Inhibitory Effects of Ginsenoside Metabolites, Compound K and Protopanaxatriol, on $GABA_C$ Receptor-Mediated Ion Currents

  • Ginsenosides, one of the active ingredients of Panax ginseng, show various pharmacological and physiological effects, and they are converted into compound K (CK) or protopanaxatriol (M4) by intestinal microorganisms. CK is a metabolite derived from protopanaxadiol (PD) ginsenosides, whereas M4 is a metabolite derived from protopanaxatriol (PT) ginsenosides. The ${\gamma}$-aminobutyric acid $receptor_C$ ($GABA_C$) is primarily expressed in retinal bipolar cells and several regions of the brain. However, little is known of the effects of ginsenoside metabolites on $GABA_C$ receptor channel activity. In the present study, we examined the effects of CK and M4 on the activity of human recombinant $GABA_C$ receptor (${\rho}$ 1) channels expressed in Xenopus oocytes by using a 2-electrode voltage clamp technique. In oocytes expressing $GABA_C$ receptor cRNA, we found that CK or M4 alone had no effect in oocytes. However, co-application of either CK or M4 with GABA inhibited the GABA-induced inward peak current ($I_{GABA}$). Interestingly, pre-application of M4 inhibited $I_{GABA}$ more potently than CK in a dose- dependent and reversible manner. The half-inhibitory concentration ($IC_{50}$) values of CK and M4 were $52.1{\pm}2.3$ and $45.7{\pm}3.9{\mu}M$, respectively. Inhibition of $I_{GABA}$ by CK and M4 was voltage-independent and non-competitive. This study implies that ginsenoside metabolites may regulate $GABA_C$ receptor channel activity in the brain, including in the eyes.

A Case of Classical Galactosemia caused by Compound Heterozygous Mutations of the GALT Gene (GALT 유전자의 복합 이형 돌연변이에 의한 전형적 갈락토오스혈증 1례)

  • Classical galactosemia is an autosomal recessive disorder of galactose metabolism, caused by a deficiency of the enzyme galactose-1-phosphate uridyltransferase (GALT). Buildup of galactose-1-phosphate is toxic at high levels and can damage the liver, brain, eyes, and other vital organs. The case presented here was that of an 11-day-old female infant who had elevated galatose levels upon initial neonatal screening test with persistent cholestatic jaundice, coagulopathy, and hepatomegaly. The patient was transferred due to aggravation of clinical symptoms including bleeding and jaundice. She had a delayed galactose free diet because of an inappropriate diagnosis. We quickly provided her with a lactose/galactose-restricted diet as per her final diagnosis. Clinical and laboratory results were improved after a few days of treatment. For confirmatory testing for classical galactosaemia, we simultaneously analyzed for GALT enzyme activity and allele-specific PCR/fragments for seven mutations and two polymorphisms in the GALT gene. We were able to find several GALT-deficient and compound heterozygous mutations of the GALT gene.

The Visual Acuity and Refraction on Students of Primary School Using Eyeglasses in Kyung-Nam Region (경남지역 안경 착용자 초등학생들의 시력과 굴절)

  • The study of the visual acuity and refraction status was performed to the students of primary school using eyeglasses in Kyung Nam region, Korea. Two hundred sixty three subjects with various styles of ametropia were analyzed the visual acuity and refraction examination by the subjective and objective tools. The results of study were as follows; 1) According to the increase of age of subjects, the visual acuity of binocular in naked eyes was decrease. However ametropia was increase. 2) The high frequency of ametropia types in the binocular was simple myopia, and the next order of high frequency was mixed astigmatism, myopic compound astigmatism, simple hyperopia, and hyperopic compound, respectively. 3) On the power of spherical refractive error of ametropia, the high frequency was -1.00 diopter, and the next order of high frequency was -2.00 diopter, +2.00 diopter, -3.00 diopter, and so on, respectively. 4) According to the increase of subjects ages, the changes of dioptric power of myopic spheric power was increase. however the frequency of the boy and girl was similar results, also the dioptric values of refractive status in binocular was similar results. As the results of this study, we conclude that the relationship of the visual acuity and refractive error status in primary students using eyeglasses was improved.

  • To investigate the visual acuity of the primary school children. the visual acuity test wan performed the object and subject method. The results were follows. 1. The eye types were 94.8% positive for myopia, 1.2% for emmetropia and 4.0% hyperopia, respectively. 2. The abnormal refraction eyes were 71.3% positive for simple myopia, 17.9% for myopic compound astigmatism, 6.9% for myopic simple astigmatism, and 2.5% for simple hyperopia, 0.6%for hyperopic compound astigmatism, 0.8%for hyperopic simple astigmatism, respectively. 3.   • To investigate the visual acuity of the women middle and high school students, the visual acuity test was performed the object and subject methods. The results were as follows; 1.   • Recently, the demand for lidar systems for autonomous driving is increasing, and research on Shortwave Infrared(SWIR) photodetectors for this purpose is being actively conducted. Most SWIR photodetectors currently being developed are based on InGaAs, and have the disadvantages of complex processes, high prices, and limitations in research due to monopoly. In addition, current SWIR photodetectors use lasers in the 905 nm wavelength band, which can pass through the pupil and cause damage to the retina. Therefore, it is required to develop a SWIR photodetector using a wavelength band of 1400 nm or more to be safe for human eyes, and to develop a material that can replace the proprietary InGaAs. PbS QDs are group 4-6 compound semiconductors whose absorption wavelength band can be adjusted from 1000 to 2700 nm, and have the advantage of being simple to process. Therefore, in this study, PbS QDs having an absorption wavelength peak of 1415 nm were synthesized, and a SWIR photodetector was fabricated using this. In addition, the photodetector's responsivity was improved by applying P3HT and ZnO NPs to improve electron hole mobility. As a result of the experiment, it was confirmed that the synthesized PbS QDs had excellent FWHM characteristics compared to commercial PbS QDs, and it was confirmed that the photodetector had a maximum current change of about 1.6 times.

Study on Anti-allergic Effect and Safety of Bangpung-galgeun-tang (방풍갈근탕(防風葛根湯)의 항알레르기효과에 관한 연구)

  • The purpose of this study was to examine the anti allergic effect in vivo and in vitro, and to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after oral administration with BGT of 0.4 g/kg and 0.8 g/kg for 8 days, and also examined MTT assay, ${\beta}-hexosaminidase$ activity, IL-4 and $TNF-{\alpha}$ from RBL-2H3 and $TNF-{\alpha}$ from Raw264.7 after pre-treatment with BGT of 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml and 2 mg/ml. To ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg /p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. BGT inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. All the concentrations of BGT from 0.25 to 2 mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosaminidase$ release, IL-4 and $TNF-{\alpha}$, and in Raw264.7, $TNF-{\alpha}$ were significantly reduced by treated all concentrations of BGT. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

The Study of Abnormal Refraction Eye on Women Population of University Students in South-East Korea (한국 동남지역 여대생들의 굴절이상 눈에 대한 연구)

  • Purpose: To research the study of abnormal refraction eye on women population of university students in South - East Korea. Methods: Between March 2007 and October 2007, the refraction test of eye glasses wearer was evaluated on women population (367 students, aged 19~22 years) of university in living on Kyung-Nam and Pusan province. Data was analysed with T-test. Results: On the abnormal refraction status, Compound Myopic Astigmatism was 76.72%, simple myopia was 10.90%, mixed astigmatism was 6.27%, respectively. On the prevalence of myopic power, low was 59.57%, moderate was 24.93%, high was 15.49%, respectively. On the myopic equivalent power, the right eye had more increase to compare to left eye. but these was not a statically significant correlation (p<0.5) between the right and left eyes. On the anisometropia of spherical equivalents and cylinder power, most subjects was under 1.0 diopter. On the other hand, the type of astigmatic axis was with-the rule (70.79%), against -the rule (18.41%), and oblique (10.80%). Conclusions: This study identify that the refraction status of abnormal refraction eye on women population in university students in South-East Korea have been more increased prevalence the myopia and astigmatism.

Polydnavirus Replication and Ovipositional Habit of Cotesia plutellae (프루텔고치벌(Cotesia plutellae) 폴리드나바이러스 복제와 산란 습성)

  • An endoparasitoid wasp, Cotesia plutellae, has been used for a biological control agent against the diamondback moth, Plutellae xylostella. It has a symbiotic polydnavirus in their reproductive tract, which is required for its successful parasitization. Here, we measured a specific replication time of the polydnavirus during female development of C. plutellae. We, also, analyzed the reproductive potentials of female C. plutellae under mating or different host conditions. At $25^{\circ}C$, pupal C. plutellae began to develop adult tissues such as compound eyes and wings since day 2. At day 5, all adult tissues including antennae were developed and were ready to emerge. With polyclonal antibody raised against C. plutellae polydnavirus, an immunobloting could confirm virus replication at day 4 during pupal stage. Virus particles could be visualized by transmission electron microscope in the oviduct lumen of day 5 pupae. After adult eclosion, venom gland and ovarian calyx increased in size, though ovarioles did not. Mated females layed large number of eggs (over $60\%$) at first 4 days during their mean longevity of ca. 8 days at $25^{\circ}C$. Unmated females showed less active ovipositional behavior, where all the eggs developed into males. C. Plutellae parasitized both P. xylostella and fall webworm, Hyphantria cunea. However, C. Plutellae developed faster and showed higher successful paarasitization in P. xylostella than in H. cunea.

A Study of Eye Refraction State on Male Old Age (노인 남성의 안굴절 상태에 대한 연구)

  • The old age may experienced the visual acuity decrease according to refraction abnormality and they have a inconvenient real life style on viewing condition. So that, This study can provide the accurate information on the treatment of visual acuity of a male old ages by test of eye refraction state. The test was performed the visual acuity test by the object and subject methods.and the subjects was the Korean male over 55 old age. The eye types were 0.8 % positive for emmetropia, 15.2 % for myopia, and 84 % for hyperopia, respectively. The abnormal refraction eyes were 1.3 % positive for simple myopic astigmatism, 13.9 % for compound myopic astigmatism. 3.4 % for simple hyperopia and 3.1 % for simple hyperopic astigmatism, 77.5 % for mixed astigmatism, respectively. The asix of astigmatisms were 62% positive for against-the-rule astigmatism, 6% for with-the rule astigmatism, and 32 % for oblique astigmatism, respectively. On the total myopic spherical power, the $-0.50{\leq}$spheric equivalent <-2.00 diopter was 84.3%, the $-2.00{\leq}$spherical equivalent <-6.00.diopter was 15.7%. On the total hyperopic spherical power, the $+0.50{\leq}$spheric equivalent <+2.00 diopter was 74.2%, the $+2.00{\leq}$spherical equivalent <+6.00 diopter was 24.8%, and anything over the +6.00 diopter was 1%. On the astigmatic power, the $0.5{\leq}$cylinder <1.00 diopter was 54.1%, the $1.00{\leq}$cylinder <2.00 diopter was 32.4%, and anything over the 2.00 cylinder diopter was 13.5%.
