Converting Panax ginseng DNA and chemical fingerprints into two-dimensional barcode

  • Background: In this study, we investigated how to convert the Panax ginseng DNA sequence code and chemical fingerprints into a two-dimensional code. In order to improve the compression efficiency, GATC2Bytes and digital merger compression algorithms are proposed. Methods: HPLC chemical fingerprint data of 10 groups of P. ginseng from Northeast China and the internal transcribed spacer 2 (ITS2) sequence code as the DNA sequence code were ready for conversion. In order to convert such data into a two-dimensional code, the following six steps were performed: First, the chemical fingerprint characteristic data sets were obtained through the inflection filtering algorithm. Second, precompression processing of such data sets is undertaken. Third, precompression processing was undertaken with the P. ginseng DNA (ITS2) sequence codes. Fourth, the precompressed chemical fingerprint data and the DNA (ITS2) sequence code were combined in accordance with the set data format. Such combined data can be compressed by Zlib, an open source data compression algorithm. Finally, the compressed data generated a two-dimensional code called a quick response code (QR code). Results: Through the abovementioned converting process, it can be found that the number of bytes needed for storing P. ginseng chemical fingerprints and its DNA (ITS2) sequence code can be greatly reduced. After GTCA2Bytes algorithm processing, the ITS2 compression rate reaches 75% and the chemical fingerprint compression rate exceeds 99.65% via filtration and digital merger compression algorithm processing. Therefore, the overall compression ratio even exceeds 99.36%. The capacity of the formed QR code is around 0.5k, which can easily and successfully be read and identified by any smartphone. Conclusion: P. ginseng chemical fingerprints and its DNA (ITS2) sequence code can form a QR code after data processing, and therefore the QR code can be a perfect carrier of the authenticity and quality of P. ginseng information. This study provides a theoretical basis for the development of a quality traceability system of traditional Chinese medicine based on a two-dimensional code.

The Effect of Lidocaine.HCl on the Fluidity of Native and Model Membrane Lipid Bilayers

  • The purpose of this study is to investigated the mechanism of pharmacological action of local anesthetic and provide the basic information about the development of new effective local anesthetics. Fluorescent probe techniques were used to evaluate the effect of lidocaine HCl on the physical properties (transbilayer asymmetric lateral and rotational mobility, annular lipid fluidity and protein distribution) of synaptosomal plasma membrane vesicles (SPMV) isolated from bovine cerebral cortex, and liposomes of total lipids (SPMVTL) and phospholipids (SPMVPL) extracted from the SPMV. An experimental procedure was used based on selective quenching of 1,3-di(1-pyrenyl)propane (Py-3-Py) and 1,6-diphenyl-1,3,5-hexatriene (DPH) by trinitrophenyl groups, and radiationless energy transfer from the tryptophans of membrane proteins to Py-3-Py. Lidocaine HCl increased the bulk lateral and rotational mobility of neuronal and model membrane lipid bilayes, and had a greater fluidizing effect on the inner monolayer than the outer monolayer. Lidocaine HCl increased annular lipid fluidity in SPMV lipid bilayers. It also caused membrane proteins to cluster. The most important finding of this study is that there is far greater increase in annular lipid fluidity than that in lateral and rotational mobilities by lidocaine HCl. Lidocaine HCl alters the stereo or dynamics of the proteins in the lipid bilayers by combining with lipids, especially with the annular lipids. In conclusion, the present data suggest that lidocaine, in addition to its direct interaction with proteins, concurrently interacts with membrane lipids, fluidizing the membrane, and thus inducing conformational changes of proteins known to be intimately associated with membrane lipid.

  • Recently, various approaches to recommendation systems have been studied in terms of the quality of recommendation system. A recommender system basically aims to provide personalized recommendations to users for specific items. Most of these systems always recommend the most relevant items of users or items. Traditionally, the evaluation of recommender system quality has focused on the various predictive accuracy metrics of these. However, recommender system must be not only accurate but also useful to users. User satisfaction with recommender systems as an evaluation criterion of recommender system is related not only to how accurately the system recommends but also to how much it supports the user's decision making. In particular, highly serendipitous recommendation would help a user to find a surprising and interesting item. Serendipity in this study is defined as a measure of the extent to which the recommended items are both attractive and surprising to the users. Therefore, this paper proposes an application of serendipity measure to recommender systems to evaluate the performance of recommender systems in terms of recommendation system quality. In this study we define relevant or attractive unexpectedness as serendipity measure for assessing recommendation systems. That is, serendipity measure is evaluated as the measure indicating how the recommender system can find unexpected and useful items for users. Our experimental results show that highly serendipitous recommendation such as item-based collaborative filtering method has better performance than the other recommendations, i.e. user-based collaborative filtering method in terms of recommendation system quality.

Present Status and Future Management Strategies for Sugarcane Yellow Leaf Virus: A Major Constraint to the Global Sugarcane Production

  • Sugarcane yellow leaf virus (SCYLV) is a distinct member of the Polerovirus genus of the Luteoviridae family. SCYLV is the major limitation to sugarcane production worldwide and presently occurring in most of the sugarcane growing countries. SCYLV having high genetic diversity within the species and presently ten genotypes are known to occur based on the complete genome sequence information. SCYLV is present in almost all the states of India where sugarcane is grown. Virion comprises of 180 coat protein units and are 24-29 nm in diameter. The genome of SCYLV is a monopartite and comprised of single-stranded (ss) positive-sense (+) linear RNA of about 6 kb in size. Virus genome consists of six open reading frames (ORFs) that are expressed by sub-genomic RNAs. The SCYLV is phloem-limited and transmitted by sugarcane aphid Melanaphis sacchari in a circulative and non-propagative manner. The other aphid species namely, Ceratovacuna lanigera, Rhopalosiphum rufiabdominalis, and R. maidis also been reported to transmit the virus. The virus is not transmitted mechanically, therefore, its transmission by M. sacchari has been studied in different countries. SCYLV has a limited natural host range and mainly infect sugarcane (Sachharum hybrid), grain sorghum (Sorghum bicolor), and Columbus grass (Sorghum almum). Recent insights in the protein-protein interactions of Polerovirus through protein interaction reporter (PIR) technology enable us to understand viral encoded proteins during virus replication, assembly, plant defence mechanism, short and long-distance travel of the virus. This review presents the recent understandings on virus biology, diagnosis, genetic diversity, virus-vector and host-virus interactions and conventional and next generation management approaches.

An efficient protocol for the production of transgenic Alstroemeria plants via particle bombardment

  • Alstroemeria plants were transformed by using an improved particle-gun-mediated transformation system. Friable embryogenic callus (FEC) induced from the leaves with axil tissues of Alstroemeria plant was used as the target tissue. Also, FEC was transformed with the bar gene was used as a selectable marker. In the case of plasmid pAHC25, 7.5% of the twice-bombarded FEC clumps showed blue foci, whereas the clumps with single bombardment showed only 2.3%. Additionally, a 90° rotation with double bombardment led to a higher frequency (6 times) of luciferase gene expression in PBL9780 than the control treatment. After 8 weeks of bombardment, more than 60 independent transgenic lines were obtained for pAHC25 and nearly 150 independent transgenic lines were obtained for PBL9780, all of which were resistant to PPT and demonstrated either GUS or luciferase activity. Regarding effect of osmotic treatment (0.2 M mannitol) with 7 different periods, the highest transient gene expression was obtained in 8 h before and 16 h after transformation in both pAHC25 and PBL9780. Compared with the control, at least three times more GUS foci and photons were observed in this treatment. With respect to different combinations of mannitol and sorbitol with 8 h before and 16 h after transformation, high numbers of transient and stable transgene expressions were observed in both 0.2 M mannitol and 0.2 M sorbitol used in the osmotic pre-culture. This combination showed the highest transformation efficiency in both pAHC25 (8.5%) and PBL9780 (14.5%). In the control treatment, only 10% of the FEC clumps produced somatic embryos. However, by using 0.2 M mannitol and 0.2 M sorbitol, the frequency of somatic embryos increased to 36.5% (pAHC25) and 22.9% (PBL9780). Of the somatic embryos produced, at least 60% germinated. Approximately 100 somatic embryos from these 210 independent transgenic lines from 2 plasmids developed into shoots, which were then transferred to the greenhouse. PCR analysis confirmed the presence of the bar gene. This is the report on the production of transgenic Alstroemeria plants by using particle bombardment with a high efficiency, thereby providing a new alternative for the transferring of gene of interests in Alstroemeria in the breeding program in the future.

The Impact of Mergers on the Experiences of Students (대학 통폐합에 대한 학생들의 적응과정 연구)

  • This article explores the impact of mergers on the experiences of students. A case design is used to gain an in-depth understanding of the students in a Korean university that was combined with a two-year college in 2007. Semi-structured interviews of 11 direct entrants, 5 transfer students, and 7 professors were conducted in 2010 for the data collection. The results suggest that mergers do not necessarily ensure the integration of the students involved. Since mergers pose enormous challenges to both transfer students and direct entrants, university leaders need to give careful attention to students' persistence and integration as the campus changes. The results also reveal that students' experience of mergers may be affected by cultural backgrounds as well as the hierarchical structure of higher education. In Korean society, where credentialism prevails, mergers can have different impacts on students who come from initial institutions of differing perceived quality, and thus mergers may incite tension among the students. In light of these concerns, this study's findings highlight the importance of student integration as a key element in mergers.

Future Scenarios of Asian Universities in a view Point of Equality (평등의 관점에서 본 아시아 대학의 미래 시나리오)

  • This study was performed in order to suggest the future model of Asian universities that could be used in the planning of the global competitive strategy. Futurologists forecasted the future of higher education using Harman Fan Scenario as like this. First, most current universities will be 'the satellite university' until 2015. Second, they also will replace 'the bookless university' until 2020. Third, they will be 'no calendar university' until 2025. And then they may be 'all have access university' until 2030. After 2030, futurologists prospected that almost universities based on off-line campus will be disappeared into the history. The analysis method of Harman fan scenario and applied scenarios were also used to "A study on the future scenario of Korean university". The predictive model and the alternative models were explored in a view point of students, enterprise, and government. Individuality with educational excellence are standardized for learner, profit and effectiveness are applied for enterpriser, and equality with welfare are adapted for national leader. Asian universities need to focus on bringing up the practical ability based on conscious and emotional education instead of knowledge based on memory. Also they need to enforce the specialized education that can create new jobs through convergence of interdisciplinary. Especially, Asian nations need to explore, to find the strengthen area of their universities compared with USA. And these area should be specialized. The convergency strategy between oriental medicine and informatics is a meaningful sample. Based on this point, a predicted with 3 alternative scenarios in a view point of equality were suggested for the future of Asian universities.

An Operating Plans for the Interconnection between Local Agencies and Promotion of Professional Human Resources on the Special Education Convergence Based on 「Department of Support for the individuals with Developmental Disabilities in Graduate school」 (특수교육 융합 기반 「대학원 발달장애인지원학과」의 전문인력 양성체제 및 지역 유관기관 간 연계 운영방안)

  • This study was conducted with the aim of exploring methods of fostering professional human resources of the "department of support for the individuals with developmental disabilities in graduate school" based on the convergence of special education and connection between local relevant institutions. To this end, research methods were constructed that combine literature exploration and expert meetings. First, the system for fostering professionals in the "department of support for the individuals with developmental disabilities in graduate school" based on the convergence of special education programs was presented interlinked with the system for fostering professional human resources in the organizational and academic categories. Next, the link operation plans between relevant local institutions under the "department of support for the individuals with developmental disabilities in graduate school" based on the convergence of special education programs were presented interlinked with core management measures for curriculum education and cooperative support. In order to conclude the preconditions for the realization of the above research results, we presented leadership of special education convergence on the support system for lifelong education for adults with developmental disabilities.

Impact of Job Stress, Organizational Culture and Professional Identity on Job Satisfaction of Post-anesthesia Care Unit Nurses (회복마취간호사의 직무만족도 영향 요인)

  • The purpose of this study was to identify the factors affecting job satisfaction of PACU (post-anesthesia care unit) nurses in the context of job stress, organizational culture and professional identity. A total of 149 nurses participated in the cross-sectional, descriptive study suing a structured questionnaire. Job stress in pay and treatment category, innovation-oriented culture, relation-oriented culture, and professional identity were positively correlated with job satisfaction (r=.373, p<.001; r=.507, p<.001; r=.559, p<.001; r=.544, p<.001). The factors related to job satisfaction were professional identity (${\beta}=.391$, p<.001), relation oriented culture (${\beta}=.302$, p<.001), innovation oriented culture (${\beta}=.209$, p=.004), which accounted for 48.7% of the variance in job satisfaction. Professional identity was the most significant predictor of job satisfaction. Thus, in order to enhance the job satisfaction of nurses in the PACU, it is necessary to develop an education program that can improve professional identity and apply strategies for fostering relation and innovation oriented organization culture.

Analysis of Factors Affecting Length Of Stay for A Serious Patients Using Medical Records (의무기록자료를 이용한 중증질환자의 재원일수에 미치는 요인 분석)

  • Objectives: In this study, we tried to analyze the factors affecting Length Of Stay for serious patients in Republic of Korea. Methods: The study included 139,172 serious patients in the 2012-2016 discharge details. Using the SPSS 23.0 program, we conducted a rank regression analysis with social and social demographic characteristics as control variables, medical institution characteristics and medical use characteristics as independent variables, and Average Length Of Stay as a dependent variable. Results: Average Length Of Stay for participants was found to be 9.92days. And the location and bed size of medical institutions were not statistically significant, the hospitalization path was more urgent(B=0.43) than the outpatient (p<0.001), and there was no secondary diagnosis(B=0.35). However, Average Length Of Stay was higher (p<0.001) than there was no main surgery(B=0.80). After discharge, Average Length Of Stay for funding(B=0.43) and death(B=0.72) was long (p<0.001). Average Length Of Stay for participants was found to be 9.92days. And the location and the bed size of the medical institution were not statistically significant, and the hospitalization pass had longer Length Of Stay for emergency patients(B=0.43) than for outpatients(p<0.001). There was a longer Length Of Stay(B=0.35) than none was diagnosed. There were longer Length Of Stay(p<0.001) than there was no major surgery(B=0.80). After discharge, the outpatients had longer Average Length Of Stay(B=0.43) and deaths(B=0.72) than those who returned home(p<0.001). Conclusion: As a result of analyzing the factors affecting Average Length Of Stay of the participants, it was confirmed that regardless of the location and bed size of medical institutions, hospitalization route, department diagnosis, main surgery, and whereabouts after discharge. Therefore, appropriate interventions and necessary support must be provided so that efficient Length Of Stay can be managed according to the medical use characteristics of serious patient.
