Comparative Study of Antioxidant Activity of Apple and Pear Peel

  • Apple and pear are popular fruits consumed in Korea and are common fruit in daily diet. In order to compare the antioxidant activity of the apple and pear peels, total polyphenol contents, total flavonoid contents, $ABTS^+$ free radical scavenging activity, and DPPH free radical scavenging activity were measured from hot water, ethanol, and methanol extracts of the two fruit peels. The total polyphenol and flavonoid contents were highest in 95% methanol extracts of the apple peelsand 70% ethanol extract of the pear peels, respectively. Total polyphenol contents of the pear peels were higher than that of apple peels, and total flavonoid contents of the apple peels were higher than that of pear peels. The apple and pear peels had the highest $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity in 95% methanol extracts and 70% ethanol extracts, respectively. $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity of pear peels was higher than that of apple peels, and the DPPH free radical scavenging activity of apple and pear peels were detected in hot water, 95% methanol, and 70% ethanol extracts, respectively. Ascorbic acid, a synthetic antioxidant used as positive control, had significantly higher scavenging activity than the apple and pear peels. In conclusion, the apple and pear peelshave great potential as natural antioxidants. Therefore, above results should be considered to provide the possibility for the development of high functional antioxidants.

The dynamics of proteinase activity at different stages of toxocariasis in mice (개회충에 감염된 생쥐에 있어서 시기별 단백분해효소 활성의 변동)

  • Toxocariasis is a parasitic zoonosis caused by infestation of humans with larvae of Toxocara canis, the common roundworm in dogs. Two syndromes have been identified : visceral larva migrans and ocular toxocariasis. In this study we were characterized proteinase activity in crude extracts from liver, lung, kidney and heart of mice infected with Toxocara canis and the dynamics of their changes in different stages of disease. The optimal pH was 5.5. In liver of mice infected with Toxocara canis, the maximun activity of proteinase was observed in 5 day post infection. In lung, the activity reached its maximun on 5th day in A group (infected with 100 embryonated eggs), and on 5th week in B group (infected with 50 embryonated eggs). In kidney, the maximum activity was shown at 6th week in A group, and in B group was shown at 10th day. In early infection, the activity reached its maximun in heart of mice infected with Toxocara canis. As we could see, the dynamics of the changes of proteinase activity in mice is similar in the case of the disease with other biochemical and immunological indices observed in toxocariasis.

Differences of Participation in Common Activities and Life Satisfaction in Swedish Senior Cohousing by Longitudinal Analyses : Focus on Differences Between the Years of 2001 and 2010 (스웨덴 노인용 코하우징 주민의 공동활동 참여도와 생활만족도 시계열 차이 : 2001년도와 2010년도 비교)

  • The purpose of this research is to find out the differences of participation in common activities and life satisfaction of Swedish senior cohousing residents between the years of 2001 and 2010 by conducting a longitudinal study. The same questionnaire were surveyed nationwide to 186 residents of 8 Senior $G{\aa}rden$ cohousings in 2001 and 127 residents of 4 +40 cohousings in 2010, resulting in total 313 respondents' data. The results show that the +40 cohousing respondents are more likely than Senior $G{\aa}rden$ cohousing respondents to participate in all common activities, especially common meals and common coffee meetings. However, the subjective evaluation of the quantity as well as the contents of participating in common activities does not statistically differ in the two groups and life satisfaction is similarly high with a few differences. The +40 cohousing respondents are less likely to stay at and more likely to move into age-mixed cohousing if they have a chance to move when compared to their Senior $G{\aa}rden$ cohousing counterparts. The result of this research reveals that the general characteristics of senior cohousing residents have been changed and they have become more likely to be active participants of various common activities and have high life satisfaction.

Vegetation Structure in Otter (Lutra lutra) Home Range of Hwacheon, Gangwon-do (강원도 화천군 수달(Lutra lutra) 서식지의 식생 구조)

  • In order to determine whether vegetation would be one of the factors for the selection of otter home range, vegetation structure and other potential factors were studied in Hwacheon, Korea. Thirteen sites, otter's activity found and not found, were investigated in North Han River and connected tributary streams of Hwacheon-gun. Three types of vegetation were classified by cluster analysis, which is short grass, tall grass and shrub type. Vegetation zone of each channel is composed of either one type, or mosaic of tall grass and shrub type. Short grass type is common in Lake Paro and upper North Han-river where water level is highly variable throughout a year. Therefore, annual species such as Persicaria nodosa, Fimbristylis dichotomam and Chenopodium ficifolium are the most dominant. Shrub type is common at the downstream sites of Jichon stream and along mainstream of North Han River down Lake Paro. A shrub species, Salix koreensis, is the most common. Tall grass type is dominant occupying the most vegetation zone of the tributary channels. Phragmites japonica is absolutely dominant. Due to its dense cover, a few plant species are co-existed. Otter activity was found in all three vegetation types and no marked activity was found at some sites of tall grass type. There is no difference in species composition and physiognomy between tall grass sites with and without otter activity, while it shows significant difference in fish availability between two groups. Overall we found that home range of otters in the region is along the mainstream and downstream of tributary streams with high fish availability in all vegetation types and in various human activity levels.

Identifying Common Daily Activities Performed by Older Adults in the United States and South Korea and Changes in Activity Participation Across the Adult Lifespan in South Korea (미국성인과 한국성인의 공통적 일상활동과 한국인의 생애주기 변동에 따른 활동참여 변화)

  • Objective : This study aimed to identify common activities with similar participation levels between community-dwelling individuals in the United States (US) and South Korea (Study 1), and analyze the changes in activity participation patterns across the adult lifespan in South Korea (Study 2). Methods : We administered the online survey-based Activity Card Sort version 3 (ACS-3) to adults living in the US and South Korea. In Study 1, we computed the average participation level and visualized 100 activities of the ACS-3 from both the US and Korean samples. The average participation level across the four age groups in Study 2 was calculated and visualized to understand the changes in patterns of involvement across the four ACS-3 domains in a Korean sample. Results : In Study 1, data from 161 Americans and 163 Koreans were analyzed. Of the 100 activities, 48 (instrumental: 20; leisure: 13; fitness/health: 6; social: 9) demonstrated similar levels of participation between the two samples. In Study 2, data from 420 Koreans were analyzed and a tendency for decreased participation with age was found in all domains, except for the instrumental domain. Conclusion : Common daily activities may be used as a means of intervention across cultures in occupational therapy. Protective approaches and support are recommended to optimize older adults' participation in daily life.

Studies on the Myofibrillar Proteins from Korean Native Goat (한국재래산양육(韓國在來山羊肉)의 근원섬유(筋原纖維) 단백질(蛋白質)에 관(關)한 연구(硏究))

  • Actomyosin and myofibril were extracted from Korean native Goat muscle with the Weber-Edsall solution. ATPase activities and physiochemical properties were measured. The results obtained were as follows; 1) Mg-activitied ATPase activity of actomyosin and myofibrill from Korean native Goat muscle exhibited a common biphasic response, a typical ATPase pattern, that is high at a low ionic strength and low at a high ionic strength. Actomyosin showed high activity than myofibrill. 2) Mg-activited ATPase activity of actomyosin from muscle increased extraction time 24 hours. 3) EDTA-enhanced ATPase activity of actomyosin was greater than myofibrill and low at the low ionic strength, high at the high ionic strength. The difference of the activity were shown great broad pattern at the after 0.3M KCI concentration. 4) Effect of EGTA on-ATPase activity of myofibrill and actomyosin from muscle was measured, the Mg-ATPase activity was markedly depressed. 5) Solubility of actomyosin from muscle began to solubilize at KCI concentration of 0.28M and solubilized completely at the KCI concentration of 0.3M.

Effects of Mulligan's Mobilization with Sustained Natural Apophyseal Glides on the Paraspinal Muscle Activity of Subjects with Chronic Low Back Pain (Mulligan의 Sustained Natural Apophyseal Glides Mobilization이 만성 요통 환자의 요부근 활성도에 미치는 효과)

  • Purpose: Low back pain occurs and frequently recurs in many people. An imbalance of paraspinal muscle activity can cause low back pain. Mulligan's concept of mobilizations with sustained natural apophyseal glides (SNAG) is a common method for treating low back pain. The purpose of this study was to investigate the effects of this method on paraspinal muscle activity in patients with chronic low back pain. Methods: Twenty-one patients with low back pain participated in this study. Patients were classified using the Oswestry disability index. The subjects' paraspinal muscles were measured by surface electromyography both before and after the SNAG mobilization. We measured the root mean square value of the paraspinal muscles during lumbar flexion and extension. Results: Paraspinal muscle activity in patients with chronic low back pain was different between the right and left sides. Importantly, paraspinal muscle activity significantly decreased after using the SNAG mobilization method. Conclusion: This study shows a difference between paraspinal muscle activity in chronic low back pain patients and finds that the Mulligan's concept of SNAG mobilization is effective at reducing imbalances in paraspinal muscle activity in low back pain patients.

Inhibitory Effect of Metabolites isolated from Streptococcus mutas sp. M-20 on Glucosyltransferase Activity from Streptococcus mutans (Streptomyces sp. M-20 균의 대사물에 의한 Streptococcus mutans의 Glucosyltransferase 활성 억제 효과)

  • Dental caries is one of the most common oral diseases in the world. Glucosyltransferase (Gtase) of Streptococcus mutans (S. mutans) plays an important role in the develo pment of dental caries. For the purpose to develop anti- caries, we examined the effect of metabolites isolated from Streptomyces sp. M-20 on Gtase and the growth of S. mutans. Streptomyces sp. M-20 isolated from Mongolian soil showed 95~96% sequence homology with that of Streptomyces lin- colnensis. The metabolites of Streptomyces sp. M-20 were partially purified by extraction with ethyl acetate, silica gel column chromatography and preparative TLC. Partially purified metabolite, red colored component (MR-20) in ethyl acetate fraction showed potent antibacterial activitiy against S. mutans and inhibitory activity against Gtase purified from S. mutans, while another isolated yellow component (MY-20) showed no activity against S. mutans. The inhibitory activity of MR-20 against Gtase was confirmed by activity staining on SDS-polyacrylamide gel electrophoresis. The concentration of MR-20 for 50% inhibition $(IC_{50})$ against Gtase activity was $60{\mu}g/ml$. These results suggest that MR-20 can be developed for antibacterial agent and anticaries.

Effects of heavy metals and albumin on lysozyme activity

  • Lysozyme is an antibacterial enzyme that is found in most of body fluids. Lysozyme in tears plays a primary role in protecting eye from harmful environments; if lysozyme is degraded or inhibited, eyes are likely to be more vulnerable to bacterial infection. In this study, lysozyme activity was evaluated according to varying concentrations of heavy metals, copper, zinc, cobalt and manganese and light metal, calcium that are frequently found in airborne particulate matters and was assayed using a dye-quenching lysozyme substrate, Micrococcus lysodeikticus. Less fluorescence intensity was observed with increasing amounts of copper, zinc, manganese and cobalt but not with calcium suggesting that these metals have some affinity with lysozyme and inhibit lysozyme activity. When albumin, the second most common protein in tears, was added on the reaction of lysozyme and metals, lysozyme activity was partially restored. This finding suggests that the albumin might protect damage caused by metals on lysozyme. To identify whether the decrease in enzymatic activity was related to structural changes of lysozyme, SDS-PAGE was conducted and only with copper did lysozyme show marked smearing bands on the SDS-gel, meaning that copper degraded lysozyme consistent with the sharpest activity decrease.

Growth Characteristics of Sprouts and Changes of Antioxidant Activities in Common Bean (Phaseolus vulgaris L.) with Cultivated Temperature (재배온도에 따른 강낭콩 싹나물의 재배특성 및 항산화 활성)

  • The changes of growth characteristics and antioxidant activity for selection of optimum germinated temperature on common bean (Phaseolus vulgaris L.). Common beans (IT100888, IT102849, and IT231267) were cultivated at $20^{\circ}C$, $23^{\circ}C$ and $25^{\circ}C$ during 5 days of germination. The range of whole length, hypocotyls length, thickness, abnormal germination and yield rate of sprouts was 7.27~27.62 cm, 3.10~18.86 cm, 1.80~2.27 mm, 5.54~18.34% and 205.95~ 618.71%, respectively. Antioxidant activities of common beans with germination temperature investigated. Common beans (IT100888, IT102849, and IT231267) germinated at $20^{\circ}C$, $23^{\circ}C$ and $25^{\circ}C$ during 5 days, and then extracted with 80% ethanol, and analyzed for total polyphenol content, DPPH and ABTS radical scavenging activity. Total polyphenol content increased from 474 mg GA eq/100 g sample for IT231267 to 1364 mg GA eq/g sample for $23^{\circ}C$ of germination. DPPH radical scavenging activity of IT102849 increased from 189mg Trolox eq./100 g sample ($20^{\circ}C$) to 1073mg Trolox eq./100 g sample ($23^{\circ}C$) also ABTS radical scavenging activity of IT234267 increased from 479 mg Trolox eq./100 g sample ($20^{\circ}C$) to 1134 mg AA eq/100 g ($23^{\circ}C$). These results suggest that germination temperature for increasing antioxidant activities may be $23^{\circ}C$.
