  • Collagen-induced arthritis (CIA) is a model for some types of human autoimmune rheumatoid arthritis (RA). In this study, we examined whether ethanol extract of potato (Solanum tuberosum) is efficacious against CIA in mice. Potato extracts (100 and 200 mg/kg) were orally administered to DBA/1J mice once daily for 49 day after initial immunization with type II collagen. Clinical assessment of disease and measurement of paw edema were conducted throughout the study. The production of CIA-related rheumatoid factor, anti-type II collagen antibody, and cytokines were examined in DBA/1J mice. Serum levels of AST, ALT, creatinine, and lipids were measured, and antioxidant enzyme activity in the spleen was also determined. The arthritis score and paw edema were markedly suppressed in the groups treated with potato extract. Levels of rheumatoid factor, anti-type II collagen antibody, interleukin (IL)-1, IL-6, LDL-cholesterol, and malondialdehyde in sera were also reduced by potato extract treatment. The activities of glutathione peroxidase and glutathione reductase were increased in the spleens of CIA mice treated with potato extract. These findings suggest that potato extract has suppressive effects on type II collagen-induced arthritis, an animal model for human RA.

  • Objectives : The present study was undertaken to determine whether the water extract of Achyranthis Radix, which is the roots of Achyranthes japonica (Achyranthis Radix, AR), is efficacious against collagen-induced arthritis (CIA) in DBA/1J mice. Methods : Mice were immunized with bovine type II collagen and orally treated with AR-W (50 and 100 mg/kg/bw) from days 21 to 42 after immunization. Arthritis was evaluated by arthritic score, histological examination of knee joint and serological markers such as TNF-${\alpha}$, $PGE_2$ and anti-type II collagen (C II)IgG. Results : The results showed that comparing with untreated CIA mice, treated with AR-W significantly suppressed the clinical score and joint tissue pathological damages, reduced the serum levels of TNF-${\alpha}$, $PGE_2$ and anti-C II IgG in CIA-mice. These results suggest that AR-W can effectively alleviate inflammatory response on CIA, and anti-inflammatoy of AR-W can be attributed, at least partially, to the inhibition of inflamamtory mediators, $PGE_2$ and TNF-${\alpha}$, in CIA. Conclusions : This study suggests that AR-W has a therapeutic potential in inflammatory joint diseases such as rheumatoid arthritis.

  • Objectives : The present study was undertaken to evaluate whether Atractylodis Rhizoma Alba ethanol (ARA-E) extract, which is the pericarp of $Atractylodes$ $japonica$ Koidz. has an effect on collagen-induced arthritis (CIA) in mice. Methods : Male DBA/1J mice were induced by intradermal injection of bovine collagen-II in Freund's incomplete adjuvant (IFA). The CIA mice in the onset of arthritis were treated daily with oral administration of ARA-E extract at dose of 50 mg/kg/bw for 28 days. Arthritis index, histopathological changes and the levels of anti-type II collagen (CII) IgG and inflammatory cytokine, TNF-${\alpha}$ in sera of mice were measured to evaluate the antiarthritic effect of ARA-E. Results : ARA-E extract significantly decreased the arthritic scores and inhibited pathological changes of knee joint tissues in CIA mice. ARA-E extract also significantly decreased the serum levels of anti-CII IgG and TNF-${\alpha}$ in CIA mice. These results indicate that ARA-E extract may effectively prevent arthritic damages in CIA mice, at least partially, by inhibiting the production of autoantibodies and inflammatory cytokine. Conclusions : This studies suggest that ARA-E has a therapeutic potential in inflammatory joint diseases such as rheumatoid arthritis.

Methods : The author performed several experimental procedures to observe the effects of the EC-HAS at the arthritis. The severity of arthritis, changes of cytokine level and antibody level, histological changes of the CIA mouse joint were analyzed. Results : 1. The incidence of arthritis and arthritis index were significantly decreased in the cases which were treated with the EC-HA. 2. Cartilage destruction and synovial cell proliferation were reduced, and the expression of the collagen fibers was similar with that of the normal group. 3. The levels of IL-6, $IFN-{\gamma}$, $TNF-{\alpha}$, $IL-1{\beta}$ in serum of the CIA mice which were treated with the EC-HA were significantly decreased compared with those of the normal group. 4. The levels of IgG, IgM and anti-collagen II in serum of CIA mice when they treated with the EC-HA were significantly decreased compared with those of the normal group. 5. The expression ratio of $CD4^+$ to $CD8^+$ cells in the EC-HA treated mice were maintained as much as the normal group of the lymph nodes in the CIA mice. 6. The $CD3e^+CD69^+$ and $CD11b^+Gr-1^+$ cell populations in the knee joint were significantly decreased in the EC-HA treated group. Conclusions : These results suggest that the EC-HA at the ST36 may be responsible roles to control on the synovial cell proliferation and to prevent the cartilage destruction in rheumatoid arthritis. These results will be important supporting evidence for the practical use of the EC-HA at rheumatoid arthritis clinic in the future.

  • Objectives : Evodia rutaecarpa (Juss.) Benth.(ER) and Chaenomeles sinensis Koehne (CS) have multiple applications and were known to have anti-inflammatory effects. In the current study, we investigated to clearly understand the mechanism of therapeutic role for CS, ER and their combination in CIA model mice. Methods : DBA/1OlaHsd mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with CS, ER and their combination once a day for 7 weeks. Cytokine production and gene expression were assessed during CIA (collagen-induced arthritis) model mice in knee joint, lymph node (LN) using ELISA and FACS analysis. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. Result : Oral administration of CS, ER and their combination (150 mg/kg) significantly suppressed the progression of CIA, and significantly suppressed the progression of CIA and inhibited the production of TNF-${\alpha}$ and IL-6 in serum. The erosion of cartilage was dramatically reduced in mouse knees after treatment with CS plus ER. Conclusion : These result suggest that CS plus ER significantly suppressed the progression of CIA and that this action was characterized by the decreased production of TNF-${\alpha}$, IL-6 and collagen II specific antibody in serum.
