Development of the Hybrid CFFT Pile (FRP-콘크리트 합성말뚝의 개발)

  • In this paper, new type CFFT (Concrete Filled FRP Tube) was suggested in order to improve the flexural stiffness. Since the existing CFFT was produced by filament winding process, re-bar for concrete may be necessary in order to ensure structural safety under flexure re-bar. In comparison with existing type CFFT, new type CFFT was reinforced by circular shaped pultrusion FRP without re-bar. Filament winding FRP was attached to the outer layer of pultrusion FRP. Structural behavior of new type CFFT filled with concrete (HCFFT) was investigated by the mechanical property test for the component element and the FE analysis. Furthermore, compressive strength of the HCFFT member based on the equation suggested in previous studies.

Bacterial Communities of Biofilms Sampled from Seepage Groundwater Contaminated with Petroleum Oil

  • The diesel-degrading activities of biofilms sampled from petroleum-contaminated groundwaters in urban subway drainage systems were examined in liquid cultures, and the microbial populations of the biofilms were characterized by denaturing gel gradient electrophoresis (DGGE) and 16S rDNA sequence analysis. Biofilm samples derived from two sites (19 K and 20 K) at subway Station N and Station I could degrade around $80\%$ of applied diesel within 20 and 40 days, respectively, at $15^{\circ}C$, and these results were strongly correlated with the growth patterns of the biofilms. The closest phylogenetic neighbor of a dominant component in the 19 K biofilm was Thiothrix fructosivorans strain Q ($100\%$ similarity). Four dominant strains in the 20 K biofilm were closely related to Thiothrix fructosivorans strain Q ($100\%$ similarity), Thiothrix sp. CC-5 ($100\%$ similarity), Sphaerotilus sp. IF14 ($99\%$ similarity), and Cytophaga-Flexibacter-Bacterioides (CFB) group bacterium RW262 ($98\%$ similarity). Three dominant members in the Station I biofilms were very similar to uncultured Cytophagales clone CRE-PA82 ($91\%$ similarity), Pseudomonas sp. WDL5 ($97\%$ similarity), and uncultured CFB group bacterium LCK-64 ($94\%$ similarity). The microbial components of the biofilms differed depending on the sampling site. This is the first report on the isolation of clones highly similar to Thiothrix fructosivorans and Thiothrix sp. from biofilms in petroleum-polluted groundwaters, and the first evidence that these organisms may play major roles in petroleum degradation and/or biofilm-development.

A High-Yielding, Generic Fed-Batch Process for Recombinant Antibody Production of GS-Engineered Cell Lines

  • An animal-component-free and chemically defined fed-batch process for GS-engineered cell lines producing recombinant antibodies has been developed. The fed-batch process relied on supplying sufficient nutrients to match their consumption, simultaneously minimizing the accumulation of by-products (lactate and osmolality). The proportionalities of nutritional consumption were determined by direct analysis. The robust, metabolically responsive feeding strategy was based on the offline measurement of glucose. The fed-batch process was shown to perform equivalently in GS-CHO and GS-NS0 cultures. Compared with batch cultures, the fed-batch technology generated the greater increase in cell yields (5-fold) and final antibody concentrations (4-8-fold). The majority of the increase in final antibody concentration was a function of the increased cell density and the prolonged culture time. This generic and high-yielding fed-batch process would shorten development time, and ensure process stability, thereby facilitating the manufacture of therapeutic antibodies by GS-engineered cell lines.

Biophysical Studies Reveal Key Interactions between Papiliocin-Derived PapN and Lipopolysaccharide in Gram-Negative Bacteria

  • Papiliocin, isolated from the swallowtail butterfly (Papilio xuthus), is an antimicrobial peptide with high selectivity against gram-negative bacteria. We previously showed that the N-terminal helix of papiliocin (PapN) plays a key role in the antibacterial and anti-inflammatory activity of papiliocin. In this study, we measured the selectivity of PapN against multidrug-resistant gram-negative bacteria, as well as its anti-inflammatory activity. Interactions between Trp2 of PapN and lipopolysaccharide (LPS), which is a major component of the outer membrane of gram-negative bacteria, were studied using the Trp fluorescence blue shift and quenching in LPS micelles. Furthermore, using circular dichroism, we investigated the interactions between PapN and LPS, showing that LPS plays critical roles in peptide folding. Our results demonstrated that Trp2 in PapN was buried deep in the negatively charged LPS, and Trp2 induced the ${\alpha}$-helical structure of PapN. Importantly, docking studies determined that predominant electrostatic interactions of positively charged arginine residues in PapN with phosphate head groups of LPS were key factors for binding. Similarly, hydrophobic interactions by aromatic residues of PapN with fatty acid chains in LPS were also significant for binding. These results may facilitate the development of peptide antibiotics with anti-inflammatory activity.

Development of Electronic Mapping System for N-fertilizer Dosage Using Real-time Soil Organic Matter Sensor (실시간 토양 유기물 센서와 DGPS를 이용한 질소 시비량 지도 작성 시스템 개발)

  • It is crucial to know spatial soil variability for precision farming. However, it is time-consuming, and difficult to measure spatial soil properties. Therefore, there are needs fur sensing technology to estimate spatial soil variability, and for electronic mapping technology to store, manipulate and process the sampled data. This research was conducted to develop a real-time soil organic matter sensor and an electronic mapping system. A soil organic matter sensor was developed with a spectrophotometer in the 900∼1,700 nm range. It was designed in a penetrator type to measure reflectance of soil at 15cm depth. The signal was calibrated with organic matter content (OMC) of the soil which was sampled in the field. The OMC was measured by the Walkeley-Black method. The soil OMCs were ranged from 0.07 to 7.96%. Statistical partial least square and principle component regression analyses were used as calibration methods. Coefficient of determination, standard error prediction and bias were 0.85 0.72 and -0.13, respectively. The electronic mapping system was consisted of the soil OMC sensor, a DGPS, a database and a makeshift vehicle. An algorithm was developed to acquire data on sampling position and its OMC and to store the data in the database. Fifty samples in fields were taken to make an N-fertilizer dosage map. Mean absolute error of these data was 0.59. The Kring method was used to interpolate data between sampling nodes. The interpolated data was used to make a soil OMC map. Also an N-fertilizer dosage map was drawn using the soil OMC map. The N-fertilizer dosage was determined by the fertilizing equation recommended by National Institute of Agricultural Science and Technology in Korea. Use of the N-fertilizer dosage map would increase precision fertilization up to 91% compared with conventional fertilization. Therefore, the developed electronic mapping system was feasible to not only precision determination of N-fertilizer dosage, but also reduction of environmental pollution.

Development of an Electronic Nose System for Evaluation of Freshness of Pork (돈육의 신선도 평가를 위한 전자코 시스템 개발)

  • The aim of this study was to develop a portable electronic nose system for freshness measurement of stored pork. An electronic nose system was constructed using seven different MOS sensor array. To determine the quality change of pork with storage time, the samples were divided into ten groups in terms of storage time with an increment of 2 day up to 19 storage days. GC-MS, total bacteria's count (TBC), thiobarbituric acid reactive substance (TBARS), and pH analyses as well as the analysis of the electronic nose system measurement were performed to monitor the freshness change of the samples. To investigate the performance of the electronic nose system for detecting the change of freshness of pork, the acquired signal values of the system were compared with those of GC-MS, TBC, TBARS, and pH analysis values. According to principal component analysis (PCA) and linear discriminant analysis (LDA) with the signals of the electronic nose system for the pork samples, the sample groups were clearly separated into two groups of 1-9 days and 11-19 days, and four groups of 1-3 days, 5-9 days, 11 days, and 13-19 days respectively. The results show that the electronic nose system has potential for evaluating freshness of pork.

Food Component Characteristics of Fish Roe Concentrates Prepared by Freeze-drying (동결건조로 제조한 어류 알 농축물의 식품성분 특성)

  • Fish roe is among the most valuable food resources produced by fisheries. Raw fish roe requires processing for conversion into hygienic, marketable, and consumer-acceptable forms. In this study, to investigate the food compositional characteristics of various types of fish roe, we applied vacuum freeze-drying to prepare fish roe concentrates (FRCs) from roe of Alaska pollack Theragra chlcogramma, bastard halibut Paralichythys olivaceus, and skipjack tuna Katsuwonus pelamis. The FRC yield ranged from 22.7 to 26.7 g/100 g roe. The major constituents of FRCs were protein (65.4-79.6%), moisture (2.8-6.2%), lipids (8.5-18.3%), and ash (4.8-7.2%). Potassium, sulfur, sodium, and phosphorus were the major mineral elements of FRCs, and the major amino acids were aspartic acid (9.0-10.4 g/100 g protein), glutamic acid (13.2-14.5 g/100 g protein), lysine (8.4-8.6 g/100 g protein), and leucine (8.3-9.7 g/100 g protein). Vacuum freeze-dried FRCs differed among fish species in terms of amino acid composition and electrophoresis protein band distribution. Therefore, FRCs are an excellent source of protein nutrition and an appropriate protein fortification material in human foods or animal feed.

  • The key technologies of GNSS receiver for GNSS sensor station are under development as a part of a GNSS ground station in ETRI. This paper presents the GNSS receiver implementation and signal processing result which is implemented based on FPGA to process the Galileo E1 and E5 signal. To verify the working and performance for GNSS receiver which is implemented based on FPGA, live signal received from GIOVE-B which is second test satellite is used. We gather GIOVE-B signal by using prototyping antenna and RF/IF units including IF-component. To verify Galileo E1 and E5 signal processing function from GIOVE-B, FPGA based signal processing module is implemented as a prototyping hardware board.

Simultaneous imaging and radiometric performance simulation for computer generated GOCI optical system with measured characteristics

  • In this study, we report a new Monte Carlo ray tracing technique for estimating GOCI (Geostationary Ocean Color Instrument) radiative transfer characteristics and imaging performance simultaneously. First, a full scale GOCI optical model was constructed with measured characteristics at the component level and placed in the geostationary orbit. An optical model of approximated GOCI target area centered at the Korean penninsular was then built using the USGS coastal line data and representative land and sea surface reflectivity data. The light rays launched from a simulated sun model travel to the Earth surface, where they are reflected and scattered. Some of the light rays that are headed to the GOCI model in the orbit were selected and traced, as they have entered into the GOCI aperture. As they pass through each GOCI optical part, the ray path and intensity are adjusted according to the measured characteristics for reflection, transmission, refractive index and surface scattering. The ray-traced imaging and radiative transfer performance indicators confirm that the computer generated GOCI optical system with measured characteristics can be used for in-orbit operation simulation following the designed measurement sequence. The computational technique and its implications as a operation support tool are discussed.

  • The use of semiconductor devices as a component of eco-friendly motor vehicles has increased and their widespread use as high voltage switches is expected. On the other hand, in the case of high-voltage switches, reliability test equipment is not localized. To test high voltage switches, this paper analyzed the relevant test standards for developing power supplies. In particular, for the automotive semiconductor reliability test, the AEC (Automotive Electronic Council) Q101 was analyzed. Based on that, the standard specifications of the power supply were determined. For the main power circuit, the pull bridge converter was adopted and based on the specification, the circuit parameters were determined and verified by simulation. The interface for the parallel and pattern operation was designed. The characteristics of the power supply were tested.
