파종심도에 따른 맥류 및 옥수수의 초엽장과 초장의 변화 (Effect of Sowing Depth on Coleoptile Length and Plant Height of Barley, Wheat, Rye and Corn)

  • 보리 4품종, 밀 4품종, 호밀 3품종, 옥수수 1품종을 가지고 파종 심도를 달리 하였을 때 이들 작물들의 초엽장과 식물체장을 조사하였고 또 이들 간의 상관을 조사한 결과를 요약하면 다음과 같다. 1. 보리, 밀, 호밀, 옥수수가 발아할 때 생기는 관 아래의 절간을 중경 또는 지하경 이라고 호칭하고 있는데 지하경으로 부르는 것이 옳을 것 같으며 보리, 밀, 호밀은 초엽이 종자로부터 지하경과 함께 나오고 옥수수는 지하경 정단의 관에서 나오는 것을 확실하게 해두는 것이 좋을 것 같다. 2. 보리의 초엽의 길이는 파종 심도가 깊어질수록 현저하게 길어지고 식물체장도 같은 경향이 있는데 보리의 2cm파종은 6cm, 10cm깊이 파종 보다 현저하게 짧게 나타났다. 품종간에는 차이를 볼 수 없었다. 3. 밀의 초엽 길이도 마찬가지로 파종 심도에 따라 현저하게 길어졌으며 2cm 파종은 6cm, 10cm 파종보다 현저하게 짧게 나타났으나 보리보다는 못하였다. 이것도 품종간 차이는 별로 나타나지 않았다. 4. 호밀과 옥수수도 파종 심도가 깊어질수록 초엽이 길어졌으며 그 변이가 보리와 밀보다 크게 나타났다. 호밀과 옥수수에서 변이가 컸다. 5. 각 작물과 품종들에 있어서 초엽과 식물체장 간에 유의성이 있는 상관을 보였고 칠보 호밀은 고도의 유의성을 나타내었다.

Evaluation for anaerobic germinability of rice germplasm for direct-seeding cultivation under submerged conditions

  • Stable stand establishment is pre-requisite in direct rice seeding system for obtaining optimal yield of rice crop in rain-fed and waterlogged areas. Anaerobic condition on waterlogged soil causes low germination which significantly reduces crop yield. Due to low availability of tolerant genetic material for anaerobic germination, there is urgent need to evaluate rice germplasm for better germinability under anaerobic conditions. Seeds of the 185 rice accessions were evaluated for germination vigor and coleoptile length under anaerobic conditions. The variation among germplasm was tested for significance using analysis of variance and various multivariate components. Significant level of variation was observed among all accessions for germination vigor and coleoptiles length. Although highest mean values for coleoptiles length (2.1cm) and germination rate (60%) were observed in japonica accessions but maximum coleoptile length (4.68cm) and germination rate (96%) was found in indica genotype CO18. A highly significant and positive correlation was also observed between germination vigor and coleoptiles length, which signify the importance of elongated coleoptile under anaerobic conditions. The PCA analysis illustrated that 97.24% variation was accounted by PC1 while PC2 and PC3 explained 2.54% and 0.24% variation for germination vigor and coleoptile length. PCA scattered plot divided the accessions in four various groups. All AG tolerant accessions were included in group I. Likewise in the case of cluster analysis, two major clades (I and II) were formed. All accessions showing >40% germination rate were included in clade I, whereas all other accessions with <40% germination rate were grouped in clade II. Further more highly tolerant accessions (>80% germination) were grouped in sub-cluster IA.

건답직파에서 토양수분조건에 따른 벼 품종의 출아특성 (Seedling Emergence of Dry-seeded Rice Varieties under Two Soil Moisture Regimes)

  • 벼 건답직파에서 벼 품종들의 출아·입모 특성과 유아의 신장 특성과의 관계를 검토하고자 국내의 재래종, 육성종 및 샤레벼, 미국 직파재배품종, 이태리 품종 등 98개 품종을 토중 4.5cm에 파종하여 적습과 과습조건하에서 출아율, 입모율, 평균출아일수, 중배축장, 하위절간장, 초엽장, 불완전엽장 등을 조사하여 얻은 결과는 다음과 같다. 1. 적습조건에서 입모율은 9∼92.5%, 평균출아일수는 12∼25일 범위였고, 과습조건에서의 출아율은 0∼90%, 평균출아일수 27∼34일로 양조건 모두 품종간 변이가 매우 컸으며, 대부분의 품종은 과습조건에서 출아율이 낮아지고 출아속도가 늦어졌다. 2. 출아·입모율과 유의한 정의 상관을 보인 형질은 적습조건에서는 중배축장, 초엽장, 불완전엽장이었고, 과습조건에서는 중배축장, 제1, 2절간장, 초엽장, 불완정엽장이었다. 3. 평균발아일수는 중배축장, 초엽장, 불완전엽장과 유의한 부의 상관을 나타내었다. 4. 표준편회귀분석결과 출아·입모의 품종간 차이에 가장 유의적으로 기여하는 형질은 적습조건에서는 중배축장과 초엽장, 과습조건에서는 중배축장과 제2절간장이었다. 5. 평균출아일수의 품종간 차에 유의적으로 기여하는 형질은 적습조건에서는 중배축장, 불완전엽장으로 회귀계수는 부의 방향이었으며, 제1,2절간장은 정의 방향이었다. 과습조건에서는 중배축장만이 유의적 기여를 하였는데 회귀계수는 부의 방향이었다. 6. 본 실험의 결과로 볼 때 건답직파조건에서 출아율이 높고 출아속도가 빠른 품종을 육성하기 위해서는 중배축, 초엽 및 불완전엽의 신장이 잘되는 특성의 선발이 바람직한 것으로 사료된다.

Morphological Difference of Rice Seedling Grown under Different Dissolved Oxygen Conditions

  • The response of dissolved oxygen (DO) concentrations caused significant change in root number, root length, coleoptile length, shoot length and leaf age of seedlings. The genotypic difference in the effect of DO also highly significant (P<0.01) for all of the seedling traits. The number and length of root were extremely inhibited at the condition of $0.39\pm0.09$ DO concentration. While the coleoptile elongated markedly in the lowest DO concentrations, the shoot did not develop. The root growth was improved slightly at the $1.39\pm0.27mg L^{-1}$, however, there were no difference among genotypes at these two low DO concentrations. As the DO concentration become higher, the growth of root and shoot was improved remarkably. Root number, root length and shoot length was significantly different between $20\;and\;30^{\circ}C$ in DO rich and normal conditions, the development of those traits were apparently accelerated in high water temperature, however those traits of seedlings in DO deficiency were not different between the two temperatures except for shoot length. On the other hand the coleoptile length was not affected by the stagnant water temperature; it was stimulated by the low DO concentration. The competition of DO was greater as the seedling density was increased in the stagnant water, therefore the seedlings grown under high density have long and white coleoptiles, and the growth of roots and shoots was retarded severely.

옥수수 자엽초 생장에서 오옥신과 $Ca^{2+}$ 상호작용 (Interaction of Auxin and $Ca^{2+}$ on Corn Coleoptile Segment Elogation)

  • We investigated the interaction of auxin and Ca2+ on coleoptile segment elongation in seedlings of Zea mays L. Seedlings imbibed and raised either in the presence of 10 mM CaCl2 (HC), or in the absence of Ca2+ (LC) were used. Exposure to 10-5M auxin of coleoptiles from either HC or LC seedlings resulted in strong promotion of elongation. However, longer latent period (90 min) of the auxin effect was observed in HC than in LC seedlings (20 min). The length of latent period observed in HC coleoptiles was proportional to the concentration of CaCl2. The latent period of auxin effect observed in HC seedlings was abolished by pretreatment of the coleoptiles with TMB-8 which inhibits IP3-induced Ca2+ release from the tonoplasts. In segments of LC seedlings, the promotive effect of IAA (10-5M) was abolished by treatment with 5 mM calcium but was reversible upon treatment of the segments with 5 mM EGTA. These results suggest that the effect of auxin on coleoptile elongation is closely related to intracellular Ca2+ level.

법씨알의 크기가 모의 초기생육에 미치는 영향 (Effect of Grain Size of Rice Seeds on the Initial Growth of Seedlings)

  • 종자의 대소가 묘의 초기생육에 미치는 영향을 알고저 재건, 농광의 2개 수도품종을 공시하고 종자를 크기에 따라 대립, 중립, 소립으로 구별하여 묘대기의 생육을 관찰한 결과는 다음과 같았다. 1. 종자가 클수록 성묘율은 높아 대립이 묘수 확보에 유리함을 알 수 있었다. 2. 발근수, 출엽수 및 분얼수 역시 대립일수록 많았으며 품종간 차는 나타나지 않았다. 자엽장은 품종적 특성에 따라 농광이 길었으며 대립일수록 역시 길었다. 4. 따라서 지상부 건물중은 대립일수록 크게 나타났으며 품종별로는 농광이 높았다. 이러한 차는 생육이 진전될수록 현저하였다. 5. 단위 자엽길이 당건물중은 이난기까지는 감소하고 그 이후부터 증가하여쓴데 종자크기에 따른 건물중은 이 시기까지는 대립이 높았으나 그 후는 곧 차를 보이지 않았고 품종간 차는 전반적으로 나타나지 않았다.

Exploring Responses to Light in the Monocot Model Plant, Brachypodium distachyon

  • Brachypodium distachyon has been developed as a monocot model plant for temperate grasses and bioenergy crops. Although B. distachyon research is moving forward rapidly, the study of photoresponses has not been explored. To extend our knowledge of responses to light in monocots, we performed photoresponse analysis of B. distachyon using two inbred lines, Bd21 and Bd21-3. In this study, we first compared growing phenotypes between the two lines and investigated coleoptile and primary leaf growths under dark, far-red, red, and white light conditions. The results showed that the growth of the two lines were similar until tillering stage, but other developmental stages from heading to senescence were much delayed in Bd21-3, which resulted in increased height and tiller numbers. Under different light conditions, primary leaf lengths were kept increasing during the growth period, whereas the coleoptile extension was inhibited 4 to 7 days after growth depending on the light conditions applied. These results suggest that the responses to light in B. distachyon can be examined by measuring coleoptile lengths approximately 7 days after seedling growth. Moreover, we selected light-responsive genes known in Arabidopsis thaliana, such as chlorophyll A/B binding protein (CAB), light-harvesting chlorophyll binding protein (Lhcb) and chalcone synthase (CHS), and confirmed their light-induced gene expression in B. distachyon. Therefore, the present study suggests that the inhibition of coleoptile growth can be used as the parameter to analyze photoresponses in the monocot model plant, and also provide the reference genes whose expression is induced by far-red and red light treatment.

Effects of Priming and Growth Regulator Treatment of Seed on Emergence and Seedling Growth of Rice

  • An experiment was carried out to determine the effects of priming and growth regulator treatment of seeds on the emergence and seedling growth of rice, Oryza sativa L. (cv. 'Ilpumbyeo'). Normal seeds were primed in a -0.6 MPa polyethylene glycol solution at 15$^{\circ}C$ for four days with air-bubbling. Then both primed and non-primed seeds were soaked in water, 100 ppm GA$_3$, 2 ppm ABA, and 10 ppm kinetin solutions for 24 hours. The seeds were planted in soil at 3 and 5 cm depths and allowed to germinate in a growth cabinet at 2$0^{\circ}C$. Generally, the emergence rate at the 3 cm seeding depth was higher and emerged faster compared with the 5 cm seeding depth. The emergence rate of primed seeds was higher and emerged faster compared to non-primed seeds. GA$_3$ and kinetin treatments were the most effective to improve the emergence rate of non-primed seeds. Coleoptile length at the 5 cm seeding depth was longer than that at the 3 cm seeding depth. Leaf number, plant height, and root length of primed seedlings were higher compared with non-primed seeds. GA$_3$ promoted the elongation of plant height and mesocotyle length.

캘리포니아 벼 품종의 수중 용존 산소 흡수와 발아 (Germination and Dissolved Oxygen Uptake of California Rices in Water Seeding.)

  • 캘리포니아 담수직파재배용 품종인 L-202, M-202, S-201, Italiconaverneco와 한국의 화성별르 공시히여 $25^{\circ}C$의 수온하에서 용존산소의 흡수 양상과 발아특성을 조사한 결과는 다음과 같다. 1. 벼 품종들의 침종후 용존산소흡수의 경시적 변화를 보면 침종 1일부터 Italiconaverneco가 가장 왕성히 흡수하였고 다음이, S-201, 화성벼, L-202, M-2-202 순이었다. 2. 초엽과 종자근이 완전히 출현한 침종 4일후의 과적 용존산소 흡수률은 Italiconaverneco 볍씨 1개가 211.7${\mu}g$을 흡수하였고 S-201이 179.5${\mu}g$, 화성벼는 144.3${\mu}g$, L-202와 M-202는 각각 140.4, 135.8${\mu}g$이었다. 3. 초엽의 출현율은 Italiconaverneco와 L-202가 가장 높았고 다음이 M-202, S-201이 었으며 화성벼가 가장 낮았다. 4. 종자근의 출현율은 Italiconaverneco가 가장 높았고 다음이 S-201, 화성벼, L-202, M-202 순으로 낮았다. 5. 초엽과 종자근의 길이는 Italiconaverneco가 월등하게 길었고 다음이 S-201, 화성벼이었고 L-202와 M-202가 가장 짧았다.

Morphological and molecular characterization of germinability related to direct-seeding in rice varieties

  • Direct-seeding cultivation of rice is increasing in Asia instead of transplanting system, because of its lower cost and operational simplicity. Low-temperature germinability (LTG) and anaerobic germinability (AG) are important characters for breeding of varieties for wide-spread adoption of direct-seeding cultivation in rice. This study was performed to characterize LTG and AG of seven rice varieties and identify varieties with strong germinability on both low-temperature and anaerobic conditions. The mean germination rate and germination vigor of seven varieties were 51.7% and 6.0 under low-temperature condition, respectively. Among these varieties, Cheongcheongjinmi and Hwanggeumnodeul had the highest germination rate of 80%, indicating that Cheongcheongjinmi and Hwanggeumnodeul have a good LTG. In anaerobic conditions, the germination rate and coleoptile length for all varieties were 47.6% and 3.2 cm, respectively. Of them, the highest germination rate and coleoptile length were observed in Subo and Hopum, respectively, suggesting that these two varieties are tolerant to anaerobic during germination stage. Molecular characterization by SDS-PAGE revealed that the protein patterns differed at 50 kDa, 40 kDa, and 22 kDa between low-temperature and anaerobic conditions. Varieties identified as good LTG or AG in this study may be used for developing new direct-seeding rice cultivars through pyramiding these traits in the breeding program.
