미니돼지에서 University of Wisconsin 용액과 Histidine-Tryptophan Ketoglutarate 용액의 신장 저온보존효과 비교 (Comparison of the Effectiveness of University of Wisconsin and Histidine-Tryptophan Ketoglutarate Solution on Renal Cold Preservation in the Miniature Pig)

  • 이식분야에서 University of Wisconsin (UW)용액과 Histidine-Tryptophan Ketoglutarate (HTK)용액은 장기 보존액으로 주로 이용되고 있다. 바이오 장기 동물모델인 미니돼지를 이용하여 신장 저온보존에서 발생하는 허혈손상을 비교함으로써 각 보존액의 효용성을 검토하고자 하였다. 이 연구에서는 12마리의 미니돼지가 이용되었으며, 각 6마리씩 무작위로 선별하여 UW군(n = 6)과 HTK군(n = 6) 두 군으로 나누었다. 미니돼지의 신장을 적출, 세척, 각각 저온 보관(0, 24, 48, 72시간, $4^{\circ}C$) 후에 조직학적 평가를 실시하였다. 저온보관시간이 증가할수록 신장의 손상이 증가하였다. 저온보관 24시간까지는 UW와 HTK용액의 저온보관 효과는 유사하였지만, 48시간 이후에는 HTK 용액의 저온보관손상이 증가하였다. 따라서, 미니돼지 신장의 저온 보존액은 UW 용액이 효과적이며 특히 장기간 보존에 적합한 용액으로 사료된다.

제주산 흥진조생 온주밀감의 저온저장 (Cold stroage of Citrus unshiu Marc. var. okitsu produced in Cheju)

  • The conditions of cold stroage of Citrus unshiu Marc. var. okitsu produced in Cheju were investigated. The changes of peel moisture contents soluble solids, total sugar, vitamin C and density were slightly occured, and decay ratio was below 20% on keeping freshness relatively till late of March during cold storage. After thats decayed citrus fruits were increased gradually mainly from cold injury with lower temperature and high humidity. The loss of fruit weight, decrease of fruit hardness, and decrease of acid content were occured gradually during cold stroage. Because of the difficulty of long term storage for Citrus unshiu, the conditions and periods of cold storage would be determined after considering the physicochemical properties of fruits every year.

시판 보냉팩 개수에 따른 진공포장 냉동 넙치(Paralichthys olivaceus) 및 고등어(Scomber japonicus) 필렛의 상온유통 중 선도유지능 비교평가 (Comparative Assessment of Freshness Preservation in Vacuum-Packed Frozen Olive Flounder Paralichthys olivaceus and Mackerel Scomber japonicus Fillets During Room Temperature Distribution Using Varied Quantities of Commercially Available Cold Packs)

  • The surge in online seafood consumption has increased parcel delivery, leading to a need to implement effective preservation methods. As the cold chain system is not fully established in Korea, styrofoam boxes and cold packs are commonly used for low-temperature seafood distribution. The impact of cold packs on product preservation depends on the number utilized. Herein, the freshness of vacuum-packed frozen fish fillets (Paralichthys olivaceus and Scomber japonicus) stored at 25±0.5℃ for up to 84 h was measured. Chemical (pH and volatile base nitrogen), microbiological (viable cell count), and physical (odor intensity) properties were assessed using 2 or 4 cold packs in a styrofoam box. Four cold packs yielded lower values, indicating superior freshness, and extended fish freshness by approximately 12 h compared with two cold packs. Therefore, it is recommended to use a minimum of 4 cold packs (-350 g/cold pack) in a styrofoam box for distributing approximately 300 g of frozen fish fillets at room temperature during the summer, considering an average delivery period of 2 days in Korea.

토끼의 대동맥 및 폐동맥 판막 동종이식편의 냉장 및 냉동 보존후 생육성 평가(I) (Viability Assay after $4^{\circ}C$ Cold Preservation & Cryopreservation of Aortic & Pulmonic Allograft Valves in Rabbits)

  • Cardiac valve allografts have been used as replacements for diseased valves and right ventricular outflow tract reconstruction, the long term follow-up of which has been reported satisfactory. For a good long-term result, it is essential that the allograft be viable at implantation. In this study, we aimed at preparing the cardiac valve allografts aseptically, preserving them at cold- and cryo-conditions, and testing the viability of the allografts after preservation by four methods. We tested the viability of the cardiac valve allografts preserved in cold refrigerated state[4$^{\circ}$C in nutrient media & in liquid nitrogen tank[cryopreservation under -149$^{\circ}$C for pre-planned time periods. The testing methods were 1 glucose utility test 2 tissue culture 3 thymidine uptake test and 4 histologic evidence by light microscopy. We observed no differences in the viability between cold- & cryo-groups and similar results among the methods for testing the viability. In conclusion, there was no difference in the viability between cold- and cryopreserved-allografts at least for 14 days of preservation. And glucose utility test and thymidine uptake test were satisfactory in the evaluation of the allograft viability, since they were easy and rapid with relatively quantitative results.

Studies on Long-term Preservation of Eggs of Indian Tropical Multivoltine Silkworm (Bombyx mori L.) Genetic Resources

  • The silkworm rearing and growth parameters of 63 multivoltine silkworm accessions under extended period of egg preservation at 5$^{\circ}C$ from 30 days to 45 days were studied. The results indicate that, nine accessions did not respond to extended period of egg preservation at low temperature and the remaining 54 accessions responded to the treatment and three rearings were conducted for comparision with the control; to estimate the effect of prolonged egg preservation at low temperature. The non-parametric tests statistics (Wilcoxon tests) was adopted for comparing the mean performance of treated batches (45 days) over the control (30 days). Highly significant variability was found among the accessions for all the parameters under study. The genetically controlled morphological characters were not altered in the treated batches, which were found to be on par with that of control. However, the total larval duration varied significantly over the control in 51 accessions. Similarly, the fifth age larval duration of 27 accessions showed decreasing trend compared to control. Altogether 41 accessions were found to be tolerant to long-term cold preservation upto 45 days, without showing any significant variation for morphological as well as essential quantitative traits. These accessions may be recommended for long-term egg preservation schedule up to 45 days, which will reduce the cost of conservation of these silkworm germplasm.

제주산 만감류 청견의 저온저장 (Cold Storage of Kiyomi Tangor Produced in Cheju)

  • Physicochemical properties and the conditions of cold storage of kiyomi clangor(Citrus unshiu x sinensis) produced in Cheju were investigated. Firmness, edible part ratio, soluble solids, and acid content were decreased gradually with incurasing fruit size. Carbohydrates in juice were consisted of 65.12% sucrose, 19.65% fructose, and 15.23% glucose The main organic acrid In juice was 57.4% of citric acid, and others were lactic acid, malic acid, oxalic acid and fumaric acid, respectively. Weight loss were occured very. slowly to about 6% till late of May, but decayed fruits were occurred very slowly to about 6% till late of May, but decayed fruits were arisen to about 8% at 3$^{\circ}C$, and about 13% at 5$^{\circ}C$ of storage temperature. The changes of peel moisture content, soluble solids, total sugar, vitaamin C, and density of fruits were slightly occured during cold storage. Cold storage at 3$^{\circ}C$ and 87% relative humidity kept freshness of citrus fruits for a long-term without damage of cold injury.

  • This study was conducted to determine the effects of storage conditions on apples treated with postharvest fungicides, benomyl and bitertanol. The fungicideds were applied to control Postharverst disease in apples during CA and cold storage. The stored Apple were tested monthly for weight loss, flesh firmness, titratable acidity, prix and free sugar. Relative to the control group, the pstharvest fungicide stoup had less disease. The fungicide treated apples stored in CA had a higher measured weight, better firmness and maintained acidity, prix and free sugar when compared to the control stoup monthly and after 200days. The fungicide treated apples in cold storage maintained their quality for 120days.

곤충병원선충(Steinernema carpocapsae)의 최적보관을 위한 환경요인 (Optimal Storage Conditions of the Entomopathogenic Nematode, Steinernema Carpocapsae)

  • Effects of temperature, pH, and salt concentration on the preservation of the infective juvenile entolopathogenic nematodes, Steinernema carpocapsae, were analyzed. The effects of acclimation to sublethal low temperature and glycerol concentrations on rapid cold hardening of the nematodes were also evaluated to set up the conditions for their cryopreservation. All tested environmental factors could change the storage effectiveness of the nematodes. These analyses indicated that the optinal storage condition for the nematodes consisted of $15^{\circ}C$, pH 8.5, and 0.5-1.0% salt concentration. The nematodes showed a rapid could hardening in response to $5^{\circ}C$ for 2h. Glycerol induced the nematodes to increase the cold hardiness. Its high concentration over 10%, however, gave a harmful effect on the nematode survival.

  • Human body fluid and humor include not only sweat, joint fluid but also every fluids, for example, blood, essence, kidney essence and marrow. Historically, in the oriental medicine, there are a lot of efforts in order to preserve fluid and humor. In "Treatise on Cold Damage and Miscellaneous Disease (傷寒雜病論)", when treating cold damage, preservation of fluid and humor is put first. I tried to find out the relation about fluid and humor and pain treatment in "Treatise on Cold Damage and Miscellaneous Disease". So, I investigated sentences related to pains in "Treatise on Cold Damage and Miscellaneous Disease" "greater yang disease part" and commentaries of several chinese medical doctors. And I divide pain treatments into four categories. (fluid and humor sufficiency-exterior pattern, fluid and humor sufficiency-interior pattern, fluid and humor deficiency-exterior pattern, fluid and humor deficiency-interior pattern) At first, when treating pains of cold damage, there are many considerations about deficiency and sufficiency of fluid and humor through pulse condition and symptoms. Second, in pain-cases of fluid and humor sufficiency, purge methods is chosen. And in pain-cases of fluid and humor deficiency, tonifying methods is chosen. Finally, one of the main objects of "Treatise on Cold Damage and Miscellaneous Disease" is preservation and supply of fluid and humor.

Effects of cold and room temperature storage on the sprouting and marketability of early-grown spring potatoes

  • Three spring potato varieties (Atlantic, Chubaek, and Superior) were cultivated for 80, 90, and 100 days and compared in terms of their total and marketable yields and specific gravities. Subsequently, changes in the quality parameters (weight loss, sprouting rate, disease incidence, dry weight, and specific gravity) of potatoes cultivated for 100 days were monitored during two-month storage in improved and semi-underground warehouses at 4℃ and room temterature. The productivities of Atlantic, Superior, and Chubaek were maximized at cultivation periods of 100, 100, and >90 days, respectively. In all cases, cold storage resulted in <5.3% weight loss and no marketability loss. However, two-month room temperature storage resulted in pronounced marketability loss due to weight loss (6.2, 7.3% and 10.9% for Atlantic, Superior, and Chubaek, respectively) and sprouting (in ~13%, >75%, and 99% of Atlantic, Superior, and Chubaek tubers, respectively). The marketability of Atlantic, Superior, and Chubaek during cold storage was maintained for >2, ~2, and <2 months, respectively. All varieties were characterized by rapid sprouting, low dry weight and specific gravity, and rapid marketability loss during storage.
