수종 곡류추출물의 효소적갈변 억제효과 (Effect of Several Cereal Extracts on Enzymatic Browning)

  • 보리가루, 엿기름, 찹쌀가루의 수용성 추출물과 농축액이 효소적 갈변에 대한 억제 효과가 있는지를 PPO 활성에 대한 저해효과를 통하여 알아보았으며, PPO에 대한 저해효과가 가장 크게 나타난 엿기름 농축액의 갈변 정도와 환원성을 측정하고 이들 성질과 PPO활성에 대한 저해효과와의 상관관계도 조사하였다. 나아가 사과 슬라이스를 엿기름 농축액에 침지한 후 냉장하고 냉장 중 사과 슬라이스의 조직감과 갈변 정도를 조사한 결과는 다음과 같다. PPO에 대한 억제 효과는 엿기름 농축액이 가장 크게 나타났다. 엿기름 농축액은 가열 시간에 따라 당도, 갈변정도와 환원성은 증가하는 경향을 나타냈으며, PPO 활성은 감소하는 경향을 보였다 이러한 변화는 가열 60분까지는 서서히 변화하였으나 70분 이후에는 급격한 변화를 나타내었다. 또한 가열시간에 따라 엿기름 농축액의 pH는 감소하였다. 한편 엿기름 농축액에 침지한 사과 슬라이스의 냉장 저장중의 갈변정도는 6.9%로, 조직감의 변화는 14.3%로 가장 낮았다. 따라서 엿기름 농축액이 사과 슬라이스의 갈변 억제와 조직감 유지에 효과가 있는 것으로 나타났다.

The Effects of Natural Antioxidants on Protein Oxidation, Lipid Oxidation, Color, and Sensory Attributes of Beef Patties during Cold Storage at 4℃

  • This study aimed to investigate the use of golden thread extract (GTE), clove extract (CE), and commercially available nitrite for retarding lipid and protein oxidation and for maintaining color stability and sensory attributes in beef patties stored at $4^{\circ}C$. GTE, CE, and nitrite treatment samples were found to be efficient in retarding lipid oxidation as all three treatments resulted in low thiobarbituric acid reactive substance (TBARS) content (p<0.05). By using GTE, CE, and nitrite into beef patties, protein oxidation was not developed. Incorporation of GTE and CE into beef patties maintained color stability by protecting against the decrease of $L^*$, $a^*$, $b^*$, chroma, and hue angle values and exhibited significant influence on sensory characteristics, including color and odor of beef patties (p<0.05). Compared to commercially available nitrite, GTE and CE were more effective as antioxidants for inhibiting lipid oxidation, and preserving color stability of fresh beef patties. The study indicated that GTE and CE could be utilized efficiently to extend the shelf life of beef patties.

Freeze Tolerance Enhanced by Antifreeze Protein in Plant

  • When plants are exposed to subfreezing temperatures ice crystals are forming within extracelluar space in leaves. The growth of ice crystal is closely related to the degree of freezing injury. It was shown that an antifreeze protein binds to an ice nucleator through hydrogen bonds to prevent growth of ice crystal and also reduces freezing damage. The antifreeze proteins in plants are similar to PR proteins but only the PR proteins induced upon cold acclimation were shown to have dual functions in antifreezing as well as antifungal activities. Three of the genes encoded for CLP, GLP, and TLP were isolated from barley and Kentucky bluegrass based on amino acid sequence revealed after purification and low temperature-inducibility as shown in analysis of the protein. The deduced amino acid of the genes cloned showed a signal for secretion into extracellular space where the antifreezing activity sup-posed to work. The western analysis using the antisera raised against the antifreeze proteins showed a positive correlation between the amount of the protein and the level of freeze tolerance among different cultivars of barely. Besides it was revealed that TLP is responsible for a freeze tolerance induced by a treatment of trinexapac ethyl in Kentucky bluegrass. Analysis of an overwintering wild rice, Oryza rufipogon also showed that an acquisition of freeze tolerance relied on accumulation of the protein similar to CLP. The more direct evidence for the role of CLP in freeze tolerance was made with the analysis of the transgenic tobacco showing extracellular accumulation of CLP and enhanced freeze tolerance measured by amount of ion leakage and rate of photosynthetic electron transport upon freezing. These antifreeze proteins genes will be good candidates for transformation into crops such as lettuce and strawberry to develop into the new crops capable of freeze-storage and such as rose and grape to enhance a freeze tolerance for a safe survival during winter.

  • 체세포배발생을 이용한 가시오갈피 대량증식체계 확립을 위하여 저온처리에 의한 체세포배의 장기저장 실험을 수행하였다. 현탁배양 체세포배의 $4^{\circ}C$에서의 저온저장에는 구형배 이전의 조기단계의 체세포배가 적합하였고 $2\;g/{\ell}$ 이하의 낮은 접종밀도에서는 36개월 이상 저장이 가능하였다. 장기간 저온 저장한 체세포배는 상온에 옮긴 후 2,4-D를 첨가한 배지에서만 성장을 회복하여 배발생캘러스를 형성하였으며 배발생캘러스의 증식속도는 배양온도를 $32^{\circ}C$로 높였을 때에 가장 효율적이였다.

항균성 동치미액의 첨가에 의한 냉면국물 중의 Listeria monocytogenes 및 Escherichia coli O157:H7 생육억제 (Growth Inhibition of Listeria monocytogenes and Escherichia coli O157:H7 in Naengmyon-Broth by addition of Antibacterial Dongchimi-Juice)

  • Juice of Dongchime a Korean traditional vegetable food fermented with lactic acid bacteria has been traditionary used as broth for Naengmyon a Korean cold noodles with broth. This study was carried out to demonstrate the growth inhibition of two food born pathogens Listeria monocytogenes and Escherichia coli O157:H7 in Naengmyon-broth containing Dongchimi-juice fermented with high antibacterial lactic acid bacteria Lactobacillus homohiochii B21 and Leuconostoc mesenteroides C16. Naengmyon-broth were made with beef broth and Dongchimi-juice fermented with lactic acid bacteria and the changes in viable cell counts of the inoculated pathogens in Naengmyon-broths were investigated during storage at 2$0^{\circ}C$ and 1$0^{\circ}C$. In Naengmyon-broth of 100% Dongchimi-juice stored at 2$0^{\circ}C$ the numbers of Listeria monocyto-genes and Escherichia coli O157:H7 were rapidly decreased from 106CFU/ml to 106CFU/ml in 8 hours and 40 hours respectively. In Naengmyon-broth containing 50% Dongchimi-juice their numbers were also rapidly decreased though the decreasing rates were not so fast as those in 1005 Dongchimi-juice. In Naengmyon-broth containing 10% Dongchimi-juice the growths of the two pathogens were markedly inhibited compared with those in 100% beef broth though some growths were occurred in early phase. But in Naengmyon-broth of 100% beef broth their growths were very fast from early. Antibacterial ac-tivity of the Dongchimi-juice was more distinct at 2$0^{\circ}C$ than at 1$0^{\circ}C$ and was more active against Listeria monocytogenes than against Escherichia coli 157:H7.

  • 대두(大豆) 및 대두가공품첨가(大豆加工品添加)에 따른 유지안정성(安定性)을 검토하기 위하여 대두유(大豆油)에 대두(大豆) 및 대두가공품(大豆加工品)의 용매추출물(抽出物)을 첨가하고 또한 Ramyon을 제조할 때 대두(大豆) 및 대두가공품(大豆加工品)을 첨가하여 그 유지(油脂)의 산가(酸價), 과산화물가(過酸化物價), TBA가(價)를 측정(測定)하였다. 1. 대두유(大豆油)에 대한 항산화효과(抗酸化?果)는 분리대두단백추출물(分離大豆蛋白抽出物)이 가장 크고 탈지대두추출물(脫脂大豆抽出物), 대두열수추출물(大豆熱水抽出物)이 그 다음가고 탈지대두(脫脂大豆) Ethanol추출물(抽出物)도 유효(有效)하였으나 대두냉수추출물(大豆冷水抽出物)은 효과(?果)가 거의 없었다. 2. Ramyon지질(脂質)에 대하여 저장기간마다 산가(酸價), 과산화물가(過酸化物價) 및 TBA가(價)의 변화는 분리대두단백가수분해물(分離大豆蛋白加水分解物)이 가장 변화(變化)가 적었고 탈지대두가수분해물(脫脂大豆加水分解物), Tempeh, Tempeh추출물(抽出物), 탈지대두분말(脫脂大豆粉末)첨가 순으로 큰 변화(變化)를 보여 대체적으로, 가수분해물(加水分解物)첨가는 매우 효과적인 산화방지작용(酸化防止作用)을 나타냈다.

부추의 항균활성 특성과 식품에의 응용 (Antimicrobial Activity of Korean Leek and Its Application to Food System)

  • 천연 항균활성 물질에 관한 대부분의 연구는 약품의 제조와 첨가물로서의 이용을 목적으로 하였으나, 물질의 효용면에서 연구결과를 실용화시키기에는 미흡한 실정이다. 이러한 사실은 천연물질의 항균성을 실제 식품에 이용하는 덴 필요한 가공적성의 연구가 따르지 못한 것이 주요한 이유라고 여겨진다. 본 연구는 우리나라 사람들이 즐겨먹는 부추의 항균활성을 연구하되, 식품에 이용할 수 있도록 항균물질의 특성규명과 더불어 가공적성을 평가하여 실제 적용하는데 목적을 두었다. 생부추즙을 agar diffusion method로 항균활성을 측정한 결과는 Bacillus cereus, Bacillus subtilis, Stayhlococcus aureus, Pseudomonas fluorescens. E. coli O157:H7 등의 세균류와 효모인 Pichia membranaefaciens등에 항균작용을 나타내었다. 생부추 착즙과 생부추 동결 후 착즙한 시료에서만 항균활성을 나타내었고 착즙한 후의 동결저장 시료나 데친 시료에서는 항균활성이 소실되었다 한편 여과와 원심분리 및 진공농축 그리고 pH변화 등에 의해서는 항균활성의 변화가 없었다. 항균활성 기초자료에 근거하여 가공식품의 모델로서 부추 마쇄시료를 첨가한 두유와 두부 및 냉면육수를 제조하여 식품응용 시험을 한 결과 부추를 첨가한 각각의 제품에서 미생물 성장 저해효과가 나타났으며, 특히 냉면육수의 경우 인위적으로 Bacillus cereus를 접종시켰음에도 불구하고 1$0^{\circ}C$, 20시간 저장시 부추를 첨가한 육수에서 미생물이 완전히 사멸되는 효과를 얻을 수 있었다.

A Modified Cryopreservation Method of Psychrophilic Chlorophyta Pyramimonas sp. from Antarctica

  • Polar psychrophiles which thrive under extreme conditions such as cold temperature, high salinity, and high dose ultraviolet light, emerge as novel targets for biotechnology. To prevent genetic drift and the possibility of contamination by subculturing, cryopreservation was employed for two psychrophilic microalgae, Porosira sp. (KOPRI AnM0008) and Pyramimonas sp. (KOPRI AnM0046), which have anti-freeze activities. Five cryoprotectants (dimethyl sulphoxide, ethylene glycol, glycerol, methanol and propylene glycol) showed toxicity at 20-30% (v/v). The optimal cryoprotectant concentration and equilibration time were less than 20% and 10 min, respectively. Cryopreservation was carried out in the presence of cryoprotectants either by direct freezing in liquid nitrogen ($LN_2$) or controlled freezing using a controlled rate freezer followed by storage in the $LN_2$ tank. As a result, Pyramimonas sp. (KOPRI AnM0046), a psychrophilic chlorophyta was revived. Cryopreserved Porosira sp. was not revived from either freezing protocols probably due to the silicic cell wall and its relatively large cell size. In the case of Pyramimonas sp. (KOPRI AnM0046), the controlled freezing method showed higher revival yield than the direct freezing method.

Transgenic Tobacco Plants Expressing the Bacterial Levansucrase Gene Show Enhanced Tolerance to Osmotic Stress

  • Fructans are polyfructose molecules that function as nonstructural storage carbohydrates in several plants. In addition, it has been suggested that, due to their solubility, they can play an important role in helping plants survive periods of osmotic stress. In order to study the effect of levan synthesis on plant growth, the coding region of the levansucrase gene, which was isolated from Zymomonas mobilis, was introduced into tobacco plants using Agrobacterium tumefaciens-mediated transformation. The presence of the levansucrase gene in transgenic plants was verified by genomic DNA gel blot analysis. RNA gel blot and immunoblot analyses showed an accumulation of the corresponding transcript and protein product of the bacterial levansucrase gene in transgenic plants. Furthermore, a thin layer chromatography analysis revealed that fructans were synthesized and deposited in transgenic tobacco plants. When $T_1$ seeds were germinated and grown under polyethylene glycol-mediated drought stress or cold stress, the transgenic seedlings displayed a substantially higher level of growth than that of untransformed plants. These results suggest that fructans may playa significant role in the tolerance of plants under osmotic stress.

흑밀복(Lagocephalus gloveri)을 이용한 레토르트파우치 복국의 제조 및 품질특성 (Processing and Quality of a Retort Pouched Soup Made from Brown-backed Toadfish Lagocephalus gloveri)

  • To obtain a value-added product from the non-toxic brown-backed toadfish Lagocephalus gloveri (pufferfish), we developed a retort pouched pufferfish soup (RPS) and characterized its processing conditions and quality metrics. We found that the most appropriate manufacturing process for the RPS consisted of detoxifying and cold-water dipping the pufferfish flesh, blanching it, and adding it to the retort pouch along with other ingredients (hot-water extract of pufferfish head and carcass, radish, bean sprouts, and garlic), after which the pouch was sealed, sterilized (120℃, F0 value 7.5-10 min.), cooled, and inspected. The moisture, crude protein, and total volatile basic nitrogen contents of the RPS were 97.2%, 1.3% and 7.7 mg/100 g, respectively. The total free amino acid content was 903.2 mg/100 g, and the main free amino acids were glutamic acid, taurine, lysine, glycine, threonine, alanine, arginine, proline, and hydroxyproline. Sterilizing the RPS for up to F0 value 10 min. did not cause any major problems in terms of chemical or sensory qualities. This RPS has good storage stability and organoleptic qualities compared with similar commercial pufferfish soups and is suitable for commercialization as a value-added instant seafood soup.
