Optimization of In Vitro Murine Embryo Culture Condition based on Commercial M16 Media

  • In vitro culture of murine embryos is an important step for in vitro production systems including in vitro fertilization and generations of genetically engineered mice. M16 is widely used commercialized culture media for the murine embryos. Compared to other media such as potassium simplex optimization medium, commercial M16 (Sigma) media lacks of amino acid, glutamine and antibiotics. In the present study, we optimized M16 based embryo culture system using commercialized antibiotics-glutamine or amino acids supplements. In vivo derived murine zygote were M16 media were supplemented with commercial Penicillin-Streptomycin-Glutamine solution (PSG; Gibco) or MEM Non-Essential Amino Acids solution (NEAA; Gibco) as experimental design. Addition of PSG did not improved cleavage and blastocyst rates. On the other hand, cleavage rate is not different between control and NEAA treated group, however, blastocyst formation is significantly (P<0.05) improved in NEAA treated group. Developmental competence between PSG and NEAA treated groups were also compared. Between two groups, cleavage rate was similar. However, blastocyst formation rate is significantly improved in NEAA treated group. Taken together, beneficial effect of NEAA on murine embryos development was confirmed. Effect of antibiotics and glutamine addition to M16 media is still not clear in the study.

Compressional Behavior of Carbon Nanotube Reinforced Mesophase Pitch-based Carbon Fibers

  • The tensile-recoil compressional behavior of the carbon nanotube reinforced mesophase pitch (MP)-based composite carbon fibers (CNT-re-MP CFs) was investigated by using Instron and SEM. The CNT-re-MP CFs exhibited improved, or at least equivalent, compressive strength as compared with commercial MP-based carbon fibers. Particularly, when CNT of 0.1 wt% was reinforced, the ratios of recoil compressive strengths to tensile strength of CNT-re-MPCFs were much higher (the difference is at least 10 % or higher) than those for the commercial counterparts and even than those for PAN-based commercial carbon fibers. FESEM micrographs showed somewhat different fractography from that of a typical shear failure as the CNT content increased.

  • Among the exhaust gas, Carbon dioxide which is a causative factor in greenhouse effect. We study for synthesis of propylene carbonate with carbon dioxide which is captured and utilized in commercially valuable. The Experiment was proceeded as pilot scale with using homogeneous organic catalyst which is able to produce propylene carbonate in commercial and reaction conditions. Optimization condition for concentration of catalyst and reaction temperature, pressure was studied. We confirm that this process is eco-friendly method and commercial application due to the mild condition and also catalyst has a competitive price, reusability.

  • Today diving activity and technology are indispensable for underwater technology and offshore engineering. And then diving business is still more important to business of subsea. This study surveyed the issue of diving activity in Korea as for compared foreign diving activity. This paper analyzed the status of diving business that include commercial and sport-leisure diving business and development of commercial divers in Korea. This paper reputed the object, the necessity and the character of commercial divers. For the purpose of this study is considered in order to analyze the environment of commercial divers and propose the planning for industrial diving business. Under the present circumstance in Korea this development planning study may be restricted. but the effective planning to activate Korean commercial diving business is abstracted as follows; First, establishment of new course, grade, certificate and licence test for diver is a pre-requisite factor for the successful enactment. Second, the political assistance of concerned government authority is needed. Third, establishment of Korean diving school and medical center for divers are strongly required. Forth, setting up the department of diving activity in university.

  • The change of shopping environment created new emerging type of shopping center after 1990's, Urban Entertainment Center(UEC) in northen America. One hand traditional shopping center was retail-centered, the other hand UEC is entertainment-centered and offers the trinity of synergy. Each components, that are retail, dining and entertainment, play a role of drawing people, extending duration of visiting and making people revisit then the synergy makes commercial profit in shopping center. As northen America many of shopping centers with complex have been built in Korea since 2000 and some projects is planning by the change of shopping environment and regenerating urban. Although the term of "UEC" is used in Korea, it seems to be added entertainment facilities to shopping center without considering on commercial strategy. This study will take a look at mix and duration of visit which ULI stresses in UEC development and comparing with those of Yongsan Station and Chyeongrangri Station which are built recently in seoul, it will get characters and situation of these UECs. Finally, the analysis is to be used as a planning data in UEC development.

Fabrication and Characterization of High-activity Pt/C Electrocatalysts for Oxygen Reduction

  • A 20 wt % Pt/C is fabricated and characterized for use as the cathode catalyst in a polymer electrolyte membrane fuel cell (PEMFC). By using the polyol method, the fabrication process is optimized by modifying the carbon addition sequence and precursor mixing conditions. The crystallographic structure, particle size, dispersion, and activity toward oxygen reduction of the as-prepared catalysts are compared with those of commercial Pt/C catalysts. The most effective catalyst is obtained by ultrasonic treatment of ethylene glycol-carbon mixture and immediate mixing of this mixture with a Pt precursor at the beginning of the synthesis. The catalyst exhibits very uniform particle size distribution without agglomeration. The mass activities of the as-prepared catalyst are 13.4 mA/$mg_{Pt}$ and 51.0 mA/$mg_{Pt}$ at 0.9 V and 0.85 V, respectively, which are about 1.7 times higher than those of commercial catalysts.

Evaluation of Recombinant SAG1, SAG2, and SAG3 Antigens for Serodiagnosis of Toxoplasmosis

  • Serologic tests are widely accepted for diagnosing Toxoplasma gondii but purification and standardization of antigen needs to be improved. Recently, surface tachyzoite and bradyzoite antigens have become more attractive for this purpose. In this study, diagnostic usefulness of 3 recombinant antigens (SAG1, SAG2, and SAG3) were evaluated, and their efficacy was compared with the available commercial ELISA. The recombinant plasmids were transformed to JM109 strain of Escherichia coli, and the recombinants were expressed and purified. Recombinant SAG1, SAG2, and SAG3 antigens were evaluated using different groups of sera in an ELISA system, and the results were compared to those of a commercial IgG and IgM ELISA kit. The sensitivity and specificity of recombinant surface antigens for detection of anti-Toxoplasma IgG in comparison with commercially available ELISA were as follows: SAG1 (93.6% and 92.9%), SAG2 (100.0% and 89.4%), and SAG3 (95.4% and 91.2%), respectively. A high degree of agreement (96.9%) was observed between recombinant SAG2 and commercial ELISA in terms of detecting IgG anti-Toxoplasma antibodies. P22 had the best performance in detecting anti-Toxoplasma IgM in comparison with the other 2 recombinant antigens. Recombinant SAG1, SAG2, and SAG3 could all be used for diagnosis of IgG-specific antibodies against T. gondii.

Antioxidative Effect of Commercial Lecithin on the Oxidative Stability of Perilla Oil (들기름의 산화안정성에 미치는 레시틴의 산화방지 작용)

  • The antioxidative effects of the six commercial lecithins, tocopherols, citric acid and ascorbyl palmitate on refined perilla oil were inverstigated by active oxygen method ($AOM,\;hrs\;at\;97.8^{\circ}C$) and oven test. Except for the lecithin I (aceton insoluble content 55%), the induction time on perilla oil treated with commercial lecithins at 5% level was longer than that of refined soybean oil. When the concentration of lecithin (0.5, 1, 2.5, 4 and 5%) in perilla oil was increased, enhanced the antioxidative effect at AOM and oven test. Lecithin also showed synergistic effect with the mixtures of tocopherol, citric acid and ascorbyl palmitate. The antioxidative effect of ${\gamma}-rich-tocopherol$ on perilla was higher than that of ${\dalta}-rich-tocopherol$ or mixed tocopherol.

Effects of commercial soils on germination, early growth, and chlorophyll content of Aspilia africana, a medicinal plant

  • Aspilia africana (Pers) C.D.Adams, a plant used for centuries in many African countries to treat diseases such as osteoporosis, malaria, tuberculosis, and diabetes, is of great pharmaceutical interest, yet there is limited scientific literature on its germination and growth. This research paper describes the effects of different commercial soils on the germination, growth, and chlorophyll content of A. africana. The germination parameters assessed included final germination percentage (FGP), mean germination time (MGT), and germination index (GI). Shoot length, leaf number, and fresh and dry weights were some of the parameters used to assess A. africana growth. The FGP was low and did not vary significantly; the MGT was 7 ~ 10 days; and the GI was significantly higher in PPS soil at 4.61 ± 0.332 days. Aspilia africana plants in HS:PPS soil showed the best overall growth, producing the highest mean leaf number (18.00 ± 1.129), longest mean shoot length (202.43 ± 13.451 mm), and highest mean fresh and dry weights (7.08 ± 1.061 g and 0.629 ± 0.112 g, respectively). The highest chlorophyll content in leaves of A. africana under HS:PPS conditions suggested a higher photosynthetic potential of plants in this soil. The best growth performance of A. africana in the HS:PPS soil could be attributed to a higher amount of certain mineral nutrients such as nitrogen, potassium, and phosphorus in the HS:PPS soil compared to the other soil categories. It is unclear why the FGP of A. africana was low and we recommend an exclusive study to investigate this further.

Analysis of Trend on the Planning Characteristics of Internal Streets within Mixed-use Commercial Complexes - Focused on the Mixed-use Commercial Complexes in Seoul City - (복합상업시설 내부가로 계획특성 경향 분석 - 서울시 복합상업시설을 중심으로 -)

  • The internal streets of mixed-use commercial complexes, which are the extension of urban streets, are being changed from a mediating space aiming at movement to a staying-type space according to the change of mixed-use commercial complexes. And they function as a resting place of user with court and plaza, and act as an urban public space, and accommodate an activity arising from the gathering of many people. As a result, a role of new relaxation and community space is played in the center of the city. And the aspect of publicness is brought into relief. Hence, this study is intended to examine the characteristics that the internal streets of mixed-use commercial complexes have, and furthermore, it is intended to show the planning characteristics in the contemporary mixed-use commercial complexes. With regard to the method of research, first, space configuration and role of internal streets was examined according to the change of mixed-use commercial complexes on the basic of precedent research and literature review. And the internal streets of mixed-use commercial complexes were classified into doorway part, traffic line, and open space. And the characteristics of internal streets in the mixed-use commercial complexes through the concept of architectural 'publicness' were derived as accessibility, amenity, perceptibility, openness, connectivity and symbolism. The internal streets of mixed-use commercial complexes should be changed by the space planning to secure continuity with the city on the basis of accessibility and connectivity and to reinforce visual openness in the internal streets in order to improve amenity and perceptibility, accordingly the mixed-use commercial complexes will be also born again as symbolic public space in the city. Variously changing elements to improve publicness very importantly act on the planning of internal streets in the mixed-use commercial complexes, and therefore researches should be continuously carried out.
