SF3B4 Depletion Retards the Growth of A549 Non-Small Cell Lung Cancer Cells via UBE4B-Mediated Regulation of p53/p21 and p27 Expression

  • Splicing factor B subunit 4 (SF3B4), a component of the U2-pre-mRNA spliceosomal complex, contributes to tumorigenesis in several types of tumors. However, the oncogenic potential of SF3B4 in lung cancer has not yet been determined. The in vivo expression profiles of SF3B4 in non-small cell lung cancer (NSCLC) from publicly available data revealed a significant increase in SF3B4 expression in tumor tissues compared to that in normal tissues. The impact of SF3B4 deletion on the growth of NSCLC cells was determined using a siRNA strategy in A549 lung adenocarcinoma cells. SF3B4 silencing resulted in marked retardation of the A549 cell proliferation, accompanied by the accumulation of cells at the G0/G1 phase and increased expression of p27, p21, and p53. Double knockdown of SF3B4 and p53 resulted in the restoration of p21 expression and partial recovery of cell proliferation, indicating that the p53/p21 axis is involved, at least in part, in the SF3B4-mediated regulation of A549 cell proliferation. We also provided ubiquitination factor E4B (UBE4B) is essential for p53 accumulation after SF3B4 depletion based on followings. First, co-immunoprecipitation showed that SF3B4 interacts with UBE4B. Furthermore, UBE4B levels were decreased by SF3B4 depletion. UBE4B depletion, in turn, reproduced the outcome of SF3B4 depletion, including reduction of polyubiquitinated p53 levels, subsequent induction of p53/p21 and p27, and proliferation retardation. Collectively, our findings indicate the important role of SF3B4 in the regulation of A549 cell proliferation through the UBE4B/p53/p21 axis and p27, implicating the therapeutic strategies for NSCLC targeting SF3B4 and UBE4B.

  • Climate change is becoming increasingly unpredictable. This has led to changes in various systems such as ecosystems, human life and hydrological cycles. In particular, the recent unpredictable climate change frequently causes extreme droughts and torrential rains, resulting in complex water resources disasters that cause water pollution due to inundation and retirement rather than primary disasters. SWAT was used as a watershed model to analyze future runoff and pollutant loads. The climate scenario analyzed the RCP4.5 climate scenario of the Meteorological Agency standard scenario (HadGEM3-RA) using the normal quantitative mapping method. Runoff and pollutant load analysis were performed by linkage simulation of climate scenario and watershed model. Finally, the results of application and verification of linkage model and analysis of future water quality change due to climate change were presented. In this study, we simulated climate change scenarios using artificial neural networks, analyzed changes in water temperature and turbidity, and compared the results of dams with artificial neural network results through W2 model, a reservoir water quality model. The results of this study suggest the possibility of applying the nonlinearity and simplicity of neural network model to Hapcheon dam water quality prediction using climate change.

  • In this paper, samples are collected for individual companies and the factors affecting worker wages (average annual salary) are analyzed in depth. In addition to the average annual salary of workers in a company, we would like to conduct an in-depth analysis of the average annual salary of first-time employees. Specifically, we tried to explain the size of companies, the production structure of capital and labor, and the impact of business on the average annual salary of first-time, college, and graduate school graduates by educational background of first-time employees. The difference in average annual salary of first-time employees of logistics companies is a decisive factor in the main project on sales, and first-time graduates and college graduates provided high salaries in the overseas passenger cargo transportation, port operation, and unloading management, and low salaries in the land cargo and warehouse industries. n addition, Korean logistics companies had relationships between the location of the headquarters, the number of employees, the proportion of labor costs to sales, and the four characteristic factors of the main business on sales. This fact is that Seoul-based foreign passenger cargo transportation, corporate logistics agency, complex transportation, and international transportation relay are large companies, with a low proportion of labor costs and a higher proportion of capital, technology, and information-intensive investments.

Impact of Microbiota on Gastrointestinal Cancer and Anticancer Therapy (미생물 균총이 위장관암과 항암제에 미치는 영향)

  • Human microbiota is a community of microorganisms, including bacteria, fungi, and viruses, that inhabit various locations of the body, such as the gut, oral, and skin. Along with the development of metabolomic analysis and next-generation sequencing techniques for 16S ribosomal RNA, it has become possible to analyze the population for subtypes of microbiota, and with these techniques, it has been demonstrated that bacterial microbiota are involved in the metabolic and immunological processes of the hosts. While specific bacteria of microbiota, called commensal bacteria, positively affect hosts by producing essential nutrients and protecting hosts against other pathogenic microorganisms, dysbiosis, an abnormal microbiota composition, disrupts homeostasis and thereby has a detrimental effect on the development and progression of various types of diseases. Recently, several studies have reported that oral and gut bacteria of microbiota are involved in the carcinogenesis of gastrointestinal tumors and the therapeutic effects of anticancer therapy, such as radiation, chemotherapy, targeted therapy, and immunotherapy. Studying the complex relationships (bacterial microbiota-cancer-immunity) and microbiota-related carcinogenic mechanisms can provide important clues for understanding cancer and developing new cancer treatments. This review provides a summary of current studies focused on how bacterial microbiota affect gastrointestinal cancer and anticancer therapy and discusses compelling possibilities for using microbiota as a combinatorial therapy to improve the therapeutic effects of existing anticancer treatments.

Analysis of Flood Reduction in Downstream Urban Areas for the Storage in Apartment Complex (하류 도심지 침수저감 분석을 통한 공동주택 단지의 우수저류조 계획)

  • Purpose: In this paper, we would like to analyze the growth rate of existing urban immersion in the downstream during large-scale urban development and the degree of reduction in existing urban immersion in the downstream when small excellent storage facilities are planned in apartment complexes. Method: A large-scale sewage model was built using the SWMM model of the U.S. Environmental Protection Agency, and the impact of flooding in existing downtown areas downstream was analyzed through simulation. The built model included the development zone, the existing downtown area downstream, and the entire river basin that discharges rainwater. Result: As a result of calculating and simulating the minimum excellent reservoir capacity for each apartment block in the study target area, it was found that the immersion of 4,893㎥ based on one hour, 25,815㎥ based on two hours, and 55,528㎥ based on three hours in the downstream urban area. Conclusion: As in this study, large-scale flooding simulation considering the existing downtown area in the downstream shows a significant increase in flooding in the downstream, and if excellent reservoir capacity is planned for each apartment block before development and the construction of excellent reservoirs is recommended.

On the Evolution of Hong Kong's Nativism and Its Public Law Solutions (论香港本土主义的流变及其公法应对)

  • Throughout history, there are three clues of dualistic structures for the development of Hong Kong's nativism. First, inward attribution and outward lookingare two paths to the formation of Hong Kong's nativism. In the dualistic framework of "self-others", nativism is formed. The formation path of "outward looking"can be seen everywhere in the construction history of Hong Kong's nativism. It is under the reflection of "two mirrors" with Britain and Chinese mainland that Hong Kong people acquire the concept of "Hong Kong's nativism". Second, there are two aspects of Hong Kong's nativism: economic and cultural aspect and political aspect. With the gradual development of Hong Kong's history, these two aspects come into being and are closely bound up, thus constituting Hong Kong's nativism today. The third clue is the most critical one. The subjectivity of colonization and decolonization are two different forms of Hong Kong people's subjectivity. These three clues run through the whole process of the construction of Hong Kong nativism, and are carried out in three stages of development: "Origin (1960s-1970s): Preliminary Construction of Hong Kong's Nativism", "Development of Hong Kong's Nativism (1980s-1997): Awakening of Political Aspect" and "Formation and Alienation of Hong Kong's Nativism (1997-present): Deformed "decolonization". Along the evolution of Hong Kong's nativism, with the disintegration of colonialism, Hong Kong people have gradually transited from the subjectivity of colonization to the subjectivity of decolonization, but the process of "decolonization" has not been completed up to now.When nativism loses its native complex from the perspective of "inherent in China", and further develops into the "separatism" of anti-constitutional system and anti-national continuity and unity, it will challenge the stability of the relationship between the central government and the Special Administrative Region under the "one country, two systems". At the same time, it will have a greater impact on the political structure and the rule of law system of Hong Kong, and trigger a series of public law problems that need to be solved urgently. In this regard, on the one hand, we should re-clarify the relationship between the central government and the region under the "one country, two systems" in light of the new situation of democratic political development in Hong Kong, and improve Hong Kong's governance mechanism on the basis of the constitution and the basic law; on the other hand, we should actively learn from the German defensive democracy system to systematically interpret, integrate and apply Hong Kong's existing legal resources so as to effectively curb the development of local separatist forces.

Development of transient Monte Carlo in a fissile system with β-delayed emission from individual precursors using modified open source code OpenMC(TD)

  • In deterministic and Monte Carlo transport codes, b-delayed emission is included using a group structure where all of the precursors are grouped together in 6 groups or families, but given the increase in computational power, nowadays there is no reason to keep this structure. Furthermore, there have been recent efforts to compile and evaluate all the available b-delayed neutron emission data and to measure new and improved data on individual precursors. In order to be able to perform a transient Monte Carlo simulation, data from individual precursors needs to be implemented in a transport code. This work is the first step towards the development of a tool to explore the effect of individual precursors in a fissile system. In concrete, individual precursor data is included by expanding the capabilities of the open source Monte Carlo code OpenMC. In the modified code - named Time Dependent OpenMC or OpenMC(TD)- time dependency related to β-delayed neutron emission was handled by using forced decay of precursors and combing of the particle population. The data for continuous energy neutron cross-sections was taken from JEFF-3.1.1 library. Regarding the data needed to include the individual precursors, cumulative yields were taken from JEFF-3.1.1 and delayed neutron emission probabilities and delayed neutron spectra were taken from ENDF-B/VIII.0. OpenMC(TD) was tested in a monoenergetic system, an energy dependent unmoderated system where the precursors were taken individually or in a group structure, and in a light-water moderated energy dependent system, using 6-groups, 50 and 40 individual precursors. Neutron flux as a function of time was obtained for each of the systems studied. These results show the potential of OpenMC(TD) as a tool to study the impact of individual precursor data on fissile systems, thus motivating further research to simulate more complex fissile systems.

Alice Springs Orogeny (ASO) Footprints Tracing in Fresh Rocks in Arunta Region, Central Australia, Using Uranium/Lead (U-Pb) Geochronology

  • This study investigates the age of the surficial rocks in the Arunta region using Uranium-Lead (U-Pb) geochronological dating. Rock samples were collected at four locations, Cattle-Water Pass (CP 1610), Gough Dam (GD 1622 and GD 1610), and London-Eye (LE 1601), within the Strangways Metamorphic Complex and crushed by selFragging. Subsequently, the zircon grains were imaged using Cathodoluminescence (CL) analysis and the U-Pb (uranium and lead) isotope ratios and the chrono-stratigraphy were measured. The imaged zircon revealed an anomalous heterogeneous crystal structure. Ellipses of the samples at locations GD1601, CP1610, and GD1622 fall below the intercept indicating the ages produced discordant patterns, whereas LE1601 intersects the Concordia curve at two points, implying the occurrence of an event of significant impact. For the rock sample at CP1610, the estimated mean age is 1742.2 ± 9.2 Ma with mean squared weighted deviation (MSWD) = 0.49 and probability of equivalence of 0.90; 1748 ± 15 Ma - MSWD = 1.02 and probability of equivalence of 0.40 for GD1622; and 1784.4 ± 9.1 Ma with MSWD of 1.09 and probability of equivalence of 0.37 for LE1601. But for samples at GD1601, two different age groups with different means occurred: 1) below the global mean (1792.2 ± 32 Ma) estimated at 1738.2 ± 14 Ma with MSWD of 0.109 and probability of equivalence of 0.95 and 2) above it with mean of 1838.22 ± 14 Ma, MSWD of 1.6 and probability of equivalence of 0.95. Analysis of the zircon grains has shown a discrepancy in the age range between 1700 Ma and 1800 Ma compared to the ASO dated to have occurred between 440 and 300 Ma. Moreover, apparent similarity in age of the core and rim means that the mineral crystallized relatively quickly without significant interruptions and effect on the isotopic system. This may have constraint the timing and extent of geological events that might have affected the mineral, such as metamorphism or hydrothermal alteration.

  • Floating LiDAR is a system that provides a new paradigm for wind condition observation, which is essential when creating an offshore wind farm. As it can save time and money, minimize environmental impact, and even reduce backlash from local communities, it is emerging as the industry standard. However, the design and verification of a stable platform is very important, as disturbance factors caused by fluctuations of the buoy affect the reliability of observation data. In Korea, due to the nation's late entry into the technology, a number of foreign equipment manufacturers are dominating the domestic market. The west coast of Korea is a shallow sea environment with a very large tidal difference, so strong currents repeatedly appear depending on the region, and waves of strong energy that differ by season are formed. This paper conducted a study examining buoys suitable for LiDAR operation in the waters of Korea, which have such complex environmental characteristics. In this paper, we will introduce examples of optimized design and verification of ship-type buoys, which were applied first, and derive important concepts that will serve as the basis for the development of various platforms in the future.

Analysis of the long-term equilibrium relationship of factors affecting the volatility of the drybulk shipping market (건화물선 해운시장의 변동성에 영향을 미치는 요인들의 장기적 균형관계 분석)

  • The drybulk shipping market has high freight rate volatility in the chartering market and various and complex factors affecting the market. In the unstable economic situation caused by the COVID-19 pandemic in 2020, the BDI plunged due to a decrease in trade volume, but turned from the end of 2020 and maintained a booming period until the end of 2022. The main reason for the market change is the decrease in the available fleet that can actually be operated for cargo transport due to port congestion by the COVID-19 pandemic, regardless of the fleet and trade volume volatility that have affected the drybulk shipping market in the past. A decrease in the actual usable fleet due to vessel waiting at port by congestion led to freight increase, and the freight increase in charting market led to an increase in second-hand ship and new-building ship price in long-term equilibrium relationship. In the past, the drybulk shipping market was determined by the volatility of fleet and trade volume. but, in the future, available fleet volume volatility by pandemics, environmental regulations and climate will be the important factors affecting BDI. To response to the IMO carbon emission reduction in 2023, it is expected that ship speed will be slowed down and more ships are expected to be needed to transport the same trade volume. This slowdown is expected to have an impact on drybulk shipping market, such as a increase in freight and second-hand ship and new-building ship price due to a decrease in available fleet volume.
