느타리버섯 원형질체내(原形質體內)에 사철느타리버섯 핵(核)의 전이(轉移) (Transfer of Isolated Nuclei from Pleurotus florida into Protoplasts of Pleurotus ostreatus)

  • 원형질체내에 외래유전물질 도입에 의한 균주 개발의 가능성 검토의 일환으로 사철느타리의 원형질체로부터 핵을 분리하여 느타리의 원형질체내로 전이하여 얻은 결과는 다음과 같았다. 1. 핵전이주는 크게 3가지 type으로 구분되었는데 type 1은 아주 빠른 균사생장과 clamp connection를 형성하지 않으며 CM+benomyl에서 균총분리가 나타남으로서 이질이배체로 사료되었다. type 2는 주된 핵전이주로서 모균주와 비슷하거나 다소 빠른 균사생장과 clamp connection을 지녀 자실체를 형성하였으며 heterokaryon으로서 원형질체 융합주와 유사하였다. 그러고 type 3은 핵 또는 염색체 단편의 전이로 clamp connection를 지니지 않고 아주 느리거나 MM에서 생육이 거의 불가능한 균주였다. 2. 핵전이주 및 모균주를 전기영동법으로 esterase 동위효소 pattern을 조사한 결과 핵전이주는 모균주와 구별되었는데 type 1인 이질이배체는 새로운 band를 나타내었으며, type 2와 type 3은 모균주들을 합한 것으로 보였다.

기관 삽관 환자의 구강간호시간에 따른 구강 건강 상태 및 세균집락의 차이 (A Comparison of Different Application Times of Oral Care on Colonies of Microorganisms and Oral Health Status on Intubated Patients)

  • Purpose: The aim of the study was to identify effects of oral care protocol on bacterial floras of the oral cavity and oral health status of intubated patients in an intensive care unit. Methods: The participants were 60 intubated patients who were recruited from an intensive care unit of a university hospital from Dec. 28, 2010 to Mar. 25, 2011. The participants were randomly assigned into 3 groups of 20 patients according to the application time of oral care (1-minute oral care, 2-minute oral care, and 3-minute oral care groups). The numbers of bacterial flora colonies in the oral cavity was assessed before and after the oral care. Oral health status was assessed using a Korean version of the Oral Assessment Guide developed by Elier et al. originally. Results: The numbers of bacterial flora colonies were less after oral care than those before the care, but there was no significant differences among the 3 groups after the care. Oral health status was better after the oral care than it was before the care, but there was also no significant differences among the 3 groups. Conclusion: If well-trained nurses perform oral care for 1 minute using a chlorhexidine swab on intubated patients, the numbers of bacterial flora colonies will be reduced and healthy oral status will be maintained.

느타리버섯과 사철느타리버섯의 종간 원형질체융합 균주의 특성 (Characteristics of Fusion Products between Pleurotus ostreatus and Pleurotus florida Following Interspecific Protoplast Fusion)

  • 식용버섯의 재배와 소비가 증가해가는 중요성에 비추어 새로운 품종 육성을 위하여 국내의 주요 재배 버섯인 느타리버섯과 사철느타리버섯의 원형질체를 융합하여 종간 체세포 잡종을 선발하였으며, 그 특성을 조사한 결과를 요약하면 다음과 같다. 1. 느타리버섯과 사철느타리버섯의 종간 원형질체 융합율은 0.18-3.80%였다. 2. 융합된 균총의 생장은 빠른 것, 중간 그리고 아주 느린 것으로 나눌 수 있었으며 그 형태는 불규칙 하였다. 3. 선발한 40개의 체세포 잡종 모두가 정상적인 자실체를 맺었으며, 자실체의 형태가 모본과는 달리 아주 다양하게 나타났다. 4. 체세포 잡종의 자실체색은 짙은 회색을 가진 느타리버섯과 연한 백색을 가진 사철느타리버섯의 중간색을 나타내었다. 5. 무포자 또는 극히 적은량의 포자를 지닌 균주를 선발하였으며 이들의 자실체 형태는 정상이었다. 6. 수량이 다소 낮은 무포자 균주를 제외하고는 대체적으로 체세포 잡종의 자실체 수량이 모본보다 증가했다.

남극 King Geroge Island 토양의 종속영양 세균 분포상과 효소 활성도 (Distribution of Heterotrophic Bacterial Flora in Soil on the King George Island (Antarctica) and Their Enzyme Activities)

  • 남극 토양생태계 세균의 분포와 세균의 생화학적 특성을 알아보기 위해 King George섬의 세종기지(한국)와 Te-niente Jubany기지(아르헨티나) 주변의 토양 17점을 남극 하계 기간(1989년 2월) 동안 채취하여 종속영양 세균의 수오 채외 효소활성도를 측정하였다. 종속영양 세균수는 시료채취지역과 배양온도가 다른 조건에서 자란 세균의 경우에도 그 평균값이 비숫한 경향을 보이고 있으나 protease는 $4^{\circ}C$에서 자란 세균의 경우 92%로서 특히 높게 나타났다. 배양온도에 따라 매우 변화가 켰으며 산술평균값은 배양온도 $37^{\circ}C$, $25^{\circ}C$, $4^{\circ}C$에서 각각 $2.5\times 10^{4}$, $2.7\times 10^{7}$, $6.9\times 10^{5}$ CFU/$cm^{3}$soil로 나타났다

한국 서식 흰개미의 특성과 방제 (Characteristic of Termite inhabits in South Korea and the control)

  • There are about 2,000 species of termite in the World, but one species of termite inhabits in the southern part of Korean peninsula. Termites are social insects that live in colonies. The colonies are composed of king, queen, soldiers and workers. Termite food consists of cellulose obtained from wood. Protozoa in their digestive tracts convert the cellulose into usable food. Korean termite is a subspecies of Reticulitermes speratus Kolbe, Rhinotermitidae. It's subterranean termite and the scientic name is R. speratus kyushensis Morimoto. The subterranean termite must nest in the soil in order to survival, and infest soft-wood which contact with the soil near the nest. There are several ways which subterranean termite infestations can be noticed. Atcertain times of the year during daylight hours, king and queen termites emerge from the colonies. The propose of these flight is to establish new colonies. The termite is a decomposer of biological ecosystem, but an invader in the preservation of cultural properties as like wood buildings. There are serveral control methods for the prevention of wood building from termite's damages. Those are biological control, ecological control, physical control and chemical control. Ecological and Physical control are the best methods in the new constructing wood-building. Fumigation which is a method of chemical control, is the best method for the building damaged by the termite. After the fumigation, we have totake wood & soil treatments for the building and nearby in order not to be reinvaded by the termites.

Conditioned medium of E17 rat brain cells induced differentiation of primary colony of mice blastocyst into neuron-like cells

  • Background: Conditioned medium is the medium obtained from certain cultured cells and contained secretome from the cells. The secretome, which can be in the form of growth factors, cytokines, exosomes, or other proteins secreted by the cells, can induce the differentiation of cells that still have pluripotent or multipotent properties. Objectives: This study examined the effects of conditioned medium derived from E17 rat brain cells on cells with pluripotent properties. Methods: The conditioned medium used in this study originated from E17 rat brain cells. The CM was used to induce the differentiation of primary colonies of mice blastocysts. Primary colonies were stained with alkaline phosphatase to analyze the pluripotency. The morphological changes in the colonies were examined, and the colonies were stained with GFAP and Neu-N markers on days two and seven after adding the conditioned medium. Results: The conditioned medium could differentiate the primary colony, beginning with the formation of embryoid-body-like structure; round GFAP positive cells were identified. Finally, neuron-like cells testing positive for Neu-N were observed on the seventh day after adding the conditioned medium. Conclusions: Conditioned medium from different species, in this case, E17 rat brain cells, induced and promoted the differentiation of the primary colony from mice blastocysts into neuron-like cells. The addition of CM mediated neurite growth in the differentiation process.

항균티슈와 물티슈 사용에 따른 시간별 세균 수 변화의 차이 (Amount of bacteria over time according to the use of antibacterial and wet wipes behavior)

  • Background: This study was conducted to analyze the time for re-detection of bacteria after surface disinfection using wet wipes, isopropyl alcohol, and benzalkonium chloride antibacterial tissue and provide standards for re-execution of surface disinfection with wet and antibacterial tissues. Methods: Seven laptops were wiped with wet tissue and isopropyl alcohol and benzalkonium chloride antibacterial tissues. Test areas were rubbed with a sterile cotton swab at baseline and after 30, 60, and 120 min. After plating on a tryptic soy agar medium, the number of colonies was counted by culturing at 36.5℃ for 24 h. Results: The average number of bacterial colonies was 5.85 ± 4.33 before isopropyl alcohol wiping and nil after wiping. The average number of bacterial colonies was 12.28 ± 14.67 benzalkonium chloride wiping and nil after wiping. Before wiping with wet wipes, the average number of bacterial colonies on laptop surfaces was 3.42 ± 5.22. Bacteria decreased after wiping with wet wipes but increased again over time. Conclusions: Wet wipes can temporarily reduce bacteria but are unsuitable for removing bacteria.

Simple Method for a Cell Count of the Colonial Cyanobacterium, Microcystis sp.

  • The cell counting of colonial Microcystis spp. is a rather difficult and error-prone proposition, as this genus forms irregularly-shaped and irregularly-sized colonies, which are packed with cells. Thus, in order to facilitate a cell count, four methods of dividing the colonies into single cells were compared, including vortexing, sonication, $TiO_2$ treatment, and boiling. As a result, the boiling method was determined to generate the greatest number of single cells from a colony, and all colonies were found to have divided completely after only 6 min of treatment. Furthermore, no significant cell destruction, which might alter the actual cell density, was detected in conjunction with the boiling method (P=0.158). In order to compute the cell number more simply, the relationship between the colony size and the cell number was determined, via the boiling method. The colony volume, rather than the area or diameter was correlated more closely with the cell number ($r^2=0.727$), thereby suggesting that the cell numbers of colonial Microcystis sp. can also be estimated effectively from their volumes.

Characterization of Alcohol Fermentation and Segregation of Protoplast Fusant of Saccharomyces cerevisiae and Pichia stipitis

  • A study was conducted to investigate the characteristics of segregation and alcohol fermentation of intergeneric fusants. The protoplast fusion of both Pichia stipitis CBS 5776 and Saccharomycess cerevisiae STV 89 was carried out. The fusion frequency was $5\times10^{-8}$ and among fusants selected, a fusant F5 showed the best results in ethanol production by sucrose and xylose fermentations. The performance of xylose fermentation by this fusant was better than that of P. stipitis CBS 5776 and fusant F5 exhibited sucrose fermentation patterns intermediate to the two parent strains. The fusant F5 was segregated into a pair of parental strains during the several culture passages. In the average, 91$%$ of colonies had a similar characteristics of P. stipitis while 7$%$ of colonies resembled S. cerevisiae. Only 2$%$ of colonies had the characteristics of the original fusants. At the sixth passage, all segregants resembled P. stipitis. From these results it is suggested that intergeneric protoplast fusion led to an integration of S. cerevisiae genes, rather than whole chromosomes, within the entire genome of P. stipitis.

Foraging behaviour and preference of pollen sources by honey bee (Apis mellifera) relative to protein contents

  • Background: Pollen is an important source of protein and lipids for many animals including honey bees. In order to understand the foraging behaviour of honey bee colonies and preference among the available floral resources, pollen collections from three experimental healthy colonies of honey bees were analysed in the month of June. Results: The amount of pollen collections were related to the colony's need which was indicated by the number of larval and adult bees present in the hive. Interesting was the sequence of pollen collection from different floral sources. All honey bee colonies collected pollens from Trifolium repens first, then Erigeron annus and the third choice was Coreopsis drummondii and Oenothera biennis flowers. Total protein content of Trifolium pollen was the highest (20.0 g/100 g DM), and the others were in the range of 8.9-11.4 g/100 g DM. Conclusion: The results indicated that the first criteria for honey bee foraging preference of pollens would be the nutritional contents of protein and the resource availability of the lesser nutritious floral sources. This information can help pollinator protection programmes of habitat manipulation using flowering plants for nectar and pollen sources.
