사과에서 분리한 Colletotrichum acutatum, C. gloeosporioides 및 Glomerella cingulata의 병원학적 특징 (Etiological Characteristivcs of Colletotrichum acutatum, C. gloeosporioides and Glomerella cingulata Isolated from Apples)

  • Pathogenic fungal species associated with apple anthracnose were studied with diseased fruits collected over two major apple growing areas from 1986 to 1988. Colletotrichum acutatum Simmonds, c. gloeosporioides(Penz.) Sacc. and Glomerella cingulata(Stonem.) Spauld. & Schrenk were identified based on their mycological characteristics. C. acutatum could be easily differentiated from C. gloeosporioides by producing their fusiform conidia and developing apricot∼salmon colony colors, non-forming an ascigerous stages. Growth rate of C. acutatum were slow in plate culture. The three species of anthracnose fungi tested caused apple rot. Symptoms produced by C. gloeosporioides were larger than those of the others. C. gloeosporioides and G. cingulata produced rot lesions on apples that appeared became brown and later black, but C. acutatum produced rot lesions appeared yellowish brown and later reddish brown. G. cingulata has the major species responsible for apple anthracnose, epidemics in Korea.

Molecular and Cultural Characterization of Colletotrichum spp. Causing Bitter Rot of Apples in Korea

  • Colletotrichum contains many important pathogens which cause economically significant diseases of crops like pepper, strawberry, tomato and apple. Forty four isolates were collected to characterize the diversity of Colletotrichum causing apple anthracnose in various regions of Korea. They were analyzed by random amplified polymorphic DNA (RAPD), internal transcribed spacer (ITS) of rDNA and partial $\beta$-tubulin gene DNA sequence, and culture characteristics on PDA and PDA-Benomyl. From the results of molecular analyses, 31 strains belonged to Colletotrichum gloeosporioides, ribosomal DNA group (RG) 4 of Moriwaki et al. (2002), 8 strains belonged to C. acutatum, A2 group of Talhinhas et al. (2005) and 5 strains to C. acutatum, A3 group of Talhinhas et al. (2005). Most isolates of C. gloeosporioides RG4 grew faster on PDA than strains of C. acutatum, A2 and A3 groups and most RG4 strains were sensitive to Benomyl. However, a few strains of RG4 grew slower and were resistant to Benomyl. On the basis of molecular characteristics, apple isolates of C. acutatum were clearly differentiated from red pepper isolates of the species, but apple isolates of C. gloeosporioides were not.

  • C. gloeosporioides와 C. acutatum의 개체군 밀도분석을 위해 기존 ITS부위를 이용한 PCR방법에 사용한 caInt2와 cgint 프라이머에 형광을 표지하여 C. acutatum에 특이적인 fcaInt2와 C. gloeosporioides에 특이적인 vcgint의 두 probe를 제작하였다. 이 두개의 프라이머와 Unicof1, Unicor1 primer를 이용 real time PCR을 수행하였을 때 C. acutatum은 fcaInt2 probe에, C. gloeosporioides는 vcgint에 특이적인 형광증폭곡선을 나타냄에 따라 delta Rn 값을 비교함으로 두 종의 구분이 가능하였다.

  • Glomerella leaf spot (GLS) caused by Colletotrichum spp. is a destructive disease of apple restricted to a few regions worldwide. The distribution and evolution of GLS symptoms were observed for two years in Uruguay. The recurrent ascopore production on leaves and the widespread randomized distribution of symptoms throughout trees and orchard, suggest that ascospores play an important role in the disease dispersion. The ability of ascospores to produce typical GLS symptom was demonstrated by artificial inoculation. Colletotrichum strains causing GLS did not result in rot development, despite remaining alive in fruit lesions. Based on phylogenetic analysis of actin, ${\beta}$-tubulin and glyceraldehyde-3-phosphate dehydrogenase gene regions of 46 isolates, 25 from fruits and 21 from leaves, C. karstii was identified for the first time causing GLS in Uruguay and C. fructicola was found to be the most frequent (89%) and aggressive species. The higher aggressiveness of C. fructicola and its ability on to produce abundant fertile perithecia could help to explain the predominance of this species in the field.

Colletotrichum spp. Agents of Anthracnose on Blueberry Leaves in Gangwon Province, Korea

  • Blueberry, which produces phenolic compounds, is one of the most popular fruits in Korea. During a survey on blueberry diseases, 16 Colletotrichum isolates were obtained from blueberry leaves in Chuncheon and Gosung, Kangwon province, Korea. Using morphological and molecular analyses, the isolates were identified as Colletotrichum aenigma, C. fioriniae, C. fructicola, C. gloeosporioides, and C. nymphaeae. C. gloeosporioides was the most frequently isolated (11 isolates), and only one or two isolates of the other species were found. After inoculation with all isolates, those leaves and fruits with wounds easily developed anthracnose; whereas, fruits without wounds became infected but leaves without wounds were infected by only two of C. gloeosporioides. Typically, around seven fungicidal agents are used to control anthracnose on blueberries in Korea. Fluzinam and prochloraz manganese complex strongly (over than 80%) inhibited the growth of all Colletotrichum species, while dithianon and mancozeb only weakly (about 80% or less) inhibited their growth.

콩, 팥 및 녹두에서 분리한 탄저병균류의 동정과 병원학적 특징 (Identification and Etiological characteristics of Anthracnose Fungi Isolated from Soybean, Small Red Bean and Green Bean)

  • Anthracnose fungi associated with soybean (Glycine max), small red bean (Vigna angularis), and green bean (Phaseolus radiatus var. aurea) were compared with respect to cultural characteristics, morphological aspects, and pathogenicity. From the legumes, four different species of Colletotrichum were identified based on their morphological characters : C. truncatum, C. destructivum, C. trifolii and C. gloeosporioides. Colonies of Colletotrichum spp.on culture media varied in colour and growth habits depending upon the media and isolates. In general, formation of conidia, setae, and perithecia on water agar leaf medium was better than that on potato dextrose agar medium under near ultraviolet light. In cross inoculation tests, isolates of C. truncatum, c. destructivum, C. trifolii and C. gloeosporioides could infect soybean, small red bean, and green bean at different growth stages. There were some differences in pathogenicity or susceptibility depending on crops tested. C. truncatum was the most virulent and major pathogenic species among the four species.

포장위생 관리가 콩 미이라병의 발생에 미치는 영향 (Effect of Field Sanitation on the Pod and Stem Blight Caused by Phomopsis spp. in Soybean)

  • The effect of field sanitation using ecological characters of the pathogen was investigated for controlling Phomopsis seed decay in soybean. Field sanitation which was eliminated the inoculum by removing host debris, abscised petioles and cotyledones out of field, reduced remarkably infection percentage of pods and seeds by Phomopsis spp. as compared to the inoculated field. Neverthless, seed infection was 28.7% in the sanitized field. The fields sanitized by benlate application around the soybean plants also decreased seed infection with Phomopsis spp. Total seed infection including that with miscellaneous pathogens occurred as much as 75∼79% to the no application and their control values were 34∼42% over the routine application schedule. Even though it was not satisfactory, field sanitation seemed to be effective in controlling Phomopsis seed decay when infection pressure was low level. Diaporthe phseolorum va. sojae, D. phaseolorum var. caulivora and Phomopsis longicolla were mostly identified from soybean seeds and Colletotrichum truncatum, Cercospora kikuchiana were also isolated in sequence. Field sanitation did not significantly increase in soybean yield over the no application, while routine application schedule did in field.
