Performance Evaluation of Hybrid SBR Aerobic Digestion combined with Ultrasonication by using a Mathematical Model (초음파 결합형 SBR 호기성 소화 모델과 영향 예측)

  • Based on the activated sludge model, a simple aerobic digestion model which represents the aerobic sludge digestion by sequencing batch reactor(SBR) equipped with ultrasonicator was composed and performed in this study. The results are as follows. Aerobic digestion efficiency can be increased by adopting ultrasonic pretreatment. For the 5 days of SRT, 24 % of particulate component is predicted to be removed by ultrasonic pretreatment and aerobic digestion. This is 7 %p higher than that of conventional aerobic digestion. A Hybrid SBR aerobic digestion combined with ultrasonication shows higher digestion efficiency than aerobic digestion and ultrasonic pretreatment system. In case of this hybrid system, the digestion efficiency was predicted up to 49 % when the ultrasonication was performed every 2 hours, repeatedly. However, excessive treatment like every hours of ultrasonication in an aerobic digestion process results in adverse effect on TCOD removal because biomass disintegrated completely and the solubilized COD can not be used for the biomass synthesis again.

  • Sugar beet pulp is an abundant industrial waste material that holds a great potential for bioethanol production owing to its high content of cellulose, hemicelluloses, and pectin. Its structural and chemical robustness limits the yield of fermentable sugars obtained by hydrolyzation and represents the main bottleneck for bioethanol production. Physical (ultrasound and thermal) pretreatment methods were tested and combined with enzymatic hydrolysis by cellulase and pectinase to evaluate the most efficient strategy. The optimized hydrolysis process was combined with a fermentation step using a Saccharomyces cerevisiae strain for ethanol production in a single-tank bioreactor. Optimal sugar beet pulp conversion was achieved at a concentration of 60 g/l (39% of dry weight) and a bioreactor stirrer speed of 960 rpm. The maximum ethanol yield was 0.1 g ethanol/g of dry weight (0.25 g ethanol/g total sugar content), the efficiency of ethanol production was 49%, and the productivity of the bioprocess was 0.29 $g/l{\cdot}h$, respectively.

Comparison of Sewage Sludge Solubilization through Different Pretreatment Methods (전처리 방법에 따른 하수슬러지 가용화 비교연구)

  • The pretreatment process was carried out to solubilize the sewage sludge for enhancing its biodegradability using alkaline treatment, ultrasonic treatment(15kHz), ozone treatment and different combination of these three methods: alkaline followed by ultrasonic as well as ozone, and ultrasonic followed by alkaline. The solubilization efficiency was evaluated based on the SCOD/TCOD ratio and VSS/TS ratio. In results, the proper condition of alkaline treatment was shown as 30meq/l of NaOH, pH 12 and 3hours of reaction time. Solubilization efficiency increased to 17% from initial 2% based on SCOD/TCOD ratio under this condition. In ultrasonic treatment, the higher ultrasonic power, the longer treatment time and the lower sludge volume resulted in higher solubilization respectively. There was a rapid increase in solubilization efficiency after 20 minute, then it was measured as 32% of SCOD/TCOD ratio in 1 hour at a ultrasonic power of 1,300W with 1/sludge. Solubilization efficiencies in combined treatment using alkaline and ultrasonic were 47-53% higher than single treatment at a sonicated time of 1 hour. Ozone treatment followed by alkaline treatment also represented the enhanced solubilization compared to ozone treatment. Therefore, ultrasonic or ozone treatment assisted by alkaline could achieve the short treatment time as well as high solubilizetion efficiency.

Synchronized Synergism Using Ethanol, L-lysine and $NaBH_4$ Glutaraldehyde Treated Porcine Pericardium (글루타르알데하이드 고정 돼지 심낭에서 Ethanol, L-lysine, $NaBH_4$ 병합 처치시 상승효과)

  • Background: Calcification is the most frequent cause of clinical failure of bioprosthetic tissues that are fabricated from Glutaraldehyde (GA)-fixed porcine valve or bovine pericardium. We recently used a multi-factorial approach of employing different mechanisms to investigate how to reduce the calcification of bioprosthetic tissues. The purpose of the present study was to evaluate the synchronized synergism using ethanol, L-lysine and $NaBH_4$ in glutaraldehyde treated porcine pericardium from the standpoint of calcification and tissue elasticity. Material and Method: Porcine pericardium was fixed with 0.625% GA (commercial fixation). An interim step of ethanol (80%; 1 day at room temperature) or L-lysine (0.1 M; 2 days at $37^{\circ}C$) or $NaBH_4$ (0.1 M; 2 days at room temperature) was followed by completion of the GA fixation (2 days at $4^{\circ}C$ and 7 days at room temperature). The tensile strength and thickness of the samples were measured. The treated pericardiums were implanted subcutaneously into three-week old Sprague-Dawley rats for 8 weeks. The calcium content was assessed by atomic absorption spectroscopy and the histology of the samples. Result: The amount of calcium in the pericardium pretreated with ethanol (13.6${\pm}$10.0 ug/mg, p=0.008), L-lysine (15.3${\pm}$1.0 ug/mg, p=0.002) and both (16.1${\pm}$11.1 ug/mg, p=0.012) was significantly reduced compared with the control (51.2${\pm}$8.5 ug/mg). However, $NaBH_4$ pretreatment (65.7${\pm}$61.8 ug/mg, p=0.653) and combined pretreatment that including ethanol, L-lysine and $NaBH_4$ (92.9${\pm}$58.3 ug/mg, p=0.288) were not significantly different from the controls(51.2${\pm}$8.5 ug/mg). Both the combined pretreatment using ethanol and L-lysine (7.60${\pm}$1.55, p=0.76) and the combined pretreatment that included ethanol, L-lysine and $NaBH_4$ (7.47${\pm}$1.85, p=0.33) increased the tensile strength/thickness ratio compared with that of the controls (4.75${\pm}$1.88). Conclusion: The combined pretreatment using ethanol and L-lysine seemed to decrease the calcification of porcine pericardium fixed with glutaraldehyde, as compared to single pretreatment, and it increase the tissue elasticity, but to the degree that showed synchronized synergism. $NaBH_4$ pretreatment seemed to increase the calcification of porcine pericardium, irrespective of whether single or combined pretreatment was used.

Kinetic Studies of Parent Compounds and Its Metabolite by Combined Treatment of Allyl Alcohol with Ethanol in vivo (Allyl Alcohol 및 Ethanol 혼합투여에 의한 혈중 농도 변화 및 독성과의 상관성)

  • Allyl alcohol is metabolized in the liver through two steps, first to reactive acrolein by alcohol dehydrogenase (ADH), subsequently to acrylic acid by aldehyde dehydrogenase (ALDH). Since ethanol could compete the same enzymes to be metabolized in the liver, we have determined the plasma concentrations of allyl alcohol and ethanol followed by combined treatment. Pretreatment of rats with 2g/kg ethanol followed by ip administration of 40mg/kg allyl alcohol increased the lethality significantly. Determination of in vivo blood concentrations revealed that ethanol pretreatment caused the apparent decrease in allyl alcohol clearance, whereas acetaldehyde level in blood increased significantly by allyl alcohol treatment, as determined by head space GC analysis. Treatment of 4-methylpyrazole, an inhibitor of ADH, delayed allyl alcohol elimination significantly and reduced its lethality. Collectively, these findings suggested that reduction of allyl alcohol clearance in the presence oj ethanol was mediated through ADH competitive inhibition.

Effects of Pretreatment Method on the Bonding Strength of 4-META/MMA-TBB Resin to Bovine Dentin (상아질 전처리방법이 4-META/MMA-TBB계 레진의 접착강도가 미치는 영향)

  • The present study investigated the effectiveness of pretreatment on dentin bonding. The adhesive resin was 5% 4-methacryloyloxyethyl trimellitate anhydride (4-META) in methyl methacrylate (MMA) combined with poly-MMA powder. Polymerization of this resin was initiated by tri-n-butyl borage (TBB). Ground bovine dentin samples were etched with either an aqueous solution of 10% citric (10-0 solution) (Group I) or aqueous solution of 10% citric acid and 3% ferric chloride(10-3 solution) (Group ll ). After etching, the primer (an aqueous solution of 35% hydroxyethyl methacrylate (HEM- A) and 5% glutaraldehyde was applied on the differently etched surfaces (Group III , Group IV). The 10-0 treatment showed the lowest tensile bond strength, followed by the 10-3 treatment, primer application after the 10-0 treatment and primer application after the 10-3 treatment. The relationship among the surface morphology after pretreatment, fractured surface morphology and tensile bond strength was examined. It revealed that the surface morphology change by different pretreatment influenced the bond strength and the resulting fractured surface morphology. The correlation of tensile bond strength with the fracture morphology was explained.

  • Reverse osmosis seawater desalination facilities can extend the cleaning cycle and replacement time of the reverse osmosis membrane by pretreatment process. Selection of pretreatment process depends on water quality. It was attempted in this study to select approriate pretreatment process for the Masan bay, which was high in particles and organic content. For this purpose, performances of pretreatment processes such as filter adsorber (FA), pore controllable fiber (PCF), and ultrafiltration (UF) were compared based on the silt density index (SDI). The SDI value of the filtrate should be less than 3. The study results showed that UF can produce the filtrate quality satisfying the requirement. However, the transmembrane pressure (TMP) of UF increased quickly, reaching 0.6 bar within 4 days. In order to secure stable operation, FA and PCF were combined with UF. The study results showed that combination of PCF and UF was able to extend the filtration duration (more than 2 months) until to reach TMP of 0.6 bar.

Pretreatment of Kenaf Core by Combined Electron Beam Irradiation and Water Steam for Enhanced Hydrolysis (향상된 가수분해율을 얻기 위한 전자선 조사와 물찜의 복합 전처리공정을 이용한 케냐프 코어 전처리)

  • We have investigated the combined pretreatment of electron beam irradiation (EBI) and water steam as a kenaf core pretreatment process. After each sample was exposed to electron beam dose ranging from 50 to 1,000 kGy, the irradiated sample was treated by water steam using an autoclave for 5-h at $120^{\circ}C$. The pretreated samples were characterized using FTIR-ATR and XRD. FTIR spectra and XRD analysis of nonpretreated and pretreated samples confirm that crystallinity changes were observed before and after the pretreatment. The crystallinity index (CrI) was increased from 50.6% for nonpretreated sample 55.0% for 500 kGy exposed sample. And then, we analyzed sugar yield that is the amount of produced mono-saccharides in pretreated sample by enzymatic hydrolysis; an enzyme activity rate was 70 FPU/mL and 40 CBU/mL, and the loading time was 24, 48 and 72-h. The highest sugar yield was 83.9% at 500 kGy after 72-h for enzymatic hydrolysis. The sugar yield of enzymatic hydrolysis for pretreatment samples was increased as doses are subsequently changed to 100, 200 and 300 kGy, allowing to give 50.8%, 58.6% and 67.9%, respectively.

Optimization of organosolv pretreatment with sulfuric acid for enhancing enzymatic hydrolysis of Pitch Pine (Pinus rigida)

  • The object is to optimize the best condition of organosolv pretreatment process with sulfuric acid as a catalyst. As a material, Pitch pine (Pinus rigida) was ground and sieved through 40-mesh screen, and Celluclast and $\beta$-glucosidase were used as enzymes for enzymatic hydrolysis. Pretreatment processes were carried out in the minibomb, and 20 g of materials with 200 ml of 50% ethanol solution (v/v) with 1% sulfuric acid as a catalyst. Pretreatment temperature was varied from $150^{\circ}C$ to $190^{\circ}C$, and time was varied from 0 to 20 min. Then, residual materials were used for enzymatic hydrolysis. The best conditions were selected by estimating followed enzymatic hydrolysis rate and degradable rates after pretreatment process. The highest value of enzymatic hydrolysis rate was obtained as 55 - 60% at 160 and at $180^{\circ}C$, but the value decreased under more severe conditions. As the residual rates decreased under severe conditions, it infered that the decrease of sugar contents limits enzymatic hydrolysis rates. Combined with enzymatic hydrolysis rate, degradable rates and H-factors, the temperatures at $160^{\circ}C$ for 20 min and at $180^{\circ}C$ for 0 min were concluded as the optimized conditions where have the lowest H-factor value for considering energy input.

  • Pretreatment process consisted of submerged hollow-fiber microfiltration(HMF) membrane and spiral-wound nanofiltration(SNF) membrane has been developed by NETEC, KHNP for the purpose of improving the impurities of liquid radioactive waste before entering Selective Ion Exchange System(SIES). The lab-scale combined system was installed at Kori NPP #2 nuclear power plant and demonstration tests using actual liquid radioactive waste were carried out to verify the performance of the combined system. The submerged HMF membrane was adopted for removal of suspended solid in liquid radioactive waste and the SNF membrane was used for removal of particulate radioisotope such as, Ag-l10m and oily waste because ion exchange resin can not remove particulate radioisotopes. The liquid waste in Waste Holdup Tank (WHT) was processed with HMF and SNF membrane, and SIES. The initial SS concentration and total activity of actual waste were 38,000ppb and $1.534{\times}10_{-3}{\mu}Ci/cc$, respectively. The SS concentration and total activity of permeate were 30ppb and lower than LLD(Lower Limit of Detection), respectively.
