Keratanase II Digestion Accompanied with a Liquid Chromatography/Tandem Mass Spectrometry for Urinary Keratan Sulfate Quantitative Analysis

  • Purpose: Mucopolysaccharidosis IV (MPS IV) is a disease characterized by deficient activity of N-acetylgalactosamine-6-sulfatase (GALNS) causing excessive lysosomal storage of keratan sulfate (KS). The identification of the relevant disaccharide units of KS after keratanase II digestion followed by liquid chromatography/tandem mass spectrometry detection (LC-MS/MS) is validated and applicable for the preliminary diagnosis of MPS IV. Methods: A total of 67 urine samples were collected and analyzed from 11 MPS IV patients comprising 10 MPS IVA and one MPS IVB patients, and 56 normal controls. Urinary glycosaminoglycan was first precipitated by the Alcian blue method followed by a digestion of keratanase II. The protonated species of the digested disaccharide products were detected by using multiple reaction monitoring experiment. Results: One particular disaccharide of KS was selected. The transition mass-to-charge (m/z) of the parent ion and its daughter ion after collision was $462.0{\rightarrow}97.0$, whereas the chondrosine used as an internal standard in this assay was m/z $353.9{\rightarrow}73.0$. The results corresponded well with the two-dimensional electrophoresis method. The quantities of urinary KS were significantly raised in confirmed MPS IV patients when comparing with those of normal controls ($170.2{\pm}81.1$ vs. $4.06{\pm}1.92{\mu}g/mL$). Conclusion: The LC-MS/MS method for MPS IVA determination is specific, sensitive, validated, and applicable for urinary KS quantification. This method can be used not only as a first-line biochemistry examination of MPS IVA, but also as an outcome survey after enzyme replacement therapy.

  • In the coastal area, many kind of vessels are operating and exposed to the various marine accidents. In particular, small-size vessels, which are not equipped with the safety equipment, are in the defenceless state to the marine accidents such as the accident according to vessel's own state caused by the diverse reasons and the collision with the neighborhood vessels and objectives. So it is necessary to resolve these situations through the inexpensive equipment. This paper implements the function that can detect vessel's own motion using Smart Phone. if the motion is over the threshold value assigned, this function decides that the current situation is dangerous for this vessel. So this function informs Smart Phone's ower in the vessel and the control center of this situation. This function can be applied to small-size vessels, such as fishing boating, passenger ship, and leasure boat, which have few the safety equipments, and then improve vessel's safety navigation.

Penetration Depth Computation for Rigid Models using Explicit and Implicit Minkowski Sums (명시적 그리고 암시적 민코우스키 합을 이용한 강체 침투깊이 계산 알고리즘)

  • We present penetration depth (PD) computation algorithms using explicit Minkowski sum construction ($PD_e$) and implicit Minkowski sum construction ($PD_i$). Minkowski sum construction is the most time consuming part in fast PD computation. In order to address this issue, we find a candidate solution using a centroid difference and motion coherence. Then, $PD_e$ constructs or updates partial Minkowski sum around the candidate solution. In contrast, $PD_i$ constructs only a tangent plane to the Minkowski sums iteratively. In practice, our algorithms can compute PD for complicated models consisting of thousands of triangles in a few milli-seconds. We also discuss the benefits of using different construction of Minkowski sums in the context of PD.

  • This paper presents an efficient modeling system of virtual pottery in which the user can deform a body of virtual clay with a haptic tool for E-learning. We propose a Circular Sector Element Method (CSEM) which represents the virtual pottery with a set of circular sector elements based on the cylindrical symmetry of pottery. Efficient algorithms for collision detection and response, interactions between adjacent elements, and GPU-based visual-haptic synchronization are designed and implemented for the CSEM. Empirical evaluation showed that the modeling system is computationally efficient with finer details and provides convincing model deformation and force feedback. The developed system, if combined with educational contents, is expected to be used as an effective E-learning platform for elementary school students.

  • Force control and collision detection for a robot are usually conducted using a 6-axis force/torque sensor mounted at the end-effector. However, this scheme suffers from high-cost and the inability to detect collisions at the robot body. As an alternative, joint torque sensors embedded in each joint were used, which also suffered from various errors in torque measurement. To resolve this problem, a robot joint module with an improved joint torque sensor is proposed in this study. In the proposed torque sensor, a cross-roller bearing and disk-type coupling are added to prevent the moment load from adversely affecting the measurement of the joint torque under consideration. This joint design also aims to reduce the stress induced during the assembly process of the sensor. The performance of the proposed joint torque sensor was verified through various experiments.

  • This paper describes a automatic video surveillance system(AVSS) with long range and 360$^{\circ}$ coverage that is automatically rotated in an elevation over azimuth mode in response to the TTM(tracked target message) signal of vessels tracked by ARPA(automatic radar plotting aids) radar. This AVSS that is a video security and tracking system supported by ARPA radar, CCTV(closed-circuit television) camera system and other sensors to automatically identify and track, detect the potential dangerous situations such as collision accidents at sea and berthing/deberthing accidents in harbor, can be used in monitoring the illegal fishing vessels in inshore and offshore fishing ground, and in more improving the security and safety of domestic fishing vessels in EEZ(exclusive economic zone) area. The movement of the target vessel chosen by the ARPA radar operator in the AVSS can be automatically tracked by a CCTV camera system interfaced to the ECDIS(electronic chart display and information system) with the special functions such as graphic presentation of CCTV image, camera position, camera azimuth and angle of view on the ENC, automatic and manual controls of pan and tilt angles for CCTV system, and the capability that can replay and record continuously all information of a selected target. The test results showed that the AVSS developed experimentally in this study can be used as an extra navigation aid for the operator on the bridge under the confusing traffic situations, to improve the detection efficiency of small targets in sea clutter, to enhance greatly an operator s ability to identify visually vessels tracked by ARPA radar and to provide a recorded history for reference or evidentiary purposes in EEZ area.

  • With the rapid development of computer hardware and invention of various game-related technologies, the quality of games has been greatly improved and the genres of games have been diversified. Yet, in comparison, puzzle games have held to the past simple way of games, which has only led to a depression of the puzzle game market. This paper is the first attempt to overcome limits of the current puzzle games, by finding a breakthrough in three-dimensional puzzle games. In order to describe a realistic puzzle assembly in a three-dimensional puzzle game, this paper aimed to design a structure assembly technique made up of three steps that used a proper process to handle structure suitable to assembly, an elaborate way of collisions between individual objects and a physical structure. Through this technique, we intend to show in a simple experiment that it is possible to minimize the calculation cost in the assembly process and at the same time to achieve an elaborate puzzle assembly between structures.

  • In this paper, the design method and algorithms of the signal processor for a multipurpose radar system are analyzed. The signal processor, operating at the two modes-collision avoidance mode and weather mode, has 4 steps of ADC, NCI, STC, CFAR. Several algorithms of NCI and CFAR are analyzed and the optimal design is proposed to the system. CVI and CMLD algorithm have good performance in decreasing the false alarm rate and increasing detection probability, Regarding processor computational capacity, K=12 for CVI, M=16~20, Ko=M-4 for CMLD is suggested. CVI processing needs much time, two or more processors need to be allocated to CVI. So, for the system with four processors, two processors should be allocated to VID of NCI with ADC input and CFAR with STC, and two processors are should be allocated to CVI.

Cycle Detection in Discrete Logarithm Using a Queue (큐를 이용한 이산대수의 사이클 검출)

  • This paper proposes a discrete logarithm algorithm that largely reduces execution times of Pollard's Rho and Brent's algorithm in obtaining ${\gamma}$ from ${\alpha}^{\gamma}{\equiv}{\beta}$(mod p). The proposed algorithm can be distinguished from the conventional Brent's algorithm by three major features: it sets an initial value as $x_0={\alpha}{\beta}$ in lieu of $x_0=1$; replaces $y=x_i$, ($i=2^k$) pointer with $y_j{\leftarrow}x_i$, ($i=2^k$, $1{\leq}j{\leq}10$) for a Queue the size 10; and detects collision of ${\beta}_{\gamma}$, ${\beta}_{{\gamma}^{\prime}}$, ${\beta}_{{\gamma}^{-1}}$ instead of ${\beta}_{\gamma}$. This Queue method has reduced the execution time of Pollard's Rho algorithm with $x_0=y_0=1$ by 65.02%, and that of Brent's algorithm with $x_0=1$ by 47.80%.

  • We present a study of citizen science performed at Korean Minjok Leadership Academy (KMLA). The importance of citizen involvements in scientific studies has been increasing, with remarkable results and performances. For instance, the discovery of an impact scar near Jupiter's south polar region (A. Wesley, 2009) led to an international campaign of professional observations to understand the asteroidal collision responsible for the scar. Citizen science at KMLA has been and will be mainly conducted by members of the astronomical observation club 'Apple-Pie' through amateur telescopes. Members of 'Apple-Pie' are specialized in various fields related to astronomy, from planetary science to cosmology. The spectrum not only includes fields that are directly related to astronomy but also fields such as computer science and astrophotography. The scheduled construction of a new observatory will further enable students to participate in higher level projects such as planetary monitoring over long timescales and the observation and detection of solar system bodies and exoplanets. In addition, a new supervisor with expertise and research experience in galactic astronomy, planetary science, and meteorology has joined the school faculty. He will supplement students with fundamental theoretical backgrounds and essential research techniques to enhance astronomical research at KMLA. KMLA's ultimate goal is to deploy a remote-controlled observatory available to aspiring scientists around the world to create a network of citizen science system. The prime observational conditions of KMLA and the willingness of the students and faculty members will provide a competitive edge for KMLA over other similar institutes in Korea.
