감압증류법의 고구마소주 제조를 위한 식이섬유 분해효소의 선별 (Screening of Dietary Fiber Degradation Enzyme for Making Sweet Potato Soju by Vacuum Distillation)

  • Sweet potato soju(SPS) has been made by vacuum distillation because sweet potato contains much fibrous materials which give high density to sweet potato mash. Generally, the SPS made by vacuum distillation has soft flavors and tastes. If the viscosity of sweet potato mash could be decreased by degradation enzyme, the process and production of SPS making by the method of vacuum distillation may be simplified and easier to distil the fermented sweet potato. Because the fibrous materials of sweet potato contains pectin with methoxyl group, methanol can be produced by fibrous degradation enzyme. For appling the fiber degradation enzymes to sweet potato mash for making SPS, the enzyme should be needed to degrade fibrous material without producing methanol. Special two fibrolytic enzymes are selected from 26 kind of commercial enzymes for the simplified and easier production of sweet potato soju by vacuum distillation, The selected enzyme A and X can degrade the fibrous material pectin of sweet potato without producing methanol. Although the different companies have produced the enzymes, same cellulase has been prepared from Trichoderma. reesei. The viscosity of sweet potato mash treated by the enzymes is decreased by 3 times with comparison to the viscosity of sweet potato mash of control group. The methanol concentration in the vacuum distilled SPS treated with the enzymes is 0.16%. The concentration is similar to that of commercially distilled SPS(0.15%). The result may suggest that the selected cellulases, A and X, can be used to make SPS by vacuum distillation.

  • Deinking is a series of unit operations designed to detach ink from cellulose fibers and separate the dispersed ink from the pulp slurry. Deinking chemicals are process aids that enable expensive mill equipment used in these unit operations to operate more efficiently - often much more efficiently. We propose the blended deinking agent with cellulolutic enzymes and synthetic collector in deinking pulp of conventional alkaline method. The deinking efficiency of old news print in alkaline pH was enhanced with enzyme treatments. The brightness of deinked pulp was increased with less residual ink particles and yield of enzymatic deinked pulp was improved compared to the deinked pulp of conventional alkaline method. Enzymes in biomass were use to Chemical Deinking for reduce environment pollution through surfactant and improve surfactants. examining into compatibility Enzymes and surfactants, these new materials are studied efficiency of deinking efficiency.

Production and Characterization of Multi-Polysaccharide Degrading Enzymes from Aspergillus aculeatus BCC199 for Saccharification of Agricultural Residues

  • Enzymatic hydrolysis of lignocellulosic biomass into fermentable sugars is a key step in the conversion of agricultural by-products to biofuels and value-added chemicals. Utilization of a robust microorganism for on-site production of biomass-degrading enzymes has gained increasing interest as an economical approach for supplying enzymes to biorefinery processes. In this study, production of multi-polysaccharide-degrading enzymes from Aspergillus aculeatus BCC199 by solid-state fermentation was improved through the statistical design approach. Among the operational parameters, yeast extract and soybean meal as well as the nonionic surfactant Tween 20 and initial pH were found as key parameters for maximizing production of cellulolytic and hemicellulolytic enzymes. Under the optimized condition, the production of FPase, endoglucanase, ${\beta}$-glucosidase, xylanase, and ${\beta}$-xylosidase was achieved at 23, 663, 88, 1,633, and 90 units/g of dry substrate, respectively. The multi-enzyme extract was highly efficient in the saccharification of alkaline-pretreated rice straw, corn cob, and corn stover. In comparison with commercial cellulase preparations, the BCC199 enzyme mixture was able to produce remarkable yields of glucose and xylose, as it contained higher relative activities of ${\beta}$-glucosidase and core hemicellulases (xylanase and ${\beta}$-xylosidase). These results suggested that the crude enzyme extract from A. aculeatus BCC199 possesses balanced cellulolytic and xylanolytic activities required for the efficient saccharification of lignocellulosic biomass feedstocks, and supplementation of external ${\beta}$-glucosidase or xylanase was dispensable. The work thus demonstrates the high potential of A. aculeatus BCC199 as a promising producer of lignocellulose-degrading enzymes for the biomass conversion industry.

인삼(人蔘) 사포닌을 비롯한 계면활성제(界面活性劑)가 위장관내(胃腸管內)의 단백질(蛋白質) 가수분해효소(加水分解酵素) 반응(反應)에 미치는 영향(影響) (The Effects of Surfactants Including Ginseng Saponins on the Gastric Enzyme-Catalyzed Hydrolysis)

  • Objectives : This study was conducted to investigate the effects of ginseng saponins and commercial surfactants such as Triton X-100, sodium deoxycholate, and sodium dodecyl sulfate on the gastric enzyme-catalyzed hydrolysis. Methods : Saponins (a surface-active plant component) from fresh ginseng root were extracted to examine its effect on the gastric enzyme-catalyzed hydrolysis. Commercial surfactants such as Triton X-100, sodium deoxycholate, and sodium dodecyl sulfate were also employed in the hydrolysis system to compare their effects with that of the ginseng saponins. The effects of surfactants on the gastric enzyme-catalyzed hydrolysis were measured by using a spectrophotometer. A spectropolarimeter was used to examine the conformational change of enzymes and substrates by the addition of ginseng saponins into the system. Results : Both the tryptic and the peptic digestion of milk casein or eggalbumin were slightly improved with an increase in the amount of ginseng saponins in the system. Triton X-100 showed an effect similar to that of ginseng saponins, while sodium dodecyl sulfate and sodium deoxycholate diminished the hydrolysis. Circular dichroism spectra of enzymes and substrates was significantly changed by the addition of ginseng saponins into the system. Conclusions : These results show that ginseng saponins affect positively the gastric enzyme-catalyzed hydrolysis, and suggest that the digestion of substrates by gastric enzymes is affected by the change of enzyme conformation by ginseng saponins.

Aspergillus niger의 Hemicellulase계 효소에 관한 연구-D-xylanase계 효소의 정제와 재조합 (Studies on Hemicellulase System in Aspergillus niger ( I ) - Purification and reconstitution of D-xylanase -)

  • 본 연구에서는 Aspergillus niger KG79에서 두종류의 D-xylanase를 분리.정제하여 그 특성을 규명하였다. 이 두 종류의 D-xylanase의 물리화학적 및 동력학적 특성은 큰 차이가 없었다. 이들 Xylanase는 D-Xylan으로부터 Xylose, Xylobiose와 Xylotriose를 분해 생성하였다. 그러나 보리짚 Xylan을 기질로 사용했을 경우에는 Xylanase I 은 II보다 측쇄 arabinose를 상당히 빨리 분해하였다. 이들 효소에 의한 Xylan의 분해도는 기질의 종류에 따라 차이가 나서 보리짚 Xylan과 larchwood Xylan의 분해도는 각각 10%와 25%(환원당량) 정도로 나타났다. 순수정제된 Xylanase와 $\beta$-Xylosidase를 사용하여 재조합한 Xylanase계의 기질분해력을 비교한 결과 최적조건에서 보리짚과 larchwood Xylan은 각각 28%와 54%씩 분해 전환되었다. 이러한 결과는 Xylan의 효소분해도의 제한요소는 기질의 물리적 특성의 차이에 기인함을 추정할 수 있었다.

탈지미강 단백질의 가수분해 및 분해물의 특성 연구 (Proteolysis of Defatted Rice Bran Using Commercial Proteases and Characterization of Its Hydrolysates)

  • 쌀부산물인 탈지미강을 상업적으로 사용되는 8가지 protease를 최적화된 조건에서 단일 혹은 혼합 처리하여 수용성 단백질을 분리하였다. 이렇게 분리된 단백질을 Lowry, Kjeldahl 그리고 Gravimetric method 등 총 3가지 방법으로 분석을 한 결과 Protamex, Alcalase, Protease N이 가장 높은 분해율을 나타냈다. 3가지 방법에서 모두 Protamex, Alcalase, Protease N이 가장 높은 분해율을 나타내었고, Gravimetric method의 경우 다른 두 분석방법인 Lowry, Kjeldahl method에 비해 더 높은 단백질 함량을 보였다. 또한 위의 단일처리결과를 바탕으로 3가지 protease를 혼합하여 처리하였을 때 단일효소처리에 비해 상승효과가 나타나는 것을 알 수 있었는데, 이것은 protease의 경우 가수분해 할 수 있는 특정 peptide 혹은 amino acid가 있는데 각각의 protease가 분해하지 못하는 peptide 혹은 amino acid를 서로 분해해줌으로써 상승효과가 나타난 것으로 생각된다. 효소처리를 하여 얻어진 단백질의 사이즈를 알아보기 위해 SDS PAGE를 한 결과 어떠한 밴드도 형성이 되지 않았고 이는 분해된 단백질이 marker의 최소 사이즈인 15 kDa보다 작기때문인 것으로 생각된다. 따라서 일반 단백질보다 사이즈가 작은 polypeptide나 amino acid로써 분해된 것을 뜻하고 실제로 섭취하였을 때는 신체에서 생성되는 단백질 분해효소인 trypsin이나 chymotrypsin의 분해 없이도 쉽게 흡수 할 수 있을 것이라 판단된다. 또한 효소의 종류가 많을수록 총 아미노산의 함량이 높아짐으로써 식품첨가물로써 활용도가 높은 단백질가수분해물로 분해되었음을 확인할 수 있었다.

상업적 응유효소의 탈지유에 대한 단백질 분해 작용 (Comparative Study of Proteolytic Activities of Some Commercial Milk Clotting Enzymes on Bovine Skim Milk)

  • 상업적 단백질 분해 효소에 0.02% $CaCl_2$를 첨가하여 응유 활성화를 시킨 탈지유에 대한 분해 작용의 결과를 요약하면 다음과 같다. 다양한 효소별 가수분해 시간에 따른 가수분해도는 미생물 유래 효소와 trypsin은 pepsin과 papain W-40보다 높은 분해도를 나타냈다. 12% TCA 용액에 가용성인 NPN의 양은 trypsin이 가장 높은 분해도를 나타내었고 rennet과 pepsin이 가장 낮은 분해도를 보였다. 전기영동에 있어서 trypsin과 protease S는 $\alpha$- lactalbumin을 분해하였고 papain w-40은 $\beta$- lactoglobulin을 미약하게 분해하였으며 neutrase 1.5는 90분 이후부터 $\alpha$-lactalbumin과 $\beta$-lactoglobulin을 분해하였다. Rennet과 비교한 전기영동상에서는 rennet에 의해 분해 되지 않은 ${\alpha}_s$- casein과 $\beta$-casein을 trypsin과 protease S가 다량 분해하였고 $\kappa$-casein은 rennet에 비해 papain W-40이 상당 수준의 분해상을 나타내었다. 이상의 결과 가수분해도 및 NPN 양은 trypsin, neutrase 1.5 및 protease S가 다른 효소에 비해 높게 나타났으며, 전기영동상에서는 pepsin과 neutrase 1.5가 rennet과 유사한 경향을 나타내었다.

Trichoderma sp. FJ1의 섬유소폐기물을 이용한 Cellulolytic enzymes의 고생산

  • Trichoderma sp. FJ1의 cellulase 생산을 위한 배지조성의 검토에서, 질소원으로서 0.1% bacto peptone을 사용하였을 때 생산성이 향상되었으며, 순수 상업용기질인 avicel과 섬유소 폐기물인 볏짚의 혼합배양에서 높은 효소생산성이 얻어졌다. 경제적인 효소생산성을 위한 기질로 섬유소 폐기물의 농도와 혼합비를 검토한 결과, 1%농도에서 볏짚과 펄프를 50:50으로 사용했을시 CMCase, xylanase, ${\beta}-glucosidase$, avicelase는 24.3, 38.7, 1.5, 0.6 U/ml가 얻어졌다. 이러한 효소생산성은 타 보고서의 결과보다 동등이상의 우위를 보여주고 있으며 섬유소 폐기물의 생물학적 당화기술에 크게 기여하리라 사료된다.

  • Ferulic acid is a phenolic compound that serves as a major biosynthetic precursor of vanillin in higher plants. We investigated the ability of the 3 commercial enzymes - Ultraflo L, Viscozyme L, and ${\alpha}-Amylase$ - to induce the release ferulic acid from the Ipomoea batatas L. (sweet potato) stem. The rate of release for ferulic acid was optimal when Ultraflo L (1.0%) was used compared with the other enzymes, whereas Viscozyme L was most effective for the release of vanillic acid and vanillin. Thus, these enzymes may be useful for the large-scale production of ferulic acid and other phenolic compounds from sweet potato stem.

Effects of Flower Thinning Formulation on Activities of Digestive Enzymes and Acetylcholine Esterase in Honey bee Apis mellifera

  • The effects of a newly developed flower thinning formulation (FTF) on the vitality of the honey bee Apis mellifera were examined by measuring the activities of various digestive enzymes in adult worker bees. First, direct spraying of the FTF solution did not cause any behavioral changes or lethal effects for the honey bees based on 24 h observation. Second, oral ingestion of a sugar solution containing the FTF did not produce any significant change in the activities of amylase, proteinases, lipase, or acetylcholine esterase (AChE) in the worker bees 6 h or 24 h after treatment. Meanwhile, a commercial formulation containing sulfur compounds showed slightly reduced activities for several digestive enzymes and AChE, although no behavioral disturbance. Thus, the results of the present study suggest that the FTF is not toxic for honey bees, in terms of contact and ingestion. Therefore, this newly developed FTF can be used for flower thinning without any detrimental effects on pollinating insects.
