Development of Mouthwash Products with Solid Fermented Oriental Medicinal Herb (고체발효 한약재 추출물을 함유한 구강세척제 개발)

  • The purpose of this study was to develop a mouthwash product with solid fermented oriental medicinal herb (OMH). Solid fermentation of magnolia, liquorice, and cnidium by Phellinus linteus mycelium was carried out successfully when 30% water was added to the medium, whereas 10% brown rice powder was required as an extra nutrient for solid fermentation of mint besides water. The amount of total phenol compounds and DPPH radical scavenging activity of OMH increased significantly (P<0.05) upon solid fermentation. Anti-microbial activities of fermented OMH also increased and were approximately 100-fold greater than those of unfermented samples. Oral pathogens such as Staphylococcus epidermis, Streptococcus pyogenes, Candida albicans, or Streptococcus mutans were used for determination of anti-microbial effects of OMH. Formulation of the mouthwash was developed based on the results of the sensory evaluation. Among seven formulas, the best formula chosen by the sensory evaluation was as follows: mouthwash prepared with 0.075% ethanol extract of solid fermented OMH as a main ingredient, 83.64% hot water extract of mint and clove (100:15, v/v) as a mouthwash base component, and other miscellaneous ingredients, including sodium fluoride, menthol, and surfactants. Data from a consumer's preference test with 30 participants, overall acceptance, and willingness to buy the product developed in this study were all significantly higher for the tested mouthwash compared to mouthwash on the market manufactured with OMH but with a different formula. Duration of freshness of the mouthwash after usage as determined by Breath Checker was not significantly different between the two samples, although the duration of our product was slightly longer than that of the commercial product mentioned above.

Evaluation of Spent Mushroom Substrates as Food for White-spotted Flower Chafer, Protaetia brevitarsis seulensis (Coleoptera: Cetoniidae) (버섯 수확 후 배지의 흰점박이꽃무지 사료화 연구)

  • The larva of the white-spotted flower chafer, Protaetia brevitarsis seulensis (Coleoptera: Cetoniidae), is known to have important medicinal properties, such as anti-cancer activity. However, the consumer market for the larvae is depressed because of its high production cost which needs to be reduced. This study was carried out to evaluate two spent mushroom substrates as food source for chafer larvae to improve the productivity and reduce production cost. The larvae were fed with spent substrates of king oyster mushroom (Pleurotus eryngii) (P-SMS) and shiitake mushroom (Lentinula edodes) (L-SMS), with a control of fermented oak sawdust. Effects of the diets on the development of P. brevitarsis seulensis and contents of minor nutrient components in the larvae produced were examined. In the P-SMS diet, the larval rearing period was shorter at 16.2 days, while the rate of larval weight gain and the cocoon weight were higher at 156.3% and 4.1 g, respectively, than that in the L-SMS diet. The pupation rate was 100% and the adult emergence was higher at 93.3% in the P-SMS diet than in the L-SMS diet with no difference to the control diet. In the P-SMS diet, the total nitrogen content during rearing was higher at 10.28% and the minor nutrient component, particularly Fe (145.8 mg/kg), was higher than that in other diets. The oviposition preference of adults showed no significant difference among the diets. Economic analysis showed that P-SMS costs approximately 667,960 Won less per 100 kg of larvae produced, compared to that of the control diet.

Hematologic Complication of Respiratory Virus Infection (호흡기바이러스 감염에 의한 혈액학적 합병증)

  • Objective: Cytopenia is a common hematologic complication of viral infections. However, information regarding hematologic effects of common respiratory virus infections is scarce. This study aimed to evaluate hematologic complications and the clinical course of patients infected with common respiratory viruses. Methods: We retrospectively analyzed 496 patients with respiratory tract infections admitted to the Department of Pediatrics, Wonkwang University Hospital from November 2011 to March 2012 using multiplex real-time polymerase chain reaction to detect the presence of respiratory viruses and hematologic abnormalities. Results: Respiratory viruses were identified in 379 patients. Respiratory syncytial virus (RSV) was most frequently detected (55.7%), followed by influenza A (Flu-A, 23.0%). Further, cytopenia was observed in 35.5% of RSV-infected patients, 25.0% of Flu-A-infected patients, and 34% of patients infected by other viruses. Each virus caused a decrease in 3 blood cell component values, which corresponded with cytopenia frequency. Of the 379 infected patients, 83 had anemia ($9.71{\pm}1.09g/dL$); 46 had neutropenia ($803.70{\pm}263.09cells/mm^3$); and 23 had transient thrombocytopenia ($142,434.78{\pm}86,835.18cells/mm^3$). However, no patient required treatment. A comparison of clinical characteristics between RSV- and Flu-A-positive patients with anemia revealed that RSV-infected patients had significantly longer duration of hospitalization. RSV was detected more commonly in young neutropenic patients, who had a shorter duration of fever. Conclusions: Our findings suggest that infections, particularly RSV and Flu-A, result in varying degrees of cytopenia, which usually improves without treatment and does not affect the clinical course of the infection.

Physical Properties of Organic Vegetable Cultivation Soils under Plastic Greenhouse (유기농 시설채소 재배지 토양의 물리적 특성변화)

  • This study was conducted to determine the effects of organic vegetable cultivation on the soil physical properties in 33 farmlands under plastic greenhouse in Korea. We were investigated 5~8 farms per organic vegetable crops during the period from August to November 2014. The main cultivated vegetables were leafy lettuce (Lactuca sativa L.), Perilla leaves (Perilla frutescens var. Japonica Hara), cucumber (Cucumis sativus L.), strawberry (Fragaria ananassa L.) and tomato (Lycopersicon spp.). We have analyzed soil physical properties. The measured soil physical parameters were soil plough layer, soil hardness, penetration resistance, three soil phase, bulk density and Porosity. The measurement of the soil plough layer, soil hardness and penetration resistance were carried out direct in the fields, and the samples for other parameters were taken using the soil core method with approximately 20 mm diameter core collected from each organic vegetable field. Soil plough layer was average 36 cm and ranged between 30 and 50 cm, and slightly different depending on the sorts of vegetable cultivation. The soil hardness was $0.17{\pm}0.15{\sim}1.34{\pm}1.02$ in the topsoil, $0.55{\pm}0.34{\sim}1.15{\pm}0.62$ in the subsoil. It was not different between topsoil and subsoil, but showed a statistically significant difference between the leafy and fruit vegetables. Penetrometer resistance is one of the important soil physical properties that can determine both root elongation and yield. The increase in density under leafy vegetables resulted in a higher soil penetrometer resistance. Soil is a three-component system comprised of solid, liquid, and gas phases distributed in a complex geometry that creates large solidliquid, liquid-gas, and gas-solid interfacial areas. The three soil phases were dynamic and typically changed in organic vegetable soils under greenhouse. Porosity was characterized as range of $54.2{\pm}2.2{\sim}60.3{\pm}2.4%$. Most measured soils have bulk densities between 1.0 and $1.6gcm^{-3}$. To summarize the above results, Soil plough layer has been deepened in organic vegetable cultivation soils. Solid hardness (the hardness of the soil) and bulk density (suitable for the soil unit mass) have been lowered. Porosity (soil spatial content) was high such as a well known in organic farmlands. Important changes were observed in the physical properties according to the different vegetable cultivation. We have demonstrated that the physical properties of organic cultivated soils under plastic greenhouse were improved in the results of this study.

  • This pot experiment was undertaken to find out the optimum fertilization ratios of total anions to total cations, ${\Sigma}A/{\Sigma}C$, and the optimum application rates of total macro-nutrients in various soil conditions. Soil samples were collected from uncultivated mountains and hills where grassland development was under consideration. 1. The optimum application ratios of ${\Sigma}A/{\Sigma}C$ and the optimum application rates of total macro-nutrients for the high yields of mixed grass-clover sward in various grassland soils were computed by the Hom$\acute{e}$s systematic variation techniqu.e. 2. With respect to the optimum application ratios of ${\Sigma}A/{\Sigma}C$ in fertilization in a mixed grass-clover sward, the grass yield and botanical composition were distinctly proportional to ${\Sigma}A$ wheras the regume yield and botanical composition were proportional to ${\Sigma}C$. 3. The optimum fertilization rates of total macro-nutrients for the high legume yields were depended upon ${\Sigma}A/{\Sigma}C$ ratios. These optimum rates were in proportional to ${\Sigma}C$ ratios and were inversely proportional to ${\Sigma}A$ ratios. 4. The efficiencies of ${\Sigma}A$ and ${\Sigma}C$ in relation to the grass and grass plus legume yields were highest with the low ratios of each other and the low fertilization rates of total macronutrients. The ${\Sigma}A$ effieiency in the legume yield tended to be similar to that of ${\Sigma}A$ related to the grass yield noted above except Daegu soil. The ${\Sigma}C$ efficiency, however, was proportional to the ${\Sigma}C$ ratio, although that was varied with the fertilization rates of total macro-nutrients and with the kinds of soils. 5. The yield of mixed forages, yield component, and botanical composition in a mixed sward were greatly influenced by the ${\Sigma}A/{\Sigma}C$ ratios, the fertilization rates of total macronutrients, and the interactions of ratio and rate noted above. In addition, these effects were generally different and opposite according to grass and legume.

Skin Improvement Effects and Development of Liposome Capsule Technology Using Centella Asiatica Extract Powder (센텔라아시아티카정량추출물의 리포좀 캡슐기술 개발과 피부개선효과)

  • In this study, we report the results of a study on the clinical evaluation of wrinkle improvement by developing a method for liposome of high-purity Centella asiatica extract used in pharmaceuticals and cosmetics, and a cream using the same. In order to make Centellasome-10EX stabilizing centella asiatica extract in liposome lamella vesicle, it could be completed using 5% hydrogenated lecithin and 2% sucrose distearate. The appearance of Centellasome-10EX was a creamy form of low viscosity, the color was pale yellow, and the odor had the inherent odor of the raw material. The pH was about 6.12, the specific gravity was 1.09, and the acid value was about 0.35. The content of the main constituents of centella asiatica extract contained in the liposome vesicle contains 10,800 ppm of asiatic acid, 10,900 ppm of asiaticoside, 6,000 ppm of madecasic acid, and 1,600 ppm of madecassoside, and long-term storage. There was no discoloration even at the time, and it was found that the main component remained stable thermodynamically. To mechanistically analyze the structure of the liposome vesicle of Centellasome-10EX, as a result of observation with a transmission electron microscope (Cryo-TEM), the multilayer vesicles are formed and filled with moisture, and there are 10 to 60 multilayers around it. It was confirmed that the liposome lamella vesicle was formed. As a clinical trial (in-vivo) test, the moisturizing effect of centellasome cream after application for 5 weeks was 28.3%, which was significantly increased compared to placebo. The skin elasticity effect was 13.6%, which significantly increased the moisturizing power than the placebo. The effect of improving fine wrinkles around the eyes was improved by 23.52% compared to placebo cream. Through the results of this study, it was possible to study the formulation and manufacturing method for encapsulation and stabilization of the developed Centellasome-10EX in the liposome vesicle. It is expected that the results obtained through clinical research on the wrinkle improvement effect of the cream using this can be widely used to study skin science in the cosmetic industry and to develop high-quality cosmetics with high efficacy.

Response to Specific Fertilizer on Chestnut Tree (II) -Deficiency of Insolation and Effect of the Compound Fertilizer with MgO Component- (밤나무 시비시험(施肥試驗)(II) -일조량부족(日照量不足)과 MgO의 시비효과(施肥効果)-)

  • During the two-years(1979 to 1980) fertilization trial on 4-year-old chestnut tree plantations, total insolation was diminished in 1980 because of unseasonable weather. In every fertilization treatment plot, especially in the NPK-fertilized plot with magnesium, growth of trees and yield of chestnuts have been increased significantly. The results obtained are as follows: 1) The growth in the NPK-fertilized plot with boron and magnesium was 20 percent higher than in plots fertilized with NPK alone. 2) Although there was some frost damage to trees on November 14, 1979, the 1979 weather was otherwise normal and daily insolation averaged 7 hours from June through september, The 1979 fertilization indices for chesnut yield were 167 for NPK with boron and 207 for NPK with magnesium, as compared with the base index of 100 for NPK alone. 3) In 1980, the second year of the fertilization trial, unseasonable weather decreased the average daily insolation from June through September to 3.8 hours. Under such conditions, the fertilization indices for chestnut yield were 620 for NPK with boron and 741 for NPK with magnesium, and Boron, as compared to the base index of 100 for NPK alone; i.e. the yields of plots treated with NPK and magnesium were 21 percent higher than for plots with NPK and boron and 7 times the yields for plots with NPK alone, But in the trial plot of NPK, yield of chestnut in 1980 decreased compared with 1979, 4) All test plots had natural magnesium levels lower than 0.8me/100gr., and the treatments with NPK and magnesium would have been less apparent on soils with higher magnesium level. The spectacular effects of treatment with NPK and magnesium during periods of low insolation may result from increased chlorophyll production and corresponding increases in active carbon assimilation, which should play an important role in carbohydrate formation.

The optimal conditions to improve retrovirus-mediated transduction efficiency to NIH 3T3 cells (레트로바이러스(retrovirus)의 NIH 3T3 세포로의 유전자 전달효율을 증가시키기 위한 적절한 조건들)

  • Purpose : We tried to assess the optimal conditions to improve low transduction efficiency and their effect on target cells. Methods : Cultured NIH 3T3 cells were incubated with retroviral vectors bearing an enhanced green fluorescent protein (eGFP) gene. We varied the ratio of viral vectors to target cells (1:1-1:8) and the number of transfections (${\times}1$, ${\times}2$), and compared transduction efficiencies. Also, the effects of polybrene on transduction efficiency and viability of target cells were assessed. Transduction of the eGFP gene was evaluated by observing NIH 3T3 cells under a fluorescence microscope and efficiencies were measured by the percentage of eGFP positive cells using FACscan. Results : As the ratio of retroviral vectors to target cells increased, transduction efficiency was greatly improved, from 7% (1:1) to 38% (1:4). However, transduction efficiency did not increase any more when the ratio increased from 1:4 to 1:8. Cells transfected twice showed higher transduction efficiencies than cells transfected once, at a ratio of 1:8. The eGFP gene transduced to NIH 3T3 cells sustained its expression during repeated passages. However, after the third passage (day 9), the percentage of eGFP positive cells began to decline. The degree of this decline in eGFP expression was lower in cells transfected twice than in cells transfected once (P<0.05). The addition of polybrene did not have any toxic effect on NIH 3T3 cells and greatly increased transduction efficiency (P=0.007). In addition to vector component, transduction efficiency was very sensitive to culture confluence. Cells cultured and transfected in 24-well plate showed higher transduction efficiency, although cells cultured in 6- well plate proliferated more (P=0.024). Conclusion : Our data could be used as a basis for retrovirus-based gene therapy. Further study will follow using human cells as target cells.

Factorial Validity of the Korean Version of the Illness Intrusive Rating Scale among Psychiatric Outpatients Mainly Diagnosed with Anxiety or Depressive Disorders (불안 및 우울장애를 주요 진단으로 하는 정신건강의학과 외래환자 대상 한국판 질병침습도 평가척도의 요인 타당도 연구)

  • Objectives : The Illness Intrusiveness Rating Scale (IIRS) is a well-validated self-report instrument for assessing negative impact of chronic illness and/or adverse effects of its treatment on everyday life domains. Although extensive literature probed its psychometric properties in medical illness, little attention was paid for its validity for psychiatric population. This study aimed to test factorial structure of the Korean Version of the IIRS (IIRS-K) in a consecutive sample of psychiatric outpatients. Methods : Data set of 307 first-visit patients of psychiatric clinic at Guri Hanyang univ. Hospital were used. Exploratory and confirmatory factor analysis, internal consistency were tested in IIRS-K. We also checked Spearman's correlation analysis between IIRS-K, Zung's self-report anxiety scale and Zung's self-report depression scale. Results : 76.9% of the patients were with anxiety disorder and depressive disorder. The principal component factor analysis of the IIRS-K extracted three-factor structure accounted for 63.2% of total variance that was contextually similar to the original English version. This three-factor solution showed the best fit when tested confirmatory factor analysis compared to the original IIRS, two-factor model of IIRS-K suggested from medical outpatients, and one-factor solution. The IIRS-K also showed good internal consistency (Cronbach's α=0.90) and good convergent validity with anxiety and depression scales. Conclusions : The IIRS-K showed the three-factor structure that was similar but not identical to original version. Overall, this study proved factorial validity of the IIRS-K and it can be used for Korean clinical population.

Effect of Brown Algae (Undasia pinnatisda)-Noodle on Inhibitory Action of Obesity in SD Rats (미역 (Undasia pinnatisda) 국수가 SD계 흰쥐의 비만 억제작용에 미치는 영향)

  • Our previous studies have demonstrated the biological effects of alginic acid as a brown algae (Undasia pinnatisda) component on inhibitory action of obesity using animal model. This study was designed to investigate the effect of brown algae (BA)-noodles ($10\%,\;20\%$ and $40\%$) on inhibitory action of obesity in Sprague-Dawley (SD) male rats for 4 weeks. Body weight gain in SD rats for 4 weeks resulted in a marked inhibition ($7\%,\;17\%\;38\%$, respectively) by administration of $10\%,\;20\%$ and $40\%$ BA-noodles compared with control group. Total energy intake and feed efficiency in SD rats for 4 weeks resulted in a marked inhibition ($6\%,\;16\%\;37\%$ and $10\%\;20\%\;40\%$, respectively) by administration of $10\%\;20\%$ and $40\%$ BA-noodles compared with control group, but gross efficiency was not observed in these BA-noodle groups. Rohrer index and TM index as an obesity markers in SD rats for 4 weeks significantly decreased ($10\%\;15\%$ and $22\%$, respectively) by administration of $10\%,\;20\%$ and $40\%$ BA-noodles compared with control group. These results suggest that inhibitory effect of obesity in SD rats for 4 weeks may be due to decreases of body fat content resulted in a decrease of feed efficiency and energy intake by these BA-noodles.
