MICROBIAL TREATMENT OF WEEVIL-INFESTED SWEET POTATO MEAL AS FEED TO BROILERS

  • A feeding trial was conducted to determine the effectivity of microbial treatment on eliminating the toxicity of weevil-infested sweet potato roots, and to assess intake level and performance of broilers fed microbiologically-treated, weevil-infested sweet potato meal. Weevil-infested sweet potato meal was treated with Aspergillus awamori (terpene-degrading fungus), dried, and mixed with other ingredients. One hundred twenty (120) broiler chicks were randomly distributed to treatment diets containing 3 types of sweet potato meal (healthy, weevil-infested, and microbiologically-treated, weevil-infested) incorporated at 2 levels (12% and 24%) in the ration, following the $2{\times}3$ factorial in CRD with 4 replicates per treatment. Voluntary intake was high with healthy sweet potato meal, even at 24% in the ration, especially at later stage of broiler development. Weevil infestation of sweet potato meal, even at 24% in the ration, especially at later stage of broiler development. Weevil infestation of sweet potato roots significantly reduced voluntary intake and broiler performance even at 12% level in the diet, much more at 24% level (p<0.01). Microbial treatment, however, was found to alleviate such problem, especially at 24% level of incorporation (p<0.01). "Toxicity" of weevil infestation, in terms of enlargement of liver and spleen, in the absence of mortality, was only apparent at 24% level of incorporation in the ration. Again, this was minimized by microbial treatment (p<0.01), and is therefore recommended at high levels of incorporating weevil-infested sweet potato meal in broiler diets. Microbial treatment constitutes an added cost, so that economic analyses should be done to find out whether increases in broiler performance, or reduction in the toxic effects of terpenoid compounds, outweigh the cost of treatment before a definite recommendation can be made for its commercial application.

Evaluation of Extruded Pellets Containing Different Protein and Lipid Levels, and Raw Fish-Based Moist Pellet for Growth of Flounder (Paralichthys olivaceus) (단백질 및 지질함량이 다른 부상배합사료와 생사료의 넙치 사육효과 비교)

  • This study was conducted to evaluate the effect of extruded pellets (EP) containing different levels of protein (51%-55%) and lipid (9%-15%) for growth of flounder (Paralichthys olivaceus) comparing with raw fish-based moist pellet (MP). Two replicate groups of 40 fish per each tank (initial mean weight 106 g) were fed one of three experimental EP (EP1, EP2 and EP3) containing different protein and lipid levels, a commercial EP (EP4) and MP for 16 weeks. Survival was not significantly different among all groups. Final mean weight of fish fed MP was significantly lower than that of fish fed EP1, EP2 and EP4 (P<0.05), but not significantly different from fish fed EP3. Feed efficiency of MP-fed fish was significantly lower than fish fed all EP formulations (P<0.05), but no significant difference was observed among the EP groups. Daily feed intake of MP-fed fish was significantly higher than fish fed all EP formulations (P<0.05). Condition factor was not significantly different among all groups. Whole body moisture and crude lipid contents were significantly affected by diet (P<0.05). Growth and feed efficiency of flounder was not affected by EP protein and lipid levels. Dietary formulation used in EP1, EP2 and EP3 can be applied to the practical feeding of flounder.

Optimum Feeding Rates for Growing and Sub-adult Olive Flounder Paralichthys olivaceus Fed Practical Extruded Pellets at High Water Temperature (고수온에서 사육한 성장기 및 미성어기 넙치(Paralichthys olivaceus)의 배합사료 적정 공급률)

  • We investigated the effects of feeding rate on the growth performance of growing and sub-adult olive flounder Paralichthys olivaceus, and the optimum feeding rate at high water temperature (25-31℃). In experiment I, two replicated groups of fish (113 g) were fed a commercial diet (CD) at rates of 0, 0.7, 1.2, 1.7, 2.2, and 2.4% (satiation) body weight (BW) per day for 4 weeks. In experiment II, two replicated groups of fish (313 g) were fed a CD feeding rates of 0, 0.3, 0.5, 0.7, 0.9, and 1.0% (satiation) BW per day for 4 weeks. In experiment I, the weight gain (WG) and specific growth rate (SGR) of fish fed at 0.7 and 0.9% BW per day was significantly higher than that of unfed fish (0%) and fish fed at 0.3, 0.5, and 1.0% (satiation) BW per day. In experiment II, the WG and SGR increased significantly as feeding rate increased from 0 to 2.2% BW per day, but decreased in the satiation group (2.4%). Broken-line regression analysis of WG showed that the optimum feeding rate of growing and sub-adult olive flounder were 1.98% and 0.55% BW per day, respectively, at the high water temperature.

Withdrawal Times of Ciprofloxacin in Oliver Flounder (Paralichthys olivaceus) after Oral Administration (양식넙치에 경구투여한 Ciprofloxacin의 근육조직 잔류량을 이용한 휴약 기간 설정 연구)

  • Ciprofloxacin is the most commonly used fluoroquinolones for treating bacterial disease in olive flounder (Paralichthys olivaceus) farming, but its withdrawal time for industrial-scale farming has not been established. The depletion of ciprofloxacin was investigated in the olive flounder under field conditions. Fish were kept in an inland fish farm and fed a commercial diet containing 5 mg/kg of ciprofloxacin for 13 days. Seven fishes per sampling time were examined during and after the treatment. Ciprofloxacin and its major metabolite, enrofloxacin, were analyzed using HPLC with a fluorescence detector. The concentrations of ciprofloxacin and enrofloxacin in muscle increased during the medication period, and then decreased rapidly. The ciprofloxacin and enrofloxacin concentration in the olive flounder peaked on days 11 and 13, respectively, with maximum concentrations in muscle of 0.58 and 0.73 mg/kg. Residual ciprofloxacin and enrofloxacin were eliminated rapidly; 6 days after treatment, the respective levels in muscle were 0.04 and 0.10 mg/kg and neither was detected 15 days post treatment. The level of ciprofloxacin accumulation at the beginning of oral administration was variable according to the farming conditions, but the overall exhaustion time was similar. In conclusion, an adequate withdrawal period for enrofloxacin is 15 days in the case of oral ciprofloxacin administration.

A Review of the Optimum Feeding Rates and Feeding Frequency in Korean Rockfish Sebastes schlegeli Reared at Seven Different Water Temperatures

  • Seven feeding trials were conducted to determine the effects of feeding rate and frequency in the Korean rockfish Sebastes schlegeli at seven different water temperatures. Two feeding-rate experiments for 5-g Korean rockfish at $17^{\circ}C$ and $20^{\circ}C$; three feeding rate experiments for 16-g Korean rockfish at $16^{\circ}C$ and $24^{\circ}C$; and finally, two feeding frequency experiments for 93- and 100-g at $15^{\circ}C$ and $20^{\circ}C$ were conducted. Twenty fish averaging $5.5{\pm}0.2$ g ($mean{\pm}SD$) were randomly distributed into 18 indoor tanks containing 40-L seawater from a semi-recirculation system. Fish were fed a commercial diet for 4 weeks at water temperatures of $17^{\circ}C$ and $20^{\circ}C$. Two feeding trials were conducted to determine the optimum feeding frequency in growing Korean rockfish reared at temperatures of $15^{\circ}C$ and $19^{\circ}C$. Broken line regression analysis of weight gain (WG) indicated that the optimum feeding rates of 5-g growing Korean rockfish were 4.48% (BW/day; BW, body weight) at $17^{\circ}C$ and 4.83% (BW/day) at $20^{\circ}C$. Broken line regression analysis of WG showed optimum feeding rates of 16-g juvenile Korean rockfish of 3.41% (BW/day) at $16^{\circ}C$, 3.75% (BW/day) at $20^{\circ}C$, and 3.34% (BW/day) at $24^{\circ}C$. The biological performance, results, along with morphological indices and serological characteristics, suggest that a feeding frequency of 1 meal/day was optimal to improve WG in growing Korean rockfish grown from 93 to 133 g at a water temperature of $15^{\circ}C$, and 100 to 132 g at $19^{\circ}C$.

  • This investigation aimed to determine the relative merit of osseointegration in immediate and delayed implantation in the dog mandible using radiography and bone scintigraphy. five adult mongrel dogs with a mean weight of 8.5 kg were used in this investigation. During the entire study period. all dogs were fed with a soft commercial diet and water ad libitum to minimize functional loading of the implant. Twenty titanium alloy systems 4 mm in diameter and 10 mm in length blasted with calcium phosphate were prepared for insertion. The second and third left mandibular premolars in each dog were extracted for the delayed implant insertion. Twelve weeks later, the second and third right mandibular premolars were extracted for the immediate implant insertion. Before the delayed and immediate implantation procedures and 0, 4, 8, and 12 weeks after the insertions, radiography and bone scintigraphy were conducted. Bone scans were obtained using a large field of view gamma camera equipped with a collimator about 3 hours after intravenous injection of Tc-99m-MDP to the dogs. All the dogs were evaluated weekly for inflammation, necrosis, and other of the bone or sort tissue. Significant macroscopic lesions were not detected. Radioisotope scintigraphy with Tc-99m-MDP hat proved to be a reliable method for measuring increased bone activity at specific skeleton tissue sites. In conclusion, osseointegration in peri-implant bone did not differ significantly between the immediate and delayed implant procedures during the experimental period. The immediate implant may be an alternative treatment of implant insertion in animals.

Improvement of Growth and Survival Rate in Larval and Juvenile Rockfish (Sebastes schlogeli) from Mother Fish in Vitellogenesis Injected with 3,5,35-triiodo-L-thyronine($T_3$) (난황형성기 모체내 3,5,35-triiodo-L-thyronine ($T_3$) 주사에 의한 조피볼락 자치어의 성장과 생존율 향상)

  • The growth and survival rate of larvae and juveniles from female rockfish (Sebastes schlegeli) broodstock in vitellogenesis by injection of 3,5,3'-triiodo-L-thyronine $T_3$ with a dose of 20 mg/kg fish wt. were examined for 30 days and compared with sham-control fish injected dime-thyl sulfoxide and control fish. Larvae were fed with rotifers for first 5 days after parturition, rotifers and Artemia nauplii for next 10 days. And then, they were fed with Artemia nauplii and commercial diet. Growth of larvae and juveniles from maternal $T_3$injection was significantly faster than that of two controls, nevertheless, the condition factor was lower than that of controls. The whole body proximate analyses indicated that there were significant effects of $T_3$ injection on protein and lipid, but no significant on moisture and ash contents. Survival rate of the fish from maternal $T_3$injection was significantly higher than that of fish from the two controls. These results indicate that thyroid hormone supplements appear to confer a distinct advantage to larval and juvenile rockfish in early, fragile development stage.

Anti-oxidative Properties of Lipids Extracted from Rainbow Trout (Oncorhynchus mykiss) Fed with Carotenoids and Conjugated Linoleic Acid

  • A commercial diet supplemented with carotenoids and conjugated linoleic acid was fed to rainbow trout (Oncorhynchus mykiss) for 8 weeks. To investigate the anti-oxidative properties of these compounds, lipids from the muscle and viscera of the fish were subjected to different assays. At $10\;{\mu}g/mL$, L-ascorbic acid exhibited 95% 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity, while the tissue lipids showed little radical scavenging activity. At 50 and $100\;{\mu}g/mL$, the lipids of the muscles and viscera showed 11.7-22.6% and 11.3-24.9% DPPH radical-scavenging activity, respectively. A lipid peroxidation inhibitory assay using the ferric thiocyanate method was also performed in comparison with $\alpha$-tocopherol at a concentration of 6.0 mg/mL. Our results indicate that the anti-oxidative property of the lipids in fish muscle, which was 85.2% compared to 85.3% for the visceral lipids, was stronger than that of $\alpha$-tocopherol (74.3%) following 3 days of storage at $40^{\circ}C$.

Carcass Fat-free Lean Gain of Chinese Growing-finishing Pigs Reared on Commercial Farms

  • Five regions and 258 pigs were selected for this study: North (Beijing), Central (Wuhan), South (Guangzhou), Southwest (Chongqing), Northeast (Harbin). Five typical genetics of growing-finishing pig were selected: Landrace${\times}$Large White${\times}$Beijing Black, Duroc${\times}$Landrace${\times}$Large White, Duroc${\times}$Large White${\times}$Landrace, Landrace${\times}$Rongchang, Landrace${\times}$Harbin White, respectively at each sites. The basal diet was a corn-soybean meal containing sufficient nutrients to meet requirements. Carcass fat-free lean gain was determined by dissecting and analyzing chemical composition of the carcass. Cubic function fitted lean moistures to live weights better than other functions. Exponential function fitted lean lipids to live weights equally to allometric function. Carcass fat-free lean gain of Duroc${\times}$Large White${\times}$Landrace, Landrace${\times}$Large White${\times}$Beijing Black, Duroc${\times}$Landrace${\times}$Large White, Landrace${\times}$Harbin White, Landrace${\times}$Rongchang from 20 to 100 kg of average body weight was 259 g/d, 261 g/d, 311 g/d, 220 g/d, 200 g/d, respectively. All are lower than intermediate fat-free lean gain in NRC (1998).

Induction of Changes in Morphology, Reactive Nitrogen/Oxygen Intermediates and Apoptosis of Duck Macrophages by Aflatoxin B1

  • The purpose of this study was to investigate the effects of aflatoxin $B_1$ ($AFB_1$) on the ultracellular morphology alteration, apoptosis induction and reactive nitrogen and oxygen intermediates production of peritoneal macrophages (DPM) from mule ducks. The ducklings were purchased from a commercial hatchery, and were fed a corn-soybean based diet. As the ducklings were grown up to 3 wk of age, the Sephadex-elicited peritoneal exudative cells (PEC) were used as the source for duck peritoneal macrophages. The ultracellular morphology study showed that significant number of cells shifted from category I (normal cell with ruffled membrane) and II (cell membrane blebbing) to category III (cell membrane blebbing and even rupture) after DPM were incubated with $AFB_1$ ($20{\mu}g/ml$) for 12 to 48 h. When DPM were exposed to $AFB_1$ in vitro, the production of NO, $H_2O_2$ and $O_2{^-}$ in macrophages was reduced after 12-48 h incubation with previous LPS stimulation. There was a DNA laddering pattern observed in DPM incubated with $AFB_1$ 5, 10, 20, 50 or $100{\mu}g/ml$ for 12 h. Evidence also revealed that the percentage of apoptotic cells was increased along with the elevation of $AFB_1$ concentration. The results suggest that $AFB_1$ exposure causes duck macrophages going on apoptotic pathway through evidence of ultracellular morphology alteration and DNA laddering in agarose electrophoresis. The production of reactive nitrogen and oxygen intermediates of duck macrophages also depressed after $AFB_1$ exposure, and this implied that $AFB_1$ could cause deteriorated functions of bacteriocidal and tumoricidal activity in duck macrophages.
