses (표면활성유리에의 결정석출을 이용한 용액중 황산염 이온 제거)

  • ${SO_4}^{2-}$ ions present in industrial waste water if combined with other metal ions in the water can cause serious scale problem in a transporting pipe. In this study therefore ${SO_4}^{2-}$ ions in an acidic solution have been removed by using surface active glasses. Glasses with various compositions of $SiO_2-Na_2O-B_2O_3-RO$ (R=Mg, Ca, Sr, Ba) system were reacted in a ${SO_4}^{2-}$ ion-containing solution with various pHs ranging from 1 to 4 for various time the reacted glass surfaces were analyzed by XRD and SEM and all ions in the reacted solution were also measured ${SO_4}^{2-}$ ions in the solution were combined with divalent ions leached out of glass and precipitated on the glass surface as sulfate crystals. In this was the surface ion could be removed from the acidic solution. The sulfate ion removal capacity is closely related to the solubility product con-stants of the newly formed sulfate crystals. Almost no sulfate crystal was formed on the MgO-containing glass while sulfate crystals were easily formed on the glass containing either SrO or BaO This indicates that those glasses have strong removal efficiency of ${SO_4}^{2-}$ ions from the solution.
