Docking Study of Cysteinyl Leukotriene 1 Receptor: Therapeutic Target for Allergy

  • Cysteinyl leukotrienes are inflammatory mediators having important role in pathophysiological conditions such as asthma and allergic rhinitis. CysLT1 receptor mediates most of the disease regulatory actions of the CysLTs and it is been implicated in a number of inflammatory conditions including gastrointestinal and cardiovascular diseases. Hence in the present study, molecular docking of CysLT1 was performed with its potent and orally efficacious antagonist CP-199330 and CP-199331. The aim of this study was to compare the interaction of CP-199330 and CP-199331 with known drugs such as Zafirlukast, Pranlukast and Montelukast which had already showed clinical efficacy in the treatment of asthma. The residues such as TYR83, GLN274, LYS311 and SER313 were found to interact with both the antagonist and the known drugs. Also, we noticed the docking scores and interaction of the antagonists were comparable with the known drugs. Hence these antagonists could serve as better drugs for the treatment of allergy.

  • It is essential to study the effects of hypergravity on the human body in the research field of aerospace medicine. Previous studies have shown that hypergravity could act in a harmful way to the human body. However, recent studies have shown that moderate degree of hypergravity can act in a beneficial way to the human body. The authors have in particular been studying the effects of hypergravity on immune diseases, especially allergic asthma and rhinitis. Therefore, this review paper will introduce the results of recent studies on hormetic (health promotion) effect of hypergravity. In addition, this review paper also examines the mechanism by which hormetic effect of hypergravity could be possible. Furthermore, we will also discuss how to apply these hormetic effects in clinical practice.

  • This study was undertaken in an attempt to provide scientific grounds in explaining the causes of environmental and respiratory diseases resulting from air pollutants in Gyeongju and its neighboring areas. In relation to heavy metals, lead (Pb) was $0.0135{\sim}0.1744\;{\mu}g/m^3$ and high in order of Pohang, Ulsan, and Gyeongju while nickel (Ni) was $0.0023{\sim}0.0115\;{\mu}g/m^3$. The concentrations of heavy metals in the investigated areas did not exceed the environmental standards or Guideline Value of Korea or other countries. However, it is considered necessary to apply intensive control to some heavy metals including cadmium (Cd) that show a relatively high level of hazard. Based on the responses to the survey, measured personal interest in environmental pollution and the basic knowledge of the causes of the respiratory diseases was higher in those with a family history of allergic reactions to metals and bronchial asthma. The incidence of allergic disease was higher in those who are currently in poor health state. In addition, the general knowledge of environmental pollutants was higher in those with higher educational level and those with a higher interest in environmental pollution. Personal interest in environmental pollution was higher in those with higher basic knowledge of environmental pollutants. Therefore, it is necessary to increase the awareness through better education and campaigns on environmental pollution.

Effect of Zedoariae rhizoma on Bronchial Inflammation and Allergic Asthma in Mice

  • There are detailed descriptions of the clinical experiences and prescriptions of asthma in traditional Korean medicine. Zedoariae rhizoma is one of the Korean herbal medicines used to treat bronchial asthma and allergic rhinitis for centuries. However, the therapeutic mechanisms of this medication are still far from clear, In this study, a house-dust-mite (Dermatophagoides pteronyssinus [Der p])-sensitized murine model of asthma was used to evaluate the immunomodulatory effect of Zedoariae rhizoma on the allergen-induced airway inflammation in asthma. Three different protocols were designed to evaluate the treatment and/or long-term prophylacitic effect of Zedoariae rhizoma in Der p-sensitized mice. Cellular infiltration and T-cell subsets in the bronchoalveolar lavage fluid (BALF)of allergen-challenged mice were analyzed. Intrapulmonary lymphocytes were also isolated to evaluate their response to allergen stimulation. When Zedoariae rhizoma was administered to the sensitized mice before AC (groups A and C), it suppressed airway inflammation by decreasing the number of total cells and eosinophil infiltration in the BALF, and downregulated the allergen- or mitogen-induced intrapulmonary lymphocyte response of sensitized mice as compared to those of controls. This immunomodulatory effect of Zedoariae rhizoma may be exerted through the regulation of T-cell subsets by elevation or activation of the CD8+ and double-negative T-cell population in the lung. However, the administration of Zedoariae rhizoma to sensitized mice 24 h after AC (group B) did not have the same inhibitory effect on the airway inflammation as Zedoariae rhizoma given before AC. Thus, the administration of Zedoariae rhizoma before AC has the immunomodulatory effect of reducing bronchial inflammation in the allergen-sensitized mice. On the other hand, to determine the potentiality of prophylactic and/or therapeutic approaches using a traditional herbal medicine, Zedoariae rhizoma, for the control of allergic disease, we examined the effects of oral administration of Zedoariae rhizoma on a murine model of asthma allergic responses. When oral administration of Zedoariae rhizoma was begun at the induction phase immediately after OVA sensitization, eosinophilia and Th2-type cytokine production in the airway were reduced in OVA-sensitized mice following OVA inhalation. These results suggest that the oral administration of Zedoariae rhizoma dichotomously modulates allergic inflammation in murine model for asthma, thus offering a different approach for the treatment of allergic disorders.

  • In order to evaluate the relationship between the bibliographical and clinical effectiveness of Socheongryoungtang, this study was carried out to investigate the effects of Socheongryoungtang on the respiratory system, cardiovascular system and isolated organ in the experimental animals. The following results of Socheongryoungtang were obtained; 1. The relaxing effect on the muscular contraction of isolated ileum induced by acetylcholine chloride, barium chloride and histamine${\cdot}$2HCl was recognized in mice, rat and guinea-pig. 2. The effect of direct vasodilatation was noted in rabbit. 3. The effect of hypotensor was recognized in rabbit. 4. The antihistamine effect was noted on both isolated ileum and tracheal strip-chain in guinea-pig. 5. The inhibitory effect on gastric ulcer induced by histamine${\cdot}$2HCl was noted in rat. 6. The inhibitory effect on vascular permeability was revealed in mice. 7. The antitussive effect was recognized in both dog and cat. 8. The effect of expectorant was recognized in rabbit. According to the above results, we has recognized that Socheongryoungtang has good efficacy for bibliographical and clinical diseases caused from respiratory system (especially allergic rhinitis etc.), cardiovascular system and digestive system.

Clinical Experiences of Stellate Ganglion Block Therapy (성상신경절차단요법에 대한 임상경험)

  • Stellate ganglion block therapy is widely practiced in pain clinics for a wide variety of diseases(numbering over 150). The precise mechanisms are unknown, but it has been said that the systemic effect of the stellate ganglion block may improve the hypothalamic circulation and thus restoring the homeostasis of the body. We have administered stellate ganglion block for the treatment and cure of the following examples of ailments idiopathic hypertension, allergic rhinitis, constipation, insomnia, irritable bowel syndrome. The causes of these diseases are unclear except that they are recognized as nervous disorders. However in clinical field, the effect of stellate ganglion block appears clearly. In this study 13 patients were treated only with stellate ganglion block; once daily for l5 days. The outcome of the treatments were as follows: 6 patients had excellent results, 5 patients had good results and the remaining 3 with poor results. We conclude that stellate ganglion block appears to be a good choice as the treatment for a number of nervous disorders.

Inhibitory Effects of Immediate-Type Allergic Reaction of Okbyungpoongsan-Gami by Anal Therapy (항장효법에 의한 옥병풍산가미의 즉각형 알레르기 반응 억제 효과)

  • Okbyungpoongsan-Gami (OG) has been used for the treatment of excessive sweating with general weakness and allergic rhinitis recently. Anal therapy is another way of taking Oriental medicine. It is an important pathway but not available in common clinical situation. This experiment was performed in order to study the inhibitory effect of immediate-type allergic reaction of OG by anal therapy. In addition, the experiment was practised by 1 H-NMR spectroscopy to examine molecular structure of OG. The results were obtained as follows : OG concentration-dependently inhibited compound 48/80- induced immediate type systemic allergic reaction with concentrations of 0.01-1.0g/kg by anal administration 1 h before injection of compound 48/80. OG also concentration-dependently inhibited compound 48/80- induced ear swelling response with concentrations of 0.01-1g/kg by anal administration 1h before injection of compound 48/80. OG also inhibited the passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE antibody concentration- dependently at concentrations ranging from 0.01 to 1g/kg. When OG was pretreated at concentrations ranging from 0.01 to 1g/ℓ, the histamine release from the rat peritoneal mast cells induced by compound 48/80 was reduced in a concentration-dependent manner. OG (0.1-1g/ℓ) had a significant inhibitory effect on histamine release from IgE-induced activated mast cells. OG is seen to be a biochemical compound certainly by 1 H-NMR spectroscopy According to above results, anal therapy of OG may be beneficial in the treatment of systemic and local immediate-type allergic reactions by inhibition of histamine release from mast cells.

Review of the Large-Scale Clinical Researches on Acupuncture in Germany: ASH, ART, ARC, and GERAC (2000년부터 독일에서 수행된 대규모 침 임상연구들에 대한 고찰: ASH, ART, ARC, GERAC)

  • Objectives : The purpose is to introduce the recent large-scale clinical researches for safety, efficacy and effectiveness of acupuncture in Germany. Results : In 2000, the German Federal Committee of Physicians and Health insurer proposed that large research initiatives on acupuncture, Acupuncture Model Projects(Modellvorhaben Akupunktur), could be conducted by health insurance companies for several pain that acupuncture is syndromes to justify the insurance-based reimbursement. Accordingly, 4 clinical researches were carried out; the Acupuncture Safety and Health economics studies(ASH), the Acupuncture Randomised Trial(ART), the Acupuncture in Routine Care studies(ARC), and the German Acupuncture trial(GERAC). Meanwhile, ASH is a prospective observational study for safety and costs. ART and GERAC are composed of RCTs for efficacy. ARC includes 6 pragmatic RCTs with additional non-randomized cohort study for effectiveness. We investigated the papers related to them and discussed about the outcomes. The researches showed that acupuncture is effective in practice for several chronic conditions such as migraine, tension-type headache, chronic low back pain, osteoarthritis of knee, dysmenorrhea, and allergic rhinitis. Based in part on them, the German health authorities decided that acupuncture would be included into routine reimbursement by social health insurance funds for chronic low back pain and chronic osteoarthritis of the knee in 2006. Conclusions : The German clinical researches may suggest the clues for establishing the evidence of acupuncture treatment.

Effects of Gamitonggyutang on the Experimental Animals on Anti-inflammatory, Analgesic, Anti-Allergic Effect and Antibacterial Activity (가미통규탕(加味通竅湯)의 항염(抗炎).진통(鎭痛).항(抗)알러지.항균(抗菌)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Gamitonggyutang has been used for treatment of rhinitis in oriental medical science. It is reported that Gamitonggyutang has a good effect on sinusitis in clinical medicine. So this study analysed the effect of Gamitonggyutang on anti-inflammatory, analgenic, anti-Allergic effect and Antibacterial Activity. The result were summerised as follows; 1. Gamitonggyutang extract decreased the edema indused by Carregennin at 200mg/kg and 400mg/kg. 2. Gamitonggyutang extract decreased the protein exudation indused by CMC-pouch at 200mg/kg and 400mg/kg. 3. Gamitonggyutang extract showed the ataralgesia at 800mg/kg by measurement of writhing syndrome, paw licking time and escape time induced by the i.p. infection of acetic acid and hot plate. 4. Gamitonggyutang extract decreased the effluent of vascular permeability indused by Evans blue at 600mg/kg and 800mg/kg. 5. Gamitonggyutang extract decreased the acute edema indused by Carregennin about 2 and 4 hours but didn't show useful effect. 6. Gamitonggyutang extract decreased the death rate, resulted from the effect of active systemic anaphylaxis reaction indused by CGG, but didn't show useful effect.. 7. Gamitonggyutang extract showed the growth inhibitory effect of each bacterias at $52mg/m{\ell}$. 8. Gamitonggyutang extract suppressed the growth of Streptococcus mutans 10449, and showed the supression of acid fabrication in case of 1:10 more than 1:100.

  • Objective : The purpose of this study is to investigite the more effective oriental medical treatment in pediatric diseases and its clinical applicability and chief complaints at oriental pediatrics in an area of busan and kyeungnam. Method : 884 new patients of theirs ages between 0 term 20 years who were treated at the pediatrics in Dong-Eui Oriental Medicine Busan Hospital from 1. 2000 to 29. Feb. 2001 Result : 1. The number of male cases was 514 (58%) and female cases 370 (42%) an so male cases 1.38 more than female. In age distribution, 0-1year: 242 patients(27.4%), 2-6years: 455 patients(51.4%), 7-20years: 187 patients(21.2%). 2. The Chief Complaint according to systemical division was Resparatory diseases (37.4%) for common cold, chronic cought, take a easy cold etc and the Digestive diseases(23.6%) for anorexia, diarrhea, abdominal pain etc and Nervous also Mental(11.8%) diseases for sezure, tic, etc and Allergic diseases(6.2%) for atopic dermititis, allergic rhinitis, etc and Physique diseases(5.4%) for short stature, leg pain, etc and Genaral symptoms(4.5%) for sweat profusely, headache, etc and Urinary diseases(3.3%) for bed wetting, frequently voiding, etc Discussion : 1. The Chief Complaint cannot be found a regional specificity 2. The oriental medicine treatment was still congnated as a way to improve the weakness by patients, also was widely noised about preventive medicine. In particular, Respiratory and Disestive diseases are dependent on the oriental medicine. 3. The new diseases and the new treatment method should be investigated as field of oriental medical treatment.
