Factos affecting the production of butanol and acetone by Clostridium acetobutylicum

  • Owing to the growing interest in the production of fuels and chemicals from biomass the well-know butanol-acetone fermentation as carried out by Clostridium acetobutylicum has been intensely studied again in recent years. Several solvent-yielding fermentation processes were established which are operated by using batch cultures or continuous cultures. 1 could be shown that under conditions of phosphate limitation an asporogenous mutant of C. acetobutylicum establishes itself in a chemostat which produces the solvents continuously. Attempts have been made to change the butanol/acetone ratio in favor of butanol production. A corresponding shift of the product spectrum can be achieved by carbon monoxide addition to the head space of the fermentation (B.H. Kim et al., App. Envioron. Microbiol. 48, 764-770 1984) or by iron limitation. Progress has been made in understanding the mechanism underlying the shift from acid to solvent prodcction. Experimental results are in agreement with the view that intracellular accumulation of acetic and butyric acid results in a shortage of phosphate and coenzyme A. This shortage may serve then as signal for the synthesis of the enzymes involved in the formation of acetone and butanol.

  • Cell-free extracts of Streptomyces neyagawaensis SL-387 grown on a chemically defined medium supplemented with DL-3-amino-3-phenylpropionic acid (APP) produced N-acetyl-APP (Ac-APP) in the presence of APP and acetyl coenzyme A. The APP obtained by acid hydrolysis of the Ac-APP was D-configuration: $[\alpha]_D+6.5^{\circ}(H_2O)\;at\;20^{\circ}C$, optical purity 92% enantiomeric excesses (ee). These results suggest that an N-acetyltransferase exists in the cell-free extract as a novel enzyme with specificity for D-APP.

Effects of Dehulled Defatted Soy Flour on Cholesterol Metabolism and Gastronitestinal Physiology in Rats (거피 탈지 콩가루 식이가 흰쥐의 콜레스테롤 대사와 소화생리에 미치는 영향)

  • This study was carried out to explore the hypocholesterolemic effect of dehyulled defatty soy flour and its possible mechanisms including endocrine status, cholesterol biosynthesis, and fecal excretion in rats. Animals fed casein were used as control and each phospholipid compared with casein feeding. Cholesterol concentrations in all lipoprotein fraction were significantly lower in defatted soy flour group compared with casein-fed control. Defatted soy flour feeding also significantly lowered hepatic total lipid, cholesteol and TG, and increaed fecal bile acid excretion by 270% compared with casein feeding. Defattd soy flour feeding had no significant effect on plasma thyroid hormone levels and hepatic 3-hydroxy-3-methyl glutary coenzyme A(HMG-CoA) reductase activity. However, plasma T4 concentration was slightly elevated and HMG-CoA reductase activity was suppressed in defatted soy flour group. These metabolic alterations partially explain the reduced plasma and hepatic cholesterol levels of rats fed defatted soy flour.

  • 아시아 및 한국에서 처음 발견된 3명의 소아 Ethylmalonic encephalopathy (EE) 환자들은 riboflavin을 이용한 치료법에 점진적인 호전을 보인 것으로 나타났다. Ethylmalonic aciduria가 3가지 효소-2-methyl-branched-chain acyl CoA dehydrogenase (2MBCAD), iso-valeryl CoA dehydrogenase (IVCAD), 그리고 short-chain acyl CoA dehydrogenase (SCAD) - 들의 부분적인 결함으로 생긴다는 가설하에 dehydrogenase (탈수효소)의 보조 효소인 riboflavin이 부분적으로 나마 치료 과정에서 어떤 역할을 수행하는지의 여부를 살펴보았다. 다음의 세 증례에서 riboflavin 혹은 coenzyme Q10을 병행하여 치료한 결과 인지 행동(cognitive behavior)과 만성 점액성 설사(chronic mucoid diarrhea)가 호전된 것으로 관찰되었다. 또한, 치료 후 운동 기능도 일부 호전되었다. EE의 질병 발생 기전이 현재까지 완전히 밝혀지지는 않았지만, 세 명의 EE 환자들의 치료경과를 관찰하여 위의 가설을 뒷받침 할 수 있었다.

  • Originally, vitamin K was defined as a factor for blood coagulation. Now more attention is focused on vitamin K for bone metabolism and bone health. Vitamin K is a coenzyme for glutamate carboxylase which converts glutamate residues to ${\gamma}$-carboxyglutamate(Gla) residues. Gla residues have calcium binding ability and bound to hydroxyapatite crystals in bone. Vitamin K promotes the carboxylation of osteocalcin and matrix Gla-protein, vitamin K-dependent proteins and improves bone mineral density and bone mass. Vitamin K deficiency causes reductions in bone mineral density and increases the risk of osteoporotic bone fractures, resulting from undercarboxylated osteocalcin. This paper is to provide a brief information of vitamin K and its role in bone health.

  • The skeletal muscle in our body is a major site for bioenergetics and metabolism during exercise. Carbohydrates and fats are the primary nutrients that provide the necessary energy required to maintain cellular activities during exercise. The metabolic responses to exercise in glucose and lipid regulation depend on the intensity and duration of exercise. Because of the increasing prevalence of obesity, recent studies have focused on the cellular and molecular mechanisms of obesity-induced insulin resistance in skeletal muscle. Accumulation of intramyocellular lipid may lead to insulin resistance in skeletal muscle. In addition, lipid intermediates (e.g., fatty acyl-coenzyme A, diacylglycerol, and ceramide) impair insulin signaling in skeletal muscle. Recently, emerging evidence linking obesity-induced insulin resistance to excessive lipid oxidation, mitochondrial overload, and mitochondrial oxidative stress have been provided with mitochondrial function. This review will provide a brief comprehensive summary on exercise and skeletal muscle metabolism, and discuss the potential mechanisms of obesity-induced insulin resistance in skeletal muscle.

Comparative Study of Red Wine and Korean Black Raspberry Wine in Adipocyte Differentiation and Cardiovascular Disease Related Gene Expression

  • Abilities of Korean black raspberry (KBR) wine to induce differentiation of 3T3 L1 adipocytes and express HepG2 cardiovascular disease-related genes were determined and compared with those of red wine. Red wine attenuated the differentiation of adipocytes faster than KBR wine and control. KBR wine decreased cholesterol concentration in HepG2 cells at a similar level to that of red wine, resulting in similar degrees of suppression in apolipoprotein B100, and enhancement in LDL receptor and 3-hydroxy-3-methylglutaryl coenzyme A reductase mRNA levels.

  • Glutaric aciduria type 1 is an inborn error of lysine, hydroxylysine, and tryptophan metabolism caused by deficiency of glutaryl-coenzyme A dehydrogenase. The disease often appears in infancy with encephalopathy episode that results in acute basal ganglia and white matter degeneration. The majority of patients develop a dystonic-dyskinetic syndrome. This reports 6year-old boy who had been done previous gastrostomy due to swallowing difficulty underwent bilateral pallidotomy with intraoperative electromyography[EMG] monitoring for disabling dystonia. Intraoperative EMG was used to assess stimulation thresholds required for capsular responses and muscle tone. Surface EMG electrodes were placed on the face and cricopharyngeal muscles. Exact target were directly modified according to MRI-visualized anatomy. EMG response was consistently seen prior to visual observation of muscle activity. The surgery improved dystonic symptoms without swallowing difficulty.

  • Atorvastatin calcium salt (1) was obtained through the preparation of lactone compound (8) from 2-((4R,6R)-6-(2-(2-(4-fluorophenyl)-5-isopropyl-3-phenyl-4-(phenylcarbamoyl)-1H-pyrrol-1-yl)-ethyl)-2-phenyl-1,3,2-dioxaborinan-4-yl)acetic acid tert-butyl ester (9) by hydrolysis in basic condition. Efficient hydrolysis of boronate compound 9 aimed at the viable synthesis for commercial production and purification of Atorvastatin calcium is reported. Detail studies of evaluation procedure are also reported.

  • Flavin-peptides were purified from pyranose oxidase (EC 1.1.3.10) after tryptic- chymotryptic and tryptic digestion. The spectral and chromatographic properties of these flavin peptides showed that the FAD of pyranose oxidase appears to be bound, by way of the 8${\alpha}$-methylene group, to the N-l position of the imidazole ring of the histidine. Automated sequence analysis showed that the amino acid sequence of the tryptic-chymotryptic flavin-peptide from pyranose oxidase is Ser-Thr-X-Trp and that of the tryptic flavin-peptide is Met-Ser-Thr-X-Trp.
