Effects of dietary supplementation with detoxified Rhus verniciflua sap on egg production, yolk lipid and intestinal microflora in laying hens

  • Objective: This study was conducted to investigate the effects of dietary detoxified Rhus verniciflua sap (RVS) on production performance, egg quality, lipid fractions of egg yolk, liver and serum, and the profile of cecal microflora in laying hens. Methods: Two hundred 52-week-old Hy-Line Brown layers were randomly divided into 4 groups with 5 replicates per group (2 hens per cage, 5 cages per replicate) and were provided with one of 4 experimental diets containing 0%, 0.05%, 0.1%, or 0.2% RVS, for 6 weeks. Due to unequal intervals of RVS doses, the interactive matrix language procedure of the SAS program was used to correct the contrast coefficients of orthogonal polynomials. Results: There were no differences in feed intake and egg weight among the groups. Egg production increased (linearly and quadratically, p<0.05) with increasing levels of RVS. Eggshell thickness increased (linear, p<0.05) as the level of RVS in diets increased. The levels of blood cholesterol and activities of glutamic-oxaloacetic transaminase and glutamic-pyruvic transaminase were not altered by dietary treatments. Increasing level of RVS increased (linear, p<0.05) the populations of cecal lactic acid bacteria. The content of yolk cholesterol decreased (linear, p<0.05) with increasing levels of dietary RVS, although there were no significant differences in each lipid fraction of the liver. Conclusion: This study indicates that dietary RVS could improve laying performance and eggshell quality, and affect cecal lactic acid bacteria in a dose-dependent manner.

  • The purpose of the study was to evaluate the cytotoxic effects of polycarboxylate cements and zinc phosphate cements in vitro. Human fibroblasts were cultured in ${\alpha}$-MEM, and each cement was manually mixed and filled in glass ring cylinder (8${\times}$8mm in diameter, in height.) Cement filled cylinders were placed in the center of the dish (35mm in diameter) containing 3ml of ${\alpha}$-MEM. Millipore filters to simulate dentinal barrier were also placed between the cylinder and the dish, then stored in 5% $CO_2$ containing chamber for 1 and 2 weeks at the temperature of $36.6^{\circ}C$. The results of the experiments were analyzed by counting the cells in the period of one week and two weeks respectively, and were assessed by calculating the cell multiplication rate and the relative growth rate. The experimental groups and the control group were compared. The results of the study were summarized as follows. 1. Durelone brand of the polycarboxylate cements showed marked cytotoxicity after one week, but after two weeks the toxicity decreased remarkably. Poly-F brand exhibited moderate cytotoxicity after one week, but after two weeks the toxicity slightly decreased. HY-BOND brand was weakly cytotoxic after one week, but after two weeks the toxicity became significant. 2. The cytotoxicity of the zinc phosphate cements was negligible after one week, but after two weeks Lee Smith brand revealed considerable cytotoxicity. 3. In general, the zinc phosphate cements were less cytotoxic than the polycarboxylate cements.

Characterization of a Fibrinolytic Enzyme Secreted by Bacillus amyloliquefaciens CB1 and Its Gene Cloning

  • Bacillus amyloliquefaciens CB1 was isolated from cheonggukjang, a Korean fermented soy food. B. amyloliquefaciens CB1 secretes proteases with fibrinolytic activities. A gene homologous to aprE of Bacillus subtilis, aprECB1, was cloned from B. amyloliquefaciens CB1, and DNA sequencing showed that aprECB1 can encode a prepro-type serine protease consisting of 382 amino acids. When aprECB1 was introduced into B. subtilis WB600 using an E. coli-Bacillus shuttle vector, pHY300PLK, transformants showed fibrinolytic activity and produced a 28 kDa protein, the size expected for the mature enzyme. The 28 kDa fibrinolytic enzyme was purified from the culture supernatant of B. subtilis WB600 transformant. AprECB1 was completely inhibited by phenylmethylsulfonyl fluoride and almost completely inhibited by EDTA and EGTA, indicating that it is a serine metalloprotease. AprECB1 exhibited the highest specificity for N-succinyl-Ala-Ala-Pro-Phe-p-nitroanilide, a known substrate for ${\alpha}$-chymotrypsin. $A{\alpha}$ and $B{\beta}$ chains of fibrinogen were quickly degraded by AprECB1, but the ${\gamma}$-chain was resistant.

  • This study was carried out to investigate the effects of spider-derived protease [Arazyme (AZ)] supplementation on egg production, egg quality, cecal microflora and viscosity of intestinal contents in laying hens at the late production. A total of two hundred, 59-wks-old, Hy-Line Brown layers were divided into four groups, placed into 5 replicates per group (50 birds per group) and fed one of four diets containing 0.05, 0.075 or 0.1% AZ or control for 5 wks. The results showed that egg weight in group fed the diet containing 0.05% AZ was significantly higher than those of other groups (P<0.05). There were no significant differences in egg interior and eggshell qualities among the groups. The serum parameters such as total cholesterol, the levels of GOT and GPT were not influenced by AZ feeding. There were no significant differences in the cecal number of total microbes, Coliforms and lactic acid bacteria among the groups. The viscosity and ammonia concentration of intestinal contents in the groups fed AZ were significantly reduced compared to those of the control (P<0.05). In conclusion, laying hens fed AZ supplemented diet improved egg weight as compared with that of control. In addition, the AZ supplemented diet reduced the viscosity and ammonia concentration of intestinal contents, reflecting a significant feed enzyme effect.

Production of Multiple Probiotics and the Performance of Laying Hens by Proper Level of Dietary Supplementation (복합 생균제의 생산과 산란계에서 생균제의 적정 급여 수준에 의한 산란 효과)

  • This experiment was conducted to evaluate the dietary effects of multiple mixture of probiotics on laying performance and the faecal examination in laying hens (Hy-line Brown) at the early (21~40 wk) and middle (41~65 wk) laying term. Multiple probiotics were produced by developing products and the properties of microorganisms were examined for detecting of acid-resistance, bile salt-resistance and antibacterial activity against pathogenic enteric bacteria. Probiotics produced to the fermenting cultures of four selected organisms and soybean meal substrates by nine steps of NK proliferating system. The most microorganisms were shown higher resistance of acidity and bile salt. High antibacterial activities against Bacillus subtilis, Lactobacillus plantarum and Enterococcus faecium were observed, but was not against Saccharomyces cerevisiae. Total egg production of the treatment was significantly higher than control group but was not statistically different between 0.1% and 0.2% treatments (P>0.05). Average egg weight of the treatment in early laying term was also significantly higher than control but was not significantly different between 0.1% and 0.2% treatments (P>0.05). But the egg weight of the treatment in middle laying term was significantly higher than control and between 0.1% and 0.2% treatments (P>0.05). The mortality of 0.2% treatment was significantly lower than control (P<0.05), and 0.2% treatment in the early laying term was tended to decreased than 0.1% treatment and control. But there was not significantly between 0.1% and 0.2% treatments in middle laying term. In feed intake, 0.2% treatment in middle laying term was significantly increased than control and 0.1% treatment (P<0.05) but not in early laying term. In faecal examination, the total number of Lactobacillus of 0.1% treatment was significantly increased than control in whole laying term (P<0.05), but Coli form of the treatment was decreased than control in middle laying term. In conclusion, dietary long term supplementation of multiple probiotics improved performance of lay hens, egg weight and mortality drop by regulating enteric bacteria.

The Effects of the Transgenic Aspergillus oryzae Supplementation on Performance, Egg Quality and Intestinal Microflora of Layers (형질 전환 Aspergillus oryzae의 첨가가 산란계의 생산성, 계란 품질 및 장내 미생물 변화에 미치는 영향)

  • An experiment was conducted to investigate the dietary effects of a transgenic Aspergillus oryzae(AO) culture on the performance, egg quality and intestinal microflora of layers. A total of 840 Hy-line Brown layers of 39wks old were assigned to one of the following 7 dietary treatments: control(C), C+0.2% AO culture, C+0.5% AO culture, C+0.2% transgenic AO culture, C+0.5% transgenic AO culture, C+0.2% transgenic mutant AO culture, and C+0.5% transgenic mutant AO culture. The transgenic AO was made by inserting Salmonella gallinarum gene to AO. And the transgenic mutant AO was made by inserting Salmonella gallinarum gene to mutant AO which was mutated by UV irradiation. Each treatment was replicated six times with 20 birds housed in 2 bird cage. Twenty birds units were arranged according to completely randomized block design. Feeding trial lasted for 8wks under 16 hour lighting regimen. Laying performance and egg quality were significantly(P<0.05) affected by the treatments. Transgenic AO culture supplementation at the level of 0.2% significantly increased egg production, while its egg weight was significantly decreased compared to that of the control. Feed intake and feed conversion ratio(FCR) were not significantly different among the AO treatments and the control. The eggshell strength of the AO treatments was significantly higher than that of the control. Transgenic mutant AO culture supplemented at the level of 0.5% significantly increased egg yolk color. Intestinal microflora were significantly(P<0.05) affected by the treatments. The cfu of Lactobacilli spp. significantly increased and those of Salmonella species and E. coli decreased in the AO treatments. The transgenic AO and transgenic mutant AO culture were more effective than the AO culture in reducing the cfu of Salmonella species and E. coli. It is concluded that supplementation of the transgenic AO culture at the level of 0.2% could be recommended for the improvement of egg production. Supplementation of transgenic AO or transgenic mutant AO culture at 0.2% level effectively controlled intestinal Salmonella species population.

  • The study was conducted to evaluate the effects of dietary microbial phytase on egg productivity, egg quality, phosphorus utilization and nutrient digestibility in laying hens. The animals used in the experiment were a total of 120 Hy-Line Brown laying hens (32 weeks old). Dietary treatments included 1) CON (basal diet), 2) LP (low phosphorus diet) 3) NP (low phosphorus diet + 0.03% normal microbial phytase) and 4) CP (low phosphorus diet + 0.03% coated microbial phytase). Six laying hens were allotted to a block (pen) with five replicated. Through the whole period of experiment, egg production was significantly increased in CON and CP treatments compared to LP treatment (P<0.05). CON, CP and NP treatments significantly increased their egg shell breaking strength and egg shell thickness compared to LP treatment (P<0.05). CON, CP and NP treatments resulted higher yolk color and Haugh unit than LP treatment (P<0.05). Calcium and inorganic phosphorus contents in blood were higher in CP treatment compared to LP treatment (P<0.05). Dry matter digestibility and nitrogen digestibility were greater in CP treatment than CON, LP and NP treatments (P<0.05). Crude ash was increased in CON, CP and NP treatments compared to LP treatment (P<0.05). Calcium digestibility and phosphorus digestibility were significantly improved in CP and NP treatments than CON and LP treatments (P<0.05). Calcium retention of CON, CP and NP treatments were higher than LP treatment (P<0.05). Phosphorus retention was increased in CP and NP treatments compared to CON and LP treatments (P<0.05). LP treatment significantly increased their nitrogen, calcium excretion compared to CON, CP and NP treatments (P<0.05). Phosphorus excretion was decreased in LP, CP and NP treatments compared to CON treatment (P<0.05). In conclusion, CP treatment improved egg production, egg quality, inorganic calcium and phosphorus in blood, calcium and phosphorus retention and nutrient digestibility and decreased nitrogen and calcium excretion in laying hens.
