Iron, Copper and Zinc Levels in Human Milk and Estimated Intake of the Minerals by Breast-Fed Infants during the Early Lactation (수유 초기 모유 중 철, 구리, 아연의 함량과 영아의 섭취량 추정)

  • The objectives of the present study were to measure the content of iron, copper and zinc in human milk and to estimate the intake of iron, copper and zinc of breast-fed infants during the early period of lactation. Twenty-five lactating women who delivered in a hospital in Seoul volunteered for the study. Milk samples were collected at day 2, 3, 4, 5, 6, 7, 15 and 30 postpartum. The contents of iron, copper and zinc were determined using the atomic absorption spectrophotometer after wet digestion. The intakes of iron, copper and zinc of infants were estimated by multiplization with the infant milk intake reported in our laboratory. The content of iron was 0.58 $\mu\textrm{g}$/g in colostrum, 0.48 $\mu\textrm{g}$/g in transitional milk and 0.39 $\mu\textrm{g}$/g in mature milk while the estimated iron intake of infants was 271, 255 and 259 $\mu\textrm{g}$/day, respectively. The content of copper in the milk was 0.45 $\mu\textrm{g}$/g in colostrum, 0.43 $\mu\textrm{g}$/g in transitional milk and 0.33 $\mu\textrm{g}$/g in mature milk while the estimated copper intake of infants was 210, 229 and 220 $\mu\textrm{g}$/day, respectively. The content of zinc in the milk was 5.24 $\mu\textrm{g}$/g in colostrum, 3.70 $\mu\textrm{g}$/g in transitional milk, 2.93 $\mu\textrm{g}$/g in mature milk while the estimated zinc intake of infants was 2452, 1968, and 1949 $\mu\textrm{g}$/day, respectively. These results suggest that copper and zinc intake of infants are met to RDA but iron is not.

Longitudinal Changes in Calcium and Phosphorus Concentrations of Korean Human Milk (수유기간의 경과에 따른 한국인 인유의 칼슘 및 인 함량의 변화)

  • The influence of stage of lactation on the moisture. total soilds, ash, calcium, phosphorus and Ca/P ratio of human milk was studied. A total number of 32 lactating mothers provided 101 samples collected at 4 consecutive stages of lactation : days o to 5 postpartum(colostrum); days 6 to 10 postpartum(transitional): days 25-35 postpartum(mature). and days 55-70 postpartum(mature). Moisture and total solids concentrations showed a little but significant difference at the 25-35 day stage as compared with those of colostrum and then remained stable, but ash levels decreased significantly with the elapse of lactation period. Calcium levels increased significantly from colostrum to transitional milk stage with a further less significant increase at mature stages. Between colostrum and transitional stages, phosphorus concentration showed a significant increase $(9.9\pm2.4$ vs $15.5\pm4.9$ mg/100 ml, p<0.001)but between the 25-35 day and the 55-70 day stage phosphorus values did not change significantly. Ca/P ratios for colostrum. transitional, the 25-35 day and the 55-70 day stages were 2.28, 1.76, 1.68 and 1.95, respectively. No significant relationship was found between ash. calcium, phosphorus or Ca/P ratio of milk and maternal parity. When modified milk formulas were reconstituted using the total solids of human mature milk as amount of the modified milk formula in 100 ml of infant feeding solution. ash, and Ca/P ratio were similar to those of human milk. However considerably higher concentrations of calcium and phosphorus exist in the reconstitution fluid than those observed in human milk.

Effect of Maternal Nutritional Status on Immunological Substances in Breast Milk of Korean Women (임산부의 영양상태와 모유의 면역물질 함량에 관한 연구)

  • The concentrations of the immunological substances in breast milk and nutritional status were studied in healthy Korean women of middle socioeconomic class. The subjects were recruited at random from obstetric clinics in Seoul. The nutrients intake, prepregnancy BMI, maternal weight gain during pregnancy were studied. The concentrations of lactoferrin(LF), lysozyme(LZ), sIgA, IgG and C3 in colostrum, in transitional milk, and in mature milk, were measured. To elucidate the effect of nutritional status on immunological substances, each components was compared on the basis of either BMI, weight gain, or protein intake. The highest concentrations of the substances were found in colostrum and decreased as lactation progressed. The decline was more prominent in IgG, C3 and sIgA, and less significant in LZ and LF. The colostrum of standard weight gain group showed higher concentrations compared to lower weight gain group. This difference became smaller as the lactation progresses. BMI and nutrient intake status had less significant effect. Lower sIgA was found in lower BMI, in lower weight gain, and lower protein intake groups compared to standard groups, which indicates sIgA is the most affected substance among the measured by nutritional status.

Stability of Immunoglobulin G(IgG) by Heat Treatment (면역단백질 G(IgG)의 열처리에 대한 안정성)

  • This study was carried out to obtain fundamental data when developing new colostrum component fortified milk products. Residual immunoglobulin G (IgG) activities of both IgG fortified milk products under different pasteurization conditions and colostrum fortified milk powder products under different dissolving temperatures were measured. In the study, residual IgG activities of raw milk and IgG (50 mg and 250 mg) fortified milk products were sharply reduced upon increasing the temperature of heat treatment. After the low temperature long time (LTLT) treatment residual IgG activities of raw milk, IgG 50 mg and 250 mg fortified milk products decreased to 79%, 30% and 21.6%, as compared to those before heat treatment respectively. However, almost no residual IgG activities were detected when IgG fortified milk was heated at 95$^{\circ}C$ for 15 sec. There was no significant change in the residual IgG activities of IgG fortified milk powder products upon different dissolving temperatures (30$^{\circ}C$, 40$^{\circ}C$, 50$^{\circ}C$ and 60$^{\circ}C$).

Amino Acid Composition of Human and Cow's Milk (모유 및 우유의 아미노산 조성에 관한 비교연구)

  • The composition of human milk was compared with that of cow's milk. The contents of amino acids were analyzed by amino acid autoanalyzer. The content of glutamic acid in cow's milk was three times as much as that in human milk. The content of essential amino acid in human colostrum was twice as much as that in mature milk.

Variations of Immunoglobulins in Colostrum and Immune Milk as Affected by Antigen Releasing Devices

  • This work was conducted to examine the variation of immunoglobulins (Igs) in serum, immune milk, normal milk and colostrum upon implantation of a new Antigen Releasing Device (ARD). The core of each ARD housed an immunostimulating complex (ISCOM) that was made of adjuvant Quil A and type XIII lipase from a Pseudomonas sp. Each ARD was coated with polylactic acid, known as polylactide, that controls antigen release. Twenty lactating Chinese Holstein cows were divided into 2 groups (n = 10): test group and control group. All cows in the test group were implanted with a single injection in the right iliac lymph node with 3 types of ARDs, which were designed to release the antigens at d 0, 14 and 28 post-implantation. Blood and milk samples were collected from both groups, and colostrum samples were also collected from other post-partum cows in the same farm. Concentrations of $IgG_1$, IgA and IgM in whey and serum were measured by sandwich ELISA. The results showed that the $IgG_1$, IgA and IgM concentrations in serum and whey from the test group were higher than from the control group. Among the three Igs measured, the $IgG_1$ concentration in serum was significantly higher at d 40 after ARD implantation, and the $IgG_1$ concentration in whey peaked at d 9, 17 and 30, which corresponded with release of the antigen. Based on Pearson's correlation between Ig concentration and production parameters, IgA concentration in normal milk was positively correlated with lactation period, which reflected IgA changes during the lactation period in immune milk. In colostrum, $IgG_1$, IgA and IgM decreased abruptly from d 0 to 3, and then decreased slightly. In conclusion, serum $IgG_1$ concentration can be affected by controlled release of the ARD, while whey IgA levels are primarily affected by lactation period. These results may be useful in future studies designed to regulate concentrations of Igs in immune milk.

Analysis of Changes in Colostrum Proteins by Mammalian Species (포유류의 종에 따른 초유 단백질의 변화에 대한 분석)

  • There have been numerous reports indicating that milk proteins influence immune functions. Colostrum refers to the breast milk of mammals, secreted starting from the fourth or fifth day after delivery. It has abundant nutrition for the survival of newborn infants. Most importantly, it contains bioactive substances with growth-stimulating and antibiotic, functions. Thus, the colostrum has various physiological roles. This study measured the differences in the composition of colostrum derived from dairy cattle, hanwoo, porcine, and goat sources. The results showed that immunoglobulin, lactoferrin, lactoperoxidase, serum albumin, IgG heavy chain, and IgG light chain were significantly higher in the colostrum of dairy cattle, hanwoo, and goats, but low in porcine colostrum. There was no significant difference in ${\alpha}_{S2}$-casein, ${\alpha}_{S1}$-casein, ${\beta}$-casein, ${\kappa}$-casein, ${\beta}$-lactoglobulin, and ${\alpha}$-lactalbumin contents until seven days after birth. However, porcine colostrum showed high contents of all proteins from the first day to the second day after delivery.

  • The importance of milk for the growth and health of a newborn offspring is well known. Milkcontains immunoglobulin G (Ig G), Ig A, lactoperoxidase, lactoferin, cytokines, and growth factors.Osteopontin, one of the multifunctional proteins, is secreted by macrophages, T cel, and epithelial cells.bovine milk have not been clarified. The aim of this study was to observe the expression of osteopontin,in bovine milk during the lactation period or bovine mamary glands..Western blot analysis detected thatosteopontin was expressed in bovine milk whey and mamary glands. The expression level of osteopontinin colostrum whey was higher than those in early milk and mature milk whey. Immunohistochemistryshowed that osteopontin was detected in the glandular epithelium and epithelial cels of intralobular ductof mamary glands. These findings suggest that osteopontin transiently shows high expression in colostrumand plays a potential role in the immunological development of breast-fed calves.

  • The fatty acid composition of human milk was determined in 20 milk samples (ten colostrum and 10 mature) obtained at different stages of lactation. Human colostrum contained a lower percentage of total lipids than mature milk. In comparison with mature milk, human colostrum was characterized by a lower percentage of saturated fatty acids, a higher percentage of monounsaturated fatty acids and a higher percentage of extra-long-chain polyunsaturated fatty acid metabolites of both linoleic acid ($\omega$ 6 series) and linolenic acid ($\omega$ 3 series). The linoleic acid levels reported here are considerably higher than those reported previously in Korea. This shift has paralleled an increase in the use of vegetable oils in Korean diet. The human mature milk differed from marketing cow's milk in fatty acid composition, as it had a lower content of short-, medium-and long-chain saturated fatty acids and a higher content of long-chain monounsaturated and polyunsaturated fatty acids.

Evaluation and Characterization of Milk-derived Microvescicle Isolated from Bovine Colostrum

  • Extracellular microvesicles are membranous nano-sized cellular organelles secreted by a variety of cells under normal and pathological conditions and heterogeneous in size ranging from 30 nm to $1{\mu}m$. They carry functional microRNAs that can influence immunity and development. For a particular application of microvesicles, choice of isolation method is particularly important; however, their isolation methods from colostrum in particular have not been described clearly. In this work, differential ultracentrifugation as a conventional method, ultracentrifugation with some modification such as additional precipitations, ultrafiltration, sucrose gradient separation and ExoQuick$^{TM}$ as a commercial reagent were compared. The goal was to compare mainly microvesicular total microRNA yield, distribution and purity among the methods then select the best isolation method for bovine colostrum microvesicles based largely on microRNA yield with the view of applying the vesicles in work where vesicular microRNA cargo is the target bioactive component. Highest yields for vesicular microRNA were obtained using conventional methods and among them, subsequent ultracentrifugation with 100,000 g and 135,000 g conventional method 2 was selected as it had the highest RNA to protein ratio indicating that it pelleted the least protein in relation to RNA an important factor for in vivo applications to assess microvesicle functionalities without risk of contaminating non-vesicular biomaterial. Microvesicles isolated using conventional method 2 were successfully internalized by cells in vitro showing their potential to deliver their cargo into cells in vitro and in vivo in case of functional studies.
