  • Objective : Citrus unshiu peel (Citri Unshius Pericarpium) has been prescribed to suppress coughing and phlegm in Korean medicine. In this study, the effect of ethanol extract of Citrus unshiu peel (CEE) on apoptosis was investigated using cadmium chloride (CdCl2) treated HepG2 cells. Methods : CEE was prepared by extracting 300 g of Citri Unshius Pericarpium in 3 L of ethanol for 72 h. Apoptosis was determined by the TUNEL assay. The mitochondrial membrane potential (MMP) was monitored using the membrane-permeable fluorescent dye Rh123. The expression level of each protein was monitored by Western blot analysis. Results : CEE protected HepG2 cells from apoptosis as determined by the TUNEL assay. A decrease in MMP was observed in cells exposed to cadmium, indicating that mitochondria are involved in the induction of apoptosis. However, CEE recovered the reduction in MMP caused by cadmium. In addition, decreased expression of B-cell lymphoma 2 (Bcl-2), procaspase, and poly(ADP-ribose) polymerase (PARP) by cadmium was increased by CEE. The anti-apoptotic effect of CEE was found to be associated with inhibition of JNK and p38 phosphorylation when examining the expression of phosphorylated MAPK by Western blot. Conclusion : This study showed that CEE exerted anti-apoptotic effects in cadmium-induced HepG2 cells by inhibiting the reduction of MMP and changes in the expression level of apoptotic proteins. These results suggest the potential for CEE to be used for heavy metal-induced liver damage.

  • In an attempt to find a feed additive showing an anti-scuticociliate effect, extracts from Citrus unshiu Peel were tested against virulent scuticociliate infection. The most effective anti-scuticociliate killing activity in vitro was observed in the extract squeezed from homogenizing water-soaked dried tangerine peel (DTP). In addition, we have investigated the effect of DTP as a feed additive on growth rate and anti-parasitic activity of olivaceus flounder. DTP extract added diets (0.1, 0.5, 1, and 5%/feed weight) were fed to flounder for 7 days for checking a growth rate and 14 days for a challenging test. As a result, the feed conversion rate was significantly improved only in 1% DPT extract group compared to the control and 0.5% DTP extract fed group showed 100% of survival rate in the challenge test, all of which indicating that DTP extract would be a potential feed additive against scuticociliatosis.

Bioconversion of Citrus unshiu peel extracts with cytolase suppresses adipogenic activity in 3T3-L1 cells

  • BACKGROUND/OBJECTIVES: Citrus flavonoids have a variety of physiological properties such as anti-oxidant, anti-inflammation, anti-cancer, and anti-obesity. We investigated whether bioconversion of Citrus unshiu with cytolase (CU-C) ameliorates the anti-adipogenic effects by modulation of adipocyte differentiation and lipid metabolism in 3T3-L1 cells. MATERIALS/METHODS: Glycoside forms of Citrus unshiu (CU) were converted into aglycoside forms with cytolase treatment. Cell viability of CU and CU-C was measured at various concentrations in 3T3L-1 cells. The anti-adipogenic and lipolytic effects were examined using Oil red O staining and free glycerol assay, respectively. We performed real time-polymerase chain reaction and western immunoblotting assay to detect mRNA and protein expression of adipogenic transcription factors, respectively. RESULTS: Treatment with cytolase decreased flavanone rutinoside forms (narirutin and hesperidin) and instead, increased flavanone aglycoside forms (naringenin and hesperetin). During adipocyte differentiation, 3T3-L1 cells were treated with CU or CU-C at a dose of 0.5 mg/ml. Adipocyte differentiation was inhibited in CU-C group, but not in CU group. CU-C markedly suppressed the insulin-induced protein expression of CCAAT/enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) as well as the mRNA levels of $CEBP{\alpha}$, $PPAR{\gamma}$, and sterol regulatory element binding protein 1c (SREBP1c). Both CU and CU-C groups significantly increased the adipolytic activity with the higher release of free glycerol than those of control group in differentiated 3T3-L1 adipocytes. CU-C is particularly superior in suppression of adipogenesis, whereas CU-C has similar effect to CU on stimulation of lipolysis. CONCLUSIONS: These results suggest that bioconversion of Citrus unshiu peel extracts with cytolase enhances aglycoside flavonoids and improves the anti-adipogenic metabolism via both inhibition of key adipogenic transcription factors and induction of adipolytic activity.

  • Effects of extraction conditions, gamma-irradiation and storage conditions on bioactivities of Citrus (Citrus unshiu) peel extract were investigated. The Hunter color $L^{*}$- and $a^{*}$-values of the extract increased but $b^{*}$-value decreased with an increase in absorbed irradiation dose. DPPH radical scavenging, tyrosinase inhibition and nitrite scavenging activities were not affected by irradiation but reduced by increased storage time. Nitrite scavenging activity of the extract was the highest at pH 1.2 followed by pH 4.2 and 6.0 and not changed by storage. Results indicated that there is potential for using citrus peel byproducts as a bioactive ingredient, and that gamma irradiation brightens the color of the extract without adversely altering its biological activity.ity.

  • 제주산 궁천조생의 저장조건에 따른 저온저장 특성을 검토하였다. 상온저장에 비해 저온저장 감귤은 저장기간 중 가용성 고형물, 총당, 비타민 C의 변화가 매우 적었으며, 부패율과 중량감소가 각각 10% 이내 수준에서 비교적 신선도를 유지한 상태에서 3월 하순까지 저장이 가능한 것으로 보였다. 또한, 중량감소, 경도의 저하, 당함량의 감소는 저장기간 중에 계속하여 서서히 일어났다. 필름포장과 wax-coating은 중량감소와 외관향상에는 도움을 주었으나 장기간 저장에는 부패율이 증가하였다. 습도조절을 하지 않은 저온저장은 습도조절한 경우와 비교하였을 때 부패율과 중량감소가 컸다. 증산작용으로 인한 껍질수분의 감소에 따라 과육으로부터 수분이동이 발생하여 저장기간 중 과육율이 감소하였으며, 저장 60일 후부터 경도저하가 많이 일어나 조직이 연화됨을 알 수 있었다. 조기수확한 감귤에 비하여 완숙한 감귤이 부패율과 중량감소가 적어 저장 100일간을 기준하여 출하하는 것이 바람직하였다. 상온저장한 감귤에 비해 저온저장한 감귤은 저장 4개월 이후 출고하였을 경우 외관 및 맛에 비교적 우수하였으며, 출고후 필름포장한 감귤의 부패율은 급속히 증가하여 취급에 주의가 필요하였다. 조생온주의 경우 저온에서 장기간 저장이 어려움을 알 수 있어서 저장용 감귤의 특성을 고려하여 저장조건을 설정할 필요가 있었다.

Effects of Dietary Fiber Extracted from Citrus (Citrus unshiu S. Marcoy) Peel on Physicochemical Properties of a Chicken Emulsion in Model Systems

  • Citrus (Citrus unshiu S. Marcoy) industry by-products were used as a source of dietary fiber, and the effects of dietary fiber extracted from citrus peel on the proximate composition, pH, color, protein solubility, cooking loss, emulsion stability, and apparent viscosity of a chicken emulsion in model systems were examined. Chicken emulsions were prepared by adding citrus peel fiber at four different concentrations (1, 2, 3, and 4%). The apparent viscosity, redness, and yellowness of the chicken emulsion with citrus peel fiber were higher than those of the control (p<0.05). The lightness values of the chicken emulsions were lower in treatments containing citrus peel fiber (p<0.05). Furthermore, moisture content, cooking loss, and emulsion stability of the chicken emulsion with 1-2% citrus peel fiber were higher than those of other treatments (p<0.05). Fat content was lower in the treatments with added citrus peel fiber than that in the control (p<0.05). Chicken emulsions with added citrus peel fiber had improved quality characteristics, and the best results were obtained for the chicken emulsion with 2% added citrus peel fiber.

Dispersal of Citrus Bacterial Canker Caused by Xanthomonas axonopodis pv. citri in Nursery Plots of Unshiu Orange

  • Dispersal of citrus bacterial canker caused by Xanthomonas axonopodis pv. citri on Unshiu orange was investigated in naturally infested nursery plot at Seogwipo in Jeju island, Korea. Based on phage detection, over 2% of the bacterial pathogen over-wintered in canker lesions and started to multiply in late May. However, symptoms were first observed 1 month after the phage detection. The disease dispersed non-directionally to nearby plants possibly because of indirect dissemination of the bacterium by rain splashes. The disease increased from late June to late August and decreased thereafter. Population of phage increased constantly, however, disease occurrence somewhat fluctuated due to environmental factors. Disease incidence and severity were correlated with rainfall with wind that occurred 14-32 days earlier from late May to late August.

  • In a bioassay-guided search for anti-allergic compounds from higher plants of Korea, poly-methoxyflavones, 3',4',5,6,7,8-hexamethoxyflavone (I), 5-hydroxy-3',4',6,7,8-pentamethoxyflavone (II) and 3',4',5,7,8-pentamethoxyflavone (III) have been isolated from the immature peels of Citrus unshiu. Structures of these compounds were elucidated on the basis of spectroscopic techniques. Compounds I and II inhibited dose-dependent histamine release from the rat peritoneal mast cells activated by compound 48/80 or anti-DNP IgE.

Characteristics of 'Hongrou Taoye', a Grafted Chimera in Sweet Orange and Satsuma Mandarin

  • The synthesis of chimeras is a breeding approach for horticultural crops. In our breeding program, a new diploid citrus chimera, named 'Hongrou Taoye' (Citrus sinensis [L.] Osbeck + Citrus unshiu Marc.), was found arising at the junction where a 'Taoye' sweet orange (C. sinensis) scion was grafted onto Satsuma mandarin (C. unshiu). As an artificial chimera, its fruit traits derived from the L1 cell layer, with juice color and carotenoid complement, in which ${\beta}$-cryptoxanthin accumulated predominantly, similar to those of Satsuma mandarin. By contrast, traits originating from the L2/L3 cell layer, including pollen, seed, and rind aroma characteristics, were the same as those of 'Taoye' sweet orange (the scion). SSR and cpSSR analyses showed that both nuclear and chloroplast genomes of the chimera were a combination of both donor parents. 'Hongrou Taoye' thus combined the valuable traits of both donor plants, and therefore has good potential in citrus fresh market.

제주산 흥진조생 온주밀감의 유통중 선도에 관한 연구 (A Study on the Post-harvest Physiology of Citrus unshiu Marc. Var. okitsu, During Transportation)

  • 제주산 감귤의 유통방법에 따른 선도 변화를 조사하고 저온유통의 효과를 검토하였다. 유통 중 성분변화는 상온(17$^{\circ}C$)유통이 저온(5$^{\circ}C$)유통에 비해서 중량감소는 6% 증가, vitamin C는 1.91mg 감소, 당산비는 20% 증가함을 보였다. 상온유통에서 중량감소로 인하여 당산비가 높아져서 (16.11에서 21.75) 감귤의 맛과 선도가 감소된다. 현행 감귤수송은 상온유통을 하고 있으나 고품질 감귤에는 부적합한 것으로 나타나며 고품질 소량생산 지향의 생산 체계에 저온유통체계를 도입하는 것이 상품성 향상에 중요 할 것으로 사료된다.
