Effect of wet treatment on the structure of various silkworm strain cocoons with different rearing seasons

  • In this study, cocoons from different silkworm strains and rearing seasons were collected and immersed in water at elevated temperatures to investigate the effect of a wet treatment on their morphology and crystallinity. Untreated cocoons exhibited different crystallinities depending on the silkworm strain and rearing season; however, no distinct changes in their morphological structure were observed. Furthermore, after wet treatment, the differences in the crystallinities of cocoons disappeared. In addition, wet treatment of degummed silk (i.e., fibroin) did not change its crystallinity. These results indicate that different crystallinities of the untreated cocoons collected from various silkworm strains and rearing seasons are due to the different characteristics of sericin in the cocoons.

Studies on the Comparative Performance of Victory-1, S-36 and Kanva-2 Mulberry Genotypes and their Impact on Silkworm Rearing under Telangana Conditions of Andhra Pradesh.

  • A new high yielding mulberry genotype, Victory-1 (V-1) was recommended to the field for irrigated areas by Central Sericultural Research and Training Institute (CSRTI), Mysore during 1996-1997. As K-2 and S-36 mulberry genotypes are already in the field, a study on the comparative performance of V-1, 5-36 and K-2 varieties, in terms of leaf yield, yield attributes, leaf moisture and moisture retaining capacity and their impact on silkworm rearing was carried out under Telangana conditions of Andhra Pradesh during 1999-2001. Among the three genotypes the leaf yield was found to be significantly (P < 0.01) higher in V-1 (64,130 kg/ha/yr) followed by S-36 (44,064 kg/ha/yr) and K-2 (23,990 kg/ha/yr). The maximum leaf moisture was also observed in V-1 (71.84%) compared to S-36 (66.86%) and K-2 (64%). The moisture retaining ability was more in V-1 (70.17%) followed by S-36 (66.20%) and K-2 (60.08%). The yield attributes were found to be significantly (P < 0.01) enhanced in V-1 over S-36 and K-2. Silkworm rearing results (bioassay) indicated a significant (P < 0.01) reduction in larval duration and improvement in larval weight, effective rate of rearing by number and weight, single cocoon and shell weights and shell ratio in the silkworm lots fed on V-1 variety followed by S-36 and K-2. Hence, in this study, V-1 was found to be superior among the three varieties studied.

Light and Electron Microscopic Study on the Development of Nosema Bombyics Naegeli in the Midgut of Silkworm Bombyx Mori L.

  • Infection effect of $Nosema$ $bombyics$ on the midgut of silkworm $Bombyx$ $mori$ and subsequent appearance of spores and the performance of larvae was studied. Autopsy of larvae showed white pustules on the surface of midgut at 5 days of post infection (pi). At later stage, important organs like midgut, silk gland and gonads reduced in size and all these organs showed white pustules. Light microscope observation of pustules revealed enormous spores. Spore multiplication was at a faster rate in young larvae. Infection of the adult larvae resulted in pebrinized pupa and moths. Larval weight, cocoon weight and cocoon shell ratio reduced as the post infection period increased. Transverse sections of midgut showed $N.$ $bombycis$ infection limited to a few columnar cells at 3-5 days of pi. At 7 days pi, cell volume increased, cells were swollen and elongated. Heavily infected cells looked like sacks filled with parasite and the apical region of certain cells were bulging into the gut lumen. Later at 8-9 days of pi, spores or its developing stages leaked into the lumen either freely or enclosed within the globules of host cytoplasm. Besides columnar cells, development of $N.$ $bombycis$ was observed in the regenerative cells and rarely in goblet cells. Development of $N.$ $bombycis$ was also observed in both longitudinal and circular muscles at the late pi period. The histopathological changes, deformities and spore production time in the host were all influenced by the spore dosage and age of the host.

  • It was found that the peptides originated from the hydrolysates of silk fibroin have in vitro immunostimulating effects in murine macrophage RAW264.7 cells. The stimulation effects on nitric oxide (NO) production resulted from treatments of acid or enzymatic hydrolysates were measured. The silk fibroin preparation isolated from cocoon was most efficiently digested by acid hydrolysis. Even though the sole treatment of acid hydrolysate stimulated the NO production in dose-dependent pattern, a part of its activity was found to be caused by the contaminated endotoxin, LPS. When each endotoxin-free hydrolysates obtained by filtering it through an ultrafiltration membrane of molecular weight (MW) cut-off 10,000 to eliminate LPS was used, the peptic hydrolysate with lowest degree of hydrolysis showed the highest activity. The fractions of peptic hydrolysate with MW ranges of 1,000∼10,000, 500∼1,000 and below 500 also showed a higher MW-higher activity correlation. From the analyses of amino acid composition of each hydrolysate, it was found that the contents of arginine, lysine, alanine and glycine residues affected the activity level of hydrolysate. The results of this study showed a possibility of utilizing fibroin as a source for immunostimulating (chemopreventive) functional peptides.

Screening of Promising Bivoltine Hybrids of Mulberry Silkworm for their Susceptibility to Bombyx mori Nuclear Polyhedrosis Virus and Bombyx mori Infectious Flacherie Virus

  • Central Sericultural Research and Training Institute, Mysore have evolved several highly productive bivoltine hybrids which can produce international grade raw silk. Among them $CSR2{\times}CSR4,\;CSR2{\times}CSR5,\;CSR3{\times}CSR6,\;CSR17{\times}CSR16,\;CSR18{\times}CSR19$ and $CSR12{\times}CSR6$ are being popularized in the field. There is a minimum difference in their economic characters but they appear to differ in survival. Though they are productive under high input management conditions, they are very susceptible to different diseases under normal rearing practices. No systematic attempts have been made to test their susceptibility status / resistance. Thus the present study is a modest attempt to screen the above six productive bivoltine hybrids to two important pathogens viz., Bombyx mori Nuclear Polyhedrosis Virus (BmNPV) and Bombyx mori Infectious Flacherie Virus (BmIFV) along with existing hybrid, $KA{\times}NB4D2$ to assess their susceptibility / resistance. The results shows that the productive hybrid $CSR2{\times}CSR4$ is the most resistant to BmNPV and it is suggested by its highest $LC_{50}$ value followed by $CSR12{\times}CSR6,\;KA{\times}NB4D2,\;CSR3{\times}CSR6,\;CSR17{\times}CSR16,\;CSR18{\times}CSR19,\;CSR2{\times}CSR5$. Based on the $LC_{50}$ value and $LT_{50}$ values for BmIFV, the hybrid $KA{\times}NB4D2$ was found to be the most resistant (1st position) one followed by $CSR3{\times}CSR6$ (2nd position) $CSR2{\times}CSR$ (3rd position) and $CSR12{\times}CSR6$ (4th position) $CSR17{\times}CSR16$, $CSR18{\times}CSR19$ (5th position) and $CSR2{\times}CSR5$ being the least. The response of 7 bivoltine hybrids to both the pathogens BmNPV and BmIFV indicates that, the hybrids $CSR2{\times}CSR4$, $CSR12{\times}CSR6$ and $KA{\times}NB4D2$ were found to be the most resistant when compared to others. Further, $KA{\times}NB4D2$ being less productive hybrid with a shell ratio of 20.08%, the other two hybrids $CSR2{\times}CSR4$ (Cocoon shell ratio, 21.44%) and $CSR12{\times}CSR6$ (cocoon shell ratio, 23.45%) can be considered to be most productive with superior quality cocoon and resistant to both BmNPV and BmIFV pathogens. The overall study indicated that the hybrid $CSR2{\times}CSR5$ is the most susceptible hybrid to both the pathogens.

The Developmental Characterization of the Sericin jam(Bombyx mori), a Mutant in the Synthesis and Secretion of Fibroin (세리신잠(Nd-s. $Nd^H$의 발육 특성)

  • This experiment was done to know developmental characteristics of Sericin jam. Incubation periods were 10 day 2 hr, and 10 day 1 hr. 11 day 1 hr for Nd-s jam, N $d^{H}$ jam, and Baegok jam, respectively. Hatching rates were 83.9, 83.3 and 96.0% for Nd-s jam, N $d^{H}$ jam, and Baegok jam. Larval periods were, 20 days 1 hr for Nd-s jam, 20 days 5 hrs for N $d^{H}$ jam, and 22 days 12 hrs for Baegok jam. Death rate of larvae was highest in N $d^{H}$ iam, followed by Nd-s jam and Baegok jam. Pupation rate was highest in Baegok Jam followed by Nd-s jam and that of N $d^{H}$ jam was the lowest among the three. Cocoon weight was 1.39, 1.08, and 2.01 g for Nd-s jam, N $d^{H}$ jam, and Baegok jam, respectively. Shell weight were 13, 3, and 48 cg for Nd-s jam, N $d^{H}$ jam, and Baegok jam. Cocoon shell ratios were 9.0% for Nd-s jam, 2.8% for N $d^{H}$ jam and 23.9% for Baegok jam. Cocoon sizes were 30.6${\times}$15.8 mm for Nd-s jam, 24.7${\times}$14.9 mm for N $d^{H}$ jam and 35.8 ${\times}$ 20.5 mm(1${\times}$w) for Baegok jam.w) for Baegok jam.

A Study on Utility of Raw Materials for Accelerating the Silkworm Mounting. (누에의 족촉진제 개발이용에 관한 연구)

  • The objective of this study is to compare the effects of raw materials as method of speeding up mounting of matured silkworm, for reducing concentrated labor during mounting periods. The results obtained from this study are summarized as fellows: (1) Quercus acutissima Carrs showed the most remarkable effect of repellent. Persicaria blumei Gross and Artemisia asiatica Nakai were followed in order. The rest repellents were also expected to be applicable. considering known material (silk-up) with lower mounting percentage. (2) The effective methods of preparation of repellent were to make distillate. Decocted solution type was considered to be the easiest way of preparation. (3) It seemed to be that there was little effect of repellent on the qualities of cocoon and silk. (4) When a repellent was applied to a silkworm bed, immatured silkworms were observed to mount. Thus it suggests that the repellent will give remarkable effects only when the degree of matured of silkworms is uniform.

Development of Artificial Diets for Green Lacewing, Chrysopa pallens (Rambur), by Addition of Natural Products (천연첨가물을 이용한 칠성풀잠자리붙이 인공사료 개발)

  • The green lacewing, Chrysopa pallens (Rambur), was reared on artificial diets supplementedwith natural products such as lyophilized beef liver, silk worm pupae powder and drone honey beepowder. The performance of the green lacewing was improved siginificantly on only the diet to whichadded 1 % of silk worm pupae powder (wt/v). Larval developement was completed in 15.6 f 0.99 days,cocoon weight was 16.4f2.94 mg, and suvival to adult was 89.3% at 27$^{\circ}$C. And another diet made bymixing 4 part of lyophilized beef liver powder, 4 part of lyophilized beef powder, 3 part of lyophilizedchicken egg, 4 part of sucrose, and 50 mg each of vitamin C and Vanderzant's vitamin B mixture waspromising. Larval development of the green lacewing on this diet was completed in 12.7 f 1.45 days at27"C, pupal weight was 18.1 f 2.24mg, suvival to adult was loo%, and 633f70.4 eggs were laid in 20days after beginning of oviposition.ginning of oviposition.

  • First of all, the properties imparted to PET fabrics are resistance to and recovery from creasing or wrinkling when wet or dry; high resistance to stretch in the filament yarns but not in the staple; high abrasion resistance; good texture and appearance; resistance to heat ageing; good chemical resistance and good resistance, behind glass, to sunlight. But, the low moisture regain of PET fabric conduces to static troubles in textile processing. Furthermore, garments made from PET may, during wear, develop electric charges which attract to the fabric particles of soil(dirt, swarf, dust) flying in the air, so that the cuffs of shirts, for example, become soiled quickly and are not easily laundered clean. The sericin constitutes 25$\sim$30% of silk protein and surrounds the fibroin fiber with sticky layer that supports the formation of a cocoon. The useful biochemical properties of sericin protein are oxidation resistant, antibacterial, UV resistant, hydrophilic property, and good affinity with hydrophobic material. These properties can be used as an improving reagent or a coating agent for natural and synthetic fibers, fabrics, and other intermediate products. The sericin is also applied to cross-link, and can be blended with other materials. In this study, we modified the surface of PET fabric by mixture of sericin finishing agent; sericin, polyuretane binder and 1,2,3,4-butanetetracarboxylic acid (BTCA) cross-link agent. Also, we investigated the finshing effect; moisture regain, stiffness, handle, drape and electrostatic. The moisture regain of PET fabric treated with sericin finishing agent was higher than that of untreated PET fabric. As a result of evaluating influence about handle of PET fabrics treated with sericin finishing agent, it was confirmed that the sericin finishing agent could be use as a linen like finishing agent.

Synthesis, Characterization and Cosmetic Application of Self-Assembled Sericin-PEG Nanoparticle

  • Silk Sericin(SS) is a natural protein extracted from cocoon of bombix mori and shows moisturizing effect to the skin due to a number of hydroxyl groups in the structure. But its application to cosmetics is limited due to its poor solubility in water. In order to solve this drawback and expand its application to cosmetics, polyethyleneglycol(PEG) was conjugated with sericin by reacting activated polyethyleneglycol(ActPEG). Reaction site of sericin is tyrosine residue, which was determined by using $^1$H-NMR. Random coil structure of sericin was transformed to beta-sheet structure by conjugating polyethyleneglycol. It was confirmed that melting point of sericin-PEG conjugate was lowered compared to that of each sericin and PEG due to the interaction between sericin and PEG in crystalline structure. Self-assembled sericin-PEG nanoparticle was obtained by dialyzing with alcohol solution of sericin-PEG conjugate against water. The particle is spherical and has 200-400nm of size. The moisturizing ability of sericin-PEG nanoparticle was much higher than that of sericin itself. Incorporation of vitamin A into sericin-PEG nanoparticle was carried out by diafiltration method. The content of incorporated Vitamin A in sericin-PEG nanoparticle was 8.9 wt%. Releasing behaviour of vitamin A incorporated into nanoparticle was tested in phosphate buffer, pH 7.4 at 37$^{\circ}C$. and half-life of Vitamin A release was 43hrs. Sericin-PEG nanoparticle exhibited higher moisturing effect than sericin itself and distilled water, respectively. No toxicity and irritation were observed in animal tests. It can be expected that the self-assembled sericin-PEG nanoparticle can be developed for cosmetics.
