Region of Interest Heterogeneity Assessment for Image using Texture Analysis

  • Heterogeneity assessment of tumor in oncology is important for diagnosis of cancer and therapy. The aim of this study was performed assess heterogeneity tumor region in PET image using texture analysis. For assessment of heterogeneity tumor in PET image, we inserted sphere phantom in torso phantom. Cu-64 labeled radioisotope was administrated by 156.84 MBq in torso phantom. PET/CT image was acquired by PET/CT scanner (Discovery 710, GE Healthcare, Milwaukee, WI). The texture analysis of PET images was calculated using occurrence probability of gray level co-occurrence matrix. Energy and entropy is one of results of texture analysis. We performed the texture analysis in tumor, liver, and background. Assessment textural features of region-of-interest (ROI) in torso phantom used in-house software. We calculated the textural features of torso phantom in PET image using texture analysis. Calculated entropy in tumor, liver, and background were 5.322, 7.639, and 7.818. The further study will perform assessment of heterogeneity using clinical tumor PET image.

Fabrication and evaluation of hydrophobic metal stent using electron beam equipment (전자빔 처리를 통한 발수성 금속 스텐트 제작 및 평가)

  • The objective of this study was to fabricate a novel hydrophobic stent for reducing restenosis by employing electron beam equipment. The stent was fabricated from a CoCr alloy tube by using a femtosecond laser and was treated with argon plasma. Subsequently, the stent's surface specification changed from hydrophilic to hydrophobic. Application of the electron beam offers several advantages such as a short processing time, whole surface reforming, and enhancement of material properties. As the surface of the stent was rendered hydrophobic, it can provide equivalent or enhanced mechanical properties and greater functionality with a higher radial force at the extended stent in a blood vessel. The obtained results corresponding to the mechanical properties indicate that the contact angle increased to approximately 130°, and the radial force increased to approximately 3 N. Furthermore, cell culture experiments were conducted for verifying whether cells were cultured on the surface-modified CoCr surface. Based on the obtained results, it is believed that an effective reduction in the restenosis of inserted vascular stents is possible.

An Experimental Study on Seismic Reinforcement of Dry Type Buckling Restrained Braces Laterally Using Buckling Restrained Rings (좌굴방지링으로 횡지지된 건식형 좌굴방지가새 내진보강에 대한 실험적 연구)

  • This study is conducted to verify the seismic reinforcement effects of internally inserted buckling-restrained braces supported laterally by buckling-restrained rings for the seismic reinforcement of existing reinforced concrete buildings with non-seismic details. First, to evaluate the performance of KDS, the hysteretic characteristics of buckling-restrained braces are verified, and it is discovered that they satisfy the conformance criteria of the displacement-dependent damping device. Three full-scale, two-story reinforced concrete framework specimens are prepared to verify the seismic reinforcement effects, and the proposed buckling-restrained braces are bolstered with single diagonal and V-shaped braces to be compared with non-reinforced specimens. By performing a comparison with non-reinforced specimens that present intensive shear cracks at the bottom of first-floor columns, it is revealed that the maximum load and energy dissipation of specimens reinforced with the proposed buckling restrained braces, in which the structural damage extends evenly throughout the system, are approximately 4 and 6.2 times higher, respectively, which proves the effectiveness of the proposed seismic reinforcement method.

Nocturnal Light Pulses Lower Carbon Dioxide Production Rate without Affecting Feed Intake in Geese

  • This study was conducted to investigate the effect of nocturnal light pulses (NLPs) on the feed intake and metabolic rate in geese. Fourteen adult Chinese geese were penned individually, and randomly assigned to either the C (control) or NLP group. The C group was exposed to a 12L:12D photoperiod (12 h light and 12 h darkness per day), whereas the NLP group was exposed to a 12L:12D photoperiod inserted by 15-min lighting at 2-h intervals in the scotophase. The weight of the feed was automatically recorded at 1-min intervals for 1 wk. The fasting carbon dioxide production rate ($CO_2$ PR) was recorded at 1-min intervals for 1 d. The results revealed that neither the daily feed intake nor the feed intakes during both the daytime and nighttime were affected by photoperiodic regimen, and the feed intake during the daytime did not differ from that during the nighttime. The photoperiodic treatment did not affect the time distribution of feed intake. However, NLPs lowered (p<0.05) the mean and minimal $CO_2$ PR during both the daytime and nighttime. Both the mean and minimal $CO_2$ PR during the daytime were significantly higher (p<0.05) than those during the nighttime. We concluded that NLPs lowered metabolic rate of the geese, but did not affect the feed intake; both the mean and minimal $CO_2$ PR were higher during the daytime than during the nighttime.

Impact of Fermentation Rate Changes on Potential Hydrogen Sulfide Concentrations in Wine

  • The correlation between alcoholic fermentation rate, measured as carbon dioxide ($CO_2$) evolution, and the rate of hydrogen sulfide ($H_2S$) formation during wine production was investigated. Both rates and the resulting concentration peaks in fermentor headspace $H_2S$ were directly impacted by yeast assimilable nitrogenous compounds in the grape juice. A series of model fermentations was conducted in temperature-controlled and stirred fermentors using a complex model juice with defined concentrations of ammonium ions and/or amino acids. The fermentation rate was measured indirectly by noting the weight loss of the fermentor; $H_2S$ was quantitatively trapped in realtime using a pre-calibrated $H_2S$ detection tube which was inserted into a fermentor gas relief port. Evolution rates for $CO_2$ and $H_2S$ as well as the relative ratios between them were calculated. These fermentations confirmed that total sulfide formation was strongly yeast strain-dependent, and high concentrations of yeast assimilable nitrogen did not necessarily protect against elevated $H_2S$ formation. High initial concentrations of ammonium ions via addition of diammonium phosphate (DAP) caused a higher evolution of $H_2S$ when compared with a non-supplemented but nondeficient juice. It was observed that the excess availability of a certain yeast assimilable amino acid, arginine, could result in a more sustained $CO_2$ production rate throughout the wine fermentation. The contribution of yeast assimilable amino acids from conventional commercial yeast foods to lowering of the $H_2S$ formation was marginal.

Molecular weight Control of Polyhydroxybutyrate (PHB) in Recombinant Escherichia coli (재조합 대장균에서의 Polyhydroxybutyrate (PHB)의 분자량 조절)

  • Two promoters (trc and P$\rho$) were inserted in PHA operon derived from Alcaligenes eutrophus to obtain high chain molecules of polyhydroxybutyrate (PHB) in recombinant Escherichia coli. Newly designed PHA operon was used to control the gene expressions of hydroxybutyric CoA and PHA polymerization, separately. Plasmids containing new synthetic operon was transformed into E. coli DH5$\alpha$ and analyzed for PHB production. Without induction of the PHA biosynthetic operon, PHA synthase which has low activity might supply low concentration of initiator during the polymerization reaction, resulting very high molecular weight of polymer. An increase of PHB average molecular weight was observed with decreased IPTG (isopropyl $\beta$ -Dithiogalactosidase) concentration. When no IPTG was added to the culture of E. coli DH5$\alpha$ /$\rho$ SJS1 which contained two promoters in PHA operon, high chain polymer having an average molecular weight of $2.5{\times}10^7$ was achieved. Analysis of the enzyme activities of PHA biosynthetic enzymes suggests that PHA synthase, the enzyme responsible for polymerizing 3-hydroxybutyric CoA, controls the molecular weight of PHB produced in vivo.

Stable Expression and Efficient Secretion of hSCF and hINFγ Protein using Binary Vectors in Chlorella vulgaris (클로렐라에서 바이너리 벡터를 이용한 hSCF와 hINFγ 단백질의 안정적인 발현과 효율적인 분비)

  • Microalgae have great potential in the biomedical and pharmaceutical industries as a new type of bioreactor that can produce proteins for specific purposes, including recombinant proteins, pharmaceuticals, and industrial enzymes. Despite the production advantages and importance of microalgae-based expression systems, studies on secretion efficiency are limited. In this study, for stable expression and efficient secretion of the heterologous protein (human SCF and human INFγ) in Chlorella vulgaris, we constructed SP:hSCF:His and SP:hINFγ:His plant binary vectors using the signal peptide (SP) of Chlamydomonas reinhardtii, and we obtained stable transformants through the effective agrobacterium-mediated transformation of these vectors. Transformants with accurately inserted hSCF and hINFγ demonstrated stably increased mRNA and protein expression using RT-PCR and western blotting under the same culture conditions. Following the analysis of the proteins secreted into the culture medium using ELISA, it was confirmed that hINFγ was effectively produced in the transformed C. vulgaris culture medium. The overall findings indicate that the combination of heterologous protein and SP may be crucial for ensuring the expression and secretion of recombinant proteins in Chlorella culture systems.

Iron Oxide-Carbon Nanotube Composite for NH3 Detection (산화철-탄소나노튜브 나노복합체의 암모니아 가스센서 응용)

  • Fabrication of iron oxide/carbon nanotube composite structures for detection of ammonia gas at room temperature is reported. The iron oxide/carbon nanotube composite structures are fabricated by in situ co-arc-discharge method using a graphite source with varying numbers of iron wires inserted. The composite structures reveal higher response signals at room temperature than at high temperatures. As the number of iron wires inserted increased, the volume of carbon nanotubes and iron nanoparticles produced increased. The oxidation condition of the composite structures varied the carbon nanotube/iron oxide ratio in the structure and, consequently, the resistance of the structures and, finally, the ammonia gas sensing performance. The highest sensor performance was realized with $500^{\circ}C/2h$ oxidation heat-treatment condition, in which most of the carbon nanotubes were removed from the composite and iron oxide played the main role of ammonia sensing. The response signal level was 62% at room temperature. We also found that UV irradiation enhances the sensing response with reduced recovery time.

Qualitative and quantitative evaluation of root injury risk potentially burdening insertion of miniscrew implants

  • Objective: Microscrew implants (MSls) offer many advantages, but some complications are known to occur during their insertion. One of the most commonly reported complications is root injury. Our aim was to identify factors associated with root injury and to evaluate their qualitative and quantitative values. Methods: Thirty-five orthodontists placed MSls (AbsoAnchor$AbsoAnchor^{(R)}$, Dentos Co. Ltd, Daegu, Korea) in the upper jaw of typodonts, labially between the second premolar and the first molar, in low and high vertical positions. Root contacts were counted, and distances between MSI apices and roots were measured. Fear level of the orthodontists was surveyed before and after the experiment. Wilcoxon's test, chi-square test, and Mann-Whitney test were used for statistical analysis. Results: Overall root contact rate of MSI insertion was 23.57%. The root contact rate was significantly higher in MSls inserted at $90^{\circ}$ (45.71%) than at $30^{\circ}$ (1.43%). The distance between the dental root and MSI also increased significantly in MSls inserted at $30^{\circ}$. Mean fear level before MSI insertion (4.6) significantly decreased after insertion (3.2); the causative factors were risk of injury to dental root and maxillary sinus or mandibular canal. Conclusions: Root injury is relatively rare, and oblique angulation reduces the risk of root and MSI contact.

Clinical Study on Safety, Clinical Indicators of Polydioxanone Sutures Inserted into Vastus Medialis Muscle in Degenerative Knee Osteoarthritis (무릎 관절염 환자에서 안쪽넓은근에 폴리디옥사논 봉합사 시술 연구)

  • Objective: Physiologically, the vastus medialis muscle is the first muscle to undergo muscle atrophy, and it was thought that pain in patients with knee osteoarthritis could be reduced if this muscle could be strengthened and stabilized. The purpose of this study was to prove the effectiveness in knee osteoarthritis using polydioxanone sutures that have been tried in other musculoskeletal areas. Method: Forty knee osteoarthritis patients voluntarily participated in the study, and divided into 30 polydioxanone suture needle (MEST-B2375 produced by Ovmedi Co.) and 10 sham needle (without suture). And the needles were inserted into the vastus medialis muscle. In all patients, safety evaluation including blood tests and ultrasonography as well as efficacy evaluation including isometric maximal contractile strength of quadriceps muscle, weight bearing pain, impression of change, quadriceps angle, rescue drug intake were evaluated up to 30 weeks after the procedure. Results: Isometric maximal contractile strength showed a significant improvement at 4 weeks after the procedure in the polydioxanone suture group, and the weight-bearing pain showed a significant improvement at every visit in the polydioxanone suture group compared with baseline values. Patient global impression of change score showed significant improvement at 20 and 30 weeks, and clinical score showed improvement at every visit. Conclusion: Insertion of polydioxanone sutures showed improvement in muscle strength and knee pain by supporting and fixation of the vastus medialis muscle in patients with degenerative knee osteoarthritis. Insertion of polydioxanone sutures is considered to have a therapeutic effect in knee osteoarthritis patients.
