Removal of Heavy Metals by Cladophora sp. in Batch Culture: The Effect of Wet-mixed Solidified Soil (loess) on Bioremoval Capacities

  • The heavy metal removal capacity of filamentous green alga Cladophora sp. cultured together with wet-mixed solidified soil (loess) was tested. A Cladophora sp. was cultured for 5d, with added Chu No. 10 medium, in stream water contaminated by high concentration of heavy metals from a closed mine effluent. Heavy metal ion concentrations of the medium and in algal tissue were measured every day during the experiment. Dissolved metals (Al, Cd, Cu, Fe, Mn, Zn) in medium were rapidly removed (over 90% elimination) within 1-2d when alga and loess were added. Dissolved heavy metals dropped by only 10% when algae were cultured without loess. The Cladophora sp. accumulated much more heavy metals when cultured with loess than when the alga was cultured alone. Cladophora sp. exhibited a maximum uptake capacity for Al ($17,000{\mu}g^{-1}$ algal dry weight). The metal bioremoval capacities of the algae were in the order Al, Fe, Cu, Mn, Zn and Cd. The heavy metal removal capacity of Cladophora sp. showed significant increases when wet-mixed solidified soil was added to culture media.

  • A filamentous green alga Cladophora vadorum (Areschoug) Kützing, bloomed at Shangrok Beach, Buan, Republic of Korea, in September 2015. This alga is currently distributed worldwide. Concentrations of total nitrogen (TN), total phosphorus (TP), dissolved inorganic nitrogen (DIN), and dissolved inorganic phosphorus (DIP) were analyzed in the bloom area and compared to those of other areas in the vicinity. DIN and DIP concentrations were similar to those of other areas. However, TN and TP were as much as six and ten times higher than in other areas, respectively. As in other Cladophora species, the bloom of C. vadorum at Shangrok Beach in 2015 appears to have depended on the TP concentration in the seawater. This suggests that blooms in the area can be controlled by reducing TP.

  • Three marine species of Cladophora (Cladophoraceae, Chlorophyta) commonly occurred on the coast of Cheju Island, Cladophora conchopheria Sakai, C. gracilis (Griff.) Kutzing and C. wrightiana Harvey were described morphologically, considering their taxonomic characters. C. conchopheria Sakai, specifically growing on the surface of Lunella coronata, is a very minute plant of 550-990 ${\mu}{\textrm}{m}$ in height and has intracuticular adventitious rhizoids borne from the lower portion of the first to fourth segments. C. gracilis occurs on calm habitat and shows a pseudosympodial growth with pectinate branchlets. c. wrightiana always produces protoplasmic protuberances at the node of middle portion of thallus. It grows to 30 cm or more in height and is dark green in subtidal bottom.

녹조 대발생종 금발대마디말(Cladophora vadorum)의 절편 생장에 온도, 조도 및 영양염 종류가 미치는 영향 (Effects of Temperature, Irradiance, and Nutrient Type on the Fragment Growth of Green Tide Alga Cladophora vadorum)

  • The green macroalga Cladophora vadorum bloomed along the coast at Sangrok Beach, Buan, South Korea, in September 2015. To elucidate the cause of bloom, the effects of environmental factors on the vegetative growth of adult fragments were examined. Growth experiments were carried out under different combinations of temperatures and irradiances, and with a single factor of nutrients (nitrogen, phosphorus). The maximal growth of C. vadorum was reported under the combination of 25°C and 100 μmol photons m−2s−1. The species grew under a wide range of N and P concentrations. The growth of C. vadorum peaked at 50 μM PO43−, 80 μM NH4+, and 100 μM NO3. Adult fragments formed holdfasts and new branches within 3 days in culture and became adults, showing polarized growth patterns, in 2 weeks. This is the first report showing the development of numerous bladelets from a segment in Cladophora species. The present results indicate that Cladophora blooms appear under growth conditions that are favorable in terms of temperatures, irradiance, and nutrients via fragment growth patterns producing rapid holdfasts and many bladelets.

  • We examined the effects of environmental factors on zoospore release and germling growth of the green tide alga Cladophora albida under various conditions of temperature${\times}$irradiance (zoospore release), temperature${\times}$irradiance${\times}$ nutrient (germling growth), and a single factor test of salinity. Zoospore release was maximized at $30^{\circ}C$ and $100{\mu}mol\;photons\;m^{-2}s^{-1}$ in the temperature irradiance experiment and at 34 psu in the salinity experiment. Maximum germling growth was observed at $25^{\circ}C$ with $100{\mu}mol\;photons\;m^{-2}s^{-1}$ and PES (Provasoli's Enriched Seawater) in the temperature irradiance nutrient experiment, and at 34 psu in the salinity experiment. Germlings grew faster at higher irradiances for a given temperature level, and also grew faster as salinity increased over the range of 5-34 psu. Overall, optimal environmental conditions for zoospore release were $30^{\circ}C$, $100{\mu}mol\;photons\;m^{-2}s^{-1}$ and 34 psu. Maximal germling growth occurred at $25^{\circ}C$, $100{\mu}mol\;photons\;m^{-2}s^{-1}$, PES, and 34 psu. C. albida blooms are most likely to occur under these optimal environmental conditions, as plentiful zoospore release and rapid germling growth lead to population growth.

충남 고정리의 하계 해조상 (Summer Algal Flora of Gojeong-Ri, West Coast of Korea)

  • The marine benthic algae of Gojeong-Ri was investigated in a floristic aspect. As a result, 34 species, 7 blue-green, 14 red, 6 brown and 7 green algae were identified as a summer flora from the area. Among them, Sirocoleum kurzii (blue-green alga), Ralfsia clavata (brown alga), Pseudulvella consociata and Cladophora speciosa (green algae) were known as new records to Korea.

경기도 수원시와 평택시의 5개 담수호소의 식물플랑크톤 분포 및 현존량 (The Distribution and Standing Crop of Phytoplankton at Five Freshwater Lakes in Suwon-si and Pyongtaek-si, Gyeonggi-do)

  • 경기도 수원시와 평택시에 위치한 광교저수지, 원천저수지, 서호, 남양호, 아산호 등 5개 호소에 대해 2003년 3월부터 11월까지 계절별로 4회의 채집을 실시하고, 식물플랑크톤의 분포 및 현존량을 분석하였다. 분석결과 본 수역에는 5문, 5강, 1아강, 16목, 26과, 1아과, 72속, 167종, 28변종, 1아종, 3품종의 총 199분류군의 식물플랑크톤이 분포하는 것으로 나타났다. 전체 식물플랑크톤 중 청정수역을 지표하는 분류군은 9월 광교저수지에서 Cladophora glomerata의 1분류군만이 출현하였으나, 오염수역을 지표하는 분류군은 Oscillatoria chlorina를 포함한 20분류군이 출현하였다. 또한 각각 9분류군과 4분류군의 이취미 및 독성물질을 분비하는 분류군들이 출현하였다. Chlorophyll-a농도에 대한 Yang and Dickman이 제안한 기준에 따르면 서호, 원천저수지, 남양호, 아산호는 과부 영화 상태이며, 광교저수지는 동계에는 빈중영양 상태이고 나머지 계절은 부영양화 상태로 나타났다.

Assessment of the macroalgal diversity of Kuwait by using the Germling Emergence Method

  • Cryptic stages of diverse macroalgae present in natural substrata, "the bank of microscopic forms", were isolated into clonal cultures and identified based on both morphological characteristics and DNA barcoding. Approximately 120 clonal isolates from 308 natural substratum samples were collected from the entire coastline of Kuwait. Amongst these isolates, 77 (64%) were identified through DNA barcoding using the nuclear ribosomal small subunit, RuBisCO spacer (ITS2, tufa, rbcL, psaA, and psbA) and sequencing. Twenty-six isolates (34%) were identified in the division Chlorophyta, 18 (23%) as Phaeophyceae, and 33 (43%) as Rhodophyta. For all DNA sequences in this study, species-level cut off applied was ≥98% homology which depend entirely on the markers used. Three putative new records of Chlorophyta new for the Arabian Gulf were made: Cladophora laetevirens (Dillwyn) Kützing, Ulva torta (Mertens) Trevisan and Ulvella leptochaete (Huber) R. Nielsen, C. J. O'Kelly & B. Wysor in Nielsen, while Cladophora gracilis Kützing and Ulva ohnoi M. Hiraoka & S. Shimada are new records for Kuwait. For Phaeophyceae, Ectocarpus subulatus Kützing and Elachista stellaris Areschoug were new records for the Gulf and Kuwait. In the Rhodophyta, Acrochaetium secundatum (Lyngbye) Nägeli in Nägeli & Cramer, Ceramium affine Setchell & N. L. Gardner, Gelidium pusillum var. pakistanicum Afaq-Husain & Shameel and Dasya caraibica Børgesen are new records for the Gulf and Kuwait, while the red alga Stylonema alsidii (Zanardini) K. Drew is a new record for Kuwait. Several isolates identified corresponded to genera not previously reported in Kuwait and / or the Arabian Gulf, such as Porphyrostromium Trevisan, a new genus from the Bangiales, and two unidentified species for the Planophilaceae Škaloud & Leliaert. The isolates cultivated from substrata enhance understanding of the marine macroalgal diversity in the region and confirmed that the Germling Emergence Method is suitable for determining the actual diversity of a given study area through isolation from cryptic life-history phases.

하계 독도 연안 해조류의 종조성 변동과 갯녹음현상 (Variation of Species Composition of Benthic Algae and Whitening in the Coast of Dokdo Island during Summer)

  • This study was to clarify the variations in a marine ecosystem through examining the species composition of benthic algae in the coast of Dokdo Island during summer (June & August, 2003). The total number of species found in benthic algae was 45 composed of 11% green algae, 29% red algae and 60% brown algae. The dominant species were Enteromorpha linza, Cladophora spp. and Codium frgile for green algae, Ecklonia cave, Eisenia bicyclis, Sargassum confusum, S. patens, S. horneri, Myagropsis myagroides for brown algae and Amphiroa cphedrace. A pusilla, Lithophyllum okamurae, Corallina spp. Laurencia pinnata, L. hamata, Chondria crassicaulit, C. dasyphylla, Gelidium amansii, Neosiphonia yendoi and Polysiphonia yendoi for red algae. The number of benthic algal species decreased more in this study than in previous researches. The whitening of the rocky shore might provoke the decrease of benthic algal biomass and species.

Twelve Undescribed Species of Macrofungi from Korea

  • A survey of the indigenous fungal resources of Korea was undertaken during 2014-2015. All specimens collected in this study were identified at the species level, based on their morphological characteristics and rDNA sequence data. Among them, 12 macrofungal species, viz. Agaricus guizhouensis, Amanita orientifulva, Armillaria cepistipes, Crepidotus inhonestus, Daldinia childiae, Elmerina cladophora, Lycoperdon scabrum, Marasmius brunneoaurantiacus, Otidea bufonia, Pluteus hongoi, Pluteus variabilicolor, and Russula grisea have not been previously reported in Korea.
