  • The udders of 28 culled dairy cows were removed at slaughter and bacteria isolated. Isolations of major known mammary pathogens comparised Staphylococcus sp. (40.7%), non-identified Gram's negative rods (33.2%) and E. coli (7.4%). In the mixed infection, Staphylococcus sp. was most frequently involved. Drug sensitivity tests were performed on several bacterial species by disc method. These tests indicated that the microorganisms isolated from clinical mastitis of dairy cows were more resistant to antimicrobial agents than the organisrms isolated from milk samples.

  • Nowadays, medical diagnostics is efficiently supported by clinical chemistry and near infrared spectroscopy is becoming a new dimension, which has shown high potential to provide valuable information for diagnosis. The investigation was carried out to study the influence of mammary gland inflammation, called mastitis, on cow´s milk spectra and milk composition measured by near infrared spectroscopy (NIRS). Milk somatic cell counts (SCC) in milk were used as a measure of mammary gland inflammation. Naturally occurred variations with milk composition within lactation and in the process of milking were included in the experimental design of this study. Time series of unhomogenized, raw milk spectral data were collected from 3 cow along morning and evening milking, for 5 consecutive months, within their second lactation. In the time of the trial, the investigated cows had periods with mammary gland inflammation. Transmittance spectra of 258 milk samples were obtained by NIRSystem 6500 spectrophotometer in 1100-2400 nm region. Calibration equations for the examined milk components were developed by PLS regression using 3 different sets of samples: samples with low somatic cell count (SCC), samples with high SCC and combined data set. The NIR calibration and prediction of individual cow´s milk fat, protein, and lactose were highly influenced by the presence of mil samples from animals with mammary gland inflammation in the data set. The best accuracy of prediction (i.e. the lower SEP and the higher correlation coefficient) for fat, protein and lactose was obtained for equations, developed when using only “healthy” samples, with low SCC. The standard error of prediction increased and correlation coefficient decreased significantly when equations for low SCC milk were used to predict examined components in “mastitis” samples with high SCC, and vice versa. Combined data set that included samples from healthy and mastitis animals could be used to build up regression models for screening. Further use of separate model for healthy samples improved milk composition measurement. Regression vectors for NIR mild protein measurement obtained for “healthy” and “mastitic” group were compared and revealed differences in 1390-1450 nm, 1500-1740 nm and 1900-2200 nm regions and thus illustrated post-secretory breakdown of milk proteins by hydrolytic enzymes that occurred with mastitis. For the first time it has been found that monitoring the spectral differences in water bands at 1440 nm and 1912 nm could provide valuable information for inflammation diagnosis.

  • Mastitis is the most costly disease results in lost milk production, decreased milk quality, milk discard, early culling of cows, drug costs and labor costs in dairy cow. Until now, a antibiotic administration at the end of lactation, dry cow therapy has been known the most effective and widely used mastitis control method. However, dry cow therapy do not control a new infection in the late dry and prepartum period because dry cow products have only persistent activity in the early dry period. Therefore, this study was conducted to evaluate clinical effect of sustained released biodegradable cephalexin microsphere using PLGA in bovine mastitis control during dry period. PLGA has been approved as controlled drug release system because of non-toxic, non-tissue reactive and bioerodible characteristics. This study revealed that cephalexin microsphere had a spherical shape with characteristic porous structure on the surface. Also, in vitro drug release studies are clearly observed that the release rate of cephalexin from PLGA microsphere decrease during the first 21 days after initial burst and then increase again between 3 and 4 weeks showing pulsatile releasing pattern. On the other hand, as tried in field the new infection rate, cure rate and mean SCC after parturition in cephalexin microsphere infused group were significantly differenced as compared to the control group. Accordingly, a sustained release of cephalexin from a biodegradable microsphere could make dry cow therapy more efficiently by preventing a new infection and decreasing the number of existing infection of mammary gland during dry period.

  • The present study was conducted to investigate isolation and antimicrobial drug susceptibility of clinical mastitic milking total 610 (897 quaters) dairy cattles from 36 dairy farms in Gyeongnam south area (Cosung, Masan) during the period from March 1999 to August 2002. The results obtained were summerized as follows . 1. Incidence of bacterial infection in four quaters was showed that right anterior quarter was 178(19.8%), right posterior quarter was 292(32.6%), left anterior quarter was 148 (16.2%), and left posterior quarter was 279 (31.1%), respectively. Isolation rate of posterior two quarters was higher 2 times than anterior two quarters. 2. Incidence of double infections of 897 clinical mastitic milk were showed that single infection was 549 (61.2%), double infection was 167(18.6%), triple infection was 9(1%) and no isolation was 172(19.2%). 3. Isolation of infected etiologic bacteria was showed that Streptococci spp., was 267(31%), Staphylococci spp., was 267(41%), S aureus, was 48(5.6%), G(-) bacillus was 126(5.6%), and Corynebactrium spy. was 52(6%), respectively, from total 861 samples. 4. The results of antimicrobial drug susceptibility of all isolates were showed that Streptococci spp., Staphylococci spp., S aureus, (G)(-) bacillus, and Corynebactrium spp. were susceptible to cefuroxime, cefoperazone, amoxicillin, cefazolin, ampicilin, penicillin, gentamicin, erythromycin, cloxacilin, ciprofloxacin, sulfamethoxasole/trimetoprim, teteracyclin, and norfloxacin (> 70%), but some of them were resistant to neomycin, streptomycin colistin, and cephalothin(> 60). 5. The results of drug susceptibility obtained from each farms had different susceptibility, even though, etiological microorganisms were same in each farms.

  • A swine farm located in the Kyungpook province (designated as farm D that have been suffering from PED for several years was selected to study the etiology and the outbreak pattern of PED by clinical and laboratory examinations. Clinical examination indicated that sows exhibited signs of mastitis resulting in an inadequate transfer of lactogenic immunity against PEDV to newborn piglets. Furthermore, serological tests revealed that all sow groups and their piglets had low levels of anti-PEDV antibody. These data suggest that improper vaccination program has been indeed performed in this farm. Remarkably, despite no symptoms of PED in weaners, the presence of PEDV was identified by RT-PCR from fecal samples of weaning piglets, indicating persistent PEDV circulation in the herd. Based on these results, the following basic control schemes were executed for the control of PEDV circulation in the farm; a) A quick removal of affected pigs and disinfection of affected sheds. b) restructuring of vaccination program and employment of consultant. c) prompt treatment of mastitis and removal of poor lactogenic sows, and d) enhancement of biosecurity of farrowing house by acquisition of additional space. We evaluated risk factors and implementation of control measures in two months and were unable to found any case related to PEDV infection. Taken together, our data indicate that the method described above is effective for the control of PED outbreak in farm persistently suffering from PEDV infection.

  • 1. The number of average milking cows, clinical forms of mastitis, mastitis-developing cows, and cows killed by mastitis a year were 25.7, 1.8(7%), 6.3(26%), and 2.7(10.1%)heads, respectively. The annual grade changes of standard plate counts(SPC) and somatic cell counts(SCC) showed the grade 1A of SPC diminished sharply from April to August, we think it was due to the lack of proper management in farming season and the grade 3 of SCC indirectly influenced increased in huge during August. 2. The average number of parturitions of farms was 2.3, but 50% of below 1 parturition were 22 farms(31%), 50% of above 3 parturitions were 16(23%) out of 71 farms. According to grades of the number of parturitions of milking cows per each farm, the farms' grades recording 3 parturitions and 50% were little bit excellent. 3. The actual situation research of foremilking CMT revealed 35 out of 74 farmer didn't do CMT Among them(35 out of 74 farmers), 80% did not test thanks to the troublesome process of the CMT. SCC grade 3, among farms who did foremilking CMT once or twice a month and who did not were 29% and 40% respectively and SPC grade 1A were 55% and 9%, respectively. 4. The research of actual situation on milking management let us know 29 farms(39%) did not do lastmilking, 37 farms(49%) usually did overmilking, and 34 farms(46%) did milking for 4 or 5 minutes. Grades according to average requiring times of milking showed SCC grade 1 of farms milking within 7 minutes was 11% and SPC grade 1A was 34%, on the other side, farms milking more than 7 minutes were 0% in SCC grade 1 and 13% in SPC grade 1A. Grades according to the starting time of milking after rubbing teats showed SPC grade 1A of farms starting milking at about 1 minute and over 2 minutes were 50% and 20%, respectively. 5. The research of actual situation on hygienic milking management uncovered 65 farms(88%) were using one towel which was used in washing teats and udders to wash more than 3 to 4 cows, and 53 farms(72%) were using one dried towel to dry udders not for each cow but for more than 3 to 4 cows after washing. Also, on milking turns disclosed 30 farms(40%) were milking cows in the order of incoming without isolation of a dominant group. According to grades of towels used in washing teats and udders, farms using a towel for each cow were 56% and a towel for over 3 cows were 31% in SPC grade 1A. According to using-or-not grades of dried towels after washing udders, farms using a towel for each cow were 79% and a towel for over 3 cows were 21% in SPC grade 1A. 6. Farms doing teat-dipping before milking were 7(10%), not doing teat-dipping after milking, or doing sometimes were 9(12%), and doing right after milking were 57(77%). And farms doing teat-dipping after dry cows and before delivery were 21(28a ). Farms using bethadine as an antiseptic solution were 70(95%), 40 farms(59%) diluted it with water as weak as 5 to 10 times, and on drying cows 64 farms(87%) slowly did it more than 2 days. Grade 1A of SPC of farms doing teat-dipping at every milking was 38%, farms doing occasionally or not was 33%, and farms doing it right after milking was 37% and doing after milking more than 5 cows was 20%. Grade 1A of SPC among farms diluting bethadine 5 times and diluting 5 to 10 times with water were 36% and 33%, respectively, and Grade 3 of SCC were 35% and 32%, respectively. 7. Studies on nonlactating period medical treatment, as the cows were on dry, 54 farms treated with their own hands.73 farms(98%) had bovine mastitis treated for themselves. And on applying medicines against mastitis, 55 farmers chose them on the basis of their own experience, 42 farms(57%) were treated more than 3 days. 41 farms(55%) dumped away the mastitis infected milk separately, 24 farms(32%) were feeding and milking at the same time. 8. Fifty-six farms(76%) always washed and disinfected milking machines after milking. Farms using the milking machines at low, or variable vacuum pressures, or at the vacuum pressure, set at the moment of its installation were 31(42%), and farms that did not know pulsation ratio were 27(37%). Farms changing liners when they were torn 8(11%), 58 farms(78%) said they checked milking system when there were wrong with them, 31 farms(42%) changed milking hoses when they found out problems, and 42 farms(57%) cleaned vacuum and milking systems when they felt dirty. • Bovine mastitis is an important disease causing serious economic loss in dairy production and food poison in public health. The major causative agents of bovine mastitis include Escherichia coli (E. coli), Streptococcus agalactiae (S. agalactiae), Staphylococcus aureus (S. aureus). These bacteria were found in milk and environmental condition such as feces, water, soil and so on. Recently, many cases of mastitis are derived from environmental contamination of micro-organisms, which important factors for the spread of this disease in farm. Ultraviolet irradiation (UV) has been used as disinfection for waste and water in clinical and industrial facilities. Moreover the UV irradiation has been used as useful bactericidal agents to remove bacterial biofilms in environmental condition. In this study, we determined the bacterial replication in different percentage of water content (PWC) in sterilized saw dust and feces complexes from farm, and results showed that slightly decreased growth pattern of E. coli and S. agalactiae but increased growth pattern of S. aureus in various PWC (200, 400 and 600%) until 144 h incubation. In the bacteriocidal effect of UV irradiation to bacteria in saw dust and feces complex, the results showed that bacteriocidal effect was depended on the UV irradiation time, irradiation distance and PWC. Especially the antibacterial activity of UV irratiation is stronger in low PWC (50%), long time irradiation (50 sec), and short distance (5 cm) than other condition of this study. Furthermore UV irradiation with stirring showed increased the bactericidal effect compared without stirring. These results suggested that bovine mastitis causing agents may survive long time in environmental condition especially saw dust and feces complexes in farm and can cause a various disease including mastitis. Moreover, these data can be used as basis for application and development of UV disinfection to control of bovine mastitis from environmental contaminated bacteria in dairy farm.

  • This study was performed to investigate Mycoplasma (M) spp. infection status of dairy cow in South Korea. Among 8,485 bulk tank milk collected from dairy farms of the 5 areas, 26 samples (0.30%) were positive for Mycoplasma by direct culture method. The isolation rates of Mycoplasma spp. according to the areas were 0.51% in Kyonggi, 0.16% in Cholla, 0.23% in Gyoungsang, 0.12% in Chungchong, and 0.08% in Kangwon. In the species identification test by indirect immunoperoxidase test, 16 out of 26 isolates were identified as M bovis (61.53%), M bovigenitalium (23.07%), M californicum (7.69%), M alkalescens and Acholeplasma laidlawii (3.84%), respectively. The isolation rate of Mycoplasma spp. from 208 quarter milk samples in culling cows due to severe clinical mastitis was 3.0%. These Mycoplasma spp. were identified as M bovis (62.0%), M bovigenitalium (12.0%), M californicaum (12.0%), and M alkalescens (12.0%). This study shows that the bovine mammary gland infected by Mycoplasma spp. is present in some dairy farms and the routine culture test of bulk tank milk samples for Mycoplasma is needed for a high quality milk promotion services.

  • Lack of hygiene and puerperal mastitis are common causes of bacterial diseases in nursing neonates. The aim of this study was to isolate microorganisms from milk samples of healthy female Jindo dogs with suckling puppies and to investigate antimicrobial susceptibility against the isolated bacteria. Milk samples were collected from 120 udders of 12 lactating Jindo dogs that were 2~4 years old without any clinical diseases including mastitis. Bacteria were isolated from 64 milk samples (53.3%), either singly (76.6%) or in combination (23.4%). Staphylococcus (S.) spp. was the most common microorganisms (74.7%) isolated from canine milk, followed by Haemophillus spp. (10.9%), Streptococcus spp. (9.6%), Gardnerella spp. (2.4%) and Moraxella spp. (2.4%). The most frequently isolated organism was S. warneri (31.3%). Antimicrobial susceptibility of these bacteria was tested with 17 antimicrobial agents by Kirbyand Bauer standardized disc diffusion method. Results indicated that bacteria isolated from healthy canine milk were mostly susceptible to amoxicillin-clavulanic acid, cephalothin and ceftiofur, but were resistant to erythromycin, neomycin and tetracycline.
