The effects of actively stimulating devices (ASD) on juvenile flatfish escape were studied to increase escape rates from the cod end by encouraging fish to approach the net wall. Two kinds of ASD were designed: a fluttering net panel, i.e., a free-end flag-like net panel, and a double conical rope array. Escape responses of juvenile bastard halibut were observed in a circulating water channel using two model cod ends, one made with diamond-shaped 43-mm-mesh-size polyethylene (PE) as a high-contrast cod end and the other with polyamid (PA) mono-ply as a low-contrast cod end. Retention rate was significantly lower with the double conical rope-array ASD in the PE cod end than with conventional PE cod ends only or the fluttering net-panel ASD inside the PE cod end. Mean retention rate with the low-contrast PA cod end was also significantly lower than that with the high-contrast PE conventional cod end. Therefore, active fluttering devices using a double conical rope array together with less visible low-contrast netting in the cod end could help to reduce the bycatch of juvenile flatfish by weakening their optomotor response and actively driving fish to the side net panel.
This study is conducted to investigate treatability by activated sludge process for wastewater from acetaldehyde manufacturing plant. The optimum hydraulic retention time in aeration tank for removal of high strength substrate were measured. The removal efficiency were checked out by hydraulic retention time : 35hr., 40hr. and 45hr., respectively. $COD_{Cr}$, like substances were removed in all hydraulic retention time zone directed for efficiency, but non-biodegradable substances were remained. $COD_{Cr}$ biomass loading was 0.81kg $COD_{Cr}/kgMLVSS$ . day at 35hr. of retention time, 0.34 kg$COD_{Cr}$/kg MLVSS . day at 40hr., and O.l9kg$COD_Cr$/kgMLVSS . day at 45hr. And the mean $COD_{Cr}$, removal efficiency was 65.5%, 81.6% and 83.0%, respectively. And also $COD_{Cr}$, volume loading was 1.01kg$COD_{Cr}/m^3$ day, 0.87kg$COD_{Cr}/m^3$ - day, and 0.79kg$COD_{Cr}/m^3{\cdot }$day, respectively. The basic design parameter obtained is as fallows. The value of Specific substrate removal rate coefficient (k), Yield coefficient(Y) and Decay coefficient($k_d$) was $0.0013day^{-1}$, $0.505kgMLVSS/kgCOD_{Cr}$ and $0.040day^{-1}$, respectively.
Polyester 감량폐수의 주성분 중 물리.화학적 처리가 어려운 EG를 생물학적으로 처리하기 위하여 jet-loop reactor를 사용하였다 Jet-loop reactor의 산소전달계수는 air-lift reactor와 비교하여 훨씬 컸다. 합성 polyester 감량폐수를 사용하여 부하율 2.64 $kgCOD_{Mn}/m^3$.day, 3.07$kgCOD_{Cr}/m^3$.day로 처리한 경우 유출수의 농도는 154 $mgCOD_{Mn}/L$, 156$mgCOD_{Mn}/L$이었으며, 이 때 각각 93, 93.9%의 제거율을 나타내었다. $COD_{Mn}$의 비부하 속도가 0.25에서 1.72$kgCOD_{Mn}/kgMLVSS$.day로 증가 함에 따라 비기질제거 속도는 0.25에서 1.60$kgCOD_{Mn}-removed/kgMLVSS$.day로 선형적으로 증가하였다. 그리고, 동력학적 계수 Y는 0.78,$K_d$는 0.1$day^{-1}$, $K_s$는 89mg/L는 k는 0.05$hr^{-1}$이었다. 실제 polyester 감량폐수를 사용하여 부하율 2.64$kgCOD_{Mn}/m^3$.day,5.24$kgCOD_{Cr}/m^3$.day로 처리한 경우 유출수의 농도는 150$kgCOD_{Mn}/L$, 3.6$kgCOD_{Cr}/L$이었으며, 이때 각각 93.2,93%의 제거울을 나타내었다.
River water and industrial waste water in Seoul were studied by means of chemical oxygen demand(COD) as an indicator for water pollution, from August 1967 to July 1968. Rivers flowing through residential and industrial areas are badly contaminated and COD of water in Han River increases as it progresses to downstream. Seasonal variation of COD showed that higher value of COD was observed in spring and lower in autumn. It is clear that the seasonal variation of COD is influenced by the precipitation. Close relationship was found between COD and population density. The lowest COD curve obtained by plotting COD values against population density and show that the curve slopes upward. The discontinuation of the curve was shown at the population density of 14,000/km$^2$; an increase in COD was acute over the population density of 14,000/km$^2$.
The three-dimensional eco-hydrodynamic model was applied to estimate the physical process in terms of COD (chemical oxygen demand) and net supply(or decomposition) rate of COD in Kamak Bay to find proper management plan for oxygen demanding organic matters. The estimation results of the physical process in terms of COD showed that transportation of COD is dominant in surface level while accumulation of COD is dominant in bottom level. In the case of surface level, the net supply rate of COD was 0 -0.50 mg/m2/day. The net decomposition rate of COD was 0 -0.04 mg/m2/day in middle level(3 -6m) and 0.05 -0.1 5 mg/m2/day in bottom level(6m -bottom). These results indicates that the biological decomposition and physical accumulation of COD are occurred predominantly at the northern part of bottom level. Therefore, it is important to consider both allochthonous and autochthonous oxygen demanding organic matters in the region.
CodY is a highly conserved protein in low G+C gram-positive bacteria that regulates genes involved in sporulation and stationary-phase adaptation. Bacillus thuringiensis is a grampositive bacterium that forms spores and parasporal crystals during the stationary phase. To our knowledge, the regulatory mechanism of CodY in B. thuringiensis is unknown. To study the function of CodY protein in B. thuringiensis, BMB171codY- was constructed in a BMB171 strain. A shuttle vector containing the ORF of cry1Ac10 was transformed into BMB171 and BMB171codY-, named BMB171cry1Ac and BMB171codY-cry1Ac, respectively. Some morphological and physiological changes of codY mutant BMB171codY-cry1Ac were observed. A comparative proteomic analysis was conducted for both BMB171codY-cry1Ac and BMB171cry1Ac through two-dimensional gel electrophoresis and MALDI-TOF-MS/MS analysis. The results showed that the proteins regulated by CodY are involved in microbial metabolism, including branched-chain amino acid metabolism, carbohydrate metabolism, fatty acid metabolism, and energy metabolism. Furthermore, we found CodY to be involved in sporulation, biosynthesis of poly-β-hydroxybutyrate, growth, genetic competence, and translation. According to the analysis of differentially expressed proteins, and physiological characterization of the codY mutant, we performed bacterial one-hybrid and electrophoretic mobility shift assay experiments and confirmed the direct regulation of genes by CodY, specifically those involved in metabolism of branched-chain amino acids, ribosomal recycling factor FRR, and the late competence protein ComER. Our data establish the foundation for in-depth study of the regulation of CodY in B. thuringiensis, and also offer a potential biocatalyst for functions of CodY in other bacteria.
현재 우리나라에서 수중의 유기물양의 지표로 사용되고 있는 $COD_{Mn}$법이 지표로서 적합한지를 알아보기 위하여 소양호의 댐앞지점과 유입수를 대상으로 유기물양과 $COD_{Mn}$과의 상관관계를 알아보았다. 또한 소양호를 표충, 중층, 심층으로 나누어 $COD_{Mn}$ 법에 의한 각 충의 유기물산화정도를 비교하였다. 소양호의 유기물농도와 $COD_{Mn}$ 비는 계절에 따라 매우 상이하였고, 수층에 따라서도 차이를 보였다. 유입수에서도 홍수기를 전후로 하여 유기물농도와 $COD_{Mn}$ 비가 유의적인 차이를 보였다. $COD_{Mn}$ 법에 의한 소양호의 유기물의 산화는 평균 40% 정도로 낮은 값을 보였다. 특히 심층의 유기물은 $COD_{Mn}$ 법에 의해 산화되는 양이 30%이하로 매우 낮았다. 이러한 결과는 $COD_{Mn}$ 법이 호수와 하천의 유기물양을 제대로 대변하고 있지 못할 뿐 아니라 계절에 따라 유기물의 조성이 변할 때 $COD_{Mn}$ 법에 의한 유기물산화력이 변한다는 단점을 보여준다.
염색가공 공정에서 배출되는 염색폐수는 염색 가공공정이나 사용하는 염료등에 따라 폐수량과 성상의 변화가 심하기 때문에 처리에 많은 어려움이 있다. 일반적인 염색폐수 처리방법으로는 물리화학절인 응집침전법과 생물학적인 활성슬러지법을 단독 또는 조합하여 주로 사용하여 왔다. 그러나 최근에는 기존시설의 처리 향상을 위한 다각적인 기술개발이 시도되고 있으며 그 일환으로 본 연구에서는 활성탄을 담지한 폴리우레탄 담체를 개발하여 염색폐수를 처리하고자 하였다. 본 연구에서는 담체를 이용한 염색폐수의 생물학적 처리 시 여러 인자들, 즉 COD/N 비, DO 농도, 담체의 충진율 등의 영향을 조사하기 위하여 회분식 실험을 수행하였다. 또한 회분식 실험에서 얻은 최적조건을 이용하여 파일럿 실험도 함께 수행하였다. COD/N 비가 COD 제거에 미치는 영향을 살펴보기 위해 COD/N 비를 조절하지 않은 경우와 COD/N 비를 2-3으로 조절한 경우에 대해서 비교하였다. 실험결과 염색폐수의 초기 COD 농도가 1,000 ppm일 때, 처리 후 COD 농도는 COD/N 비를 조절하지 않았을 경우 860 ppm (제거효율 = 18%), COD/N 비가 2일 경우 550ppm (제거효율 = 52%), COD/N 비가 3의 경우 390 ppm (제거효율 = 57%)으로 각각 나타났다. DO 농도가 COB 제거에 미치는 영향을 알아보고자 DO 농도를 1, 6, 7 mg/L로 변화시켰다. 그 결과 DO의 농도가 1 mg/L일 경우 COD 제거효율은 20% 정도였으나, DO 농도가 6 mg/L이상에서는 모두 80%에 가까운 제거효율을 나타내었다. 담체 충진율의 경우, 10% 충진시 40%, 20% 충진시 50%, 30% 충진시 70%의 제거효울을 각각 나타내었다. 회분식 실험에서 얻은 최적조건으로 파일럿 반응기를 100일 이상 운전한 결과 염색폐수의 COD 농도변화는 1,000ppm에서 380ppm로 평균 60%이상 제거되는 높은 효율을 보였다. 또 담체 첨가의 영향을 살펴보기위해, 담체를 충진하지 않은 조건에서 파일럿 반응기를 운전한 결과 평균 COD 제거율은 50%로 비교적 낮은 효율을 보였다.
Seasonal and stational variation of SS and COD were investigated from February 2008 to December 2010 and the relationship between them was discussed. During three years monitoring, SS decreased significantly (46% decline) possibly due to the increase of precipitation and thereby resulting salinity drop. COD on average was the highest in 2009. SS was the highest in autumn and the lowest in winter, and over 72% of SS was FSS. While SS is high in the upper sampling stations of the bay with shallow water, COD values do not show any relationship to the geomorphological characteristics. $COD_{ins}$, which was defined as COD after filtration, ranged 56%(winter) ~ 44.6%(summer) and showed no correlation with SS. It indicates that high SS concentration is not necessarily related to the high $COD_{ins}$. The seasonal $COD_{ins}$/SS data, which can be interpreted as COD density in SS, shows that SS in winter contains the dense COD materials compared to the other seasons.
A conventional thermophilic anaerobic digester was converted into a thermophilic high-rate hybrid anaerobic reactor (THAR) for treating distillery wastewater. The THAR has been operating successfully since May 1995 at a loading rate of 5.45 to $11.52{\;}kg/\textrm{m}^3/d$ (maximum of 15.02). The THAR has demonstrated a soluble Chemical Oxygen Demand (sCOD) removal efficiency of 85 to 91% and a total COD (tCOD) removal efficiency of as much as 72 to 84%. Product gas had a methane content of 59 to 68%. The tCOD removal rates were 4.31 to 5.43, 6.26 to 6.89, and 9.03 to $9.78kg{\;}tCOD/\textrm{m}^3/d$ for tapioca, com, and naked-barley wastewater, respectively. The sCOD removal rates ranged from 3.75 to 4.79,3.28 to 4.89, and 5.57 to 6.21kg $sCOD/\textrm{m}^3/d$ for tapioca, com, and naked-barley wastewater, respectively. There were unknown substances in a naked-barley distillery wastewater that were identified as being toxic for microorganisms. However, the THAR treated naked-barley wastewater continuously for 26 days, operating at an average tCOD loading of $11.08{\;}kg/\textrm{m}^3/d$without any signs of deterioration in either COD removal efficiency or gas production rate. During this period, the average removal efficiencies of tCOD and sCOD were 84% and 91%, respectively, and the gas production rate averaged 6.61 to $7.57{\;}\textrm{m}^3/\textrm{m}^3$ reactor/d which produced 0.57 to $0.69{\;}\textrm{m}^3{\;}biogas/kg{\;}tCOD_{rem}$. From tapioca and com wastewater, the reactor showed an average gas production rate of 3.18 to 3.46 and 4.91 to $5.22{\;}\textrm{m}^3/\textrm{m}^3$ reactor/d which produced 0.53 to 0.69 and 0.62 to $0.71{\;}\textrm{m}^3/kg{\;}tCOD_{rem}$, respectively.
