옥수수(Zea may L.)재배 토양에서 제지슬러지퇴비의 시용효과: 무기태인산을 중심으로(II) (Influence of paper Mill Sludge Compost to Corn(Zea may L.) cultivated Soil(Incase of Inorganic Phosphorus))

  • 본 연구는 제지슬러지퇴비를 농업분야에 활용할 때 시용효과를 분석하고자 수행하였고, 대상작물은 옥수수였다. 퇴비는 사질토양과 일정비율로 혼합하였으며, 시용효과를 분석하기 위해 인산의 가용화, 작물체의 양분흡수. 이에 따른 토양중 무기인산의 변화를 평가했다. 포트 시험은 비닐하우스에서 이루어졌으며, 인산의 분획물과 작물의 수량을 조사하였다. 포토실험에서 시간에 따른 변화중 Saloid-P의 양은 다소 증가하는 경향이었으나, Fe-P는 감소하였고, 시용량이 많았던 100Mg/ha 처리구에서는 감소의 폭이 뚜렷하였다. 제지슬러지퇴비를 시용한 포트에서 옥수수를 재배한 결과, 퇴비의 시용량이 많을수록 수량은 증가하였으며, 질소의 흡수는 증가한 반면, 식물체중 인산의 농도는 감소하는 경향이었다. 그러나 퇴비의 시용량 증가로 식물체중 인산의 농도가 감소하는 경향이었음에도 불구하고 인산의 결핍은 일어나지 않았다.

국내 옥수수 순계주에서 CP4 5-Enol- Pyruvylshikimate-3- Phosphate Synthase 유전자의 발현 (Expression of CP4 5-Enol-Pyruvylshikimate-3- Phosphate Synthase Transgene in Inbred Line of Korean Domestic Maize (Zea may L.))

  • 국내 옥수수 순계주에서 Agrobacterium 공동배양으로 CP4 5-Enol-pyruvylshikimate-3-phosphate synthase (CP4 EPSPS) 유전자가 도입된 제초제저항성식물체를 개발하였다. 5개의 순계주 (HW1, KL103, HW3, HW4, HW7)의 미숙배를 Ubiquitin promoter-CP4 EPSPS 유전자와 CaMV35S promoter-nptII 유전자가 발현되도록 제조된 pCAMBIA2300 벡터를 C58C1 Agrobacterium에 형질전환하여 공동 배양하였다. 항생제로 paromomycin이 첨가된 배지에서 선발된 옥수수 형질전환체를 PCR, RT-PCR 및 Northern 분석을 통하여 유전자의 도입과 발현을 확인하였다. 또한 형질전환 식물체의 glyphosate 처리에 따른 shikimate 축적반응을 확인하였다. Paromomycin 저항성 캘러스 형성빈도는 옥수수 각 순계주 HW1, KL103, HW3, HW4, HW7에서 각각 0.37%, 0.03%, 2.20%, 2.37%, 0.81%로 나타났으며, PCR분석을 통하여 최종적으로 2개의 옥수수 순계주 (HW3, HW4)의 paromomycin 저항성 캘러스로부터 분화된 식물체에서 확인하였다. 이러한 형질전환체중에서 RT-PCR 및 Nothern blot 분석을 통하여 CP4 EPSPS 유전자가 발현되는 2개의 계통 (M266, M104) 을 선발하였고, shikimate 축적반응을 통하여 glyphosate에 대한 저항성을 갖는 계통 (M266)을 최종적으로 선발하였다. 이러한 결과는 국내 옥수수 순계주에서 제초제저항성을 갖는 옥수수 형질 전환체를 개발할 수 있음을 시사한다.

인체지방유래 간질세포의 부착 및 연골분화유도를 위한 PLGA 지지체의 플라즈마 처리 효과 (The Effect of the Plasma Treatment on PLGA Scaffold for Adhesion and Chondrogenic Differentiation of Human Adipose-derived Stromal Cells)

  • High-density micromass culture was needed to take three dimensions culture with ASCs(adipose derived stromal cells) and chondrogenesis. However, the synthetic polymer has hydrophobic character and low affinity to cells and other biomolecules. Therefore, the surface modification without changes of physical and chemical properties is necessary for more suitable condition to cells and biomolecules. This study was performed to investigate the effect of surface modification of poly (lactic-co-glycolic acid)(PLGA) scaffold by plasma treatment (P(+)) on the adhesion, proliferation and chondrogenesis of ASCs, and not plasma treatment (P(-)). ASCs were isolated from human subcutaneous adipose tissue obtained by lipectomy and liposuction. At 1 hour 30 minutes and 3days after cell seeding onto the P(-) group and the P(+) group, total DNA amount of attached and proliferated ASCs markedly increased in the P(+) group (p < 0.05). The changes of the actin under confocal microscope were done for evaluation of cellular affinity, at 1 hour 30 minutes, the shape of the cells was spherical form in all group. At 3rd day, the shape of the cells was fiber network form and finely arranged in P(+) group rather than in P(-) group. RT-PCR analysis of cartilage-specific type II collagen and link protein were expressed in 1, 2 weeks of induction. Amount of Glycoaminoglycan (GAG) markedly increased in P(+) group(p < 0.05). In a week, extracellular matrix was not observed in the Alcian blue and Safranin O staining. However in 2 weeks, it was observed that sulfated proteoglycan increased in P(+) group rather than in P(-) group. In conclusion, we recognized that plasma treatment of PLGA scaffold could increase the hydrophilic property of cells, and provide suitable environment for high-density micromass culture to chondrogenesis

채취 방법에 따른 소 난포란의 회수율 및 수정란의 발달율 (Rates of Recovery and of Development In vitro of Follicular Oocytes Collected by Aspiration and Slicing Method in Cattle)

  • 본 연구에서 도축된 한우 난소를 aspiration법과 면도날 장착으로 제작된 기구를 이용해서 slicing 법으로 난포란을 회수하여 그에 따른 난포란의 회수율과, 난포란을 체외에서 22시간 성숙시킨 후 난포란의 핵 성숙을 및 체외수정 후 수정율과 발달율을 요약하면 다음과 같다. 1. 난포란의 회수율에 있어서 각 난소당 회수는 aspiration법이 6.7개. slicing법이 15.1개의 결과를 나타내어 난소에 대한 난포란의 회수율은 slicing법이 유의적 (P<0.05)으로 높은 결과를 나타내었다. 2 Aspiration법과 slicing법으로 채란된 난자를 체외에서 22시간 성숙시킨 후, 제 2감수분열 중기까지의 핵 성숙율은 각각 83%와 62%로 유의적인(P<0.05) 차이를 보였으나, 난소 한 개당 제 2감수분열 중기까지 성숙된 난포란수는 aspiration과 slicing법이 각자 5.6개와 9.4개로 slicing법에 의해서 유의적 (P<0.05)으로 많았다. 3. Aspiration법과 slicing법으로 채란된 난포란의 발달을 조사결과를 살펴보면 분할율과 배반포기까지의 발달율에서 aspiration법이 유의적 (P<0.07)으로 높게 나타났으나, 이와 상반되게 난소 한 개당 생산된 배반포기 수정란의 수는 slicing법에 있어서 2.8개로 aspiration법의 2.1개보다 유의적인 (P<0.05) 증가를 보였다. 이상의 결과에 따라 많은 난자를 이용하기 위한 목적과 혹은 이식 가능한 다수의 수정란 생산을 위해서는 본 연구에서 고안 제작된 기구를 이용하여 slicing법으로 난포란을 채란하는 것이 효과적인 방법이 될 것으로 사료된다.

  • 1. 항온기간중(恒溫期間中) $N_2O$의 경시적(徑時的) 발생량(發生量)은 항온(恒溫) 1일(日)과 10일경(日傾) 그리고 $N_2$의 발생량(發生量)은 1일(日)과 30일경(日傾)에 많았다. 2. 토양유기물함량별(土壤有機物含量別) 탈질량(脫窒量)은 현저(顯著)한 차이(差異)를 보이지 않았으며 시용유기물간(施用有機物間)에는 녹비(綠肥)>볏짚>퇴비(堆肥)>무시용(無施用)의 순서(順序)로 높았다. 3. 질소시비량(窒素施肥量)이 증가(增加)할수록 탈질량(脫窒量)은 증가(增加)되었으나 토양간(土壤間)에는 뚜렷한 차이(差異)가 없었음. 4. 탈질반응속도(脫秩反應速度)(계수(係數))는 토양유기물함량(土壤有機物含量)이 많고 역분해성유기물(易分解性有機物)을 시용(施用)할수록 증가(增加)되었음. 5. 토양중(土壤中) 탄소발생량(炭素發生量)과 탈질량(脫窒量)과의 관계(關係)에서 아산화질소(亞酸化窒素)는 정상관(正相關) 그리고 분자상질소(分子狀窒素)는 부상관관계(負相關關係)를 보였음. 6. 탄소(炭素) 1 mg이 소모(消耗)될때 약 4 mg의 $N_2O$와 3 mg의 $N_2$가 생성되었음.

오디(Mulberry) 추출물의 성분분석 및 생리활성에 관한 연구 (Studies on the Chemical Constituents Biological Activities of Mulberry Extracts)

  • 동결 오디(Mulberry) 10 kg을 80% 에탄올(EtOH)로 추출하고 분획깔때기 상에서 헥산(n-hexane), 에틸아세테이트(EtOAc) 및 물 가용부로 분획하였다. 에틸아세테이트 가용부(22 g)를 Sephadex LH-20으로 충진한 칼럼에서 MeOH-$H_2O$ (2 : 1~1 : 3, v/v)을 용출용매로 사용하여 칼럼크로마토그래피를 실시하였다. 단리된 화합물들은 셀룰로오스 박층크로마토그래피(TLC)로 확인한 후 $^1H-$, $^{13}C$-NMR 등의 스펙트럼을 사용하여 정확한 구조를 구명하였고 FAB 및 EI-MS로써 분자량을 측정하였다. 오디의 에틸아세테이트 가용부에서 quercetin-3-O-${\beta}$-D-glucopyranoside (화합물 1), protocatechuic acid (화합물 2), p-hydroxybenzoic acid (화합물 3)을 단리할 수 있었다. 항산화 시험 결과 에틸아세테이트와 물 가용부에서 BHT보다 우수한 활성을 나타내었으며 MTT assay에 의한 세포생존율 측정결과 세포독성은 없는 것으로 나타났다. 또한, 미백활성 및 항암활성은 낮았지만 세포신장 효과가 우수하여 주름개선 화장품 소재로서 충분히 이용 가능성이 있는 것으로 판단되어 진다.

자궁경부암(子宮頸部癌) 방사선치료(放射線治療) 전후(前後) Renogram의 의의(意義) (The Value of Isotope Nephrography in Carcinoma of Cervix - Follow up Studies of Pre and Post Irradiation)

  • It is a great value to find an early detection of involvement of ureteric obstruction in the carcinoma of cervix. Little or no knowledge of the condition of the kidneys or the lower urinary tract are able to elucidate by the biochemical studies such as blood nitrogen or urine creatinine in carcinoma of cervix. Findings of urography delineates the condition of urinary tract stasis in the renal pelvis and ureters, however, slight stasis maybe difficult to demonstrate. On the other hand isotope nephrography is accepted as a sensitive method to observe renal function especially in regarding to the excretory function of kidney. It was attempted to analysis the findings of urography conjunction with isotope nephrography in 50 cases of unselected patients with invasive carcinoma of cervix through pre and post irradiation follow up studies. Urography was done as a routine procedure and.analysed emphasising changes of collecting systems and ureter condition. Isotope nephrography was carried out by means of $15{\mu}ci\;I^{131}$-Hippuran injected intravenously and the curves were analysed as follows. Parameter were; time of maximum amplitude ($T_{max}$), half time of maximum amplitude ($T\frac{1}{2}$), Kac and Kex value calculated from these two parameters in Tobe's method. The excretion index by Aurell defines the ratio between the maximum activity and the activity measured on the slope of the third phase ten minites after it has reached its maximum. Results: 1. 28.8% had an abnormal IVP suggestive of ureteric involvement before irradiation therapy and the patient of stage III and IV were the great part. 2. 21.7% had abnormal findings of per-irradiation renogram whom showed normal IVP. The other group showed normal IVP which group also showed normal renogram prior irradiation. 3. The more severe the ureteric involvement, the change of excretion index was greater. 4. Even in stage I and II patient, abnormal renogram was revealed in 12 cases (39.4%) among 31 cases. 5. All cases of TAH showed abnormal findings of IVP and renogram. 6. No. definite change of renogram was obtained just after the irradiation therapy (point $A:8000{\sim}9000rads,\;B:5000{\sim}6000rads,\;Co:11000{\sim}13000rads$). Each 3 month follow up study was performed and comparing with preirradiation study which showed significant changes of excretion index of renogram were 42.8% in $6{\sim}9$ month follow-up and 75% in $9{\sim}12$ month, respectively. 7. It seems to be important to observe the parameter Kex and excretion index of renogram to determine early abnormality of kidney excretory function by means of post-irradiation follow up study.

뇌졸중(腦卒中)에 관(關)한 임상통계적(臨床統計的) 연구(硏究) (Clinical Observation on C.V.A.)

  • Clinical observation was done on 1092 cases of cerebrovascular disease which were confirmed by Brain CT scan in Oriental Medical Hospital in Kyung Hee Univ. from May 1987 to May 1988. Specially, clinical prognosis of 250 patients who had been hospitalized for over 4 weeks, were obserbed. The results were obtained as follows; 1. In this study, Occlusive CVD was 77.9%, Cerebral hemorrhage was 18.8%, Subarachnoid hemonhage was 0.8%. 2. The ratio of male to female was 1.4:1. In the age distribution, 60th decade, 50th decade, 40th decade, 30th's, 20th's were in order of frequency and specially 60th decade was 35.53% over 70th decade was 17.1% in ratio. 3. The most common preceding disease of CVD was hypertension (54.21%) and diabetes mellitus (19.96%) was second. 4. Almost, the duration of hospitalization was 2-4 weeks in 34.8%, within 4 weeks in 78.02%. 5. Primary attack was 75.7%, 2nd attack was 17.9%, over 3rd attack was 3.1% in ratio of recurrence. 6. The level of consciousness was Grade I in 96.4%, Grade II in 3.2%, Grade III in 1% at attack. 7. A few complications of C.V.A. were observed in the studies: pneumonia was noted frequently in 3.2%, bed sore, urinary tract infection, gastro intestinal bleeding in order of frequency. 8. The ratio of neurologic deficiency in occlusive CVD decreased from 51.9% to 29.3% in upper limb, 52.6% to 24.4% in lower limb, and that in cerebral hemorrhage decreased from 69.5% to 25% in upper limb, 50% to 20% in lower limb. 9. The ratio of left side hemiplegia to right was 1.04:1 in male, 1:1.18 in female. 10. The herb medications for C.V.A. were various Chungg-Paesagantang, Sunghanggeonggisan were used most frequently to Chungyeold, Geopung, Soongi, Haldam and Chungsimtang, the drugs for Bogiheol were used as discharge. In these oriental medical therapy of C.V.A. objective diagnosis and more various therapeutic method must be obtained through east-west medical co-operation.

중.저준위 방사성폐기물 처분 부지의 지하수 유동에 대한 수치 모사: 2. 처분 안전성 평가 인자 (Numerical simulation of groundwater flow in LILW Repository site:II. Input parameters for Safety Assessment)

  • 중 저준위 방사성폐기물 처분 부지 건설될 지하 시설물을 고려하여 지하수 유동 모델링을 수행하였으며, 처분 안전성 평가를 위해 건설 중 지하 시설물로의 지하수 유입량, 처분장 폐쇄 후 처분 사일로를 통과하는 지하수 유동량 및 처분 사일로에서 배출 지역까지의 지하수 유동로에 대한 이동 거리 및 단위 유량에 대한 입력 인자를 도출하였다. 처분장 건설 중 지하수 유입량 및 폐쇄 후 지하수의 유동량은 처분 심도에서 존재하는 다수의 투수성 구조 영역에 의해 영향을 받아 10가지 경우에서 각각 상이한 결과를 나타내었다. 지하수 유동로에 대한 지하수의 이동 거리 및 단위 유량에 대한 결과도 지하수의 유동로에 존재하는 투수성 구조 영역에 영향을 받는 것으로 분석된다. 가상의 인간 침입 시나리오를 위해 우물 시나리오를 가정하여 지하수 유동 모델링을 수행하였으며, 그 결과를 이용하여 처분 안전성 평가와 관련된 입력 인자를 도출하였다.

$Smartpeg^{TM}$의 고압멸균소독 및 재사용이 임플랜트 안정성 지수(ISQ) 측정에 미치는 영향 (THE EFFECT OF AUTOCLAVE STERILIZATION AND REUSE OF $SMARTPEG^{TM}$ ON THE IMPLANT STABILITY QUOTIENT (ISQ) MEASUREMENT)

  • Statement of problem: Resonance frequency analysis is evaluated as the non-invasive and objective method for the evaluation of implant stability and has been increasingly used. It is necessary to evaluate the factors affect the ISQ measurement stability. Purpose: This study was performed to evaluate the effect of the autoclave sterilization and reuse of $Smartpeg^{TM}$ on ISQ measurement. Material and methods: $SmartPeg^{TM}$ (Integration Diagnostics Ltd., $G\ddot{o}teborg$, Sweden) of autoclave group (A) was autoclave sterilized 9 times and $Smartpeg^{TM}$ of reuse group (R) was reused 9 times. Ten $SmartPeg^{TM}s$ were allocated to each group and after each autoclave sterilization and reuse, implant stability quotient (ISQ) values were measured 3 times from the two directions a and b at a right angle. $Osstell^{TM}$ mentor (Integration Diagnostics Ltd. $G\ddot{o}teborg$, Sweden) was used and type 1 (article no. 100353) $Smartpeg^{TM}$ was selected according to $Smartpeg^{TM}$ reference list. Osstem Implant US II future (Osstem Co., Seoul, Korea) in $4.0mm{\times}11.5mm$ was embedded in the self-curing acrylic resin ($Orthojet^{(R)}$, Lang Dental, U.S.A.). Data was statistically analyzed by one-way ANOVA $({\alpha}=.05)$ and scheffe test was done where a significant difference exist. Correlation test was also done between ISQ value and the number of autoclave sterilization or reuse. Results: 1. In autoclave group, the means and sd. of ISQ value before autoclave sterilization were $84.97{\pm}0.41,\;84.93{\pm}0.74$ at direction a and b. There was significant differences between autoclave groups at direction a and b (P=.000). 2. In reuse group, the means and sd. of ISQ value before reuse were $85.40{\pm}0.62,\;85.50{\pm}0.57$ at direction a and b. There was no significant difference between reuse groups at direction a and b (P>.05). 3. There was a weak positive correlation between the number of reuse and ISQ value at direction a and b (${\gamma}=.207$ and .246, P<.01). Conclusion: Within the limitations of this study, the following conclusions were drawn. Till ninth reuse of $Smartpeg^{TM}$, the ISQ measurement stability did not be affected. After twice autoclave sterilization of $Smartpeg^{TM}$ the ISQ measurement stability was affected.
