UV-induced Photodamage - attenuating Properties of Water Extract from Lentinuls edodes (피부각질형성세포에서 표고버섯 물 추출물의 피부노화 억제 효과)

  • Lentinuls edodes has been used for traditional food and medicine around Asia, and a variety of biological effects have been reported. In this study, L. edodes water extract (LWE) was investigated for its anti-photodamage effect in HaCaT keratinocytes. To perform the necessary assays, L. edodes was extracted with distilled water for 8 hr at 40℃ in an extract tank. Anti-photodamage activity was assessed using a scratch wound healing assay, cell proliferation, and a reactive oxygen species (ROS) scavenging test and by measuring the mRNA and protein expression levels of matrix metalloproteinases (MMPs) and type I procollagen. MMPs and collagen expression are major markers of UV-induced photodamage in skin. Prior to photodamage analysis, the total polyphenol and β-glucan contents of the LWE were evaluated and found to be 4.64 mg GAE/g DW and 165.96 mg/g, respectively. Treatment with LWE induced cell migration and cell proliferation in UV-irradiated HaCaT cells, and LWE effectively scavenged the ROS induced by H2O2 and UVB irradiation in HaCaT cells. UVB irradiation induced ROS generation and led to increased production of MMP-1 and MMP-9 and to decreased collagen production in human keratinocytes. Treatment with LWE upregulated the expression levels of MMP-1, MMP-9, and type I procollagen in UVB-irradiated HaCaT cells. This study suggests that LWE could be used to develop cosmetic materials with anti-photodamage effects.

Physicochemical and Sensory Characteristics of Beef-bone Broths Prepared under Atmospheric Pressure and Overpressure (상압과 가압조건에서 제조한 소뼈 용출액의 이화학 및 관능 특성)

  • This study was conducted to compare the physicochemical and sensory characteristics of beef-bone broths prepared under atmospheric pressure (BBAP) and overpressure (BBOP). Beef-bone was boiled in water (bone/water=1:2, w/w) for 1, 2, 4, 6, and 12 h under atmospheric pressure and overpressure ($121^{\circ}C$, 1.25 atm). The BBOP broth samples were found to contain significantly higher amounts of solid, crude protein, crude ash, collagen, and chondroitin sulfate than the BBAP broth samples for a given boiling time (p<0.05). In addition, the Ca and Mg contents in the BBAP samples were higher than those in the BBOP samples, whereas the P, Na, and K contents were higher in BBOP than BBAP. The L value of BBAP was also significantly higher than that of BBOP (p<0.05). Further, as the boiling time increased, turbidity increased in the BBAP samples. In the quantitative descriptive analysis, the BBOP samples exhibited stronger brownness, transparency, meaty off-odor, meaty off-flavor, and sulfuric odor than the BBAP samples.

  • In order to compare and study the effects of Kamipalmijiwhang tang(加味八味地黃湯) & Kamigangwhalsokdantang(加味羌活續斷湯) on inhibition of osteoporosis induced by ovariectomy, the author administered Kamipalmijiwhangtang & Kamigangwhalsokdantang to the sample groups per orally before and after ovariectomy. The author observed the activities of cathepsin L, K and the contents of calcium, calcitonin, osteocalcin and phosphorus in the serum for the biochemical index, the contents of IL-6 in the serum and bone marrow for immunological index, the state of bone loss in the metaphysis of tibia by histological study, the expression of ELAM-1 &collagen IV by immunohistochemical analysis, the expression of fos by immunocytochemical analysis. The results were as follows: 1. The activities of cathepsin L, K in the serum had a significant inhibitive effects in all sample groups compared with the control group. The inhibitive effects of Sample A-1 & B-1 was better than those of A-2 & B-2 and the inhibitive effects of sample B-1 & B-2 was better than those of A-1 & A-2. 2. The content changes of IL-6 in the serum and bone marrow had a significant inhibitive effects in all sample groups compared with the control group. The inhibitive effects of sample A-1 & B-1 was better than those of A-2 & B-2 and the inhibitive effects of sample B-1 & B-2 was better than those of A-1 & A-2. 3. In the histological study of tibial metaphysis all the sample groups inhibited bone loss significantly compared with the control group and expressed trabecula shape of bone similar to the normal group. 4. The expression of ELAM-1 & collagen IV decreased a little bit in sample A-2. It also decreased to a similar degree in sample B-2 like normal group and disturbed itself. 5. The expression of fos was inhibited in all sample groups compared with control group. The inhibitive expression of sample B-1 & B-2 was more remarkable than those of A-1. & A-2. 6. The serum calcium had a significant increasing effects in an sample groups compared with control group. Each sample groups didn't have any significant difference. 7. The serum calcitonin had a significant increasing effects in sample A-1, A-2 and B-1 groups compared with control group. Each sample groups didn't have any significant difference. 8. The serum osteocalcin and phosphorus didn't have any significant difference in all sample groups compared with control group.

Physicochemical Properties and Biological Activities of Collagens with Different Molecular Weights from Alaska Pollack (Theragra chalcogramma) Skin (명태 껍질 유래 콜라겐의 분자량에 따른 이화학적 특성 및 생리활성)

  • This study was conducted to investigate the physicochemical properties and biological activities of collagens with different molecular weights from Alaska pollack (Theragra chalcogramma) skin as well as their efficacies as functional materials. The molecular weights of collagens were between 1~10 kDa (below 1 kDa (AP1), 1~3 kDa (AP2), 3~10 kDa (AP3), and above 10 kDa (AP4). The protein content of AP4 (40.19 g/100 g) was the highest. Collagen contents of AP1, AP2, AP3, and AP4 were 36.43, 32.23, 19.23, and 14.89%, respectively. The free amino acid and essential amino acid contents of AP1 were higher than those of AP2, AP3, and AP4. Fourier transform infrared spectroscopy spectra of collagens with different molecular weights showed wavenumbers representing the regions of amide I, amide II, amide III, and amide A, respectively. The electron-donating ability (29.51%) and SOD-like activity (38.45%) of AP1 were higher than those of AP2, AP3, and AP4. Tyrosinase inhibition activity of AP1 improved with higher treatment concentration. The rate of inhibition of MMP-1 production in HS68 cells exposed to UVB was suppressed by treatment with AP1 (29.78%) and AP2 (26.49%) at 1 mg/mL. Furthermore, there was a strong correlation between DPPH, superoxide dismutase, tyrosinase activity, and MMP-1 inhibition rate of collagens with different molecular weights.

Suppressing Effects of Sowhalrack-dan(Xiǎohuóluò-dān) on Freund's Complete Adjuvant Induced Arthritic Rats (Adjuvant 유발(誘發) 류마티스 관절염(關節炎)에 대한 소활락단(小活絡丹)의 억제(抑制) 효과(效果))

  • Objectives : The object of this study is to observe the favorable anti arthritic effects of Sowhalrack-dan($Xi\check{a}ohu\acute{o}lu\grave{o}-d\bar{a}n$)(SWRD), which has been traditionally used in Korean medicine to treat rheumatoid arthritis on Freund's complete adjuvant(FCA) induced arthritic Wistar rats. Methods : Rheumatoid arthritis was induced by intradermal injection of FCA(10 mg in 1 ml paraffin oil 0.1 ml/rats). Each of 8 rats showing regular ankle circumferences per group were selected in 14 days after FCA treatment to confirm the induction of rheumatoid arthritis. 300, 150 or 75 mg/kg of SWRD was orally administered once a day for 14 days from 14 days after FCA treatments. Dexamethasone was intraperitoneally administered 15 mg/kg, once a day for 14 days from 14 days after FCA treatments. Rats were sacrificed after 14 days of continuous oral treatment of SWRD or intraperitoneal administration of dexamethasone, and changes were observed; the body weight, knee circumferences, gross arthritis score, inflammatory tissue $prostaglandin(PG)E_2$ levels and cartilage collagen, glucosaminoglycans compositions - chondroitin sulphate, heparin sulphate and hyaluronic acid in the present study. Results : As results of FCA treatment, classic rheumatoid arthritis featuring dramatical decreases on the body weights, cartilage collagen contents and bone glucosaminoglycans - chondroitin sulphate, heparin sulphate and hyaluronic acid contents. Also, it increases the knee circumferences, gross arthritis scores and inflammatory tissue $PGE_2$ levels. However, these changes from FCA induced rheumatoid arthritis were clearly reduced due to the dexamethasone and both two different dosages of SWRD, 300 and 150 mg/kg in the present study. Although FCA induced arthritis were more favorably inhibited by treatment of dexamethasone 15 mg/kg compared to SWRD 300 mg/kg, marked decreases of body weights were detected in dexamethasone 15 mg/kg treated rats. Conclusions : The results obtained in this study suggest that over 150 mg/kg of SWRD showed favorable anti-arthritic effects on the FCA induced arthritis mediated by suppression of $PGE_2$. However, detailed mechanism studies are needed with the screening of the biological active compounds in SWRD. Although FCA induced arthritis were more favorably inhibited by treatment of dexamethasone 15 mg/kg compared to SWRD 300 mg/kg, marked decreases of body weights were detected in dexamethasone 15 mg/kg treated rats, in the present study.

Comparison of Quality Traits of Thigh Meat from Korean Native Chickens and Broilers

  • The quality attributes of thigh meat from a certified meat-type commercial Korean native chicken (WoorimatdagTM, WM) and a commercial broiler (Ross, CB) raised under the same environmental conditions, were compared. One-d-old, mixedsex WM (200), and 200 broilers (Ross, CB) each were randomly assigned to floor pens (20 chickens per pen). The production stages (I, II, and III) were selected based on similar live weight. The crude fat contents of WM thigh meat were lower than those of CB. WM thigh meat contained higher stearic and arachidonic acid contents but lower palmitoleic and oleic acid contents compared to those of CB. The thigh meat color CIE $L^*$- and CIE $b^*$-values of WM were lower, but inosine-5'-monophosphate content was higher in WM than CB in whole production stages. WM thigh meat showed higher collagen content, hardness, springiness, gumminess, and chewiness than CB. In sensory evaluation, odor, taste, and overall acceptance were higher while color was lower in WM. Based on these results, it can be concluded that even if commercial broilers are raised under the same environmental conditions as WM, the unique quality attributes of WM thigh meat are distinctively unique.

Comparison of Fatty Acid and Nutritional Composition of Korean Native Black Cattle and Hanwoo (한국 재래흑우 및 한우의 지방산 조성 및 영양성분 비교)

  • The nutritional composition of Korean native black cattle (KNBC) and Hanwoo beef were compared. According to the results, there were no significant differences in moisture, protein, ash, calorie and collagen content, but the fat content of KNBC was significantly higher than that of Hanwoo (p<0.05). The P, Na and Cu content of KNBC were significantly higher than that of Hanwoo (p<0.05). The KNBC and Hanwoo had total mineral amounts of 4,052.34 and 3,214.44 ppm, respectively, with the KNBC being significantly higher than Hanwoo (p<0.05). The vitamin B1 content of KNBC was significantly higher than that of Hanwoo (p<0.05), but there was no significant difference in B2 content between the samples. The total structural amino acid contents was not significantly different between KNBC (12.35%) and Hanwoo (12.58%). The KNBC and Hanwoo had total free amino acid amounts of 752.698 and 661.795 ppm, respectively, with the KNBC being significantly higher than Hanwoo (p<0.05). The stearic acid (10.631%), linoleic acid (2.271%), linolenic acid (0.065%) and tricosanoic acid (0.038%) contents of KNBC were lower than those of Hanwoo (p<0.05). However, their palmitoleic acid (5.292%), oleic acid (48.815%), ${\gamma}-linolenic$ acid (0.094%), eicosenoic acid (0.042%) and docosadienoic acid (0.099%) contents were higher than those of Hanwoo (p<0.05). There was no significant difference between the KNBC and Hanwoo in the saturated fatty acid : unsaturated fatty acid ratio.

Effect of Parity of Hanwoo Cow on Physico-Chemical, Sensory and Nutritional Characteristics of Sullungtang (한우 암소의 산차가 설렁탕의 이화학적, 관능적 및 영양적 특성에 미치는 영향)

  • This study was performed to provide scientific information for consumers and to increase the quality and consumption of Sullungtang prepared with bone from Hanwoo. The physico-chemical, sensory, nutritional properties of Sullungtang by different parity were investigated. The extracting extents of blood were significantly higher for heifer among Hanwoo cows with different parity(none, the second or the fourth) and for back bones among different bone portions (round, hind shank, arm, fore shank or back bone) (p〈0.05). For Sullungtang extracted from bones of heifer was higher in collagen contents than those extracted from cows with the second or the fourth parity. Sullungtang extracted from bones of heifer and cows with the second parity had significantly higher contents of condroitin and viscosity than those of cows with the fourth parity, however there was no significant difference between heifer and cows with the second parity. In color, L values were significantly high, and a and b values were significantly low for Sullungtang extracted from bones of heifer when compared to those extracted from the other cows with the second parity. The contents of total nitrogen, sodium, or calcium were higher for Sullungtang extracted from heifer than those from the cows with the second or the fourth parity. In amino acid compositions, the percentages of glycine were highest and proline and glutamic acid were followed fro all Sullungtang samples from Hanwoo. The results showed that the overall quality of Sullungtang significantly decreased as the parity increased for Hanwoo cows. The Sullungtang extracted from bones of heifer had the best sensory scores as well as nutritional quality when compared to those extracted from the cows with parity. Therefore, the labeling and price for cow bones should be differently evaluated by their parity and age in marketing system.

Physicochemical Meat Quality and Fatty Acid Compositions of Striploin, Chuck Tender, Eye of Round Muscles from Holstein Steer Beef Slaughtered at Different Fattening Periods (홀스타인 거세육우의 출하시기에 따른 채끝, 꾸리, 홍두깨육의 이화학적 육질특성 및 지방산조성)

  • This study was performed to investigate the physicochemical properties of striploin (m. longissimus dorsi), chuck tender (m. supraspinatus) and eye of round (m. semitendinosus) of Holstein steer beef produced from different fattening groups (18, 21, 24 mon-old). The intramuscular fat contents were significantly higher in striploin (9.14%) and eye of round (4.31%) from 24 mon-old groups when compared to the same cuts (6.53% and 2.63%) from 18 mon-old groups, respectively. Three muscles from 18 mon-old group had significantly higher moisture contents (%) than those from the other groups (p<0.05). The protein contents were significantly higher in chuck tender (19.39%) and eye of round (21.09%) from 24 mon-old group than 18- and 21 mon-old groups (p<0.05). There were not significantly different in collagen contents among three fattening groups. In meat color, striploin, chuck tender and eye of round from 21 mon-old group had significantly higher CIE $L^*$ (41.77), $a^*$ (20.98) and $b^*$ (10.87) values than those from the other groups (p<0.05). Warner-Bratzler shear force values (WBS) for three muscles were significantly lower in 24 mon-old group than the other groups (p<0.05). The oleic acids (C18:1n9) contents of three muscles from 24 mon-old group were highest (p<0.05) among three groups. Total contents of MUFA were significantly higher and total contents of PUFA including arachidonic acids (C20:4n6) were significantly lower in chuck tender and eye of round muscles from 24 mon-old group (p<0.05).

Effect of Ledebouriella seseloides Extracts on Lipid Parameters in Ovariectomized Rats (방풍나물 추출물이 난소절제 흰쥐의 혈중 지질 변화에 미치는 영향)

  • This study was investigated the improvement effects of Ledebouriella seseloides (LS) ethanol extracts on lipid parameters in an ovariectomized animal model. Sixty, nine-week old female Sprague Dawley rats were randomly assigned to four groups as follows: sham-operated rats (SHAM), ovariectomized rats (OVX-CON) and ovariectomized rats that were treated with LS ethanol extracts (50 mg/kg/day and 200 mg/kg/day, respectively). The diets were fed to the rats for six weeks after their operation. The total-cholesterol and triglyceride contents on serum increased in the OVX-CON group compared to the SHAM group, but supplementation with the LS extract caused these factors to decrease. Notably, the serum LDL-cholesterol concentration in the supplemented 200 mg/kg/day LS ethanol extract group was significantly more reduced than the OVX-CON group. In addition, the platelet aggregation ability was lower in groups treated with LS than in the OVX-CON group. The alkaline phosphatase (ALP) activity was lower in the LS extract group compared to the OVX-CON group. Collagen content, in bone and cartilage, were reduced by ovariectomy, but the supplemented LS extract groups exhibited higher concentrations in their bones. According to these results, the improvement effects of LS extract on serum lipid parameters and osteogenesis in ovariectomized rats were illuminated.
