Studies of the Effects of Hexachlorophene on the $C^{14}-Glucose$ and $C^{14}-Glucose$ Metabolism of Clonorchis Sinensis (Hexachlorophene의 간흡충(肝吸?)의 $C-^{14}$ Glucose 및 $C-^{14}$ Glycine 대사(代謝)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • Hexachlorophene was reported previously to have a powerful parasiticidal effects on Clonorchis sinensis in vitro and in vivo, but the mechanism of its effect was not known. In the present report it was observed that there was an influence of hexachlorophene on the oxygen consumption, the glycolysis, the glycogenesis and the protein synthesis of C. sinensis. A hundred mg. of C. sinensis collected from the biliary tracts of the infested rabbits was incubated in 2 ml of K.R.P. medium with vavious concentration of hexachlorophene, $glucose-1-C^{14}$ and $glycine-1-C^{14}$ in a 25 ml incubation flask with central well. The oxygen consumption was observed by Warburg manometer, the glycogenolysis by measurement of radioactivities of extracted glycogen and protein from C. sinensis incubated with $C^{14}-glucose$ or$C^{14}-glycine$. 1) The oxygen consumption by C. sinensis was markedly inhibited during all stages of incubation in concentration of $10^{-4}$ and $10^{-5}g/ml$ of hexachlorophene, but in $10^{-6}$, slightly increased initially and gradually decreased after 3 hours of incubation. 2) Hexachlorophene inhibited the glycolysis by C. sincnsis markedly in the concentration of $10^{-4}$, $10^{-5}$, $10^{-6}$ and $10^{-7}g/ml$. 3) The protein synthesis by C. sinensis from glycine was inhibited in the concentration of $10^{-5}$, $10^{-6}$ and $10^{-7}g/ml$ of hexachlorophene. 4. The glycogen synthesis by C. sinensis in each concentration of $10^{-4}$, $10^{-5}$ and $10^{-6}g/ml$ of hexachlorophene was inhibited markedly. The speed of inhibition was more rapid in high concentration than in low, and in low concentration even the glycogen itself which had synthesized in their stage in their body was consumed in later stage. 5) The effects of oxygen consumption, glycolysis and glycogen synthesis were not influenced in the concentration of $10^{-5}g/ml$ of chloroquine phosphate, whereas hexachlorophene and dithiazanine inhibited markedly in same concentration, and the former was more potent than the latter.

Lipofuscin pigment in adult Clonorchis sinensis (간흡충에 나타나는 리포푸신 색소)

  • 실험동물에서 짧은 기간 감염시킨 후 얻은 간흡충(Clonorchis sinensis)에서는 거의 볼 수 없지만, 간, 담도 수술 또는 부검을 실시할 때 사람에서 얻은 간흡충중에는 검은 색소를 갖는 것을 관찰할 때가 있다. 이 색소는 살아 있는 간흡충에서도 명백하며, 색소가 없는 간흡충은 색이 분홍빛이고 투명한데 비하여 색소가 있는 것은 검은색을 띠고 불투명하다. 사람에서 얻은 간홉충 모두가 검은 색소를 갖고 있는 것은 아니며, 이제까지의 경험으로는 30세 이상 어른에서 얻은 간흡충중 그 길이가 약 1cm이상인 것에서 나타난다. 이 색소의 성분에 대하여 과거에는 빌리루빈, 또는 숙주 혈구세포에서 유래한 철을 포함하는 색소일 것이라고 생각하고 있었다. 저자등은 이 색소가 간흡충의 노쇠현상과 관련된 것으로 생각하고, 그렇다면 lipofuscin일 것이라고 생각하였다. 위의 가정을 확인하기 위하여 육안적으로 분명히 검은 색소가 있은 충체의 조직절편을 만들고, H & E, prussian blue, bilirubin, Fontina-Masson, PAS, amylase-PAS, Ziehi-Neelsen 및 AFIP method for lipofuscin등 8가지 조직화학적 염색을 실시하였다. 색소는 H & E, prussian blue, bilirubin, montana-Masson염색으로 착색되지 않았으며, PAS, amylase-PAS, Ziehi-Neelen, 및 AFIP method for lipofuscin에 염색되어 이것이 조직화학적으로 리포푸신 색소임을 확인하였다. 색소를 다시 전자현미경으로 관찰한 바 색소과립안에는 모양이 등글거나 타원형이고 크기가 $0.05~0.240{\mu}m$인 세포막구조가 산재하였음을 발견할 수 있어 간흡충의 검은 색소는 리포푸신 색소임을 다시 확인할 수 있었다.

  • A follow-up stool examination was done to know the status of intestinal parasitic infections in Army soldiers who were stationing in Whachon-gun, Kangwon-do, Korea in 1993. Of 233 examined soldiers, Infections with Clonorchis sinensis (0.4%), Metagonimus spp. (0.9%), Echinostomc honene (0.4%), Neoniplostomum seodenis (0.4) and Ginrdin lomblic (3/233) were found. S. honene and N. seouLensis infections were each 75th, and 27th cases in Korea. Regular control program of intestinal parasitic Infections might be able to contribute to diminish the prevalence rate in young Army soldiers in Korea.

  • In a paleo-parasitological analysis of soil samples obtained from V-shaped pits dating to the ancient Baekje period in Korean history, we discovered Ascaris lumbricoides, Trichuris trichiura, and Clonorchis sinensis eggs. In light of the samples' seriously contaminated state, the V-shaped pits might have served as toilets, cesspits, or dung heaps. For a long period of time, researchers scouring archaeological sites in Korea have had difficulties locating such structures. In this context then, the present report is unique because similar kind of the ancient ruins must become an ideal resource for successful sampling in our forthcoming paleoparasitological studies.

  • The experimental rabbits were infested with 300 of metacercariae of Clonorchis sinensis. The therapeutic effects of following 6 kinds of hexachlorophene derivatives on infected rabbits were compared with those of hexachlorophene and 2,2-methylenebis (3,4,6-trichlorophenoxy acetic acid) (MTPA). The drugs were medicated by mouth for 7 days after infection of 40 days, and the following results were obtained. 1) The therapeutic effects of following four hexachlorophene derivatives are less potant than those of hexachlorophene and MTPA. 2) Among hexachlorophene derivatives, 2,2-methylenebis (3,4,6-trichlorophenoxy benzoylate) and 2,2-methylenebis (3,4,6-trichlorophenoxy ${\beta}-ethly$ alcohol) are more effective than 1,1-dihydroxy ethyl ether, 2,2-methylenebis (3,4,6-trichlorobenzene) (Table 1).

  • Purpose: The purpose of this research was to describe clonorchis sinensis infection experience in high risk populations living in riverside areas. The research question was "How do local residents perceive Clonorchiasis and how are they infected with the parasite". Methods: Qualitative data were collected by focus group interviews with 16 participants from January to February, 2009. All the interviews were tape-recorded, transcribed, and analyzed by the content analysis method. Results: Five main categories were conceptualized, which were "lacking in understanding of Clonorchiasis," "culture of the rural community," "life style," "recovering experience from infection" and "change of health behavior." Conclusion: The result of this study indicates that residents are infected with Clonorchiasis through interaction between individual and group risky factors. Therefore, it is important to develop effective health education programs on both individual and group levels to prevent infection with Clonorchiasis.

  • The present study was undertaken to evaluate the present status of intestinal parasitic infection in Seoul area, Korea. During the period from June 1985 to July 1986, a total of 5,251 stool samples were collected in Department of Clinical Pathology, Hanyang University Hospital and examined by formalin-ether sedimentation technique once for helminth ova and protozoan cysts. The results were summarized as follows: 1. The overall egg positive rate of intestinal helminthes was 2.53%; and 1.43% for Clonorchis sinensis, 0.7% for Trichuris trichiura, 0.13% for Metagonimus yokogawai, 0.06% for hookworm, and 0.02% for Ascaris lumbricoides, Diphyllobothrium latum and Hymenolepis nana, respectively. 2. The overall cyst positive rate of intestinal protozoa was 1.07%. Cyst positive rate was 0.06% for Entamoeba histolytica and 0.13% for Giardia lamblia, respectively. 3. Higher prevalence of intestinal parasitic infection was observed in male and in $21{\sim}50$ year-old groups. 4. The highest prevalence of Clonorchis sinensis infection was observed in 40th age group (3.4%), and male (2.3%) was more infected than female (0.7%) in general.

  • Oxygen consumption by Clonorchis sinensis (C.S.) was observed in a medium of valuable pH of KRP, lack of certain electrolytes of KRP and of glucose, acetic acid or propionic acid was added to KRP. The results were as follows. 1. In a different solutions, KRP with or without $0.4%-glucose$, $0.9%-saline$ solution, the variation of oxygen consumption by C.S. was not significant. 2. Oxygen consumption by C.S. was inhibited by the solution of KCI, $CaCl_2$, or $MgSO_4$ in a 0.9% solution of saline. 3. Under the acidity, oxygen consumption was enhanced but under the alkality, it was inhibited. 4. Oxygen consumption by C.S. was inhibited by acetic acid or propionic acid in KRP solution. 5. C.S. keeping in KRP solution for a long duration, oxygen cosumption was reduced in the course of time.

  • The inhibitory action of oxygen consumption and parasiticidal action of hexachlorophene or MTPA combined with or without sodium cholate in Clonorchis sinensis (C.S) have been observed. The results were obtained as follows. 1. In low concentration of MTPA or Hexachlorophene, they inhibited the oxygen consumption of C.S and in high concentration, inhibition of oxygen consumption and parasiticidal action was more remarkable. 2. The oxygen consumption was inhibited also in the concentration of $10^{-2}$ and $2{\times}10^{-3}g/ml$ of sodium cholate. 3. In the presence of sodium cholate, oxygen consumption of C.S was more remarkably inhibited by MTPA or Hexachlorophene than the absence of sodium cholate. 4. From those results, it may be considered that sodium cholate combined with MTPA or Hexachlorophene produced potentiation in its action.

  • The prevalence of Intestinal parasite Infections In Inhabitants at the reaches of the Hantan River, Chorwon-gun, Korea, was observed from August 12 to September 14, 1993. Of 465 people observed by cellophane thick smear and formalin-ether concentration method, 2 Ascots lumbricoides (unfertilized), 1 Trichwis Dichturn, 39 Clonorchis sinensis and 16 Metagonimus egg positive cases were found. After treatment, the Metogonimus egg positive cases passed out flukes of Metogonimus Mlyata type. Of 68 fish caught In the Hantan River, 14 (20.6%) were Infected with metacercanae of Metogontmn, while no metacercana of C. sinenis was found. At this area, soul-transmitted nematodes are very low, but clonorchlasis and metagonlmlasis are prevalent by modernate endemicity.
