Improvement of an Analytical Method for Methoprene in Livestock Products using LC-MS/MS (LC-MS/MS를 이용한 축산물 중 살충제 메토프렌의 잔류분석법 개선)

  • The research aims to develop a rapid and easy analytical method for methoprene using liquid chromatography-tandem mass spectrometry (LC-MS/MS). A simple, highly sensitive, and specific analytical method for the determination of methoprene in livestock products (beef, pork, chicken, milk, eggs, and fat) was developed. Methoprene was effectively extracted with 1% acetic acid in acetonitrile and acetone (1:1), followed by the addition of anhydrous magnesium sulfate (MgSO4) and anhydrous sodium acetate. Subsequently, the lipids in the livestock sample were extracted by freezing them at -20℃. The extracts were cleaned using MgSO4, primary secondary amine (PSA), and octadecyl (C18), which were then centrifuged to separate the supernatant. Nitrogen gas was used to evaporate the supernatant, which was then dissolved in methanol. The matrix-matched calibration curves were constructed using 8 levels (1, 2.5, 5, 10, 25, 50, 100, 150 ng/mL) and the coefficient of determination (R2) was above 0.9964. Average recoveries spiked at three levels (0.01, 0.1, and 0.5 mg/kg), and ranged from 79.5-105.1%, with relative standard deviations (RSDs) smaller than 14.2%, as required by the Codex guideline (CODEX CAC/GL 40). This study could be useful for residue safety management in livestock products.

Evaluation of Temperature and Precipitation over CORDEX-EA Phase 2 Domain using Regional Climate Model HadGEM3-RA (HadGEM3-RA 지역기후모델을 이용한 CORDEX 동아시아 2단계 지역의 기온과 강수 모의 평가)

  • This study evaluates the temperature and precipitation results in East Asia simulated from the Hadley Centre Global Environmental Model version 3 regional climate model (HadGEM3-RA) developed by the UK Met Office. The HadGEM3-RA is conducted in the Coordinated Regional climate Downscaling Experiment-East Asia (CORDEX-EA) Phase II domain for 15 year (2000-2014). The spatial distribution of rainbands produced from the HadGEM3-RA by the summer monsoon is in good agreement with the Asian Precipitation Highly Resolved Observational Data Integration Towards Evaluation of water resources (APRODITE) data over the East Asia. But, precipitation amount is overestimated in Southeast Asia and underestimated over the Korean Peninsula. In particular, the simulated summer rainfall and APRODITE data show the least correlation coefficient and the maximum value of root mean square error in South Korea. Prediction of temperature in Southeast Asia shows underestimation with a maximum error during winter season, while it appears the largest underestimation in South Korea during spring season. In order to evaluate local predictability, the time series of temperature and precipitation compared to the ASOS data of the Seoul Meteorological Station is similar to the spatial average verification results in which the summer precipitation and winter temperature underestimate. Especially, the underestimation of the rainfall increases when the amounts of precipitation increase in summer. The winter temperature tends to underestimate at low temperature, while it overestimates at high temperature. The results of the extreme climate index comparison show that heat wave is overestimated and heavy rainfall is underestimated. The HadGEM3-RA simulated with a horizontal resolution of 25 km shows limitations in the prediction of mesoscale convective system and topographic precipitation. This study indicates that improvement of initial data, horizontal resolution, and physical process are necessary to improve predictability of regional climate model.

A preliminary assessment of high-spatial-resolution satellite rainfall estimation from SAR Sentinel-1 over the central region of South Korea (한반도 중부지역에서의 SAR Sentinel-1 위성강우량 추정에 관한 예비평가)

  • Reliable terrestrial rainfall observations from satellites at finer spatial resolution are essential for urban hydrological and microscale agricultural demands. Although various traditional "top-down" approach-based satellite rainfall products were widely used, they are limited in spatial resolution. This study aims to assess the potential of a novel "bottom-up" approach for rainfall estimation, the parameterized SM2RAIN model, applied to the C-band SAR Sentinel-1 satellite data (SM2RAIN-S1), to generate high-spatial-resolution terrestrial rainfall estimates (0.01° grid/6-day) over Central South Korea. Its performance was evaluated for both spatial and temporal variability using the respective rainfall data from a conventional reanalysis product and rain gauge network for a 1-year period over two different sub-regions in Central South Korea-the mixed forest-dominated, middle sub-region and cropland-dominated, west coast sub-region. Evaluation results indicated that the SM2RAIN-S1 product can capture general rainfall patterns in Central South Korea, and hold potential for high-spatial-resolution rainfall measurement over the local scale with different land covers, while less biased rainfall estimates against rain gauge observations were provided. Moreover, the SM2RAIN-S1 rainfall product was better in mixed forests considering the Pearson's correlation coefficient (R = 0.69), implying the suitability of 6-day SM2RAIN-S1 data in capturing the temporal dynamics of soil moisture and rainfall in mixed forests. However, in terms of RMSE and Bias, better performance was obtained with the SM2RAIN-S1 rainfall product over croplands rather than mixed forests, indicating that larger errors induced by high evapotranspiration losses (especially in mixed forests) need to be included in further improvement of the SM2RAIN.

A Study on Evaluation Method for Structural Suitability of Constructed Wetlands in Dam Reservoirs as an Ecological Water Purification System (생태적 수질정화시설로서 댐 저수지 인공습지의 구조 적정성 평가방안)

  • Many constructed wetlands have been installed in dam reservoirs nationwide for ecological purification of watershed pollutants, but aging and reduced efficiency are becoming issues. To improve the management of constructed wetlands, an objective evaluation of structural suitability is required. This study evaluated 39 constructed wetlands of 15 dams. First, through fogus group interview(FGI), survey analysis, and analytic hierarchy process(AHP), eight evaluation items in the physical and vegetative aspects were selected and the evaluation criteria applied with weights were prepared. Second, as a result of the structural suitability evaluation, the average score of the overall constructed wetlands was 80.8, with 10 sites rated as 'good grade(91~100)', 22 sites rated as 'normal grade(71~90)' and 7 sites rated as 'poor grade(70 or less)'. The average score of physical structure evaluation was 52.6, with 14 sites rated as 'good', 21 sites as 'normal' and 4 sites as 'poor'. The suitability of location was good level in most constructed wetlands, but the water supply system, depth of water, ratio of length-to-width, and slope of flow channel were evaluated as 'normal' or less in constructed wetlands of 50% or more. Therefore, it was found that overall improvement was necessary for stable flow supply and flow improvement in the constructed wetland. The average score of vegetative structure evaluation was 28.2, and about 84% of them were identified as 'normal' or lower. As a result of analyzing the Spearman's correlation coefficient between the physical structure evaluation score and the vegetation structure evaluation score, there was a significant correlation(r = 0.728, p < 0.001), and it was found that each evaluation factor also influences each other. As a result of the case review of 6 constructed wetlands, the appropriateness of the evaluation results was confirmed, and it was found that the location, flow rate supply, and type of wetland had a great influence on the efficiency and operation of the wetland. Through this study, it will be possible to derive structural weaknesses of constructed wetlands in dam reservoirs as a nature-based solution, to prepare types and practical alternatives for improved management of each constructed wetland in the future, and to contribute to enhancing various environmental functions.

Optimization for Ammonia Decomposition over Ruthenium Alumina Catalyst Coated on Metallic Monolith Using Response Surface Methodology (반응표면분석법을 이용한 루테늄 알루미나 메탈모노리스 코팅촉매의 암모니아 분해 최적화)

  • As a result of the recent social transformation towards a hydrogen economy and carbon-neutrality, the demands for hydrogen energy have been increasing rapidly worldwide. As such, eco-friendly hydrogen production technologies that do not produce carbon dioxide (CO2) emissions are being focused on. Among them, ammonia (NH3) is an economical hydrogen carrier that can easily produce hydrogen (H2). In this study, Ru/Al2O3 catalyst coated onmetallic monolith for hydrogen production from ammonia was prepared by a dip-coating method using a catalyst slurry mixture composed of Ru/Al2O3 catalyst, inorganic binder (alumina sol) and organic binder (methyl cellulose). At the optimized 1:1:0.1 weight ratio of catalyst/inorganic binder/organic binder, the amount of catalyst coated on the metallic monolith after one cycle coating was about 61.6 g L-1. The uniform thickness (about 42 ㎛) and crystal structure of the catalyst coated on the metallic monolith surface were confirmed through scanning electron microscopy (SEM) and X-ray diffraction (XRD) analysis. Also, a numerical optimization regression equation for NH3 conversion according to the independent variables of reaction temperature (400-600 ℃) and gas hourly space velocity (1,000-5,000 h-1) was calculated by response surface methodology (RSM). This model indicated a determination coefficient (R2) of 0.991 and had statistically significant predictors. This regression model could contribute to the commercial process design of hydrogen production by ammonia decomposition.

Rice Yield Estimation Using Sentinel-2 Satellite Imagery, Rainfall and Soil Data (Sentinel-2 위성영상과 강우 및 토양자료를 활용한 벼 수량 추정)

  • Existing domestic studies on estimating rice yield were mainly implemented at the level of cities and counties in the entire nation using MODIS satellite images with low spatial resolution. Unlike previous studies, this study tried to estimate rice yield at the level of eup-myon-dong in Gimje-si, Jeollabuk-do using Sentinel-2 satellite images with medium spatial resolution, rainfall and soil data, and then to evaluate its accuracy. Five vegetation indices such as NDVI, LAI, EVI2, MCARI1 and MCARI2 derived from Sentinel-2 images of August 1, 2018 for Gimje-si, Jeollabuk-do, rainfall and paddy soil-type data were aggregated by the level of eup-myon-dong and then rice yield was estimated with gamma generalized linear model, an expanded variant of multi-variate regression analysis to solve the non-normality problem of dependent variable. In the rice yield model finally developed, EVI2, rainfall days in September, and saline soils ratio were used as significant independent variables. The coefficient of determination representing the model fit was 0.68 and the RMSE for showing the model accuracy was 62.29kg/10a. This model estimated the total rice production in Gimje-si in 2018 to be 96,914.6M/T, which was very close to 94,470.3M/T the actual amount specified in the Statistical Yearbook with an error of 0.46%. Also, the rice production per unit area of Gimje-si was amounted to 552kg/10a, which was almost consistent with 550kg/10a of the statistical data. This result is similar to that of the previous studies and it demonstrated that the rice yield can be estimated using Sentinel-2 satellite images at the level of cities and counties or smaller districts in Korea.

Modification and Validation of an Analytical Method for Dieckol in Ecklonia Stolonifera Extract (곰피추출물의 지표성분 Dieckol의 분석법 개선 및 검증)

  • This study was to investigate an analytical method for determining dieckol content in Ecklonia stolonifera extract. According to the guidelines of International Conference on Harmonization. Method validation was performed by measuring the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) of dieckol using high-performance liquid chromatography-photodiode array. The results showed that the correlation coefficient of calibration curve (R2) for dieckol was 0.9997. The LOD and LOQ for dieckol were 0.18 and 0.56 ㎍/mL, respectively. The intra- and inter-day precision values of dieckol were approximately 1.58-4.39% and 1.37-4.64%, respectively. Moreover, intra- and inter-day accuracies of dieckol were approximately 96.91-102.33% and 98.41-105.71%, respectively. Thus, we successfully validated the analytical method for estimating dieckol content in E. stolonifera extract.

A Longitudinal Validation Study of the Korean Version of PCL-5(Post-traumatic Stress Disorder Checklist for DSM-5) (PCL-5(DSM-5 기준 외상 후 스트레스 장애 체크리스트) 한국판 종단 타당화 연구)

  • The aim of this study is to examine the psychometric properties of the Korean version of the Post-traumatic Stress Disorder Checklist for DSM-5(PCL-5). For this purpose, online surveys were conducted for two times with a one year interval using the data from 1,077 Korean adults at time 1, and 563 Korean adults at time 2. First, from the result of the confirmatory factor analysis, comparing the model fit of the 1, 4, 6, and 7-factor model, the 4, 6, and 7-factor model showed a acceptable fit, and the best fit was seen in the order of the 7, 6, 4-factor model. Second, the internal consistency, omega coefficient, construct validity, average variance extracted, and test-retest reliability results were all satisfactory.. Third, a correlation analysis with the K-PC-PTSD-5 and the sub-factors of BSI-18 was conducted to check the validity of the Korean Version of PCL-5. As a result, a positive correlation was seen with both K-PC-PTSD-5 and BSI-18. Fourth, a hierarchical multiple regression was performed to examine whether the Korean Version of PCL-5 predicts future PTSD, depression, anxiety, and somatization. As a result, the Korean Version of PCL-5 measured at time 1 significantly predicted PTSD, depression, anxiety, and somatization symptoms at time 2. Fifth, by analyzing the ROC curve, the discriminant power of PCL-5 for screening PTSD symptom groups was confirmed, and the best cut-off score was suggested. As a result of the longitudinal validation of Korean version of PCL-5, it was found that this scale is a reliable and valid measure for Korean adults. By looking into the predictive validity of the scale, it was found that the Korean version of PCL-5 can predict not only PTSD symptoms but also PTSD-related symptoms such as depression, anxiety, and somatization. Also, this study differs from previous validation studies measuring PTSD symptoms in that it suggested a cut-off score to help differentiate PTSD symptom groups.

Validation of an Analytical Method for Deacetylasperulosidic acid, Total Sugar and Monosaccharide Analysis in Fermented Morinda citrifolia Polysaccharide Powder (발효노니 다당체 분말의 deacetylasperulosidic acid, 총당 및 단당류 분석법 검증)

  • This study was aimed at validating the analysis methods for deacetylasperulosidic acid (DAA), total sugar, galacturonic acid, glucose, and galactose, which are the indicator components of fermented Morinda citrifolia polysaccharide extract (Vitalbos). We modified the previously reported methods for validating the analytical methods. The specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) were measured using phenol-sulfuric acid method and high-performance liquid chromatography (HPLC). The retention time and spectrum of the standard solution of Vitalbos coincided, confirming the specificity. The calibration curve correlation coefficient (R2), of five indicator components, ranged from 0.9995-0.9998, indicating excellent linearity of 0.99 or more. The intra-day and inter-day precision range of the assay was 0.14-3.01%, indicating a precision of less than 5%. The recovery rate was in the range of 95.13-105.59%, presenting excellent accuracy. The LOD ranged from 0.39 to 0.84 ㎍/mL and the LOQ ranged from 1.18 to 2.55 ㎍/mL. Therefore, the analytical method was validated for DAA, total sugar, galacturonic acid, glucose, and galactose, in Vitalbos. The indicator component content in Vitalbos was determined using a validated method. The contents of DAA, total sugar, galacturonic acid, glucose, and galactose were 2.31±0.06, 475.92±5.95, 72.83±1.05, 71.63±2.44, and 67.30±2.31 mg/g of dry weight, respectively. These results suggest that the developed analytical method is efficient and could contribute to the quality control of Vitalbos, as a healthy functional food material.

Reliability Analysis on Stability of Armor Units for Foundation Mound of Composite Breakwaters (혼성제 기초 마운드의 피복재 안정성에 대한 신뢰성 해석)

  • Probabilistic and deterministic analyses are implemented for the armor units of rubble foundation mound of composite breakwaters which is needed to protect the upright section against the scour of foundation mounds. By a little modification and incorporation of the previous empirical formulas that has commonly been applied to design the armor units of foundation mound, a new type formula of stability number has been suggested which is capable of taking into account slopes of foundation mounds, damage ratios of armor units, and incident wave numbers. The new proposed formula becomes mathematically identical with the previous empirical formula under the same conditions used in the developing process. Deterministic design have first been carried out to evaluate the minimum weights of armor units for several conditions associated with a typical section of composite breakwater. When the slopes of foundation mound become steepening and the incident wave numbers are increasing, the bigger armor units more than those from the previous empirical formula should be required. The opposite trends however are shown if the damage ratios is much more allowed. Meanwhile, the reliability analysis, which is one of probabilistic models, has been performed in order to quantitatively verify how the armor unit resulted from the deterministic design is stable. It has been confirmed that 1.2% of annual encounter probability of failure has been evaluated under the condition of 1% damage ratio of armor units for the design wave of 50 years return period. By additionally calculating the influence factors of the related random variables on the failure probability due to those uncertainties, it has been found that Hudson's stability coefficient, significant wave height, and water depth above foundation mound have sequentially been given the impacts on failure regardless of the incident wave angles. Finally, sensitivity analysis has been interpreted with respect to the variations of random variables which are implicitly involved in the formula of stability number for armor units of foundation mound. Then, the probability of failure have been rapidly decreased as the water depth above foundation mound are deepening. However, it has been shown that the probability of failure have been increased according as the berm width of foundation mound are widening and wave periods become shortening.
